[Pathogenesis and prevention of Alzheimer's disease: when and in what way does the pathological process begin?].

Abnormal tau lesions (e.g., pretangles, neuropil threads, and neurofibrillary tangles) that develop in a few types of nerve cells in the central nervous system are essential to the pathogenesis of Alzheimer's disease. Pretangles begin to occur in puberty and even during early childhood in the locus coeruleus. Evolutionally speaking, the propensity to develop tau lesions may be related to late developing and maturing nerve cell types in the human brain.

Electrical stimulation of primary olfactory nerve induces two types of variations in the extracellular potassium activity within the glomerulus of the rat olfactory bulb in vivo.

Primary olfactory nerve stimulation induces a two-step increase in extracellular potassium, as recorded by K+-sensitive microelectrodes inserted into single glomeruli of the rat olfactory bulb. The second rise in K+ (K-peak) occurs in a graded, activity-dependent manner at the basal pole of the glomerular neuropile. This phenomenon appears to be directly related to the rate of extracellular K+ accumulation and is accompanied by a substantial depression of the mitral cell firing rate.

Spatially Mapping Neuropeptide Isomers via MALDI Trapped Ion Mobility MS Imaging.

Imaging endogenous peptides with the chiral selectivity of their component amino acids remains an unmet analytical challenge. d-amino acid-containing peptides (DAACPs), formed by post-translational isomerization of l- to d-amino acids, make up a functionally important class of neuropeptides whose spatial distribution remains poorly understood. Here, we introduce untargeted imaging of neuropeptide stereoisomers using matrix-assisted laser desorption/ionization-trapped ion mobility mass spectrometry imaging (MALDI-TIMS-MSI). We mapped neuropeptides and their DAACP forms in the central nervous sy

Expression profiles of the autism-related SHANK proteins in the human brain.

BACKGROUND: SHANKs are major scaffolding proteins at postsynaptic densities (PSDs) in the central nervous system. Mutations in all three family members have been associated with neurodevelopmental disorders such as autism spectrum disorders (ASDs). Despite the pathophysiological importance of SHANK2 and SHANK3 mutations in humans, research on the expression of these proteins is mostly based on rodent model organisms. RESULTS: In the present study, cellular and neuropil SHANK2 expression was analyzed by immunofluorescence (IF) staining of post mortem human brain tissue from four male individua

Polyglutamine tract expansion of the androgen receptor in a motoneuronal model of spinal and bulbar muscular atrophy.

Spinobulbar muscular atrophy (SBMA) is a late-onset disorder characterized by progressive muscle loss, degeneration of motoneurons in the spinal cord and brainstem, and partial androgen insensitivity. SBMA is directly correlated with the expansion of CAG repeats encoding a polyglutamine tract (polyQ) of extended length. The identification of polyQ expansion in SBMA led to the discovery of an entire class of neurodegenerative disorders. In fact, at least eight different diseases, including Huntington's disease, share a common molecular mechanism involving an expansion of a polyQ tract within di

Anatomy and physiology of neurons with processes in the accessory medulla of the cockroach Leucophaea maderae.

The accessory medulla (AMe), a small neuropil in the insect optic lobe, has been proposed to serve a circadian pacemaker function analogous to the role of the suprachiasmatic nucleus in mammals. Building upon considerable knowledge of the circadian system of the cockroach Leucophaea maderae, we investigated the properties of AMe neurons in this insect with intracellular recordings combined with dye injections. Responses of neurons with processes in the AMe to visual stimuli, including stationary white light, moving objects, and polarized light were compared with the responses of adjacent medul

Chondroitin sulfate proteoglycan expression pattern in hippocampal development: potential regulation of axon tract formation.

A variety of molecular influences in the extracellular matrix (ECM) interact with developing axons to guide the formation of hippocampal axon pathways. One of these influences may be chondroitin sulfate proteoglycans (CSPGs), which are known to inhibit axonal extension during development and following central nervous system injury. In this study, we examined the role of CSPGs and cell adhesion molecules in the regulation of axon tract formation during hippocampal development. We used indirect immunofluorescence to examine the developmental pattern of CSPG expression relative to axon tracts tha

Developmental expression and biochemical analysis of conulin, a protein secreted from a subset of neuronal growth cones.

In this report, we analyze the developmental pattern of expression of a new grasshopper protein, Conulin, using the monoclonal antibody 7D2 on whole-mount embryos and dissociated neurons. We also have examined its biochemical properties by immunoblot analysis. Conulin is a protein expressed by a subset of neurons in the grasshopper embryo. The monoclonal antibody 7D2 recognizes Conulin as an M(r) 190 x 10(3) protein that is found in both the soluble and membrane-bound fractions of embryonic proteins. The membrane association is disrupted by alkaline pH and high ionic strength. Conulin first is

Cellular and regional distribution of reduced glutathione in the nervous system of the rat: histochemical localization by mercury orange and o-phthaldialdehyde-induced histofluorescence.

Differences in the cellular distribution of antioxidant defense mechanisms in heterogeneous tissue such as the nervous system are likely critical determinants of differential sensitivity to toxicants. Regional and cellular localization of reduced glutathione (GSH) in central and peripheral nervous tissue was determined from the pattern of fluorescence observed in tissue sections stained with mercury orange; localization was confirmed using a novel histofluorochromatic staining method, o-phthaldialdehyde (OPT). Excellent concordance between the distribution of fluorescence obtained with mercury

Senile plaque neurites fail to demonstrate anti-paired helical filament and anti-microtubule-associated protein-tau immunoreactive proteins in the absence of neurofibrillary tangles in the neocortex.

Although much work has been directed recently towards unravelling the protein chemistry of neurofibrillary tangle (NFT) and senile plaque (SP) components in Alzheimer's disease, the pathogeneses of these lesions remains largely unknown and the problem of their relationship is unresolved. In particular, although paired helical filaments (PHF) have long been documented in SP neurites, we do not know if they are of pathogenetic relevance for the formation of the SP. To investigate the relationship between NFT and SP, we examined antigenic properties of proteins in SP neurites in neocortical tissu

Organization of the filum terminale in the frog.

The histological organization of the filum terminale of the spinal cord in Rana catesbeiana and Rana pipiens was characterized to determine if this region possessed an organized neuropil or whether it was merely a glial remnant that persisted after absorption of the larval tail. The excised filum was maintained in vitro. Intracellular electrophysiological recording was performed with horseradish peroxidase injection. Tyrosine hydroxylase and serotonin distribution were revealed by immunocytochemical methods. Astroglia were the dominant cell type and displayed an elaborate variety of forms. The

Mixed pituitary adenoma/pituitary neuroendocrine tumor-gangliocytoma: Immunohistochemical insights.

Mixed pituitary adenoma/PitNET-gangliocytomas (PA/PitNET-GC) have been reported in small series over the past 20 years; some had limited immunohistochemistry (IHC) data. We interrogated our experience over 20 years, focusing on patterns of the GC component and IHC results for anterior pituitary hormones, transcription factors, NFP, and CAM5.2. A search of cases from 2002 to 2023 yielded 20 cases: 7M:13F, ages 20-71 years; 17 macroadenomas, 1 microadenoma, 2 ectopic. GC was co-associated with 4 corticotroph, 2 densely granulated lactotroph, 5 mixed lactotroph-somatotroph, 1 immature PIT1-lineag

Intestinal Region-Dependent Alterations of Toll-Like Receptor 4 Expression in Myenteric Neurons of Type 1 Diabetic Rats.

Toll-like receptor 4 (TLR4) can activate pro-inflammatory cascades in the gastrointestinal tract. Our aim was to determine TLR4 expression in myenteric neurons of different gut regions using a type 1 diabetic model. Ten weeks after the onset of hyperglycemia, myenteric whole-mount preparations from the duodenum, ileum and colon of streptozotocin-induced diabetic, insulin-treated diabetic and control rats were prepared for TLR4/peripherin double-labelling fluorescent immunohistochemistry. Immunogold electron microscopy was applied to evaluate TLR4 expression in the myenteric perikaryon and neur

Laser microbeam axotomy and conduction block show that electrical transmission at a central synapse is distributed at multiple contacts.

Touch (T) sensory neurons in the leech innervate defined regions of skin and synapse on other neurons, including other T cells, within the ganglionic neuropil. The cells' receptive fields in the periphery are comprised of a central region, innervated by thick axons, and adjoining regions (minor fields) innervated by thinner axons. Secondary branches, known to be sites of synapses, emerge from the thinner and thicker axons. Pairs of T cells appear to make up to 200 separate contacts distributed within the neuropil. When the T cell is hyperpolarized, as occurs during natural stimulation of the c

Central nervous system of Rhipicephalus sanguineus ticks (Acari: Ixodidae): an ultrastructural study.

This study performed the ultrastructural description of the synganglion of Rhipicephalus sanguineus males and females, aiming to contribute to the understanding of the cellular organization of this organ. The results show that the central nervous system of these individuals consists of a mass of fused nerves, named synganglion, from where nerves emerge towards several parts of the body. It is surrounded by the neural lamella, a uniform and acellular layer, constituted by repeated layers of homogeneous and finely granular material. The perineurium is just below, composed of glial cells, which e

Glioneuronal tumors of the central nervous system.

Advances in the immunohistochemical detection of neuron-specific and neuronal-associated antigens have resulted in the discovery of neuronal elements in certain primary human brain tumors. The results have been not only to expand what neuropathologists commonly recognize as gangliogliomas, including the tumors now known as glioneurocytic tumor with neuropil rosettes and papillary ganglioneuroma, but also to expand the spectrum of tumor types to now include tumors such as central neurocytoma, dysembryoplastic neuroepithelial tumor, and desmoplastic infantile ganglioglioma. These discoveries hav

Tracing neural pathways in snail olfaction: from the tip of the tentacles to the brain and beyond.

The anatomical organization of the olfactory system of terrestrial snails and slugs is described in this paper, primarily on the basis of experiments using the African snail Achatina fulica. Behavioral studies demonstrate the functional competence of olfaction in mediating food finding, conspecific attraction, and homing. The neural substrate for olfaction is characterized by an extraordinarily large number of neurons relative to the rest of the nervous system, and by the fact that many of them are unusually small. There exist multiple serial and parallel pathways connecting the olfactory orga

Late-onset Lafora's disease with typical intraneuronal inclusions.

We describe a patient with progressive myoclonus epilepsy (late-onset Lafora's disease). Onset was in early adult life, and death was at age fifty-four. The initial symptoms were epileptic seizures and progressive dementia, with later occurrence of myoclonus. Lafora bodies were ubiquitous and in neuronal perikarya in many areas. Dust-like granular bodies predominated in the neuropil of cerebral cortex, substantia nigra, and striatum. Abnormal deposits were also found in the myocardium.

Neurotropic melanoma of the vulva.

A spindle cell melanoma of the clitoris was associated with a large area of lentiginous melanocytic dysplasia in the vulva of a 51-year-old woman. There was extensive neoplastic of the dermis, a desmoplastic reaction, and replacement of perineurial and Schwann cells by tumor cells (neurotropism). The superficial dysplastic components were pigmented, but melanosomes were not identified in a lymph node metastasis. Here the internuclear areas strongly resembled neuropil. Vulvar melanoma is rate, and this is the first report of neurotropic melanoma in this site.
