Neurovascular coupling alterations related to cognitive impairment in cerebral small vessel disease: A multiscale brain network perspective.

Evidence suggests that neurovascular coupling (NVC) dysfunction in cerebral small vessel disease (CSVD) may precede typical clinical and imaging manifestations. Here, we explored the underlying brain alterations of multiscale networks in CSVD patients related to cognitive impairment based on the method of NVC. We investigated 124 CSVD patients, including 70 patients with mild cognitive impairment (MCI) and 54 patients with no cognitive impairment (NCI). Functional MRI and arterial spin labeling were employed to estimate the coupling of spontaneous neuronal activity and cerebral blood perfusion

Neurovascular coupling assessment by transcranial doppler in acute stroke could be informative of long-term cognitive status.

To assess whether Neurovascular Coupling (NVC) status in acute ischemic stroke predicts long-term cognitive impairment, this prospective study included consecutive ischemic stroke patients, without prior cognitive deficits, recruited between September 2021 and August 2022. Transcranial Doppler (TCD) monitoring was performed within 72 hours post-stroke to measure NVC, determined by peak relative increase in cerebral blood velocity (CBv) in the posterior cerebral artery (PCA) during a visual stimulus. Age-, gender-, and vascular risk factor-matched controls were included. The primary outcome was

EEG Spectral Power and Neurovascular Coupling as Early Predictors of Neurodevelopmental Outcome in Neonatal Hypoxic-Ischemic Encephalopathy.

Hypoxic ischemic encephalopathy (HIE) remains one of the leading causes of morbidity and mortality in newborns. There is a strong need to predict their neurodevelopmental impairment (NDI) within early hours of life, tailoring treatment strategies accordingly. This study aims to explore the discriminatory capabilities of electroencephalogram (EEG) delta power (DP) and total power (TP), along with neurovascular coupling (NVC) to predict NDI. The study evaluates the relationships of single biomarkers (DP, TP, and NVC) with NDI using univariate logistic regression models. The predictive accuracy o

Assessment of neurovascular coupling and cortical spreading depression in mixed mouse models of atherosclerosis and Alzheimer's disease.

Neurovascular coupling is a critical brain mechanism whereby changes to blood flow accompany localised neural activity. The breakdown of neurovascular coupling is linked to the development and progression of several neurological conditions including dementia. In this study, we examined cortical haemodynamics in mouse preparations that modelled Alzheimer's disease (J20-AD) and atherosclerosis (PCSK9-ATH) between 9 and 12 m of age. We report novel findings with atherosclerosis where neurovascular decline is characterised by significantly reduced blood volume, altered levels of oxyhaemoglobin and

Brain hemodynamic changes mediated by dopamine receptors: Role of the cerebral microvasculature in dopamine-mediated neurovascular coupling.

The coupling between neurotransmitter-induced changes in neuronal activity and the resultant hemodynamic response is central to the interpretation of neuroimaging techniques. In the present study, MRI experiments showed that dopamine transporter blockers such as cocaine and dopamine releasers such as amphetamine and D1 receptor agonists induced large positive increases in relative cerebral blood volume (rCBV) that were not sensitive to nitric oxide synthase inhibition. However, D1/D5 receptor antagonism with SCH-23390 prevented or blocked the hemodynamic response without any concomitant effect

Amyloid pathology drives cognitive decline in Alzheimer's disease through impairing neurovascular coupling: An [¹⁸F]-Florbetapir PET/MRI Study.

BACKGROUND: Neurovascular coupling (NVC) impairment is implicated in Alzheimer's disease (AD), but its relationship with amyloid-β (Aβ) pathology and diagnostic value remain unclear. We examined Aβ-related NVC changes and their mediating role in cognitive decline. METHODS: Using [¹⁸F]-florbetapir PET/MRI, we assessed Aβ deposition, neural activity, and cerebral blood flow (CBF) in 96 participants (24 Aβ-negative mild cognitive impairment [MCI⁻], 30 Aβ-positive MCI [MCI⁺], and 42 Aβ-positive AD [AD⁺]). Neural activity was quantified using amplitude of low-frequency fluctuation (ALFF), fraction

Neurovascular-metabolic coupling links hearing loss to cognitive impairment: Evidence from GABA and cerebral blood flow in presbycusis.

Hearing loss is recognized as the leading modifiable risk factor for dementia. However, the neurobiological pathways bridging auditory dysfunction and cognitive decline remain poorly understood. We hypothesized that a neurovascular-metabolic cascade, involving neurotransmitter imbalances and perfusion deficits, underlies this link. We recruited 124 patients with presbycusis and 91 age-matched health controls. Auditory GABA and glutamate levels were quantified using macromolecule-suppressed MEGA-PRESS, and cerebral blood flow (CBF) was quantified using pseudo-continuous arterial spin labeling.

Periodontitis Increases Gingival, Serum and Hippocampus β-Amyloid Expressions but Reduces Neurovascular Coupling in Basilar Artery of Rats.

AIM: Neurodegenerative diseases are characterized by early increased beta-amyloid (Aβ) and decreased cerebrovascular reactivity. We investigated Aβ in gingiva, serum or hippocampus and neurovascular reactivity in basilar artery (BA) of periodontitis rats, to test the impact of Aβ on BA vasoreactivity ex vivo. MATERIALS AND METHODS: Periodontitis was induced in 32 rats using silk-ligation. Rats were sacrificed at weeks 0, 1, 2 and 4. Gingival TNF-α, IL-1β and Aβ were quantified via immunoblotting. Alveolar bone destruction was examined by micro-computed tomography. Serum and hippocampus Aβ val

Altered dynamic regional homogeneity and neurovascular coupling in multi-frequency bands in patients with cerebral small vascular disease.

BACKGROUND: Cerebral small vessel disease (CSVD) can lead to neurologic deficits and cerebral perfusion injuries. However, the dynamic characteristics of neural function homogeneity and their coupling with cerebral perfusion are rarely elucidated in the sub-frequency bands. The purpose of this study is to explore these features for further understanding of the neural mechanisms of CSVD. METHODS: In this cross-sectional study, we obtained magnetic resonance imaging (MRI) data and a series of cognitive scores from 27 CSVD patients with mild cognitive impairment (CSVD-MCI), 32 CSVD patients with

Psychedelic 5-HT2A receptor agonism alters neurovascular coupling and differentially affects neuronal and hemodynamic measures of brain function.

Human neuroimaging studies report that psychedelics induce serotonin-2A receptor-dependent changes in functional brain reorganization, presumably reflecting neuromodulation. However, these studies often overlook the potent vasoactive effects of serotonin. Here we identified psilocybin-induced alterations in hemodynamic response functions during human functional magnetic resonance imaging, suggesting potential disruptions in neurovascular coupling. We then used wide-field optical imaging in awake Thy1-jRGECO1a mice to determine whether psychedelic-induced changes in hemodynamics arise from neur

Effect of inflammation on neurovascular coupling, microperfusion, and clinical outcomes in ischemic stroke patients: a case series report.

INTRODUCTION: Ischemic stroke leads to a range of sequelae that affect daily functioning. In many cases, such as wake-up strokes or late hospital arrivals, the therapeutic window for reperfusion is missed for the patient, and functional outcomes remain uncertain. The inflammatory response to ischemia plays a pivotal role in the initiation, progression, and recovery phase of stroke. Yet, a gap remains in understanding its impact on neuroimaging and clinical outcomes. This prospective case series investigates the relationship between inflammation, neuroimaging findings in the first 48 h after st

Physiological influences on neurovascular coupling: A systematic review of multimodal imaging approaches and recommendations for future study designs.

In this review, we have amalgamated the literature, taking a multimodal neuroimaging approach to quantify the relationship between neuronal firing and haemodynamics during a task paradigm (i.e., neurovascular coupling response), while considering confounding physiological influences. Original research articles that used concurrent neuronal and haemodynamic quantification in humans (n ≥ 10) during a task paradigm were included from PubMed, Scopus, Web of Science, EMBASE and PsychINFO. Articles published before 31 July 2023 were considered for eligibility. Rapid screening was completed by the fi

Abnormal neurovascular coupling exists in patients with peritoneal dialysis and hemodialysis: evidence from a multi-mode MRI study.

BACKGROUND: Neurovascular coupling (NVC), as indicated by a comprehensive analysis of the amplitude of low-frequency fluctuation (ALFF) and cerebral blood flow (CBF), provides mechanistic insights into neurological disorders. Patients undergoing peritoneal dialysis (PD) and hemodialysis (HD) often face cognitive impairment, the causes of which are not fully understood. METHODS: ALFF was derived from functional magnetic resonance imaging, and CBF was quantified using arterial spin labeling in a cohort comprising 58 patients with PD, 60 patients with HD and 62 healthy controls. Neurovascular coupling and cerebrovascular hemodynamics are modified by exercise training status at different stages of maturation during youth.

Neurovascular coupling (NVC) is mediated via nitric oxide signaling, which is independently influenced by sex hormones and exercise training. Whether exercise training differentially modifies NVC pre- versus postpuberty, where levels of circulating sex hormones will differ greatly within and between sexes, remains to be determined. Therefore, we investigated the influence of exercise training status on resting intracranial hemodynamics and NVC at different stages of maturation. Posterior and middle cerebral artery velocities (PCAv and MCAv) and pulsatility index (PCAPI and MCAPI) were assessed

The relationship between neurovascular coupling, vision and sensory performance, and concussion history in Special Operations Forces combat soldiers.

OBJECTIVE: To determine the relationship between neurovascular coupling (NVC), vision and sensory performance in Special Operations Forces (SOF) combat soldiers with and without concussion history. METHODS: We studied 61 SOF combat soldiers (male, age = 33.8 ± 3.7 years, n = 40 with concussion history [Median = 3; range = 1-10+]). We instrumented our participants with transcranial Doppler to quantify NVC response during reading and visual search tasks. All participants completed vision and sensory performance testing (Senaptec Sensory Station). We performed separate multiple regressions to de

Where's Waldo? The utility of a complicated visual search paradigm for transcranial Doppler-based assessments of neurovascular coupling.

BACKGROUND: The concept of neurovascular coupling has been postulated since the late 1800s and has been demonstrated most commonly in humans using visual stimuli (e.g. reading, checkerboards). These traditional paradigms evoke only a moderate cerebral blood flow response due to the relative simplicity of the visual stimuli. NEW METHOD: Forty subjects completed three visual paradigms each challenging the visual processing areas to a different extent: reading text, complicated visual searching (new method: Where's Waldo) and viewing coloured dots. Posterior and middle cerebral artery (PCA, MCA)

Investigating the neurovascular coupling across multiple motor execution and imagery conditions: a whole-brain EEG-informed fMRI analysis.

The complementary strengths of electroencephalography (EEG) and functional magnetic resonance imaging (fMRI) have driven extensive research into integrating these two noninvasive modalities to better understand the neural mechanisms underlying cognitive, sensory, and motor functions. However, the precise neural patterns associated with motor functions, especially imagined movements, remain unclear. Specifically, the correlations between electrophysiological responses and hemodynamic activations during executed and imagined movements have not been fully elucidated at a whole-brain level. In thi
