Anti-listeria activity of newly isolated lactic acid bacteria from fermented foods and their application to smoked salmon(Coho).

The objective of this study was to evaluate the anti-listeria activities of lactic acid bacteria (LAB) isolated from Korean fermented foods and to assess the effect of fermentate (cells and cell-free supernatant [CFS]), CFS or cells in controlling L. monocytogenes on smoked salmon, packaged either in vacuum- and air-packaging. One L. lactis (CJNU3001) strain and two Enterococcus faecium (CJNU2524 and CJNU2008) strains appeared to have higher anti-listeria activity than the other 15 LAB strains. They also exhibited anti-listeria activity against 24 L. monocytogenes strains of various serotypes.

Application of nanotized formulation in the control of snail intermediate hosts of schistosomes.

Schistosomiasis continues to pose significant public health problems in many developing countries. Mass drug administration (MDA) is the most adopted control option but there is increasing evidence for the development of praziquantel-resistant Schistosoma strains. This shortcoming has necessitated the search for other effective methods for the control of schistosomiasis. The breaking of Schistosoma transmission cycles through the application of molluscicides into snail infested freshwater bodies has yielded positive outcomes when integrated with MDA in some countries. However, few of such effe

A Standardised Combinational Method for Evaluating Antimicrobial Compounds Against Biofilm Attachment, Development and Eradication.

Biofilm-mediated antimicrobial resistance remains a significant challenge for healthcare and patient safety. Currently, there are gaps in standardised methods for assessing antimicrobials against biofilm formations such as (1) assessment of initial bacterial attachment inhibition, as well as (2) assessment of antimicrobial compounds against both the external biofilm mass and biofilm-embedded metabolically active bacteria. The aim of this study is to address these gaps by combining several anti-biofilm techniques. In the procedure96-well anti-biofilm assessments were performed using plate well

Membrane-targeting antimicrobial compounds have differential effects on living and artificial yeast membrane models.

The stability of the plasma membrane is crucial for cell viability and disruptions in membrane stability can significantly impact cell function. Antimicrobial compounds targeting fungal membranes are required as novel alternatives to current resistance-prone fungicides. Six antimicrobials were assessed using the yeast Saccharomyces cerevisiae in living and artificial membrane models to gain insight into their efficacy and mechanistic activity. Antimicrobial-treated yeast cultures were monitored for growth inhibition and cell membrane permeability. Liposomes prepared from yeast polar lipids wer

D-cycloserine increases the effectiveness of vancomycin against vancomycin-highly resistant Staphylococcus aureus.

Vancomycin is a widely used clinical drug to treat for infection by methicillin-resistant Staphylococcus aureus. Some patients show a weak response to vancomycin treatment. We previously reported that β-lactams increase the susceptibility to vancomycin by vancomycin-highly resistant S. aureus (VRSA) strains obtained following repeated in vitro mutagenesis and vancomycin selection. Here we found that the susceptibility of the VRSA strains to vancomycin was remarkably increased by combined treatment with D-cycloserine. On the other hand, VRSA did not show increased susceptibility to vancomycin i

Terminal complement complexes with or without C9 potentiate antimicrobial activity against Neisseria gonorrhoeae.

UNLABELLED: The complement cascade is a front-line defense against pathogens. Complement activation generates the membrane attack complex (MAC), a 10-11 nm diameter pore formed by complement proteins C5b through C8 and polymerized C9. The MAC embeds within the outer membrane of Gram-negative bacteria and displays bactericidal activity. In the absence of C9, C5b-C8 complexes can form 2-4 nm pores on membranes, but their relevance to microbial control is poorly understood. Deficiencies in terminal complement components uniquely predispose individuals to infections by pathogenic Neisseria, includ

Are bacteriocins underexploited? Novel applications for old antimicrobials.

Bacteriocins are ribosomally synthesized (poly)peptides produced by almost all prokaryotic lineages. Bacteriocins from lactic acid bacteria (LAB) and bacteriocin-producer probiotic organisms have been thoroughly studied due to their wide spectra of action, the long-term use in food fermentations and the consideration of these microorganisms as beneficial for human beings. Most of the studies on the biotechnological application of diverse bacteriocins have been focused on their use as food preservatives, nisin being the prototype successfully used in alimentation. However, bacteriocins from LAB

Susceptibility of Listeria monocytogenes strains isolated from dairy products and frozen vegetables to antibiotics inhibiting murein synthesis and to disinfectants.

The susceptibility of 96 strains of Listeria monocytogenes isolated from food to antibiotics and disinfectants currently used in human therapy, veterinary, medicine and food industry was determined by a standard operating procedure--broth dilution method. Antimicrobial agents included the beta-lactams ampicillin and penicillin, the lantibiotic nisin, and the disinfectants benzalkonium chloride and chlorhexidine gluconate. Among the studied strains we found 13 strains with 8-fold, 7 strains with 16-fold and 2 strains with 32-fold decreased susceptibility to ampicillin, as determined by MIC, com

Oral vaccine of Lactococcus lactis harbouring pandemic H1N1 2009 haemagglutinin1 and nisP anchor fusion protein elevates anti-HA1 sIgA levels in mice.

OBJECTIVE: An oral lactococcal-based vaccine which haboured the haemagglutinin1 (HA1) antigen fused to nisP anchor protein for the purpose of surface displaying the HA1 antigen was developed against H1N1 virus. RESULTS: Recombinant L. lactis strains expressed HA1-nisP fusion proteins when induced with nisin, as confirmed through western blotting. However, immunofluorescense did not detect any surface-displayed proteins, suggesting that the protein was either unsuccessfully translocated or improperly displayed. Despite this, oral administration of recombinant L. lactis strains to BALB/c mice r

Cysteine-Rich Peptides in Food Systems: Structure, Sources, Functional Properties, Potential Applications, and Future Perspectives.

Cysteine-rich peptides (CRPs) are structurally diverse bioactive peptides in which cysteine residues form disulfide bonds or, in some lanthipeptides, cysteine-derived thioether bridges. These constraints can improve resistance to thermal, chemical, and enzymatic stress, although stability and activity remain peptide- and matrix-dependent. CRPs have attracted interest as antimicrobial, antioxidant, and immunomodulatory agents in food preservation, functional foods, and active packaging. Nisin provides the clearest regulatory and commercial precedent: this lanthipeptide is used in selected food

Biomimetic chitosan-derived bifunctional lubricant with superior antibacterial and hydration lubrication performances.

The lubrication deficiency in joints is a major cause of osteoarthritis. One of the most commonly used treatment means is to inject artificial lubricants, but there is a potential risk of infection during the injection process. Therefore, developing artificial lubricants with dual functions of friction-reduction and antibacterial is urgent. In this work, a novel polysaccharide-derived lubricant with simultaneous anti-bacteria and water-lubrication properties, called CS-MPC-N, is developed by grafting 2‑methacryloyloxylethyl phosphorylcholine (MPC) and nisin peptide onto backbone of chitosan (C

A microplate assay for the coupled transglycosylase-transpeptidase activity of the penicillin binding proteins; a vancomycin-neutralizing tripeptide combination prevents penicillin inhibition of peptidoglycan synthesis.

A microplate, scintillation proximity assay to measure the coupled transglycosylase-transpeptidase activity of the penicillin binding proteins in Escherichia coli membranes was developed. Membranes were incubated with the two peptidoglycan sugar precursors UDP-N-acetyl muramylpentapeptide (UDP-MurNAc(pp)) and UDP-[(3)H]N-acetylglucosamine in the presence of 40 μM vancomycin to allow in situ accumulation of lipid II. In a second step, vancomycin inhibition was relieved by addition of a tripeptide (Lys-D-ala-D-ala) or UDP-MurNAc(pp), resulting in conversion of lipid II to cross-linked peptidogly

Controlled production of stable heterologous proteins in Lactococcus lactis.

The use of Lactococcus lactis (the most extensively characterized lactic acid bacterium) as a delivery organism for heterologous proteins is, in some cases, limited by low production levels and poor-quality products due to surface proteolysis. In this study, we combined in one L. lactis strain use of the nisin-inducible promoter P(nisA) and inactivation of the extracellular housekeeping protease HtrA. The ability of the mutant strain, designated htrA-NZ9000, to produce high levels of stable proteins was confirmed by using the staphylococcal nuclease (Nuc) and the following four heterologous pr

Bacteriocins for Control of Listeria spp. in Food.

Foodborne outbreaks of listeriosis caused by Listeria monocytogenes have contributed to public consciousness about bacterial pathogens involved with foodborne disease. Major concerns with L. monocytogenes are its high mortality rate, wide distribution on raw products, growth at low temperatures, and its ability to establish itself in various food-processing environments. These concerns have prompted the examination of novel approaches, including the use of antimicrobial peptides, or bacteriocins, to combat its survival in foods. Bacteriocins from lactic acid bacteria have received much attenti

Complement-dependent outer membrane perturbation sensitizes Gram-negative bacteria to Gram-positive specific antibiotics.

Gram-negative bacteria are refractory to the action of many antibiotics due to their impermeable outer membrane. An important player of the immune system is the complement system, a protein network in serum that directly kills Gram-negative bacteria through pore-formation by the Membrane Attack Complexes (MAC). We here show that the MAC rapidly perforates the outer membrane but that inner membrane damage, which is essential for killing, is relatively slow. Importantly, we demonstrate that MAC-induced outer membrane damage sensitizes Gram-negative bacteria to otherwise ineffective, Gram-positiv

Antimicrobial peptides of the vaginal innate immunity and their role in the fight against sexually transmitted diseases.

Some antimicrobial peptides (AMPs) are produced in the vaginal innate immune system and play an important role in protecting this organ against pathogenic agents. Moreover, sexually transmitted diseases have become a major problem in human societies and are rapidly spreading. The emergence of antibiotic-resistant microbes (superbugs) can pose a major threat to human societies and cause rapid spread of these diseases. Finding new antimicrobial compounds to fight superbugs is therefore essential. It has been shown that AMPs have good potential to become new antibiotics. The most important AMPs i

Chemistry and biology of the ramoplanin family of peptide antibiotics.

The peptide antibiotic ramoplanin factor A2 is a promising clinical candidate for treatment of Gram-positive bacterial infections that are resistant to antibiotics such as glycopeptides, macrolides, and penicillins. Since its discovery in 1984, no clinical or laboratory-generated resistance to this antibiotic has been reported. The mechanism of action of ramoplanin involves sequestration of peptidoglycan biosynthesis Lipid intermediates, thus physically occluding these substrates from proper utilization by the late-stage peptidoglycan biosynthesis enzymes MurG and the transglycosylases (TGases

Recombination of miR-146b by Lactococcus lactis for Remolding Macrophages and the Microbiome in the Treatment of Murine Colitis.

MicroRNA 146b (miR-146b) mitigates the progression of inflammatory bowel disease (IBD) by reprogramming macrophage polarization through the induction of interleukin-10 (IL-10), thereby eliciting an anti-inflammatory response, but its application is currently hindered by the lack of safe delivery systems that allow sustained miR-146b expression. Inspired by the probiotic Lactococcus lactis, which can produce extracellular vesicles (EVs) that carry a variety of biomolecules, we successfully constructed a genetically modified L. lactis strain that expresses miR-146b (LL-miR-146b) in a nisin-depen
