Deliberations on the microplastic-guided rare heavy metal toxicity in agricultural crops grown around nuclear reactors: molecular regulation and strategies for remediation.

Rapid growth in the nuclear energy sector has led to increased construction of nuclear power plants (NPPs). Although this promotes the generation of alternate sources of "clean" energy that does not harm the environment, potential concerns regarding soil and water pollution with microplastics and multiple rare heavy metals (HMs) used in NPPs usually do not grab the required attention. The group of rare HMs comprises of uranium, cadmium, mercury, cobalt, germanium, and indium, which are known ecological toxins affecting agricultural quality and consumer safety. When expunged as nuclear waste di

Mito-nuclear incompatibility disrupts ACOX1/ACOX3-mediated mitochondria-peroxisome metabolic coordination in black soldier flies.

Mito-nuclear coadaptation is a fundamental evolutionary process linking genomic interactions to metabolic performance and organismal fitness. Although mito-nuclear incompatibility is known to impair mitochondrial function and contribute to hybrid breakdown, its effects on metabolic coordination between cellular organelles remain poorly understood, particularly in insects. Here, we used the black soldier fly (Hermetia illucens L.) as an experimental model and established a mito-nuclear incompatibility system by introducing heterologous mitochondrial genomes into a common nuclear background. The multi-faceted role of retinoid X receptor in cardiovascular diseases.

Retinoid X receptors (RXRs) are members of ligand-dependent transcription factors whose effects on a diversity of cellular processes, including cellular proliferation, the immune response, and lipid and glucose metabolism. Knock out of RXRα causes a hypoplasia of the myocardium which is lethal during fetal life. In addition, the heart maintains a well-orchestrated balances in utilizing fatty acids (FAs) and other substrates to meet the high energy requirements. As the master transcriptional regulators of lipid metabolism, RXRs become particularly important for the energy needs of the heart. Ac

Fourth-order perturbative model for photoinduced internal conversion processes.

Essential to the functionality of numerous biological and synthetic molecular systems is the ability to rapidly convert electronic excitation energy into heat. Such internal conversion (IC) transitions often cannot be described by traditional second-order kinetic theories because of time-coincident electronic and nuclear relaxation processes. Here, we present a perturbative fourth-order phenomenological model for photoinduced IC that incorporates effects associated with finite laser bandwidths and nonequilibrium nuclear motions. Specialized knowledge of first-principles computational methods i

RIP140 deficiency enhances cardiac fuel metabolism and protects mice from heart failure.

During the development of heart failure (HF), the capacity for cardiomyocyte (CM) fatty acid oxidation (FAO) and ATP production is progressively diminished, contributing to pathologic cardiac hypertrophy and contractile dysfunction. Receptor-interacting protein 140 (RIP140, encoded by Nrip1) has been shown to function as a transcriptional corepressor of oxidative metabolism. We found that mice with striated muscle deficiency of RIP140 (strNrip1-/-) exhibited increased expression of a broad array of genes involved in mitochondrial energy metabolism and contractile function in heart and skeletal

Transcriptional Integration of Distinct Microbial and Nutritional Signals by the Small Intestinal Epithelium.

BACKGROUND & AIMS: The intestine constantly interprets and adapts to complex combinations of dietary and microbial stimuli. However, the transcriptional strategies by which the intestinal epithelium integrates these coincident sources of information remain unresolved. We recently found that microbiota colonization suppresses epithelial activity of hepatocyte nuclear factor 4 nuclear receptor transcription factors, but their integrative regulation was unknown. METHODS: We compared adult mice reared germ-free or conventionalized with a microbiota either fed normally or after a single high-fat m

Mitochondrial energy production genes affecting reactive oxygen species and ATP synthesis, inflammatory pathways, and neuron viability are differentially expressed in brains of Alzheimer's disease patients.

BackgroundWe have previously reported that a subset of nuclear-encoded mitochondrial genes involved in mitochondrial biogenesis was differentially expressed in Alzheimer's disease (AD) brains. These were associated with compromised biological pathways of mitochondria such as mitochondrial morphology, fragmentation, transmembrane potential and neuronal cell death.ObjectiveTo use an array of energy production genes to determine whether expression changes compromise mitochondrial function and other important biological processes or pathways impacting the development of AD.MethodsRT2-PCR arrays we

Vitamin D and energy homeostasis: of mice and men.

The vitamin D endocrine system has many extraskeletal targets, including adipose tissue. 1,25-Dihydroxyvitamin D₃, the active form of vitamin D, not only increases adipogenesis and the expression of typical adipocyte genes but also decreases the expression of uncoupling proteins. Mice with disrupted vitamin D action--owing to gene deletion of the nuclear receptor vitamin D receptor (Vdr) or the gene encoding 1α-hydroxylase (Cyp27b1)--lose fat mass over time owing to an increase in energy expenditure, whereas mice with increased Vdr-mediated signalling in adipose tissue become obese. Effects of radiation exposure and storage on the energy metabolome of platelets in whole blood.

BACKGROUND: Exposure to radiation through battlefield use of nuclear weapons, terrorist attacks or accidents at nuclear power plants is a current concern for the military. Beyond the risk of exposure to personnel is the intentional or accidental irradiation of our blood banking supply system. It is unknown how large doses of ionizing radiation affect storage of blood and blood products, including platelets. The major function of platelets is clot formation which includes aggregation, shape change, vesicle release, and fibrinogen attachment; these tasks require a significant amount of energy. H

HACD3 integrates central leptin sensitivity and peripheral thermogenesis to oppositely regulate energy balance.

The enzyme 3-hydroxyacyl-CoA-dehydratase 3 (HACD3) is involved in fatty acid synthesis, but its systemic role in metabolism is unknown. This study investigated the physiological function of HACD3 in energy homeostasis. Systemic HACD3 deficiency protected mice from diet-induced obesity by increasing energy expenditure and activating adipose thermogenesis. Conversely, deleting HACD3 specifically in leptin receptor-expressing neurons caused obesity, hyperphagia, and leptin resistance. In adipocytes, HACD3 cell-autonomously repressed thermogenesis by forming a complex with Keratin 1 (KRT1), which

Research on New and Traditional Energy Sources in OECD Countries.

To mitigate the problems associated with climate change, the low-carbon economy concept is now being championed around the world in an effort to reduce greenhouse gas emissions and ensure sustainable economic growth. Therefore, to reduce the dependence on traditional energy sources, the Organization for Economic Co-operation and Development (OECD) has been actively promoting the use of renewable energy. Past research has tended to neglect the influence of other pollutants such as fine particulate matter (PM2.5) and sulfur dioxide (SO₂) and have mainly been based on static analyses. To make up

Mechanobiology of the Nucleolus.

Ribosome biogenesis is an essential and energy-demanding process required to maintain cellular proteostasis. The nucleolus, which orchestrates this vital process, assembles as a nuclear condensate and exhibits remarkable plasticity in its structure, function, and protein composition. This dynamic membraneless organization of the nucleolus contributes to its involvement in diverse signaling pathways, influencing cell cycle regulation, proliferation, apoptosis, differentiation, and cellular stress response. Here, we focus on how mechanical cues influence nucleolar biology. Although classical mec

circ_0043837 attenuates macrophage mitochondrial damage through XPO1-associated HuR transport in atherosclerosis.

Macrophage mitochondrial damage is implicated in the pathogenesis of atherosclerosis. Circular RNAs (circRNAs) regulate mitochondrial energy metabolism and contribute to the development of various diseases, but the mechanism in atherosclerosis regulation remains to be elucidated. This study aimed to investigate the underlying mechanisms by which circRNAs modulate mitochondrial function in the progression of atherosclerosis. We established a macrophage-derived foam cell model to define the role of circ_0043837 in mitochondrial dysfunction during atherosclerosis. Foam cells showed loss of mitoch

Current density functional framework for spin-orbit coupling.

Relativistic two-component density functional calculations are carried out in a non-collinear formalism to describe spin-orbit interactions, where the exchange-correlation functional is constructed as a generalization of the non-relativistic density functional approximation. Contrary to non-relativistic density functional theory (DFT), spin-orbit coupling, however, leads to a non-vanishing paramagnetic current density. Density functionals depending on the kinetic energy density, such as meta-generalized gradient approximations, should therefore be constructed in the framework of current DFT (C

Mitochondrial Neurodegeneration.

Mitochondria are cytoplasmic organelles, which generate energy as heat and ATP, the universal energy currency of the cell. This process is carried out by coupling electron stripping through oxidation of nutrient substrates with the formation of a proton-based electrochemical gradient across the inner mitochondrial membrane. Controlled dissipation of the gradient can lead to production of heat as well as ATP, via ADP phosphorylation. This process is known as oxidative phosphorylation, and is carried out by four multiheteromeric complexes (from I to IV) of the mitochondrial respiratory chain, ca

The impact of the International Atomic Energy Agency (IAEA) program on radiation and tissue banking in Mexico.

Tissue banking started in Mexico in 1948-1949, when two bone banks were established, one at the Infantile Hospital of Mexico and other at the Central Military Hospital. Mexico has benefited for the implementation of the IAEA program since through it has been able to settle down and to consolidate the Tissue Bank at the Instituto Nacional de Investigaciones Nucleares ININ (National Institute for Nuclear Research). This is the only bank in Latin America that has a Quality Management System in force, certified under ISO 9001:2000 since August 1, 2003. The first tissue processed was amnion. The ma

High Accuracy ab Initio Calculations of Rotational-Vibrational Levels of the HCN/HNC System.

Highly accurate ab initio calculations of vibrational and rotational-vibrational energy levels of the HCN/HNC (hydrogen cyanide/hydrogen isocyanide) isomerising system are presented for several isotopologues. All-electron multireference configuration interaction (MRCI) electronic structure calculations were performed using basis sets up to aug-cc-pCV6Z on a grid of 1541 geometries. The ab initio energies were used to produce an analytical potential energy surface (PES) describing the two minima simultaneously. An adiabatic Born-Oppenheimer diagonal correction (BODC) correction surface as well

Massively parallel implementation of gradients within the random phase approximation: Application to the polymorphs of benzene.

The Random-Phase approximation (RPA) provides an appealing framework for semi-local density functional theory. In its Resolution-of-the-Identity (RI) approach, it is a very accurate and more cost-effective method than most other wavefunction-based correlation methods. For widespread applications, efficient implementations of nuclear gradients for structure optimizations and data sampling of machine learning approaches are required. We report a well scaling implementation of RI-RPA nuclear gradients on massively parallel computers. The approach is applied to two polymorphs of the benzene crysta
