The role of inner nuclear membrane protein emerin in myogenesis.

Emerin, a ubiquitously expressed inner nuclear membrane protein, plays a central role in maintaining nuclear structure and genomic organization, and in regulating gene expression and cellular signaling pathways. These functions are critical for proper myogenic differentiation and are closely linked to the pathology of Emery-Dreifuss muscular dystrophy 1 (EDMD1), a laminopathy caused by mutations in the EMD gene. Emerin, along with other nuclear lamina proteins, modulates chromatin organization, cell signaling, gene expression, and cellular mechanotransduction, processes essential for muscle de

The emerging role of estrogen's non-nuclear signaling in the cardiovascular disease.

Sexual dimorphism exists in the epidemiology of cardiovascular disease (CVD), which indicates the involvement of sexual hormones in the pathophysiology of CVD. In particular, ample evidence has demonstrated estrogen's protective effect on the cardiovascular system. While estrogen receptors, bound to estrogen, act as a transcription factor which regulates gene expressions by binding to the specific DNA sequence, a subpopulation of estrogen receptors localized at the plasma membrane induces activation of intracellular signaling, called "non-nuclear signaling" or "membrane-initiated steroid signa

Loss of β-catenin promotes chondrogenic differentiation of aortic valve interstitial cells.

OBJECTIVE: The Wnt/β-catenin signaling pathway has been implicated in human heart valve disease and is required for early heart valve formation in mouse and zebrafish. However, the specific functions of Wnt/β-catenin signaling activity in heart valve maturation and maintenance in adults have not been determined previously. APPROACH AND RESULTS: Here, we show that Wnt/β-catenin signaling inhibits Sox9 nuclear localization and proteoglycan expression in cultured chicken embryo aortic valves. Loss of β-catenin in vivo in mice, using Periostin(Postn)Cre-mediated tissue-restricted loss of β-cateni

Posttranslational modification of β-catenin is associated with pathogenic fibroblastic changes in bronchopulmonary dysplasia.

Bronchopulmonary dysplasia (BPD) is a common complication of premature birth. The histopathology of BPD is characterized by an arrest of alveolarization with fibroblast activation. The Wnt/β-catenin signaling pathway is important in early lung development. When Wnt signaling is active, phosphorylation of β-catenin by tyrosine kinases at activating sites, specifically at tyrosine 489 (Y489), correlates with nuclear localization of β-catenin. We examined fetal lung tissue, lung tissue from term newborns, and lung tissue from infants who died with BPD; we found nuclear β-catenin phosphorylation a

The Vascular Endothelial Growth Factor A-Vascular Endothelial Growth Factor Receptor 2 Axis Drives Malignant Potential in Oral Squamous Cell Carcinoma by Promoting Stemness and Cell Proliferation.

Oral squamous cell carcinoma (OSCC) is an aggressive malignancy with poor clinical outcomes driven by metastasis and recurrence. Vascular endothelial growth factor receptor 2 (VEGFR2), a key mediator of vascular endothelial growth factor A (VEGFA)-dependent angiogenesis, has recently been implicated in tumor cell-intrinsic signaling; however, its role in OSCC remains unclear. This study investigated VEGFA-VEGFR2 signaling in OSCC using a 4-nitroquinoline 1-oxide-induced mouse carcinogenesis model and human tissue specimens. Spatial transcriptomic analysis of murine tongues demonstrated enrichm

Distinct mechanisms of nuclear accumulation regulate the functional consequence of E2F transcription factors.

Transcription factor E2F plays an important role in coordinating and integrating early cell cycle progression with the transcription apparatus. It is known that physiological E2F arises when a member of two families of proteins, E2F and DP, interact as E2F/DP heterodimers and that transcriptional activity is regulated through the physical association of pocket proteins such as pRb. However, little information is available regarding the mechanisms which control the levels of functional E2F. In this study, we have characterised one such mechanism which regulates the nuclear accumulation and acti

Translational control mechanisms in cutaneous malignant melanoma: the role of eIF2α.

BACKGROUND: Melanoma cells develop adaptive responses in order to cope with particular conditions of tumor microenvironment, characterized by stress conditions and deregulated proliferation. Recently, the interplay between the stress response and the gene expression programs leading to metastatic spread has been reported. METHODS: We evaluated levels and localization of eIF2α/peIF2α in V600BRAF and wtBRAF metastatic melanoma cell lines by means of western blot and confocal microscopy analyses. Furthermore, we performed a sequence analyses and structure and dynamics studies of eIF2α protein to

Close but distinct regions of human herpesvirus 8 latency-associated nuclear antigen 1 are responsible for nuclear targeting and binding to human mitotic chromosomes.

Human herpesvirus 8 is associated with all forms of Kaposi's sarcoma, AIDS-associated body cavity-based lymphomas, and some forms of multicentric Castleman's disease. Herpesvirus 8, like other gammaherpesviruses, can establish a latent infection in which viral genomes are stably maintained as multiple episomes. The latent nuclear antigen (LANA or LNAI) may play an essential role in the stable maintenance of latent episomes, notably by interacting concomitantly with the viral genomes and the metaphase chromosomes, thus ensuring an efficient transmission of the neoduplicated episomes to the daug

Enhanced Precision of Fluorescence In Situ Hybridization (FISH) Analysis Using Neural Network-Based Nuclear Segmentation for Digital Microscopy Samples.

Introduction: Accurate nuclear segmentation is essential for the reliable diagnostic interpretation of fluorescence in situ hybridization (FISH) results. However, automated 2D digital algorithms often fail in samples with dense or overlapping nuclei, such as lymphomas, due to the loss of spatial depth information. Here, we tested if AI-based 3D nuclear segmentation can improve the accuracy, reproducibility, and diagnostic reliability of FISH reading in critical situations. Materials and Methods: Formalin-fixed follicular lymphoma sections were FISH-labeled for BCL2 gene rearrangements and digi

Nuclear ubiquitin proteasome degradation affects WRKY45 function in the rice defense program.

The transcriptional activator WRKY45 plays a major role in the salicylic acid/benzothiadiazole-induced defense program in rice. Here, we show that the nuclear ubiquitin-proteasome system (UPS) plays a role in regulating the function of WRKY45. Proteasome inhibitors induced accumulation of polyubiquitinated WRKY45 and transient up-regulation of WRKY45 target genes in rice cells, suggesting that WRKY45 is constantly degraded by the UPS to suppress defense responses in the absence of defense signals. Mutational analysis of the nuclear localization signal indicated that UPS-dependent WRKY45 degrad

The differential osmoregulation and localization of taurine transporter mRNA and Na+/myo-inositol cotransporter mRNA in rat eyes.

We studied the cellular localization and osmotic regulation of taurine transporter (TauT) mRNA in the rat eyes using in situ hybridization. TauT mRNA signals were expressed in the ciliary body, and the outer part of the inner nuclear layer (INL), the outer nuclear layer (ONL) and the inner segment (IS) of the adult rat retina. Chronic hypernatrema, induced by gavaging with 1 ml/100 g body weight of 5% NaCl every other day for 7 days, markedly increased in TauT mRNA in the retina compared with control rats. However, there was little change in TauT mRNA in the eyes in acute hypernatremic state t

Symmetric chromosomal order in leukocytes indicated by DNA image cytometry and fluorescence in situ hybridization.

OBJECTIVE: To study if the symmetry observed in the nuclear structure of polymorphonuclear cells (PNC) could be verified functionally by determining DNA and chromosome distributions using image cytometry (ICM) and fluorescence in situ hybridization (FISH) STUDY DESIGN: In murine and human leukocytes the proportion of PNCs with apparently bilaterally symmetric nuclei was determined. By ICM, total nuclear DNA and DNA content in each half of these nuclei were measured. For chromosomal localization the FISH technique was applied. RESULTS: About 13% of PNC nuclei appeared to be bilaterally symmetr

Liver Stiffness Rises Early in MASLD and Drives Inflammation, Lipid Dysmetabolism, and Fibrosis via Piezo1-YAP Mechanotransduction.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is shaped by metabolic injury and tissue mechanics. This study investigated whether liver stiffening occurs early in MASLD and how extracellular matrix (ECM) mechanics interact with lipid droplet (LD) overload to promote inflammation, fibrogenesis, and lipid dysmetabolism. Clinical data, mouse models, and in vitro experiments are integrated. Liver stiffness shifted modestly with steatosis but increased substantially in the presence of inflammation. In a diet-induced mouse model, liver stiffness increased before overt fibrosis. In

New role for hPar-1 kinases EMK and C-TAK1 in regulating localization and activity of class IIa histone deacetylases.

Class IIa histone deacetylases (HDACs) are found both in the cytoplasm and in the nucleus where they repress genes involved in several major developmental programs. In response to specific signals, the repressive activity of class IIa HDACs is neutralized through their phosphorylation on multiple N-terminal serine residues and 14-3-3-mediated nuclear exclusion. Here, we demonstrate that class IIa HDACs are subjected to signal-independent nuclear export that relies on their constitutive phosphorylation. We identify EMK and C-TAK1, two members of the microtubule affinity-regulating kinase (MARK)

Histones, protamine, and polylysine but not poly(E:K) enhance transfection efficiency.

Liposomal gene delivery has a great potential for the treatment of cancer and other human diseases. In this work we have investigated the optimal conditions for liposome-mediated transfer of the luciferase gene to human erythroleukemia K562 cells. DDAB:DOPE liposomes were more efficient than lysyl-DOPE:DOPE (1:2) and DDAB:cholesterol for transfection. Total histones from bovine thymus, salmon sperm protamine, and polylysine at an optimal ratio of 0.5 mg protein/mg DNA enhanced up to 7-fold the transfection efficiency of luciferase plasmids; on the contrary, the synthetic polymer poly(E:K), con

Karyopherin α 3 and karyopherin α 4 proteins mediate the nuclear import of methyl-CpG binding protein 2.

Methyl-CpG binding protein 2 (MeCP2) is a nuclear protein with important roles in regulating chromatin structure and gene expression, and mutations in MECP2 cause Rett syndrome (RTT). Within the MeCP2 protein sequence, the nuclear localization signal (NLS) is reported to reside between amino acids 255-271, and certain RTT-causing mutations overlap with the MeCP2 NLS, suggesting that they may alter nuclear localization. One such mutation, R270X, is predicted to interfere with the localization of MeCP2, but recent in vivo studies have demonstrated that this mutant remains entirely nuclear. To cl

Nuclear envelope localization of Ran-binding protein 2 and Ran-GTPase-activating protein 1 in psoriatic epidermal keratinocytes.

The nuclear localization signal (NLS)-containing proteins LEDGF and STAT3 localize to the nucleus in both the spinous and basal layers of the epidermis in psoriatic skin, where they function as transcription factors or co-factors to activate epidermal keratinocytes (KCs). However, the mechanism underlying the localization of these proteins remains to be elucidated. We investigated the differential nucleocytoplasmic transport of NLS-containing proteins as a potential pathogenic mechanism for psoriasis vulgaris. Nucleoporins play an important role in the Ran-GTP-dependent nucleocytoplasmic trans

KRIT1 nuclear localization does not impair KRIT1-dependent regulation of endothelial barrier permeability and cell homeostasis.

KRIT1 plays a key role in regulating the barrier function of endothelial cells, where it localizes to the adherens junctions, cytoplasm, and nucleus. However, how subcellular localization may regulate KRIT1 remains unclear. Here, we investigate what effect nuclear localization has on its ability to stabilize the endothelial barrier. We generated a KRIT1 mutant lacking the endogenous nuclear localization signal (NLS) and a nuclear-targeted KRIT1 isoform created by attaching the NLS of simian virus 40 and expressed these constructs in cells depleted of endogenous KRIT1. After confirming the rela

Genome-Wide Identification and Functional Characterization of the TIFY Gene Family Associated with Abiotic Stress and Hormone Response in Carthamus tinctorius L.

Jasmonate-ZIM domain (JAZ) proteins act as core negative regulators of jasmonic acid signaling and mediate plant stress adaptation, developmental progression, and secondary metabolism. Safflower (Carthamus tinctorius L.) is an economically important Asteraceae crop with medicinal, edible oil, and ornamental values; however, the evolutionary characteristics and molecular functions of its TIFY gene family remain poorly characterized. In the present study, we performed a genome-wide identification of the safflower CtTIFY family, followed by systematic analyses of phylogenetic relationships, chrom

Serotype-Specific Regulation of Dengue Virus NS5 Protein Subcellular Localization.

Dengue virus (DENV) nonstructural protein 5 (NS5), consisting of methyltransferase and RNA-dependent RNA polymerase (RdRp) domains, is critical for viral RNA synthesis within endoplasmic reticulum-derived replication complexes in the cytoplasm. However, a significant proportion of NS5 is localized to the nucleus of infected cells for DENV2, 3, and 4, whereas DENV1 NS5 is localized diffusely in the cytoplasm. We still have an incomplete understanding of how the DENV NS5 subcellular localization is regulated. Within NS5, two putative nuclear localization signal (NLS) sequences have been identifi
