AEG-1 on cancer drug resistance: From molecular insights to therapeutic challenges.

Astrocyte elevated gene-1 (AEG-1), also known as metadherin (MTDH), has emerged as a multifunctional oncogene implicated in cancer progression, metastasis, immune evasion, and notably, drug resistance across diverse malignancies. AEG-1 exerts its effects by modulating key signaling cascades, including PI3K/Akt, NF-κB, and Wnt/β-catenin, and by regulating genes associated with epithelial-mesenchymal transition, apoptosis suppression, cancer stemness, and multidrug resistance. Its interactions with molecular partners such as SND1, USP10, and nucleolin further amplify its oncogenic potential, esp

An integrative meta-analysis of SARS-CoV-2 RNA-protein interactomes identifies conserved host factors shared with other RNA viruses.

RNA viruses cause substantial global disease burden and depend on host RNA-binding proteins and translation machinery. However, it remains unclear which host factors are robustly engaged across independent Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) RNA interactome studies and to what extent these factors are shared with other RNA viruses. Here, we perform an integrative meta-analysis of eight published SARS-CoV-2 RNA-protein interactomes and compare them with corresponding Influenza A virus, Zika virus, and Dengue virus datasets to define conserved host networks and prioritiz

SARS-CoV-2 and HSV-1 Induce Amyloid Aggregation in Human CSF Resulting in Drastic Soluble Protein Depletion.

The corona virus (SARS-CoV-2) pandemic and the resulting long-term neurological complications in patients, known as long COVID, have renewed interest in the correlation between viral infections and neurodegenerative brain disorders. While many viruses can reach the central nervous system (CNS) causing acute or chronic infections (such as herpes simplex virus 1, HSV-1), the lack of a clear mechanistic link between viruses and protein aggregation into amyloids, a characteristic of several neurodegenerative diseases, has rendered such a connection elusive. Recently, we showed that viruses can ind

Uterine fluid proteome changes during diapause and resumption of embryo development in roe deer (Capreolus capreolus).

The uterine microenvironment during pre-implantation presents a pro-survival milieu and is essential for embryo elongation in ruminants. The European roe deer (Careolus capreolus) pre-implantation embryo development is characterised by a 4-month period of reduced development, embryonic diapause, after which the embryo rapidly elongates and implants. We investigated the uterine fluid proteome by label-free liquid chromatography tandem mass spectrometry at four defined stages covering the phase of reduced developmental pace (early diapause, mid-diapause and late diapause) and embryo elongation.

A ribonucleoprotein complex protects the interleukin-6 mRNA from degradation by distinct herpesviral endonucleases.

During lytic Kaposi's sarcoma-associated herpesvirus (KSHV) infection, the viral endonuclease SOX promotes widespread degradation of cytoplasmic messenger RNA (mRNA). However, select mRNAs escape SOX-induced cleavage and remain robustly expressed. Prominent among these is interleukin-6 (IL-6), a growth factor important for survival of KSHV infected B cells. IL-6 escape is notable because it contains a sequence within its 3' untranslated region (UTR) that can confer protection when transferred to a SOX-targeted mRNA, and thus overrides the endonuclease targeting mechanism. Here, we pursued how

Cellular adaptation to hypoxia and p53 transcription regulation.

Tumor suppressor p53 is the most frequently mutated gene in human tumors. Meanwhile, under stress conditions, p53 also acts as a transcription factor, regulating the expression of a series of target genes to maintain the integrity of genome. The target genes of p53 can be classified into genes regulating cell cycle arrest, genes involved in apoptosis, and genes inhibiting angiogenesis. p53 protein contains a transactivation domain, a sequence-specific DNA binding domain, a tetramerization domain, a non-specific DNA binding domain that recognizes damaged DNA, and a later identified proline-rich

Aptamer-Functionalized DNA Frameworks as Targeted Drug Buffers for Concentration Regulation of Maytansine.

Traditional chemotherapy often suffers from nonspecific distribution and poor drug concentration control, leading to compromised therapeutic efficacy and adverse effects. Mertansine (DM1), a potent cytotoxic agent with poor selectivity, dose-limiting toxicity, and a narrow therapeutic window, demands targeted delivery and precise concentration regulation for safety and efficacy. Herein, we constructed two Janus DNA triangular prism-based molecular buffers (3A-JTP-3S2 and A-MTP-3S2) for targeted DM1 delivery and concentration regulation. Both of the buffers integrate nucleolin-targeting aptamer

Paclitaxel-loaded nanostructured lipid carriers surface-functionalized with N6L for pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most aggressive and lethal malignancies, with limited therapeutic options. Despite advances in chemotherapy, such as FOLFIRINOX and Abraxane® (nab-paclitaxel) combined with gemcitabine, these treatments show limited efficacy due to systemic toxicity, poor tumor specificity, and drug resistance. Nanomedicine offers a promising strategy to improve drug delivery, reduce off-target effects, and enhance therapeutic efficacy. In this study, we developed and physicochemically characterized paclitaxel (PTX)-loaded nanostructured lipid carriers

(Non)canonical Wnt signaling, cytoarchitecture and stemness: new insights from primary nonmetastatic, primary metastatic, regional and distant metastatic models of adrenocortical carcinoma.

Tumor cells rearrange cytoskeletal networks, harmonize signaling pathways and modulate cell fate to initiate tumor growth and metastasis. Activating mutations in the canonical Wnt-β-catenin pathway are associated with aggressiveness and poor prognosis in adrenocortical carcinoma (ACC). However, the contribution of noncanonical Wnt pathways, which are known to regulate cell polarity and migration, is still widely unknown. Here we comprehensively investigated canonical and noncanonical Wnt signaling along with cytoskeleton networks, stemness programs and HOX gene expression in recently developed

The longevity-associated BPIFB4 gene supports cardiac function and vascularization in ageing cardiomyopathy.

AIMS: The ageing heart naturally incurs a progressive decline in function and perfusion that available treatments cannot halt. However, some exceptional individuals maintain good health until the very late stage of their life due to favourable gene-environment interaction. We have previously shown that carriers of a longevity-associated variant (LAV) of the BPIFB4 gene enjoy prolonged health spans and lesser cardiovascular complications. Moreover, supplementation of LAV-BPIFB4 via an adeno-associated viral vector improves cardiovascular performance in limb ischaemia, atherosclerosis, and diabe

PIKE mediates EGFR proliferative signaling in squamous cell carcinoma cells.

One of the key drivers for squamous cell carcinoma (SCC) proliferation is activation of the epidermal growth factor receptor (EGFR), a known proto-oncogene. However, the mechanism of EGFR-dependent SCC proliferation remains unclear. Our previous studies indicate that epidermal growth factor (EGF)-induced SCC cell proliferation requires the SH3 domain of phospholipase C-γ1 (PLC-γ1), but not its catalytic activity. The SH3 domain of PLC-γ1 is known to activate the short form of nuclear phosphatidylinositol 3-kinase enhancer (PIKE) that enhances the activity of nuclear class Ia phosphatidylinosit

c- and N-myc regulate neural precursor cell fate, cell cycle, and metabolism to direct cerebellar development.

Separate murine knockout (KO) of either c- or N-myc genes in neural stem and precursor cells (NSC) driven by nestin-cre causes microcephaly. The cerebellum is particularly affected in the N-myc KO, leading to a strong reduction in cerebellar granule neural progenitors (CGNP) and mature granule neurons. In humans, mutation of N-myc also causes microcephaly in Feingold Syndrome. We created a double KO (DKO) of c- and N-myc using nestin-cre, which strongly impairs brain growth, particularly that of the cerebellum. Granule neurons were almost absent from the Myc DKO cerebellum, and other cell type

Enhanced targeted liposomal delivery of imiquimod via aptamer functionalization for head and neck cancer therapy.

The incidence of head and neck cancers, particularly those associated with Human Papillomavirus (HPV) infections, has been steadily increasing. Conventional therapies exhibit limitations and drawbacks, prompting the exploration of new strategies over the years, with nanomedicine approaches, especially liposomes gaining relevance. Additionally, the functionalization of liposomes with aptamers enables selective delivery to target cells. For instance, AT11 can serve as a targeting moiety for cancer cells due to its high affinity for nucleolin, a protein overexpressed on the cancer cell's surface.

Oxidative damage-induced hyperactive ribosome biogenesis participates in tumorigenesis of offspring by cross-interacting with the Wnt and TGF-β1 pathways in IVF embryos.

In vitro fertilization (IVF) increases the risk of tumorigenesis in offspring. The increased oxidative damage during IVF may be involved in tumor formation. However, the molecular mechanisms underlying this phenomenon remain largely unclear. Using a well-established model of oxidatively damaged IVF mouse embryos, we applied the iTRAQ method to identify proteins differentially expressed between control and oxidatively damaged zygotes and explored the possible tumorigenic mechanisms, especially with regard to the effects of oxidative damage on ribosome biogenesis closely related to tumorigenesis

Tumor microenvironment-responsive dynamic self-assembly of DNAzyme nanowires for high-contrast imaging of multiple messenger RNAs and chemotherapy.

Messenger RNAs (mRNAs) are a category of protein-coding RNA, and their dysregulation is closely implicated in diverse cancers. Static DNA nanodevices have been engineered for the imaging and therapy, but they rely on the pre-assembly of DNA components and are limited by the low imaging contrast. Herein, we demonstrate the tumor microenvironment-responsive dynamic self-assembly of DNAzyme nanowires for high-contrast imaging of multiple mRNAs and chemotherapy. We design a cruciform DNA module (C-model) as a basic structural unit of DNAzyme nanowire, which consists of two DNAzyme strands and two

Stabilization of a DNA aptamer by ligand binding.

G-rich aptamers such as AS1411 are small oligonucleotides that present several benefits comparatively to monoclonal antibodies, since they are easier to manufacture and store, have small size and do not stimulate an immune response. We analyzed AT11-B1, a modified sequence of AT11 (itself a modified version of AS1411), in which one thymine was removed from the bulge region. We studied G-quadruplex (G4) formation/stabilization using PhenDC3, PDS, BRACO-19, TMPyP4 and 360A ligands by different biophysical techniques, namely circular dichroism (CD), Förster resonance energy transfer (FRET-melting

Novel Naturally Occurring Dipeptides and Single-Stranded Oligonucleotide Act as Entry Inhibitors and Exhibit a Strong Synergistic Anti-HIV-1 Profile.

INTRODUCTION: The availability of new classes of antiretroviral drugs is critical for treatment-experienced patients due to drug resistance to and unwanted side effects from current drugs. Our aim was therefore to evaluate the anti-HIV-1 activity of a new set of antivirals, dipeptides (WG-am or VQ-am) combined with a single-stranded oligonucleotide (ssON). The dipeptides were identified as naturally occurring and enriched in feces and systemic circulation in HIV-1-infected elite controllers and were proposed to act as entry inhibitors by binding to HIV-1 gp120. The ssON is DNA 35-mer, stabiliz

HDGF stimulates liver tumorigenesis by enhancing reactive oxygen species generation in mitochondria.

Hepatoma-derived growth factor (HDGF) overexpression and uncontrolled reactive oxygen species (ROS) accumulation are involved in malignant transformation and poor prognosis in various types of cancer. However, the interplay between HDGF and ROS generation has not been elucidated in hepatocellular carcinoma. Here, we first analyzed the profile of HDGF expression and ROS production in newly generated orthotopic hepatomas by ultrasound-guided implantation. In situ superoxide detection showed that HDGF-overexpressing hepatomas had significantly elevated ROS levels compared with adjacent nontumor t
