Hydrogen sulfide affects the performance of a methanogenic bioelectrochemical system used for biogas upgrading.

Methanogenic bioelectrochemical systems (BESs) can convert carbon dioxide (CO2) to methane (CH4) and may be used for anaerobic digester biogas upgrading. However, the effect of hydrogen sulfide (H2S), a common biogas component, on BES performance is unknown. Thus, the objective of this study was to assess the effect of H2S addition to the cathode headspace on BES performance at a range of initial gas-phase H2S concentrations (0-6% v/v), as well as its effect on the anode and cathode microbial communities. As the initial cathode headspace H2S increased from 0 to 2% (v/v), biocathodic CH4 produc

A MALDI-TOF MS database with broad genus coverage for species-level identification of Brucella.

Brucella are highly infectious bacterial pathogens responsible for a severely debilitating zoonosis called brucellosis. Half of the human population worldwide is considered to live at risk of exposure, mostly in the poorest rural areas of the world. Prompt diagnosis of brucellosis is essential to prevent complications and to control epidemiology outbreaks, but identification of Brucella isolates may be hampered by the lack of rapid and cost-effective methods. Nowadays, many clinical microbiology laboratories use Matrix-Assisted Laser Desorption Ionization-Time Of Flight mass spectrometry (MALD

Diversity, biofilm formation and antimicrobial susceptibility of aerobic heterotrophic bacteria isolated from cooling towers.

Cooling towers (CTw) are essential for industrial refrigeration but provide favourable conditions for microbial growth and biofilm formation, potentially facilitating the spread of antimicrobial-resistant bacteria (ARB). This study examines bacterial diversity, biofilm potential and antimicrobial susceptibility in two industrial CTw in Brazil using culture-based and metataxonomic (16 S rRNA gene amplicon sequencing) approaches. Fourteen bacterial genera and 22 species were identified, including Bacillus spp. (60% of isolates), Acinetobacter spp. (14%) and others like Pseudomonas, Serratia, and

Insights into the alteration of vaginal microbiota and metabolites in pregnant woman with preterm delivery: prospective cohort study.

Disruptions in vaginal microbiota and metabolites during pregnancy may be the most important risk factor for preterm delivery, thus the difference in vaginal microbiota and metabolites between women who subsequently delivered at term and who eventually experienced preterm birth. In this study, 63 participants were enrolled before the cervical cerclage surgery (namely pre-cerclage), comprising women who subsequently delivered at term and who eventually experienced preterm birth. The cervical-vaginal fluid (CVF) was collected two days prior to the cervical cerclage surgery. Compared with the ter

Diversity of Bacterial Endophytes of Maize (Zea mays) and Their Functional Potential for Micronutrient Biofortification.

Microorganisms due, to their immense metabolic diversity, have the potential to augment the uptake of iron and zinc in addition to other important nutrients in plants. In the present work, 129 different strains of endophytic bacteria were retrieved from stems and leaves of maize. Qualitative screening of these endophytes showed that 24.5% of these isolates were siderophore producers, while 14% could solubilize insoluble zinc compounds and 33% of them had phytase activity. Based on zinc solubilization efficiency and siderophore production ability, 10 isolates each from zinc solubilizers and sid

Brucellosis: an overview.

Brucellosis remains a major zoonosis worldwide. Although many countries have eradicated Brucella abortus from cattle, in some areas Brucella melitensis has emerged as a cause of infection in this species as well as in sheep and goats. Despite vaccination campaigns with the Rev 1 strain, B. melitensis remains the principal cause of human brucellosis. Brucella suis is also emerging as an agent of infection in cattle, thus extending its opportunities to infect humans. The recent isolation of distinctive strains of Brucella from marine mammals has extended its ecologic range. Molecular genetic stu

Wild felids as sentinels of antimicrobial resistance: susceptible Escherichia coli isolates from a protected area in southeastern Brazil.

Antimicrobial resistance (AMR) is a major global health concern, and its monitoring in wildlife is essential from a One Health perspective. Here, we obtained bacterial isolates and assessed the AMR profiles and extended-spectrum β-lactamase (ESBL) production of Escherichia coli isolated from fecal samples of wild felids in a protected Atlantic Forest area in southeastern Brazil. We collected 22 fecal samples and identified depositor species through trichological analysis (ocelot, Leopardus pardalis = 18; jaguar, Panthera onca = 1; puma, Puma concolor = 1; Leopardus sp. = 1; unidentified felid

The new strains Brucella inopinata BO1 and Brucella species 83-210 behave biologically like classic infectious Brucella species and cause death in murine models of infection.

BACKGROUND: Recently, novel atypical Brucella strains isolated from humans and wild rodents have been reported. They are phenotypically close to Ochrobactrum species but belong to the genus Brucella, based on genetic relatedness, although genetic diversity is higher among the atypical Brucella strains than between the classic species. They were classified within or close to the novel species Brucella inopinata. However, with the exception of Brucella microti, the virulence of these novel strains has not been investigated in experimental models of infection. METHODS: The type species B. inopin

Phlebotomus duboscqi gut microbiota dynamics in the context of Leishmania infection.

INTRODUCTION: The manipulation of the gut microbiota of disease vectors has emerged as a new approach to use in the integrated control of vector-borne diseases. For this purpose, a deep knowledge of their gut microbial communities is essential. To our knowledge, to date, no study has documented the gut microbiome dynamics of Phlebotomus duboscqi sand flies over the entire time-period required for the maturation of a Leishmania infection. Here, we address this limitation. METHODS: P. duboscqi midguts were dissected both before and at different days after L. major infection and subjected to gen

Raman spectroscopy as a potential tool for detection of Brucella spp. in milk.

Detection of Brucella, causing brucellosis, is very challenging, since the applied techniques are mostly time-demanding and not standardized. While the common detection system relies on the cultivation of the bacteria, further classical typing up to the biotype level is mostly based on phenotypic or genotypic characteristics. The results of genotyping do not always fit the existing taxonomy, and misidentifications between genetically closely related genera cannot be avoided. This situation gets even worse, when detection from complex matrices, such as milk, is necessary. For these reasons, the

Development of a PCR assay for typing and subtyping of Brucella species.

In the course of this study, examinations were carried out to develop a PCR-based test which allows discrimination of Brucella species and biovars not targeted by the currently established gel-based PCR assays. Appropriate primers were designed based on specific deletions and insertions in the different Brucella genomes as determined by RAPD-PCR and whole-genome comparisons. After testing the specificity of the primers with a set of 22 Brucella reference strains of all species and biovars, they were used to supplement the existing PCR assays resulting in a 19-primer multiplex PCR. In addition

Use of amplified fragment length polymorphism to identify and type Brucella isolates of medical and veterinary interest.

Amplified fragment length polymorphism (AFLP) is a whole-genome fingerprinting method that relies on the selective PCR amplification of restriction fragments. The potential of this approach for the discrimination of Brucella isolates at the species and intraspecies level was assessed. A number of different combinations of restriction enzymes and selective primers were examined, and one, using EcoRI and MseI with additional selective TC bases on the MseI primer, was selected for full assessment against a panel of Brucella isolates. The technique could readily differentiate Brucella spp. from al

Manganese residue harmlessness with the combination of manganese mineralizing microbes, CaO and CO2.

Manganese residue, including electrolytic manganese residue (EMR) and electrolytic manganese dioxide residue (EMDR), have complicated compositions and are easily generated heavy metals (HMs) and ammonia nitrogen (NH4+-N) pollution. In this study, HMs and NH4+-N in manganese residue were stabilized and solidified using a combined remediation system of quicklime (CaO), carbon dioxide (CO2), and manganese mineralizing microbes. The sequencing of the manganese mineralizing microbes assemblage revealed that Pseudomonas geniculata, Leclercia adecaarboxylata, Ochrobactrum spp., and Delftia tsuruhaten

Microbial degradation as a powerful weapon in the removal of sulfonylurea herbicides.

Sulfonylurea herbicides have been widely used worldwide and play a significant role in modern agricultural production. However, these herbicides have adverse biological effects that can damage the ecosystems and harm human health. As such, rapid and effective techniques that remove sulfonylurea residues from the environment are urgently required. Attempts have been made to remove sulfonylurea residues from environment using various techniques such as incineration, adsorption, photolysis, ozonation, and microbial degradation. Among them, biodegradation is regarded as a practical and environment

Phenotypic and Molecular Characterization of Brucella microti-Like Bacteria From a Domestic Marsh Frog (Pelophylax ridibundus).

Several Brucella isolates have been described in wild-caught and "exotic" amphibians from various continents and identified as B. inopinata-like strains. On the basis of epidemiological investigations conducted in June 2017 in France in a farm producing domestic frogs (Pelophylax ridibundus) for human consumption of frog's legs, potentially pathogenic bacteria were isolated from adults showing lesions (joint and subcutaneous abscesses). The bacteria were initially misidentified as Ochrobactrum anthropi using a commercial identification system, prior to being identified as Brucella spp. by MALD

Analysis of ten Brucella genomes reveals evidence for horizontal gene transfer despite a preferred intracellular lifestyle.

The facultative intracellular bacterial pathogen Brucella infects a wide range of warm-blooded land and marine vertebrates and causes brucellosis. Currently, there are nine recognized Brucella species based on host preferences and phenotypic differences. The availability of 10 different genomes consisting of two chromosomes and representing six of the species allowed for a detailed comparison among themselves and relatives in the order Rhizobiales. Phylogenomic analysis of ortholog families shows limited divergence but distinct radiations, producing four clades as follows: Brucella abortus-Bru

Nasopharyngeal microbiome of COVID-19 patients revealed a distinct bacterial profile in deceased and recovered individuals.

The bacterial co-infections in SARS-CoV-2 patients remained the least explored subject of clinical manifestations that may also determine the disease severity. Nasopharyngeal microbial community structure within SARS-CoV-2 infected patients could reveal interesting microbiome dynamics that may influence the disease outcomes. Here, in this research study, we analyzed distinct nasopharyngeal microbiome profile in the deceased (n = 48) and recovered (n = 29) COVID-19 patients and compared it with control SARS-CoV-2 negative individuals (control) (n = 33).
