Olfactory Function and Olfactory Disorders.

The sense of smell is important. This became especially clear to patients with infection-related olfactory loss during the SARS-CoV-2 pandemic. We react, for example, to the body odors of other humans. The sense of smell warns us of danger, and it allows us to perceive flavors when eating and drinking. In essence, this means quality of life. Therefore, anosmia must be taken seriously. Although olfactory receptor neurons are characterized by regenerative capacity, anosmia is relatively common with about 5 % of anosmic people in the general population. Olfactory disorders are classified accordin

Does intranasal application of zinc sulfate produce anosmia in the mouse? An olfactometric and anatomical study.

Mice pre-trained in an olfactometer were tested daily on odor detection and discrimination tasks after irrigation of their olfactory epithelium in each naris with 50 microl of 5% zinc sulfate or saline. Anterograde transport of a wheatgerm agglutinin-horseradish peroxidase (WGA-HRP) conjugate from the epithelium to the olfactory bulb was used to assess anatomical connectivity in these and in mice that were used only for histological analyses. One day after treatment, saline controls performed at high levels of accuracy in detecting vapor from solutions of 5-0.01% ethyl acetate and in an odor d

Cadaver dogs: unscientific myth or reliable biological devices?

Dogs are commonly used to detect explosives, narcotics, and other illegal materials. In the forensic setting, cadaver dogs are trained to detect and locate concealed human remains or fluids due to the high sensitivity and selectivity of the canine olfactory system and the relative ease with which dogs can be trained and handled. The need for international and scientifically validated standards has long been outlined by the literature. It is important, therefore, to establish the reliability of the handler/dog team. Our study aimed to detect the real effectiveness of dogs trained to locate huma

Olfactory receptors and human diseases.

Olfaction plays a crucial role in distinguishing odors, enabling organisms to seek benefits and evade hazards. Olfactory receptors (ORs), characterized by highly variable binding pockets, facilitate the detection of diverse odorants from both external and internal environments. Nasal ORs, expressed in olfactory sensory neurons (OSNs), are critical for olfactory cognition and associated neuronal plasticity. In contrast, extra-nasal ORs, expressed in extra-olfactory tissues, detect specific chemicals and modulate cellular processes such as proliferation, migration, inflammation, and apoptosis. A

Olfactory and gustatory perception in adults with long COVID.

INTRODUCTION: Olfactory dysfunction is a common symptom in COVID-19 and Long COVID. However it is unclear how well perceived impairment of olfactory and gustatory dysfunction correlate with the objectively measured intensity of olfactory dysfunction among adults with Long COVID compared to healthy never-SARS-CoV-2-infected and previously infected control cohorts. MATERIALS AND METHODS: We recruited 154 participants from an online Long COVID survey (DEFEAT Corona) for in-person examinations. Participants were allocated to three groups: Long COVID (persistent symptoms >4 weeks post-infection, n

Post-viral olfactory loss and parosmia.

The emergence of SARS-CoV-2 has brought olfactory dysfunction to the forefront of public awareness, because up to half of infected individuals could develop olfactory dysfunction. Loss of smell-which can be partial or total-in itself is debilitating, but the distortion of sense of smell (parosmia) that can occur as a consequence of a viral upper respiratory tract infection (either alongside a reduction in sense of smell or as a solo symptom) can be very distressing for patients. Incidence of olfactory loss after SARS-CoV-2 infection has been estimated by meta-analysis to be around 50%, with mo

Excitable properties of olfactory receptor neurons.

Action potential-generating properties of olfactory receptor neurons in the olfactory epithelium of the salamander, Ambystoma tigrinum, were studied in control animals, and 2 and 4 weeks after olfactory nerve transection. The threshold for impulse generation in response to injected current was extremely low (74 +/- 46 pA). In addition, the discharge frequencies of the receptor neurons were exquisitely sensitive to small increments of injected current. These high sensitivities may be characteristic of small neurons and stand in contrast to the much lower sensitivities reported for large neurons

Therapies for Olfactory Dysfunction - an Update.

PURPOSE OF REVIEW: Olfactory dysfunction is a prevalent condition affecting 5-15% of the general population, with significant impact on quality of life. This review summarizes the most recent and relevant literature in the treatment of olfactory dysfunction. RECENT FINDINGS: Current evidence supports the short-term use of topical corticosteroids and systemic therapy. These treatments may occur in conjunction with olfactory training, which is well supported by the literature. While there are several additional treatments currently under investigation, meaningful conclusions are not yet able to

Olfactory and gustatory disorders associated with SARS-CoV-2 infection in children and adults: a topic review.

This paper is aimed at comprehensively reviewing olfactory and gustatory disorders caused by SARS-CoV-2 in children and adults. An electronic and manual search was done on three databases: MEDLINE, Embase, and Web of Science. Inclusion criteria included publications written in English, involving humans in the age range of 0 to 99 years that were captured by a controlled vocabulary of thesaurus terms. Olfactory and gustatory disorders rates in COVID-19 ranged from 22% to 71.9% in adults and 16.6% to 25.8% in children. Olfactory and gustatory disorders might appear as the first symptom, and in a

Olfactory Decline in Elderly at High Altitudes: A Narrative Review.

Olfactory dysfunction is highly prevalent in the elderly population, severely affecting nutritional status, environmental safety perception, and quality of life, and is recognized as an early warning marker of neurodegenerative diseases. Compared with populations living at low altitudes, residents at high altitudes are chronically exposed to combined extreme environmental conditions including hypoxia, low temperature, and low humidity, which may lead to more complex and severe degenerative changes in the olfactory system. However, existing evidence remains fragmented, and a systematic synthesi

Phase-dependent filtering of sensory information in the oscillatory olfactory center of a terrestrial mollusk.

With electrophysiological techniques, we found phase-dependent modification of the efficacy of signal transmission in the procerebrum (PC), the oscillatory olfactory center, of the terrestrial mollusk Limax marginatus and elucidated its neuronal mechanism. Previous studies have indicated that about 10(5) PC neurons can be classified into only two types: bursting (B) neurons and nonbursting (NB) neurons, and both types of neurons have ongoing and phase-locked periodic oscillation of their membrane potentials. On olfactory nerve stimulation, excitatory postsynaptic potentials (EPSPs) were evoked

Intact olfactory memory in the 5xFAD mouse model of Alzheimer's disease from 3 to 15 months of age.

Alzheimer's disease (AD) is an age-related neurodegenerative disorder that causes profound cognitive dysfunction. Deficits in olfactory memory occur in early stages of AD and may be useful in AD diagnosis. The 5xFAD mouse is a commonly used model of AD, as it develops neuropathology, cognitive and sensori-motor dysfunctions similar to those seen in AD. However, olfactory memory dysfunction has not been studied adequately or in detail in 5xFAD mice. Furthermore, despite sex differences in AD prevalence and symptom presentation, few studies using 5xFAD mice have examined sex differences in learn

An olfactory-based Brain-Computer Interface: electroencephalography changes during odor perception and discrimination.

Brain-Computer Interfaces (BCIs) are devices designed for establishing communication between the central nervous system and a computer. The communication can occur through different sensory modalities, and most commonly visual and auditory modalities are used. Here we propose that BCIs can be expanded by the incorporation of olfaction and discuss the potential applications of such olfactory BCIs. To substantiate this idea, we present results from two olfactory tasks: one that required attentive perception of odors without any overt report, and the second one where participants discriminated co

Involvement of Adult-born and Preexisting Olfactory Bulb and Dentate Gyrus Neurons in Single-trial Olfactory Memory Acquisition and Retrieval.

The production of new neurons and their incorporation into preexisting neuronal circuits occur throughout adulthood in the olfactory bulb and the hippocampal dentate gyrus of the mammalian brain. To determine whether the adult-born neurons are engaged in the acquisition and retrieval of olfactory associative memory, we developed and validated a single-trial olfactory fear conditioning protocol in mice which allows to detect activation of newborn neurons during a specific episode of memory acquisition. Using c-Fos mapping of neuronal activity, we then examined the activation of new and preexist

Olfactory implant: Demand for a future treatment option in patients with olfactory dysfunction.

OBJECTIVES: Therapeutic options in olfactory dysfunction (OD) are limited. Numerous studies have shown impact of OD on quality of life. Lately, various studies support benefits of olfactory training, but therapy-refractory cases leave doctors and patients locked in a stalemate. An olfactory implant (OI), in analogy to the widely successful cochlear implant, still seems far away from realization. The present study sought to evaluate the demand of OI in patients with OD. METHODS: Sixty-one patients (28 females and 33 males, mean age/standard deviation 54.9/17.6 years) with OD were recruited. We

Interhemispheric transfer of olfactory information in homing pigeon.

The role of the anterior commissure in discrimination of and habituation to olfactory stimuli was studied in pigeons by measuring changes in the heart rate. In order to exclude any trigeminal response, we presented odorous stimuli to groups of pigeons with one olfactory nerve cut and either the ipsilateral or the contralateral nostril plugged. When the nostril involved in the experiments was on the same side of the cut olfactory nerve, the pigeons did not show any response, whereas they displayed changes in heart rate when the nostril tested was on the opposite side. In habituation transfer ex

Enhancement of odorant-induced mucosal activity patterns in rats trained on an odorant identification task.

Previous studies have demonstrated that there are intrinsic spatial patterns of odorant sensitivity across the rat olfactory mucosa. The question of how these patterns are determined and whether they are modifiable with experience remains open. Therefore, the present study examined whether the odorant-induced spatial activity patterns which are characteristic of different odorants would be altered by experience. Odorant exposure was achieved as a consequence of training and testing on a five odorant identification task in which rats were trained to differentially report (i.e. identify) the odo
