Polymerization-dependent gut microbiome responses to curdlan β-1,3-glucan and its oligosaccharides.

β-1,3-Glucan is a dietary fiber with microbiome-modulating potential, yet the role of polymerization degree on microbial composition and metabolism remains unclear. Curdlan is an insoluble linear β-1,3-glucan widely used as a food hydrocolloid that provides a model to examine structure-dependent microbiome responses. We compared glucose, curdlan (CBG), and curdlan-derived β-1,3-glucan oligosaccharides (OBG) using anaerobic in vitro fermentation of pooled human feces. A multi-omics approach integrating 16S rRNA sequencing, shotgun metagenomics, short-chain fatty acid (SCFA) quantification, and

Conformation and solvation shell of stearoyl-modified hyaluronic-acid oligosaccharides in binary solvents.

Hyaluronic acid (HA) is a natural hydrophilic polysaccharide with wide applications in healthcare and cosmetics, often substituted by non-polar aliphatic residues, giving it an amphiphilic nature. The stability of secondary structures of these molecules depends on the length of the HA chain, the type of the substituent, the degree of substitution and the composition of the used solvent - its influence is investigated in this molecular-dynamics study. Sole 18-carbon saturated aliphatic chains and their couples are simulated in three solvent systems: water, water : 1,4-dioxane and water : tert-b

Chemoenzymatic Synthesis of Heparan Sulfate Oligosaccharides by a Covalent Catch-and-Release Approach.

A covalent catch-and-release approach is described for the chemoenzymatic synthesis of HS oligosaccharides. It exploits that a glycan modified by a hydrazide tag can under mild acidic conditions efficiently be reacted with an aldehyde-containing resin to give an immobilized hydrazone product. The hydrazone is stable under neutral conditions, allowing stringent washing conditions to remove all other components. The product can be released by transiminolysis using aqueous hydroxylamine, which, after evaporation, yields a compound that can be immediately employed in the next cycle of modification

Enzymatic depolymerization, structural characterization, and anticoagulant activity evaluation of chondroitin sulfate A oligosaccharides.

Chondroitin sulfate (CS) polysaccharides exhibit intrinsic structural heterogeneity, limiting precise studies of their bioactivities and therapeutic applications. To obtain homogeneous CS oligosaccharides and elucidate structure-activity relationships, we developed a controlled enzymatic method for preparing well-defined chondroitin sulfate A (CSA) oligosaccharides and investigated their chain length-dependent anticoagulant mechanisms. Herein, hyaluronidase was used to depolymerize CSA and subsequently purified by sequential ultrafiltration and anion-exchange chromatography, yielding CSA oligo

Agaro-Oligosaccharides Modulate Gut Microbiota to Improve Lipid Metabolism in Obesity Model Rats.

Obesity is a global health concern, prompting extensive research to identify food components with potential anti-obesity properties. Agaro-oligosaccharides (AOS) have been reported to have anti-obesity effects; however, the precise mechanisms underlying these effects, particularly in relation to the gut microbial metabolites, remain unclear. This study investigated the anti-obesity effects of AOS using Otsuka Long-Evans Tokushima Fatty (OLETF) rats, an established animal model of obesity. Seven-week-old Long-Evans Tokushima Otsuka (LETO) and OLETF rats were fed a control diet or a diet supplem

Oligosaccharides: a comprehensive review of various types, biological activities and emerging health benefits.

Oligosaccharides (OS's) have gained significant attention as functional food ingredients due to their diverse health-promoting properties and broad industrial applicability. OS support the maintenance and proliferation of beneficial gut microbiota, thereby contributing to improved gastrointestinal health. Beyond their prebiotic functions, OS serve as low-calorie sweeteners and stabilizers while aiding in the prevention of dental caries, modulation of immune responses, regulation of blood glucose and lipid profiles, enhancement of mineral bioavailability, and exertion of antioxidant effects. Human Milk Oligosaccharides Mediate the Host-Microbe Interface in a Model Vaginal Community.

Group B Streptococcus (GBS) is an opportunistic bacterium that can cause severe infection during gestation, leading to adverse pregnancy outcomes and neonatal disease. As current treatments only decrease chances of early onset neonatal disease without impacting the risk of chorioamnionitis, preterm birth, or late-onset disease, novel therapeutics are needed. Here, we demonstrate that human milk oligosaccharides (HMOs) positively modulate cocultures of GBS and Lactobacillus spp., common inhabitants of a healthy vaginal microbiome, across in vitro, ex vivo, and in vivo experiments. HMOs shift th

Chitosan Oligosaccharides Prevent Alcohol-Induced Liver Disease by Attenuating Inflammation and Oxidative Stress.

Alcoholic liver disease (ALD) is a liver disorder resulting from excessive alcohol intake, and currently, there are no therapeutics approved by the FDA for its treatment. This study investigates the protective effects and underlying pharmacological mechanisms of two chitosan oligosaccharides, COST (MW ≤ 1000 Da) and COSM (MW ≤ 3000 Da), in mitigating alcohol-induced liver disease (ALD). In animal models, we evaluated the changes in ALD following treatment with COST and COSM. Histopathological analysis revealed that both COST and COSM interventions mitigated hepatic steatosis and inflammatory i

Vaginal prevention of Candida albicans: synergistic effect of lactobacilli and mannan oligosaccharides (MOS).

Vulvovaginal candidiasis (VVC) affects approximately 30-50% of women at least once during their lifetime, causing uncomfortable symptoms and limitations in their daily quality of life. Antifungal therapy is not very effective, does not prevent recurrencies and usually causes side effects. Therefore, alternative therapies are urgently needed. The goal of this work was to investigate the potential benefits of using mannan oligosaccharides (MOS) extracts together with a Lactobacillus sp. pool, composed by the most significant species present in the vaginal environment, to prevent infections by Ca

Oligosaccharides act as the high efficiency stabilizer for β-galactosidase under heat treatment.

β-Galactosidase (β-Gal) as dietary supplement has the ability to alleviate symptoms of lactose intolerance. This study investigated the ability of oligosaccharides to protect β-Gal against heat stress. Four kinds of oligosaccharides including Isomalto-oligosaccharides (IMO), Xylo-oligosaccharides (XOS), Konjac-oligosaccharides (KOS), and Mycose significantly increased the activity retention of β-Gal under heat treatment. The results of three assays including circular dichroism, fluorescence, and Fourier transform infrared spectroscopy (FTIR) illustrated that these oligosaccharides could stabil

Effects of galacto-oligosaccharides on growth and gut function of newborn suckling piglets.

BACKGROUND: Cervical cancer is a major public health problem. In 2018, globally 569,847 cervical cancer were diagnosed and 311,000 deaths were projected due to this preventable disease. Worldwide, therefore, the cervical cancer disease ranks as the fourth most frequently diagnosed cancer and the fourth leading cause of cancer death in women in 2018.The high rate of dysplasia in Senegal and the absence of well-organized screening programs informed this study, which aims to determine the prevalence of cervical dysplasia and its relationship to biological and socio-demographic characteristics. Preparation, characterization and immunogenicity of HIV-1 related high-mannose oligosaccharides-CRM197 glycoconjugates.

The dense glycan shield on the surface of human immunodeficiency virus type 1 (HIV-1) gp120 masks conserved protein epitopes and facilitates virus entry via interaction to glycan binding proteins on susceptible host cells. The broadly neutralizing monoclonal antibody 2G12 binds a cluster of high-mannose oligosaccharides on the gp120 subunit of HIV-1 Env protein. This oligomannose epitope is currently being considered for the design of a synthetic vaccine. The cluster nature of the 2G12 epitope suggests that a multivalent antigen presentation is important to develop a carbohydrate-based vaccine

Beta1,6-branched oligosaccharides regulate melanin content and motility in macrophage-melanoma fusion hybrids.

In previous studies, fusion of peritoneal macrophages or blood monocytes with mouse melanoma cells produced hybrids with upregulated expression of the glycosyltransferase beta1,6-N-acetylglucosaminyltransferase V (GnT-V) and its enzymatic product, beta1,6-branched oligosaccharides. This correlated with marked increases in motility, metastatic potential and, surprisingly, melanin content. This study was designed to establish direct roles for beta1,6-branched oligosaccharides in melanogenesis and motility. The levels of beta1,6-branched oligosaccharides were lowered by transfecting beta1,4-N-ace

Endoplasmic reticulum-to-cytosol transport of free polymannose oligosaccharides in permeabilized HepG2 cells.

Free polymannose oligosaccharides have recently been localized to both the vesicular and cytosolic compartments of HepG2 cells. Here we investigated the possibility that free oligosaccharides originating in the lumen of the endoplasmic reticulum (ER) are transported directly into the cystosol. Incubation of permeabilized cells in the absence of ATP at 37 degrees C led to the intravesicular accumulation of free Man9GlcNAc2 which was generated from dolichol-linked oligosaccharide in the ER. This oligosaccharide remained stable within the permeabilized cells unless ATP was added to the incubation

[Thin-layer chromatography of urine oligosaccharides in diagnosis of some lysosomal storage disorders].

Inherited lysosomal storage disorders are caused by the deficiency or importantly lowered activity of one of the lysosomal enzymes, leading to the storage in the lysosomes the not degraded high-molecular substrates, among others: mucopolysaccharides, glycolipids, oligosaccharides and glycoproteins. Thin-layer chromatography of urine oligosaccharides allows reliable and fast diagnosis of some lysosomal storage disorders e.g. alpha-mannosidosis, fucosidosis, sialidosis, galactosialidosis, Schindler disease, GM1-gangliosidosis, GM2-gangliosidosis (Sandhoff type), Pompe disease, Salla disease, muc

Evidence for processing of dolichol-linked oligosaccharides in patients with neuronal ceroid-lipofuscinosis.

In agreement with reports from other laboratories, we have shown that patients with the juvenile or late infantile forms of neuronal ceroid-lipofuscinosis (NCL) have greatly increased levels (5-fold to 20-fold) of dolichyl pyrophosphoryl oligosaccharides in their cerebral gray matter. Oligosaccharides containing 2 GlcNAc residues and 3 to 9 mannose residues were liberated by mild acid hydrolysis. The oligosaccharide profile given by brain tissue from 2 patients with infantile NCL was markedly different from that of late infantile and juvenile NCL brain, with Man9GlcNAc2 as the most abundant co

Synthesis of Human Milk Oligosaccharides-Fluoroquinolone Conjugates and Investigation of Their Antimicrobial Activity.

The copper-catalyzed azide-alkyne cycloaddition reaction was applied to conjugate four human milk oligosaccharides (HMOs) with antimicrobial fluoroquinolones (FQs) (ciprofloxacin and sparfloxacin) to obtain a total of 12 per-O-acetylated and unprotected HMO-FQ conjugates. These compounds underwent antimicrobial testing against Gram-positive (Streptococcus agalactiae, Enterococcus faecalis, and Staphylococcus aureus) and Gram-negative (Escherichia coli and Pseudomonas aeruginosa) bacteria. The HMO-FQ conjugates exhibited the most significant antimicrobial activity against E. coli and S. aureus

Diversity-Oriented Enzymatic Synthesis of Human Milk Oligosaccharides Bearing a 3-Fucosyllactose Core.

A notable subset of human milk oligosaccharides (HMOs) bearing an α1,3-fucosylated lactose (3FL) moiety at the reducing end constitutes one of the most abundant and representative classes of HMOs, referred to as 3FL-related HMOs (3FL-HMOs). Despite their significant biological relevance, the systematic synthesis of 3FL-HMOs has remained challenging due to their structural diversity and complexity, which has hindered comprehensive functional studies. Herein, we report a highly efficient enzymatic modular assembly strategy for the collective synthesis of 32 3FL-HMOs, encompassing 5 core backbone

Systematic Virtual Screening of Heparan Sulfate Oligosaccharides and Mechanistic Insights into Heparanase Inhibition.

Heparanase (Hpse) is an endo-β-glucuronidase that specifically cleaves heparan sulfate (HS) and plays critical roles in the pathogenesis of multiple diseases, making it a well-validated therapeutic target. To identify novel Hpse inhibitors, we developed a rule-based computational pipeline to construct a rigorously curated database of HS oligosaccharides ranging from disaccharides to hexasaccharides. This database systematically encompasses all possible sulfation patterns and stereochemical configurations of the three monosaccharide species in HS. Using a multistage virtual screening strategy a
