BACKGROUND: Eimeria are obligate protozoan parasites, and more than 1,500 species have been reported. However, Eimeria genomes lag behind many other eukaryotes since obtaining many oocysts is difficult due to a lack of sustainable in vitro culture, highly repetitive sequences, and mixed species infections. To address this challenge, we used whole-genome amplification of a single oocyst followed by long-read sequencing and obtained a chromosome-level genome of Eimeria tenella.
RESULTS: The assembled genome was 52.13 Mb long, encompassing 15 chromosomes and 46.94% repeat sequences. In total, 7,

Recent molecular characterizations of Cryptosporidium parasites make it possible to differentiate the human-pathogenic Cryptosporidium parasites from those that do not infect humans and to track the source of Cryptosporidium oocyst contamination in the environment. In this study, we used a small-subunit rRNA-based PCR-restriction fragment length polymorphism (RFLP) technique to detect and characterize Cryptosporidium oocysts in 55 samples of raw surface water collected from several areas in the United States and 49 samples of raw wastewater collected from Milwaukee, Wis. Cryptosporidium parasi

OBJECTIVE: To evaluate the infectivity of Cryptosporidium parvum oocysts in NMRI suckling mouse.
METHODS: Four-day-old SPF NMRI suckling mice were inoculated with different amounts of oocysts by oral gavage. On day 7 after inoculation, suckling mice were sacrificed, and a suspension was prepared by homogenizing the intestinal tract from pylorus to anus. A mouse was considered infected when oocysts were found in smears of the intestinal content suspension stained with carbo lfuchsin solution. The infectivity of oocysts was evaluated as measured by the percentage of infected mice in each group.

Cryptosporidium parvum infections of domestic animals can have a considerable economic impact and as oocysts are voided in the faeces of infected hosts, environmental contamination with agricultural waste has also become a matter of concern. Since only viable oocysts are potentially infectious, the numbers of oocysts excreted during infection can have important implications for both veterinary and public health. During the course of infection in experimentally infected lambs, oocyst viability was assessed by a fluorogenic vital dyes assay and by a maximized in vitro excystation assay. The excr

Cryptosporidium spp. oocyst shedding was observed in calves from approximately 1 to 30 days of age. Oocysts were detected by either the Kinyoun acid-fast staining technique (microscopic examination--ME) or a commercially produced enzyme immunoassay EIA). Test concordance between the two detection methods was determined. The mean (+/- SD) number of days to detection of cryptosporidial oocysts was 9.52 +/- 1.92 for the ME and 9.83 +/- 3.19 for the EIA. No significant difference between the means was found (P = 0.17). The period prevalence of cryptosporidiosis was 100% in calves from 1 to 30 days

Demineralized water was enriched with a known number of Cryptosporidium parvum oocysts obtained from fresh calf feces, which were purified and exposed to ultraviolet (UV) light (15,000 mW/sec) for different lengths of time. Coccidium-free litters of CD-1 neonatal mice then were inoculated intragastrically with the treated water. Seven days postinoculation mice were killed and C. parvum infection prevalence and intensity determined. In mice inoculated with C. parvum-enriched water that had been exposed to UV light for at least 150 min, no infection occurred.

Infection by Cryptosporidium was detected in 94 (71.75%) asymptomatic adult cattle from 131 fecal samples examined microscopically. In two cases Cryptosporidium oocysts were observed which were distinctly larger (5.5-6.5 microns x 6.6-7.0 microns) than those we had seen in the majority of feces examined (4.0-4.5 microns x 4.0-4.5 microns) and these specimens were considered to be Cryptosporidium muris; it is possible that the other oocysts should be considered as Cryptosporidium parvum. The seroprevalence of IgG antibodies to Cryptosporidium was 63.35% as detected by indirect fluorescent antib

Four hundred fecal specimens which had been received for routine ova and parasite examination were concentrated by Formalin-ether sedimentation. Sediments were examined as saline and iodine-stained wet preparations and were stained with rhodamine-auramine O and a commercially available monoclonal fluorescent-antibody stain for oocysts of Cryptosporidium species. Examination with the fluorescent stains detected cryptosporidia in both positive specimens (0.5% prevalence), and routine direct wet-preparation examination detected cryptosporidia in one of them. Detection of only low numbers of posit

Features are described of a new staining method for the detection of Cryptosporidium oocysts in faeces. Smears, fixed in acid-methanol, are stained with heated safranin and counterstained with methylene blue. Oocysts stain a vivid orange-pink and are easily recognized. The method is rapid and simple with little source of error. The method is more sensitive than currently recommended Ziehl-Neelson methods; of 26 cases of cryptosporidiosis diagnosed by the new method only 19 were detected by acid-fast and only 11 by acid-alcohol-fast methods. Oocysts can be stained by safranin-methylene blue aft

Information on the viability of Toxoplasma gondii oocysts is crucial to establish the public health significance of this environmental transmission stage that can contaminate water and foods. Interest for molecular-based methods to assess viability is growing and the aim of our study was to assess, for the first time, a propidium monoazide (PMA)-qPCR approach to determine the viability of T. gondii oocysts. Untreated and heat-killed (99 °C, 5 min) oocysts were incubated with PMA, a photoreactive DNA binding dye, and analyzed by confocal microscopy and flow cytometry to characterize oocysts' dy

Avian coccidiosis is the most important parasitic disease in poultry production, which inflicts numerous losses to the industry. The extensive use of anticoccidial drugs leads to parasite resistance and drug residue in poultry products. In the present study, we aimed to investigate the effects of three famous essential oils (EOs) and their combination on inactivation of mixed oocysts of Eimeria adenoides, Eimeria dispersa, Eimeria meleagrimitis, and Eimeria meleagridis. The EOs of Thymus vulgaris, Artemisia sieberi, and Mentha pulegium were prepared. After inoculation of each turkey with 7&

The objective was to describe the probability of Cryptosporidium parvum fecal oocyst shedding at different magnitudes of exposure, the pattern of fecal shedding over time, and factors affecting fecal shedding in dairy calves. Within the first 24 h of life, 36 calves were experimentally challenged with C. parvum oocysts at one of four possible magnitudes of oral exposure (1 × 10(3), 1 × 10(4), 1 × 10(5), and 1 × 10(6) oocysts), and 7 control calves were sham dosed. Fecal shedding occurred in 33 (91.7 %) experimentally challenged calves and in none of the control calves. There was a difference i

The survival kinetics of purified Cryptosporidium parvum oocysts of both human and ovine origin, immersed in four still natural mineral waters (total dissolved salts ranging from 91 mg/liter to 430 mg/liter) and reverse osmosis water was assessed by inclusion or exclusion of the fluorogenic vital dyes 4',6-diamidino-2-phenylindole and propidium iodide over a 12-week period. Semipermeable chambers were used to contain the oocysts while immersed in each mineral water type, permitting both intimate interactions between oocysts and matrices and straightforward sampling for viability assessments. The dispersion and initial transport of Cryptosporidium oocysts from fecal pats were investigated during artificial rainfall events on intact soil blocks (1,500 by 900 by 300 mm). Rainfall events of 55 mm h(-1) for 30 min and 25 mm h(-1) for 180 min were applied to soil plots with artificial fecal pats seeded with approximately 10(7) oocysts. The soil plots were divided in two, with one side devoid of vegetation and the other left with natural vegetation cover. Each combination of event intensity and duration, vegetation status, and degree of slope (5 degrees and 10 degrees ) was evaluated twi

Two commercially valuable mollusc species ( Ostrea edulisand Tapes decussatus) were experimentally contaminated with Cryptosporidium parvum oocysts. A direct immunofluorescent antibody technique and inclusion/exclusion of the fluorogenic vital dye propidium iodide were used to test for the presence and viability of the oocysts, showing that transmission of contamination occurred between coexisting species. There was a decrease in the viability of oocysts in the initially uncontaminated molluscs as well as a large decrease in the number of oocysts retained when dead molluscs were used as the so

There is no paucity of methods for diagnosing Cryptosporidium spp. infection. The merits of immunoassays notwithstanding, microscopic identification of Cryptosporidium spp. oocysts in fecal samples remains an important diagnostic procedure. It owes the persistence of its use to such characteristics as dispensing with expensive equipment and kits, requiring only basic laboratory facilities, and having a low probability of false positive results when permanent slides are prepared, which can be re-examined in case of doubt. Cryptosporidium spp. oocysts can be readily identified in fecal smears pr

Several in vitro surrogates have been developed as convenient, user-friendly alternatives to mouse infectivity assays for determining the viability of Cryptosporidium parvum oocysts. Such viability assays have been used increasingly to determine oocyst inactivation following treatment with chemical, physical, or environmental stresses. Defining the relationship between in vitro viability assays and oocyst infectivity in susceptible hosts is critical for determining the significance of existing oocyst inactivation data for these in vitro assays and their suitability in future studies. This study evaluated the effects of the human serine protease inhibitor alpha-1-antitrypsin (AAT) on in vitro excystation and infectivity of Cryptosporidium parvum. Excystation was monitored at 37 C in RPMI medium in the presence of 0, 100, 500, or 1,000 micrograms/ml AAT. AAT significantly inhibited (P < 0.05) excystation of bleach-decontaminated oocysts in a concentration-dependent manner at incubation intervals from 15 to 90 min but did not alter the excystation dynamics of unbleached oocysts. Bleach-treated oocysts, suspended in RPMI containing 0, 1, 10, 100, 500, or 1,000 micrograms/ml AA
