Fungicide action is generally assumed to be dependent on an antibiotic effect on a target pathogen, although a role for plant defense mechanisms as mediators of fungicide action has not been excluded. Here, we demonstrate that in Arabidopsis, the innate plant defense mechanism contributes to the effectiveness of fungicides. In NahG and nim1 (for noninducible immunity) Arabidopsis plants, which normally exhibit increased susceptibility to pathogens, the fungicides metalaxyl, fosetyl, and Cu(OH)2 are much less active and fail to control Peronospora parasitica. In contrast, the effectiveness of t

Durian cultivation has become a key development industry in China in recent years, and durian root rot (DRR) constitutes a major threat to production. In this study, symptomatic root and stem base tissues and leaves were systematically collected from the main durian-producing regions of China, yielding 202 fungal and oomycete isolates. Isolates were identified based on morphological characterization and multi-locus phylogenetic analysis. Fusarium spp. and Phytophthora spp. were the most frequently recovered taxa, accounting for 39.11% and 16.34% of the isolates, respectively. Koch's postulates

BACKGROUND: Cucumber plants suffer from a serious threatening disease, downy mildew, throughout the growing seasons irrespective of the weather temperature. The causal agent, Pseudoperonospora cubensis, tends to evolve rapidly upon sequential applications of chemical fungicides and generate new progeny possessing tolerance to such fungicides. Glycoproteins represent an environmentally safe alternative for chemically synthetized fungicides and do not trigger fungicide resistance. We studied the antifungal activity of four glycoproteins namely soybean β-conglycinin, chickpea vicilin, duck egg ov

Salicylic acid (SA), jasmonic acid (JA), and ethylene (ET) are each involved in the regulation of basal resistance against different pathogens. These three signals play important roles in induced resistance as well. SA is a key regulator of pathogen-induced systemic acquired resistance (SAR), whereas JA and ET are required for rhizobacteria-mediated induced systemic resistance (ISR). Both types of induced resistance are effective against a broad spectrum of pathogens. In this study, we compared the spectrum of effectiveness of SAR and ISR using an oomycete, a fungal, a bacterial, and a viral p

A 31-year-old female presented with a 3-week history of right eye redness, ocular pain, photophobia and decreased vision after tap water exposure. Slit-lamp examination revealed corneal lesions with pseudopodia and hill-shaped hypopyon. In vivo confocal microscopy (IVCM) detected filament-like structures in the corneal stroma. The patient was initially suspected of having fungal keratitis, but the condition deteriorated despite standardized antifungal treatment. Repeated corneal scraping culture and targeted DNA sequencing confirmed the infection of Pythium insidiosum. After switching to linez

Pseudoperonospora cubensis, the causal agent of cucurbit downy mildew, causes severe annual losses in cucurbit production. Management relies on frequent fungicide applications, with oxathiapiprolin (OXTP; FRAC 49) being a cornerstone of current programs. Because OXTP is classified as high risk for resistance development, monitoring for resistance alleles is critical. Although OXTP resistance mutations have been reported in other oomycetes, none had previously been documented in P. cubensis. In this study, we reconstructed the P. cubensis oxysterol-binding protein (OSBP) gene, the target of OXT

Fusarium proliferatum is a fungal pathogen with an exceptionally broad host range, infecting ornamental plants and forest trees such as Populus and Cedrus deodara, and causing heart rot, root rot, and other diseases that lead to severe yield losses. It can also produce mycotoxins, including fumonisins, which threaten human and animal health. Conventional detection relies on isolation, culture, and morphological identification, which are time-consuming and prone to misidentification. Here, we established a rapid, visual molecular detection method by combining recombinase polymerase amplificatio

BACKGROUND: To develop novel natural product-derived fungicides, four series of myrtenal oxadiazole/thiadiazole-hydrazide/amide derivatives were designed, synthesized and evaluated for their inhibitions toward several crop pathogenic fungi/oomycetes.
RESULTS: In the in vitro antifungal experiments, some myrtenal derivatives showed good antifungal activity against Botryosphaeria dothidea, Sclerotinia sclerotiorum and Fusarium graminearum. Among them, compound 6d exhibited the best antifungal activity against B. dothidea, with EC50 value of 0.052 mg/L, which was significantly stronger than carb

Invasive fungal infections in cats can result in sinonasal or sino-orbital disease, with brachycephalic breeds reported to be predisposed. However, a subset of cases exhibits isolated orbital signs without clinically evident sinonasal involvement, raising questions about whether the sinonasal component is genuinely absent or subclinical at the time of presentation. This retrospective study describes 11 cats with orbital mycosis lacking overt sinonasal disease, identified through the Comparative Ocular Pathology Laboratory of Wisconsin between 1998 and 2024. All 11 cats were mesocephalic breeds

Fungi and oomycetes found in vineyards cause diseases such as powdery and downy mildew. Consequently, conventional and alternative agronomical practices are widely used prior to harvest to protect grapes. Alternative products are considered more eco-friendly and environmentally sustainable in comparison to conventional chemical products. However, the effect of these alternative products on yeast ecology, from the vineyard to the winery, is poorly understood. This study compared the effect of alternative and conventional chemical antifungal compounds (copper and sulphur based) on grapes' mycobi

Due to their high specificity and efficacy, triazoles have become versatile antifungals to treat fungal infections in human healthcare and to control phytopathogenic fungi in agriculture. However, azole resistance is an emerging problem affecting human health as well as food security. Here we describe the synthesis of 10 novel {2-(3-R-1H-1,2,4-triazol-5-yl)phenyl}amines. Their structure was ascertained by liquid chromatography-mass spectrometry, 1 H and 13 C NMR, and elemental analysis data. Applying an in vitro growth assay, these triazoles show moderate to significant antifungal activity aga

Crayfish plague is the most important disease of freshwater crayfish with a significant impact on European species. We compared the analytical test sensitivity and specificity of three published PCR assays for the detection of Aphanomyces astaci, the causative agent of crayfish plague: a conventional PCR assay targeting the ITS region and two TaqMan(®) real time assays, targeting either the ITS region or the chitinase gene. We also tested a variation of the conventional assay, by changing one of the primers. Test specificity was assessed using DNA from a range of A. astaci strains and an array

Here we present the development and first validation of a TaqMan minor groove binder (MGB) real-time polymerase chain reaction (RT-PCR) method for quantitative and highly specific detection of Aphanomyces astaci, the causative agent of crayfish plague. The assay specificity was experimentally assessed by testing against DNA representative of closely related oomycetes, and theoretically assessed by additional sequence similarity analyses comparing the primers and probe sequences to available sequences in EMBL/GenBank. The target of the assay is a 59 bp unique sequence motif of A. astaci found i

Numerous pathogens invade plants through stomata, and plants, on the other hands, could close the stomata after sensing the pathogens PAMPs. Subsequently, pathogens could overcome this plant immune response with their phytotoxins and effectors. A previous study showed that Plasmopara viticola, a strictly obligate biotrophic oomycete, could inhibit the Vitis vinifera stomatal closure to promote infection. However, the underlying mechanism stimulating the stomata movement is still poorly understood. Here we demonstrate that PvRxLR24, an effector from P. viticola, functions analogously to a phosp

Public metabolites such as vitamins play critical roles in maintaining the ecological functions of microbial community. However, the biochemical and physiological bases for fine-tuning of public metabolites in the microbiome remain poorly understood. Here, we examine the interactions between myxobacteria and Phytophthora sojae, an oomycete pathogen of soybean. We find that host plant and soil microbes complement P. sojae's auxotrophy for thiamine. Whereas, myxobacteria inhibits Phytophthora growth by a thiaminase I CcThi1 secreted into extracellular environment via outer membrane vesicles (OMV

Crayfish plague, a devastating disease of freshwater crayfish, is caused by an oomycete organism, Aphanomyces astaci. Currently five genotypes of A. astaci are known, but variable features between the strains or genotypes have not been studied extensively. This study analysed 28 isolates of the As genotype and 25 isolates of the Ps1 genotype and reveals that the radial growth rate is significantly (P < 0.001) different between these two genotypes, although highly variable inside the genotype As. Two Ps1 genotype isolates and two As genotype isolates with different radial growth rates were test

A 7-month-old, male jaguar presented with dyspnea and leukocytosis unresponsive to antibiotic therapy. Radiographs revealed unilateral pulmonary consolidation. An exploratory thoracotomy was performed, and the left lung, which contained a large multilobular mass with extensive fibrosis and numerous caseonecrotic foci, was removed. Microscopically, eosinophilic granulomatous inflammation surrounded broad (4.4-8.3 microm) rarely septate hyphae. A diagnosis of Pythium insidiosum infection was confirmed by immunohistochemistry, immunoblot serology, culture, and polymerase chain reaction. Dyspnea r

The plant apoplast is integral to intercellular signalling, transport and plant-pathogen interactions. Plant pathogens deliver effectors both into the apoplast and inside host cells, but no computational method currently exists to discriminate between these localizations. We present ApoplastP, the first method for predicting whether an effector or plant protein localizes to the apoplast. ApoplastP uncovers features of apoplastic localization common to both effectors and plant proteins, namely depletion in glutamic acid, acidic amino acids and charged amino acids and enrichment in small amino a

Vacuolar H+-ATPase (V-ATPase) is essential for vacuolar acidification, ion homeostasis, and development in various organisms, including plant pathogens. The V-ATPase a-subunit has been recently identified as the target protein of the oomycete inhibitor fluopicolide, highlighting the favorable druggability of V-ATPase. In this study, we propose a structure-based virtual screening approach to identify novel bioactive compounds targeting the a-c subunit interface of V-ATPase. Following multistep docking and molecular mechanics/generalized born surface area (MM/GBSA) analysis, compound a10 was ide

Rising atmospheric CO2 has significant implications for crop productivity and food security. Based on studies in C3 plants, elevated CO2 (eCO2) can shape plant-pathogen interactions, although the outcomes are often variable. The question of how eCO2 influences immunity and disease development in C4 plants, such as the globally important cereal crop maize (Zea mays L.), has not been systematically examined. We challenged maize plants grown under ambient CO2 (aCO2, 420 ppm) and eCO2 (550 ppm) with bacterial, viral, fungal, and oomycete pathogens. Plants grown in eCO2 were more susceptible to sug
