Hearing loss related to ototoxicity in children with cancer.

Children undergoing treatment for cancer often receive agents that put them at risk for ototoxicity. Platinum-based chemotherapy, aminoglycoside antibiotics, loop diuretics, and radiotherapy are all capable of inducing inner ear damage, which may result in significant sensorineural hearing loss. Frequent audiological monitoring is necessary for the early detection of changes in hearing thresholds. Age-appropriate modification of audiological testing is essential to obtain accurate results and provide maximum comfort for pediatric patients. When hearing loss is identified promptly, consideratio

[A model for the study of the ototoxic effects of drugs in an experiment].

The authors compared sensitivity of the acoustic analyzer elements to ototoxic drugs in cat experiments. The model proposed assesses primary responses of the brain cortical acoustic zone (BCAZ) to sound stimulation. The drugs effect on the analyzer's periphery was determined from changes in bioelectric responses of the cochlea to sound stimulation (microphone potential, potential of the acoustic nerve action). The arguments are presented for feasibility of using primary BCAZ responses evoked by electric stimulation of thalamocortical radiation fibers coming from the medial geniculate body for

Ototoxicity of topical ticarcillin and clavulanic acid in the chinchilla.

BACKGROUND: Currently available topical otic preparations contain a variety of antibiotics and other ingredients that are potentially damaging to the middle and inner ear. There is therefore a need to identify agents that are safe as well as effective for topical otologic use. In pursuit of that goal, we used an animal model to evaluate the ototoxic potential of the broad-spectrum, penicillin derivative ticarcillin--both alone and combined with clavulanic acid (a beta-lactamase inhibitor). METHODS: Twenty chinchillas served as subjects. Ten of the animals were given a single middle ear applic

Trimethyltin ototoxicity: evidence for a cochlear site of injury.

The environmental contaminant, trimethyltin (TMT), produces a profound elevation in tone intensity necessary to inhibit the acoustic startle reflex in laboratory animals which recovers over a prolonged period except at very high frequencies. The recovery that is observed does not begin until 3 to 5 weeks after a single acute administration depending upon dosage. As opposed to the very temporary threshold shifts by the salicylates and loop diuretics or the permanent and progressive ototoxicity resulting from aminoglycoside antibiotics the time course for recovery of acoustic startle reflex inhi

Potentiating effects of cisplatin and ethacrynic acid in ototoxicity.

Seven milligrams per kilogram of body weight of cisplatin and 50 mg/kg of body weight of ethacrynic acid were given intravenously to guinea pigs. The threshold for Preyer's reflex, the cochlear microphonics (CM), and the endocochlear dc potential (EP) were measured during a four-day period after the injection. The changes in Preyer's reflex audiometry and the CM were more severe than that of the EP. These changes are greater than those that occur with the administration of each agent independently. The ototoxic interaction seemingly has a greater effect on the hair cells than on the stria vasc

Potentiation of kanamycin ototoxicity by a history of noise exposure.

Chinchillas were exposed to a noise band (1,414 to 5,656 Hz, 100-dB sound pressure level [SPL] for one hour) and treated with kanamycin (150 mg/kg a day until hearings loss was noted at 6.0 kHz) either separately, simultaneously, or sequentially. Simultaneous noise and kanamycin resulted in interactive potentiation of threshold shift and cochlear pathologic condition. Kanamycin treatment two months after noise exposure produced similar potentiation. No interaction was seen when noise exposure occurred one month after kanamycin treatment.

The ototoxic potential of propylene glycol in guinea pigs.

Propylene glycol, previously indicated to be ototoxic and to produce "deafness," was studied in the middle ear space of guinea pigs. Ninety percent propylene glycol produced conductive middle ear problems, but there was no loss of hair cells above that found in normal untreated animals. Ten percent propylene glycol produced no negative effects in the middle ear or elsewhere. Topical ear drops containing high concentrations of propylene glycol are contraindicated in cases with perforation of the tympanic membrane.

[On a model of cisplatin-induced ototoxicity in Wistar rats].

UNLABELLED: An experimental in vivo model of cisplatin-induced ototoxicity (CO) is a prerequisite for developing methods to prevent hearing loss resulting from cisplatin (CP) chemotherapy. However, there are no clear guidelines regarding its design in a research model as widely used as the Wistar rat. OBJECTIVE: To develop and validate a model of auditory analyzer injury in Wistar rats that reflects the specific nature of cisplatin-induced ototoxicity. MATERIAL AND METHODS: The study was conducted on male Wistar rats of SPF status, n=24, weighing 300-340 g. CP was administered in doses of 6,

Hair Cell Protection from Ototoxic Drugs.

Hearing loss is often caused by death of sensory hair cells (HCs) in the inner ear. HCs are vulnerable to some ototoxic drugs, such as aminoglycosides(AGs) and the cisplatin. The most predominant form of drug-induced cell death is apoptosis. Many efforts have been made to protect HCs from cell death after ototoxic drug exposure. These mechanisms and potential targets of HCs protection will be discussed in this review. And we also propose further investigation in the field of HCs necrosis and regeneration, as well as future clinical utilization.

The Ototoxicity of Antimalarial Drugs-A State of the Art Review.

This review summarizes current knowledge about the occurrence of hearing and balance disorders after antimalarial drugs treatment. It also examines the clinical applications of antimalarials, their mechanisms behind this ototoxicity and how it can be monitored. It includes studies with larger numbers of patients and those in which auditory function was assessed using audiological tests. Some antimalarials have been repurposed for other conditions like autoimmune disorders, rheumatic diseases, some viral diseases and cancers. While old antimalarial drugs, such as quinoline derivatives, are know

Circadian vulnerability of cisplatin-induced ototoxicity in the cochlea.

The chemotherapeutic agent cisplatin is renowned for its ototoxic effects. While hair cells in the cochlea are established targets of cisplatin, less is known regarding the afferent synapse, which is an essential component in the faithful temporal transmission of sound. The glutamate aspartate transporter (GLAST) shields the auditory synapse from excessive glutamate release, and its loss of function increases the vulnerability to noise, salicylate, and aminoglycosides. Until now, the involvement of GLAST in cisplatin-mediated ototoxicity remains unknown. Here, we test in mice lacking GLAST the

Necroptosis and Apoptosis Contribute to Cisplatin and Aminoglycoside Ototoxicity.

Ototoxic side effects of cisplatin and aminoglycosides have been extensively studied, but no therapy is available to date. Sensory hair cells, upon exposure to cisplatin or aminoglycosides, undergo apoptotic and necrotic cell death. Blocking these cell death pathways has therapeutic potential in theory, but incomplete protection and lack of therapeutic targets in the case of necrosis, has hampered the development of clinically applicable drugs. Over the past decade, a novel form of necrosis, termed necroptosis, was established as an alternative cell death pathway. Necroptosis is distinguished

Ototoxicity of acetic acid on the guinea pig cochlea.

BACKGROUND: To evaluate the ototoxicity of acetic acid solutions. METHODS: Compound action potentials (CAPs) of the eighth nerve were measured in guinea pigs before and after the application of acetic acid in the middle ear cavity. The pH values of the acetic acid solutions were pH 3.0, 4.0, and 5.0, and the application times were 30 min, 24 h, and 1 week. RESULTS: Acetic acid solution (pH 3.0, N = 3) for 30 min caused no significant elevation in CAP threshold at 4 kHz, but a significant elevation in the threshold was noted for 8 kHz and clicks. Acetic acid solutions (pH 4.0 N = 6, 5.0 N = 5
