Mechanistic Insight into Caffeine-Oxalic Cocrystal Dissociation in Formulations: Role of Excipients.

Caffeine-oxalic acid cocrystal, widely reported to be stable under high humidity, dissociated in the presence of numerous pharmaceutical excipients. In cocrystal-excipient binary systems, the water mediated dissociation reaction occurred under pharmaceutically relevant storage conditions. Powder X-ray diffractometry was used to identify the dissociated products obtained as a consequence of coformer-excipient interaction. The proposed cocrystal dissociation mechanism involved water sorption, dissolution of cocrystal and excipient in the sorbed water, proton transfer from oxalic acid to the exci

Enteric oxalate nephropathy in the renal allograft: an underrecognized complication of bariatric surgery.

Enteric hyperoxalosis is a recognized complication of bariatric surgery, with consequent oxalate nephropathy leading to chronic kidney disease and occasionally end-stage renal failure. In patients with prior gastrointestinal bypass surgery, renal allografts are also at risk of oxalate nephropathy. Further, transplant recipients may be exposed to additional causes of hyperoxalosis. We report two cases of renal allograft oxalate nephropathy in patients with remote histories of bariatric surgery. Conservative management led to improvement of graft function in one patient, while the other patient

Hereditary renal hypouricemia: a cause of calcium oxalate urolithiasis in a young female.

BACKGROUND: The impact of cervical sagittal parameters on patient-reported outcomes (PRO) after posterior cervical fusion (PCF) remains inconclusive. We sought to evaluate whether normative sagittal cervical alignment was independently associated with improved PRO. METHODS: Retrospective review of consecutive patients who underwent PCF for degenerative conditions from January 2016 to December 2020 at a single institution with multiple surgeons. Cervical sagittal vertical axis (cSVA), McGregor's slope (McGS), and T1 slope minus C2-7 lordosis (T1S-CL) were measured using standing neutral latera

Rare calcium oxalate monohydrate calculus attached to the wall of the renal pelvis.

Most renal calculi can be classified using well-established criteria in a manner that reflects both composition and fine structure under specific pathophysiological conditions. However, when a large patient population is considered, rare renal calculi invariably appear, some of which have never been classified; careful study is required to establish stone etiology in such cases. The patient in the present case report formed two types of calculi. One was attached on the wall of the renal pelvis near the ureter and part of the calculus was embedded inside pelvic renal tissue. The calculus develo

Characterization of Ceriporiopsis subvermispora bicupin oxalate oxidase expressed in Pichia pastoris.

Oxalate oxidase (E.C. 1.2.3.4) catalyzes the oxygen-dependent oxidation of oxalate to carbon dioxide in a reaction that is coupled with the formation of hydrogen peroxide. Although there is currently no structural information available for oxalate oxidase from Ceriporiopsis subvermispora (CsOxOx), sequence data and homology modeling indicate that it is the first manganese-containing bicupin enzyme identified that catalyzes this reaction. Interestingly, CsOxOx shares greatest sequence homology with bicupin microbial oxalate decarboxylases (OxDC). We show that CsOxOx activity directly correlates

Calcium oxalate stone formation in the inner ear as a result of an Slc26a4 mutation.

Calcium oxalate stone formation occurs under pathological conditions and accounts for more than 80% of all types of kidney stones. In the current study, we show for the first time that calcium oxalate stones are formed in the mouse inner ear of a genetic model for hearing loss and vestibular dysfunction in humans. The vestibular system within the inner ear is dependent on extracellular tiny calcium carbonate minerals for proper function. Thousands of these biominerals, known as otoconia, are associated with the utricle and saccule sensory maculae and are vital for mechanical stimulation of the

Regressive course of oxalate deposition in primary hyperoxaluria after kidney transplantation.

Primary hyperoxaluria (PH) is a rare autosomal recessive disease caused by the functional defect of alanine-glyoxylate aminotransferase (AGT) enzyme in the liver and it is characterized by the deposition of diffuse calcium oxalate crystals. A 38-year-old male patient presented with history of recurrent nephrolithiasis and has received chronic hemodialysis treatment for 2 years. Cadaveric renal transplantation was applied to the case. The patient was reoperated on postoperative day 13 because of the collection surrounding the urethra. During this operation, kidney biopsy was made due to late de

Parenteral ambroxol treatment causes xanthine and calcium oxalate stones in rats.

INTRODUCTION: Ambroxol (ABX) is known to promote bronchial secretion and is used as an expectorant. This study was undertaken to document the connection between ambroxol parenteral treatment and bladder stones in rats. MATERIAL AND METHODS: Forty-five wild rats (Rattus sp.) were divided into three equal groups. Rats from the first and second groups received ABX s.c. during 2 weeks in total doses of 30 mg/kg per 24 h and 60 mg/kg per 24 h, respectively. Rats from the control group received 1 mL of injection solution s.c. One month after the treatment termination, animals were sacrificed and ur

Characteristics of the transport of oxalate and other ions across rabbit proximal colon.

In order to characterize oxalate handling by the P2 segment of the rabbit proximal colon, the fluxes of [14C]oxalate, 22Na+, and 36Cl- were measured in vitro using conventional short-circuiting techniques. In standard buffer the proximal colon exhibited net secretion of Na+ (-2.31 +/- 0.64 mu equiv cm-2 h-1), negligible net Cl- transport, and net secretion of oxalate (-12.7 +/- 1.6 pmol cm-2 h-1). Replacement of buffer Na+ or Cl- abolished net oxalate secretion, while HCO(3-)-free media revealed a net absorption of oxalate (19.3 +/- 4.2 pmol cm-2 h-1) and stimulated NaCl absorption. Crystal growth of calcium oxalate in urine of stone-formers and normal controls.

In this study, the relative crystal growth rate (Vcr) of calcium oxalate (Caox) and a number of other parameters were determined in 17-h daily (d) and 7-h nocturnal fractions (n) of whole urine from 20 recurrent Caox stone formers (SF) and 29 age-matched male normal controls (NC). Vcr, which was determined by the gel crystallization method (GCM), showed the largest difference between SF and NC among all parameters under investigation. Mean values (+/- SD) obtained for Vcr were: 0.73 +/- 0.58 (SF-d)/0.21 +/- 0.22 (NC-d; P less than 0.001) and 0.63+/- 0.58 (SF-n)/0.24 +/- 0.25 (NC-n; P less than

Oxalate crystal deposits in the heart in chronic renal failure: an experimental study.

The present study was undertaken to evaluate three factors which may contribute to the myocardial deposition of calcium oxalate in 5/6 nephrectomised rats: (1) increased plasma oxalate, (2) elevated plasma ionised calcium, and (3) local myocardial tissue damage. A simple increase in plasma oxalate concentrations produced by injection of sodium oxalate did not cause deposits in the heart. Increased plasma ionised calcium combined with elevated plasma oxalate for 60 days was likewise unimportant with regard to cardiac deposits. However, when local tissue damage was added by heterotopic cardiac t

[Effect of cocoa on excretion of oxalate, citrate, magnesium and calcium in the urine of children].

Cocoa is a strong carrier of oxalic acid (average: 400 mg per 100 g). In three calcium oxalate stone formers clinical observation had been suggestive of excessive intake of cocoa products contributing to calculus formation. We studied the effect on renal oxalate excretion of an oral cocoa load (30 g per m2 body surface given on 2 consecutive days) in 12 former stone formers (group 1), 14 children with isolated microscopic haematuria (group 2), 13 healthy boys (group 3), and 12 healthy girls (group 4). A new enzymatic method was used to measure oxalic acid in cocoa products as well as in urine

The influence of poly(aspartic acid) on the deformation behavior of calcium oxalate crystals.

In this study, we engineered a modular microfluidic device to grow individual synthetic calcium oxalate monohydrate (COM) crystals in the presence or absence of polyelectrolytes directly onto a glass substrate. The modular design allowed the glass substrate and attached crystals to be removed from the device, which enabled individual COM crystals to be identified and targeted by microscopy for subsequent characterization. Qualitative (morphology, Raman spectroscopy) and quantitative (surface roughness and nanomechanical properties) measurements were performed on these COM crystals to evaluate

Electrocatalytic C─C Coupling of Dimethyl Oxalate to C4 Dicarboxylate on Cu.

Electrocatalytic conversion of CO to value-added multi-carbon chemicals enables carbon resource recycling and emission reduction. Building on the industrial process of dimethyl oxalate (DMO) production from CO and methanol, we report in this work a selective electrochemical pathway to convert DMO to dimethyl tartarate (DMT), a precursor to multiple C4-dicarboxylic acids. On a Cuα catalyst dominated by Cu(111)-like sites, Faradaic efficiency and partial current density for DMT reach 74.7% and 152 mA/cm- 2, respectively, which significantly outperforms Cuβ dominated with Cu(100)/(110)-like sites

Electroreduction of carbon dioxide (CO2) at oxalate and polypyrrole modified copper surfaces.

The electrochemical conversion of CO2 into useful chemicals remains an active area of investigation, especially towards higher order C2 and C3 products such as ethylene, ethanol and n-propanol. Herein, we demonstrate that oxalate modified copper surfaces on porous gas-diffusion electrodes (GDEs) can be used to modify the selectivity of the electrochemical CO2 reduction reaction (eCO2RR) towards higher order chemicals using a flow-type electrochemical cell at near-neutral pH and industrially viable current densities. These multi-layer composite cathodes consisting of a Teflon substrate, coated

Oxalate nephropathy from untreated pancreatic insufficiency in a neuroendocrine tumour patient.

Pancreatic exocrine insufficiency (PEI) is an under-recognised cause of chronic diarrhoea in patients with neuroendocrine tumours (NETs). If left untreated or undertreated, PEI can lead to complications such as oxalate nephropathy (ON), which may result in loss of kidney function and potentially limit oncologic treatment options, including peptide receptor radionuclide therapy (PRRT). We present a man in his late 60s with a small bowel NET who developed persistent, uncontrolled diarrhoea for several years. The diarrhoea was attributed to multiple factors, including secretory NET (ie, carcinoid
