Stable oxygen isotopes of hominin enamel carbonate (δ(18)OEC) provide a window into aspects of past drinking behavior and diet, body size, breastfeeding and weaning, mobility, and paleoclimate. It is tempting to compare all hominins across time and space in order to gauge species-level adaptations to changing environments and niche separation between those living sympatrically. Basinal, sub-basinal, and micro-environmental differences, however, may exert an influence on variation in enamel carbonate isotopic values that must be reconciled before hominin species across Africa can be meaningfull

Sauropod dinosaurs were the largest vertebrates ever to walk the Earth, and as mega-herbivores they were important parts of terrestrial ecosystems. In the Late Jurassic-aged Morrison depositional basin of western North America, these animals occupied lowland river-floodplain settings characterized by a seasonally dry climate. Massive herbivores with high nutritional and water needs could periodically experience nutritional and water stress under these conditions, and thus the common occurrence of sauropods in this basin has remained a paradox. Energetic arguments and mammalian analogues have b

CADD (chlamydia protein associating with death domains) is a p-aminobenzoate (pAB) synthase involved in a noncanonical route for tetrahydrofolate biosynthesis in Chlamydia trachomatis. Although previously implicated to employ a diiron cofactor, here, we show that pAB synthesis by CADD requires manganese and the physiological cofactor is most likely a heterodinuclear Mn/Fe cluster. Isotope-labeling experiments revealed that the two oxygen atoms in the carboxylic acid portion of pAB are derived from molecular oxygen. Further, mass spectrometry-based proteomic analyses of CADD-derived peptides de

Monsoon precipitation affects natural and social systems in East Asia, one of the most densely populated regions in the world. Monsoon precipitation variability is strongly influenced by El Niño-Southern Oscillation (ENSO) and may be related to the phase of the Pacific Decadal Oscillation (PDO). However, a collective understanding of the long-term PDO-ENSO-monsoon relationship remains limited because related studies are almost exclusively based on short instrumental records. Although paleoclimate proxies for PDO and ENSO are currently available, there is a lack of high-quality proxies for East

The evolution of the extinct megatooth shark, Otodus megalodon, and its close phylogenetic relatives remains enigmatic. A central question persists regarding the thermophysiological origins of these large predatory sharks through geologic time, including whether O. megalodon was ectothermic or endothermic (including regional endothermy), and whether its thermophysiology could help to explain the iconic shark's gigantism and eventual demise during the Pliocene. To address these uncertainties, we present unique geochemical evidence for thermoregulation in O. megalodon from both clumped isotope p

Chemically reinforced essential fatty acids (FAs) promise to fight numerous age-related diseases including Alzheimer's, Friedreich's ataxia and other neurological conditions. The reinforcement is achieved by substituting the atoms of hydrogen at the bis-allylic methylene of these essential FAs with the isotope deuterium. This substitution leads to a significantly slower oxidation due to the kinetic isotope effect, inhibiting membrane damage. The approach has the advantage of preventing the harmful accumulation of reactive oxygen species (ROS) by inhibiting the propagation of lipid peroxidation

The global sulfur cycle plays a critical role in the redox evolution of Earth's surface and upper mantle, yet the distribution and origin of sulfur in the mantle remains largely unconstrained. El Hierro is a volcanic island in the Canary archipelago that is fed by sulfur-rich magmas. To constrain the origin of sulfur in these melts, we combine in situ sulfur isotope analyses with regression modeling. We calculate that undegassed El Hierro melts have δ34S values of 0 ± 2‰. The average δ34S of undegassed El Hierro melts is 0.3‰ to 1‰ higher than magmas erupting at mid-ocean ridges. Mass balance

The cyclization of 2-(hydroxypropyl)-4-nitrophenyl phosphate (HpPNP) catalyzed by the dinuclear zinc complex of 1,3-bis(1,4,7-triazacyclonon-1-yl)-2-hydroxypropane (1) proceeds by a transition state that is different from that of the uncatalyzed reaction. Kinetic isotope effects (KIEs) measured in the nucleophilic atom and in the leaving group show that the uncatalyzed cyclization has a transition state (TS) with little phosphorus-oxygen bond fission to the leaving group ((18)k(lg) = 1.0064 +/- 0.0009 and (15)k = 1.0002 +/- 0.0002) and that nucleophilic bond formation occurs in the rate-determ

Cyanobacteria are the most ancient oxygenic photosynthetic organisms on Earth and play a pivotal role in the global carbon cycle. Despite their ecological and evolutionary significance, the mechanisms of carbon acquisition and fixation in this phylum remain largely unexplored beyond a few model species. Here, we examined representative taxa spanning the full phylogenetic breadth of Cyanobacteria, assessing in vivo carbon acquisition pathways, the role and effectiveness of CO2-concentrating mechanisms (CCMs), as well as conducting in vitro biochemical characterizations of the kinetic traits and

The processes leading to high levels of arsenic (As), iron (Fe), and manganese (Mn) in groundwater, in a naturally reducing aquifer at a controlled municipal landfill site, are investigated. The challenge is to distinguish the natural water-rock interaction processes, that allow these substances to dissolve in groundwater, from direct pollution or enhanced dissolution of hydroxides as undesired consequences of the anthropic activities above. Ordinary groundwater monitoring of physical-chemical parameters and inorganic compounds (major and trace elements) was complemented by environmental isoto

Marine subseafloor sediments underlying open ocean regions are predominantly low-energy environments; however, they harbor vast microbial populations. How these communities persist and whether they retain anabolic capacity during transient oxygen depletion remain unclear. We herein investigated oxic sediments from the South Atlantic Gyre (SAG; IODP Site U1559D, core 7X-1) using paired incubations under oxic and anoxic (oxygen-depleted) conditions with defined C/N amendments. Sediments were amended with a 13C-amino acid mix+15N-ammonium, 13C-bicarbonate+15N-ammonium, or 15N-ammonium only and th

Cribra orbitalia (CO), or porotic hyperostosis (PH) of the orbital roof, is one of the most common pathological conditions found in archaeological subadult skeletal remains. Reaching frequencies higher than 50% in many prehistoric samples, CO has been generally attributed to a variety of factors including malnutrition (e.g., megaloblastic anemia) and parasitism. In this study, we tested the relationship between CO, trace element concentrations, and stable isotope values (δ13C, δ15N, δ18O) in subadult skeletons from a 17th to 18th century cemetery in the historic town of Jēkabpils, Latvia. Reactive capture and conversion (RCC) is a process intensification approach that integrates CO2 capture and hydrogenation within a single unit, removing the CO2 purification and storage steps of traditional process flow schemes. This alters the catalytic step from a traditional steady-state (SS) flow process to a transient capture and conversion cycle, which could lead to product distributions distinct from those observed in conventional SS experiments. Such differences are investigated in the combined capture and hydrogenation of carbon dioxide to methanol over a ZnZrO2 catalyst and a ZnZrO2

Hydrogeological, hydrochemical and isotopic traits of the groundwater in the Quaternary aquifer system in an urban-periurban locality within and encircling the Kolkata-Howrah twin city in the south Bengal Basin have been synthesised to explain the present- and paleo-hydrological processes, surface and groundwater interaction and mixing dynamics of contamination of groundwater. Rock-weathering, evaporation, ion-exchange and active mineral dissolution are the key processes commanding the groundwater chemistry. Freshwater flushing from the recharge zones had thinned the entrapped sea water which

RATIONALE: Applications of high-precision isotope analysis of ecohydrological water samples such as soil and plant waters with infrared (IR) laser spectrometry have been hampered by spectral interferences of volatile organic compounds.
METHODS: In this study, three methods were tested to remove methanol (MeOH) and/or ethanol (EtOH) added to water for δ2H, δ18O, and 17O-excess analysis with Picarro L2140-i: microcombustion module (MCM), pre-treatment with solid-phase extraction (SPE), and pre-treatment with simple MeOH/EtOH combustion (SMEC) method.
RESULTS: The Picarro MCM was not able to co

BACKGROUND: Cerebral glucose hypometabolism is consistently observed in individuals with Alzheimer's disease (AD), as well as in young cognitively normal carriers of the Ε4 allele of Apolipoprotein E (APOE), the strongest genetic predictor of late-onset AD. While this clinical feature has been described for over two decades, the mechanism underlying these changes in cerebral glucose metabolism remains a critical knowledge gap in the field.
METHODS: Here, we undertook a multi-omic approach by combining single-cell RNA sequencing (scRNAseq) and stable isotope resolved metabolomics (SIRM) to def

Positron emission tomography (PET) with (15)O tracers provides essential information in patients with cerebral vascular disorders, such as cerebral blood flow (CBF), oxygen extraction fraction (OEF), and metabolic rate of oxygen (CMRO(2)). However, most of techniques require an additional C(15)O scan for compensating cerebral blood volume (CBV). We aimed to establish a technique to calculate all functional images only from a single dynamic PET scan, without losing accuracy or statistical certainties. The technique was an extension of previous dual-tracer autoradiography (DARG) approach, but ba

Palaeoloxodon is the largest known terrestrial mammal in the history of Taiwan. However, little is known about the palaeoecology and palaeoenvironment of these extinct proboscideans. In this study, we investigate the stable carbon and oxygen isotopic ratios of sequential enamel samples taken along the growth direction of molars from three Palaeoloxodon specimens of different ontogenetic ages from the Pleistocene of Taiwan. Our results suggest that these proboscideans relied extensively on a C4 diet, a dietary niche distinct from the Eurasian Palaeoloxodon antiquus but similar to the predominan
