Converging infectious disease threats in the post-COVID era: surveillance fragility, pandemic risk, and global preparedness.

The COVID-19 pandemic exposed profound weaknesses in global infectious disease surveillance yet, in its aftermath, pandemic-prone threats have intensified rather than receded. This narrative review synthesizes contemporary evidence on three converging drivers of pandemic risk zoonotic emergence, antimicrobial resistance, and climate-sensitive disease expansion and examines how structural failures in surveillance and global health governance amplify their impact. Drawing on recent epidemiological analyses, policy reports, and One Health evaluations, the review maps post-COVID surges in zoonotic

Optimizing Pandemic Preparedness and Response Through Health Information Systems: Lessons Learned From Ebola to COVID-19.

Strengthening health systems and maintaining essential service delivery during health emergencies response is critical for early detection and diagnosis, prompt treatment, and effective control of pandemics, including the novel coronavirus disease 2019 (COVID-19). Health information systems (HIS) developed during recent Ebola outbreaks in West Africa and the Democratic Republic of the Congo (DRC) provided opportunities to collect, analyze, and distribute data to inform both day-to-day and long-term policy decisions on outbreak preparedness. As COVID-19 continues to sweep across the globe, HIS

Relative contributions of pre-pandemic factors and intra-pandemic activities to differential COVID-19 risk among migrant and non-migrant populations in the Netherlands: lessons for future pandemic preparedness.

BACKGROUND: Although risk factors for differences in SARS-CoV-2 infections between migrant and non-migrant populations in high income countries have been identified, their relative contributions to these SARS-CoV-2 infections, which could aid in the preparation for future viral pandemics, remain unknown. We investigated the relative contributions of pre-pandemic factors and intra-pandemic activities to differential SARS-CoV-2 infections in the Netherlands by migration background (Dutch, African Surinamese, South-Asian Surinamese, Ghanaians, Turkish, and Moroccan origin). METHODS: We utilized

Defining Optimal Vaccine Features for Pandemic Preparedness: an Individual-Based Model Bridging Within- and Between-Host Dynamics.

Disease X, a yet-to-be-identified pathogen of pandemic potential, underscores the urgency of proactive surveillance and preparedness. Developing prototype vaccine for representative pathogens is key to this effort. In alignment with the "100 Day Mission" to ensure equitable vaccine access, this study aims to identify desirable vaccine features needed to control future pathogens outbreaks, possibly SARS-CoV-2 type. We developed an individual-based transmission model integrating viral load and antibody kinetics to examine combinations of the virus's basic reproduction number (R0) and vaccine cha

Lessons learnt from the COVID-19 pandemic: Middle East and North Africa regional perspective for future preparedness.

The COVID-19 pandemic revealed critical gaps in preparedness and response capacities globally. These gaps were evident in the Middle East and North Africa (MENA) region, as the region faces unique challenges due to ongoing humanitarian crises, political instability and large-scale religious gatherings, which further exacerbate the risk of spread of infectious diseases. In April 2024, a conference hosted by the US Centres for Disease Control and Prevention MENA Regional Office and National Institutes of Allergy and Infectious Diseases, in collaboration with the Mohammed Bin Rashid University of

A Revised Public Health Emergency Preparedness Logic Model Reflecting the European Experience in the COVID-19 Pandemic.

In 2017, the European Centre for Disease Prevention and Control developed a public health emergency preparedness logic model for European Union, differentiating between capacities (resources and infrastructure) and response capabilities (actions during emergencies). Analysis of the COVID-19 experience suggests that the main features of this logic model, including the 5 high-level categories of capabilities, remain valid. Some changes are needed, however, in 3 areas. In the detection and assessment area, "laboratory analysis" was integrated with "testing" to better represent their combined role

Nationwide survey for household preparedness and vaccination acceptance during a novel influenza pandemic scenario in Japan, 2025.

Pandemics and associated mitigation measures can disrupt children's daily lives and shift substantial burden to households. However, families' actionable preparedness for future pandemics remains insufficiently understood, including caregiving capacity and vaccination acceptance. We conducted an anonymous nationwide online cross-sectional survey in November 2025 among Japanese parents residing with at least one child aged 0-18 years (N = 4961). Respondents were presented with standardized hypothetical influenza pandemic scenarios and asked to report anticipated impacts of school/childcare clos

Interactys-AI: Toward AI-Driven Structural Mapping of Virus-Host Interfaces for Antiviral Repurposing and Pandemic Preparedness.

Understanding how viruses engage host cell surfaces is fundamental to infection biology and therapeutic development. While vaccines remain central to prevention, recent global crises have emphasized the need for complementary antiviral strategies that can be mobilized rapidly against both known and emerging pathogens. In this context, artificial intelligence (AI) systems for biomolecular structure prediction, culminating in AlphaFold 3, are reshaping what is experimentally and conceptually achievable. Here, we present "Interactys-AI", a framework designed to exploit AI-based structural modelin

'Bern, get ready', BEready, a household-based cohort study for pandemic preparedness research in Switzerland: pilot study.

OBJECTIVES: Research for pandemic response needs to be timely to inform evidence-based decision making. The lack of epidemiological data at the start of the COVID-19 pandemic led experts to call for cohorts that could rapidly supply data about newly emerging infectious diseases. The 'Bern, get ready' (BEready) study aims to establish a prospective 'pandemic preparedness cohort' in the canton of Bern, Switzerland. This cohort can be pivoted to the needs of a new pandemic pathogen. The aim of this pilot study was to investigate the potential response and to test the feasibility of procedures for

Filling the gap: artificial intelligence-driven one health integration to strengthen pandemic preparedness in resource-limited settings.

Emerging zoonotic pathogens like SARS-CoV-2 and Nipah virus demonstrate the critical need for integrated surveillance systems connecting human, animal, and environmental health. This review examines how artificial intelligence can address One Health integration gaps in pandemic surveillance, focusing on resource-limited settings. While global digitization levels now support Artificial Intelligence (AI)-powered platforms, LMICs face barriers including limited resources and fragmented data systems. Current AI tools remain domain-specific and designed for high-income settings, limiting its applic

Respiratory pandemic preparedness learnings from the June 2020 COVID-19 outbreak at San Quentin California State Prison.

PURPOSE: This study aims to characterize the June 2020 COVID-19 outbreak at San Quentin California State Prison and to describe what made San Quentin so vulnerable to uncontrolled transmission. DESIGN/METHODOLOGY/APPROACH: Since its onset, the COVID-19 pandemic has exposed and exacerbated the profound health harms of carceral settings, such that nearly half of state prisons reported COVID-19 infection rates that were four or more times (and up to 15 times) the rate found in the state's general population. Thus, addressing the public health crises and inequities of carceral settings during a r
