Neuregulin-1 (Nrg1) is mainly expressed in rat pituitary gonadotroph cells and possibly regulates prolactin (PRL) secretion in a juxtacrine manner.

The binding of Neuregulin-1 (Nrg1) to its cognate receptors ErbB-3 and -4 mediates intercellular and intracellular communication. In vitro, this interaction has been shown to control prolactin (PRL) secretion from pituitary tumour cells. However, Nrg1/ErbB signalling and its function in vivo are not well understood. In the present study, we demonstrated that type I and III Nrg1 isoforms were expressed in the rat anterior pituitary. We observed that Nrg1 positive gonadotrophs can form contacts with lactotrophs, which are positive for ErbB-3 receptor. In addition, we show that gonadotroph cell-d

Intercellular Signalling Cross-Talk: To Kill, To Heal and To Rejuvenate.

Intercellular cross-talk is a fundamental process for spreading cellular signals between neighbouring and distant cells to properly regulate their metabolism, to coordinate homeostasis, adaptation and survival as a functional tissue and organ. In this review, we take a close molecular view of the underpinning molecular mechanisms of such complex intercellular communications. There are several studied forms of cell-to-cell communications considered crucial for the maintenance of multicellular organisms. The most explored is paracrine signalling which is realised through the release of diffusibl

Sinusoidal communication in chronic liver disease.

The liver sinusoid, mainly composed of liver sinusoidal endothelial cells, hepatic macrophages and hepatic stellate cells, shapes the hepatic vasculature and is key to maintaining liver homeostasis and function. During chronic liver disease (CLD), the function of sinusoidal cells is impaired, being directly involved in the progression of liver fibrosis, cirrhosis, and main clinical complications including portal hypertension and hepatocellular carcinoma. In addition to their roles in liver diseases pathobiology, sinusoidal cells' paracrine communication or cross-talk is being studied as a mech

Dual fluorescence reporter mice for Ccl3 transcription, translation, and intercellular communication.

Chemokines guide immune cells during their response against pathogens and tumors. Various techniques exist to determine chemokine production, but none to identify cells that directly sense chemokines in vivo. We have generated CCL3-EASER (ErAse, SEnd, Receive) mice that simultaneously report for Ccl3 transcription and translation, allow identifying Ccl3-sensing cells, and permit inducible deletion of Ccl3-producing cells. We infected these mice with murine cytomegalovirus (mCMV), where Ccl3 and NK cells are critical defense mediators. We found that NK cells transcribed Ccl3 already in homeosta

Intra-Islet Paracrine Regulation of Glucagon Secretion During Hypoglycemia, Euglycemia, and Hyperglycemia.

The regulation of glucagon and insulin secretion, from pancreatic α- and β-cells, respectively, is essential for maintaining blood glucose within the physiological set point. Insulin secretion is stimulated as glucose rises from euglycemia, with incretin hormones modulating the secretion in a glucose-dependent manner. Glucagon secretion is elevated with hypoglycemia but also with hyperglycemia. Within the pancreatic islet, α-cell glucagon output is regulated by complex neuroendocrine and paracrine signals from insulin-releasing β-cells and somatostatin-secreting δ-cells. This network of intra-

A preoptic neuronal population controls fever and appetite during sickness.

During infection, animals exhibit adaptive changes in physiology and behaviour aimed at increasing survival. Although many causes of infection exist, they trigger similar stereotyped symptoms such as fever, warmth-seeking, loss of appetite and fatigue1,2. Yet exactly how the nervous system alters body temperature and triggers sickness behaviours to coordinate responses to infection remains unknown. Here we identify a previously uncharacterized population of neurons in the ventral medial preoptic area (VMPO) of the hypothalamus that are activated after sickness induced by lipopolysaccharide (LP

The potential role of antioxidants in mitigating skin hyperpigmentation resulting from ultraviolet and visible light-induced oxidative stress.

Oxidative stress is an integral element that influences a variety of biochemical reactions throughout the body and is known to play a notable role in melanogenesis. Exogenous triggers of oxidative stress, such as ultraviolet radiation (UVR) and visible light (VL), lead to pigment formation through somewhat different pathways, but both share a common endpoint-the potential to generate cosmetically undesirable hyperpigmentation. Though organic and inorganic sunscreens are available to protect against the UVR portion of the electromagnetic spectrum, coverage is lacking to protect against the VL s

The role of brain natriuretic peptide in maintaining myometrial quiescence during pregnancy.

Myometrial quiescence is a physiological stage of the myometrium during pregnancy. It is a period of active relaxation of the myometrial smooth muscle cells; myometrial quiescence is responsible for maintaining pregnancy. The precise mechanisms underlying myometrial quiescence have not been completely elucidated, although many mediators and cellular pathways have been described as playing a role. Fetal membranes (chorion and amnion) produce and release one or more substances that inhibit myometrial contractions, playing a central role in the maintenance of myometrial quiescence. Brain natriure

Glomerular structure and function require paracrine, not autocrine, VEGF-VEGFR-2 signaling.

VEGF is a potent vascular growth factor produced by podocytes in the developing and mature glomerulus. Specific deletion of VEGF from podocytes causes glomerular abnormalities including profound endothelial cell injury, suggesting that paracrine signaling is critical for maintaining the glomerular filtration barrier (GFB). However, it is not clear whether normal GFB function also requires autocrine VEGF signaling in podocytes. In this study, we sought to determine whether an autocrine VEGF-VEGFR-2 loop in podocytes contributes to the maintenance of the GFB in vivo. We found that induced, whole

Oxygen-dependent osteocyte transcriptomic adaptations occur without modulation of endothelial gene expression via osteocyte paracrine signaling.

Bone regeneration remains a significant clinical challenge, requiring a deeper understanding of the cellular and molecular mechanisms underlying bone repair. Osteocytes, which represent 90%-95% of bone cells, form a connective dendritic network and play a key role in skeletal homeostasis. Embedded within the mineralized matrix, they experience unique metabolic constraints, including reduced oxygen availability. Hypoxia may, therefore, influence their function, but its impact and potential role in regulating angiogenesis during bone regeneration remain poorly understood. This study aimed to inv

Hypoxic CA9-high mesenchymal-like glioma stem cells promote ANXA1-FPR1-dependent macrophage polarization in recurrent glioblastoma.

BACKGROUND: Recurrent glioblastoma (rGBM) is characterized by marked myeloid remodeling and a profoundly immunosuppressive microenvironment. Although carbonic anhydrase 9 (CA9) is a canonical hypoxia-inducible molecule linked to aggressive glioblastoma (GBM) behavior, its cell-state-specific distribution in rGBM and its role in glioma stem cell (GSC)-macrophage crosstalk remain incompletely understood. METHODS: We integrated public transcriptomic cohorts, single-cell RNA sequencing, quantitative proteomics, paired clinical specimens, and in vitro/in vivo functional assays to define the role o

Cancer neuroscience: State of the field, emerging directions.

The nervous system governs both ontogeny and oncology. Regulating organogenesis during development, maintaining homeostasis, and promoting plasticity throughout life, the nervous system plays parallel roles in the regulation of cancers. Foundational discoveries have elucidated direct paracrine and electrochemical communication between neurons and cancer cells, as well as indirect interactions through neural effects on the immune system and stromal cells in the tumor microenvironment in a wide range of malignancies. Nervous system-cancer interactions can regulate oncogenesis, growth, invasion a

[Exosomal microRNAs: an emerging player in systemic lupus erythematosus].

Exosomes are nanometer-sized membranous extracellular vesicles that can be secreted by almost all types of cells in the body. Exosomes are involved in cell-to-cell communication through autocrine and paracrine forms. Exosomal microRNAs (miRNAs) are stable in plasma, urine and other body fluids, and have various biological functions. They play an irreplaceable role in the occurrence, development, immune regulation of systemic lupus erythematosus (SLE). Recent studies have proposed that exosomal miRNAs have promising application prospects in the pathogenesis, early diagnosis, and treatment of SL

Cardiac nonmyocytes in the hub of cardiac hypertrophy.

Cardiac hypertrophy is characterized by complex multicellular alterations, such as cardiomyocyte growth, angiogenesis, fibrosis, and inflammation. The heart consists of myocytes and nonmyocytes, such as fibroblasts, vascular cells, and blood cells, and these cells communicate with each other directly or indirectly via a variety of autocrine or paracrine mediators. Accumulating evidence has suggested that nonmyocytes actively participate in the development of cardiac hypertrophy. In this review, recent progress in our understanding of the importance of nonmyocytes as a hub for induction of card

AMP-activated protein kinase influences metabolic remodeling in H9c2 cells hypertrophied by arginine vasopressin.

Substrate use switches from fatty acids toward glucose in pressure overload-induced cardiac hypertrophy with an acceleration of glycolysis being characteristic. The activation of AMP-activated protein kinase (AMPK) observed in hypertrophied hearts provides one potential mechanism for the acceleration of glycolysis. Here, we directly tested the hypothesis that AMPK causes the acceleration of glycolysis in hypertrophied heart muscle cells. The H9c2 cell line, derived from the embryonic rat heart, was treated with arginine vasopressin (AVP; 1 microM) to induce a cellular model of hypertrophy. Rat

Injury-induced paracrine effects on the podocyte's transcriptome.

Although injury in glomerular disease might only damage a subset of podocytes in any given glomerulus, the response of the healthy neighboring podocytes to the injured podocytes oftentimes determines the course of the disease. To investigate this relationship, we designed a dual-chamber open microfluidic coculture device to specifically examine paracrine signaling from podocytes undergoing targeted injury by either adriamycin, puromycin aminonucleoside, or a cytopathic antipodocyte antibody to healthy podocytes. Global transcriptomic analysis measured by RNA sequencing revealed shared and uniq

Integrative analysis of corticotrophs and folliculostellate cells: aging and paracrine regulation.

BACKGROUND: Aging of the human anterior pituitary is accompanied by structural remodeling of hormone-producing and non-endocrine cellular components. The coordinated morphometric relationship between adrenocorticotropic hormone (ACTH)-producing corticotrophs and folliculostellate (FS) cells during aging remains insufficiently characterized. MATERIALS AND METHODS: Twenty-nine human anterior pituitaries (33-95 years, both sexes) obtained at autopsy were examined using immunohistochemistry for ACTH and S-100 protein. Volume densities (VVACTH, VVFS) were quantified using calibrated digital morpho

An atlas of early human mandibular endochondral and osteogenic paracrine signaling regions of Meckel's cartilage.

The mandible, also known as the lower jaw, is the only bone in the skull that can move and is essential for speaking and chewing. Meckel's cartilage (MC) is a temporary structure that supports the formation of the mandible, but how MC is involved in the ossification of the mandible is poorly understood. Through the use of single-cell RNA sequencing and single-cell spatial transcriptomics analyses, a spatiotemporal atlas of MC in human fetuses from 7 to 15 wk postconception was established, highlighting the role of MC in the ossification of the mandible. Importantly, we revealed that two popula

Paracrine Signals in Calcified Conditioned Media Elicited Differential Responses in Primary Aortic Vascular Smooth Muscle Cells and in Adventitial Fibroblasts.

Our goal was to determine if paracrine signals from different aortic layers can impact other cell types in the diabetic microenvironment, specifically medial vascular smooth muscle cells (VSMCs) and adventitial fibroblasts (AFBs). The diabetic hyperglycemic aorta undergoes mineral dysregulation, causing cells to be more responsive to chemical messengers eliciting vascular calcification. Advanced glycation end-products (AGEs)/AGE receptors (RAGEs) signaling has been implicated in diabetes-mediated vascular calcification. To elucidate responses shared between cell types, pre-conditioned calcifie
