Left ventricular assist device explantation using a new double-patch technique†.

OBJECTIVES: There are several surgical approaches for explanting a left ventricular assist device (LVAD) after recovery of cardiac function. Thus, remaining ventricular assist device components may bear significant risks of infection or thrombosis. We hereby report our technique and two-center experience with explantation of LVADs using a new double-patch technique. METHODS: From March 2019 to April 2021, five patients underwent LVAD explantation after myocardial recovery (HVAD, n = 2; HeartMate 3, n = 3). The mean patient age was 50.3 years (100% male); the mean time on the LVAD was 23.1 ± 2

Furosemide alters nonlinear capacitance in isolated outer hair cells.

The outer hair cell (OHC) from the organ of Corti plays a crucial role in hearing through its unique voltage-dependent mechanical responses. Furosemide, one of the loop diuretics, disrupts normal cochlear function. Here we report on direct effects of furosemide on OHC motility-related, voltage-dependent capacitance using the whole-cell patch-clamp technique. Extracellularly applied furosemide reversibly shifted the voltage at peak capacitance (V(pkC(m))) to positive levels. The shift, whose maximum approached 90 mV, evidenced a Hill coefficient of 1.5 and K(1/2) of 10 mM. Changes in the magnit

Magnitude and modulation of pancreatic beta-cell gap junction electrical conductance in situ.

The parallel gap junction electrical conductance between a beta-cell and its nearest neighbors was measured by using an intracellular microelectrode to clamp the voltage of a beta-cell within a bursting islet of Langerhans. The holding current records consisted of bursts of inward current due to the synchronized oscillations in membrane potential of the surrounding cells. The membrane potential record of the impaled cell, obtained in current clamp mode, was used to estimate the behavior of the surrounding cells during voltage clamp, and the coupling conductance was calculated by dividing the m

Serum of Isaacs' syndrome suppresses potassium channels in PC-12 cell lines.

Blockage of K+ channels in nerve terminals by immunoglobulin is the speculated pathomechanism of Isaacs' syndrome. Using patch-clamp technique (whole-cell clamp), we investigated the effects on K+ current of serum taken from 2 patients with Isaacs' syndrome employing the clonal cell line PC-12. The addition of a patient's serum to the perfusion solution had little effect on the K+ current of P-12 cells. In contrast, K+ current was reduced by 25-80% when cells were cultured for 3-6 days with 2% serum as compared to control serum values. Suppression of the K+ current appears to develop gradually

Evidence for a PO2-sensitive K+ channel in the type-I cell of the rabbit carotid body.

Type-I cells of rabbit carotid bodies were studied with the patch-clamp technique in the whole-cell and on the cell-attached configuration. Cells exhibiting resting potentials of about -40 mV under normoxic conditions (PO2: 20 kPa), depolarized during hypoxia (PO2: 3.7 kPa). Hypoxia did not affect inward Ca2+ currents but inactivated outward K+ currents in voltage-clamp experiments. Single-channel currents recorded for the cell-attached mode showed a slope conductance of about 137 pS and a 0 mV reversal potential under symmetrical K+ concentration (140 mM). The open-probability (PO) of the sin

Functional heterogeneity of periglomerular cells in the rat olfactory bulb.

The periglomerular cells of the rat olfactory bulb, a virtually unknown population of interneurons, have been studied applying the whole-cell patch-clamp technique to thin slices. A prominent result, obtained under current-clamp conditions, is that these cells appear to be functionally heterogeneous, and show distinct excitability profiles. Voltage-clamp analysis allows the identification of the ionic basis of these differences and suggests a division into at least two classes, based on the characteristics of the K+ conductances. The first group displays two K+ conductances (delayed rectifier,

Thiamine Deficiency Increases Intrinsic Excitability of Mouse Cerebellar Purkinje Cells.

Thiamine deficiency is associated with cerebellar dysfunction; however, the consequences of thiamine deficiency on the electrophysiological properties of cerebellar Purkinje cells are poorly understood. Here, we evaluated these parameters in brain slices containing cerebellar vermis. Adult mice were maintained for 12-13 days on a thiamine-free diet coupled with daily injections of pyrithiamine, an inhibitor of thiamine phosphorylation. Morphological analysis revealed a 20% reduction in Purkinje cell and nuclear volume in thiamine-deficient animals compared to feeding-matched controls, with no

Contribution of calcium-sensitive potassium channels to NS1619-induced relaxation in human pregnant myometrium.

Large-conductance, calcium-sensitive potassium channels (BK(Ca)) are found at high density in the pregnant human myometrium. We have investigated, using isolated myometrial strips and freshly dispersed human myometrial cells, the action of a novel drug (NS1619) which has potassium channel opening activity. Isometric tension experiments demonstrated that NS1619 has a potent, relaxant effect on the pregnant human myometrium. Using both the inside-out and outside-out configurations of the patch-clamp technique, NS1619 appears to act directly on myometrial BK(Ca) channels to stimulate channel acti

[Inhibitory effect of tocainide on calcium current and potassium current in guinea pig ventricular myocytes].

In whole cell voltage clamp experiment with isolated guinea pig ventricular myocytes, tocainide was shown to exhibit a concentration-dependent inhibition of calcium current (ICa) and delayed rectifier potassium current (IK). Tocainide with a therapeutic concentration of 50 mumol.L-1 significantly inhibited the ICa and Itail (deactivating tail current of IK) by 16% and 3% respectively. Inhibition of ICa by tocainide was also shown to be dependent upon stimulating frequencies, with a greater blockade occurring at 2.0 Hz (57%) than at 0.2 Hz (17%). Tocainide was found to inhibit the IK,ATP by 74%

Ginsenoside Rg3 Alleviates Antithyroid Cancer Drug Vandetanib-Induced QT Interval Prolongation.

Источник пометил публикацию как отозванную

Inhibition of human ether-a-go-go-related gene (hERG) potassium channel is responsible for acquired long QT syndromes, which leads to life-threatening cardiac arrhythmia. A multikinase inhibitor, vandetanib, prolongs the progression-free survival time in advanced medullary thyroid cancer. However, vandetanib has been reported to induce significant QT interval prolongation, which limits its clinical application. Some studies have showed that ginsenoside Rg3 decelerated hERG K(+) channel tail current deactivation. Therefore, in this study, we aim to confirm whether ginsenoside Rg3 targeting hERG

Enantiospecific Positive Allosteric Modulation of α4β2 Nicotinic Receptor Subtypes.

Nicotinic acetylcholine receptors (nAChRs) are integral to brain function, playing critical roles in cognition and reward pathways. Among these, α4β2 nAChRs are key targets for developing therapeutics to address nicotine addiction and cognitive disorders. Here, we report the synthesis, stereochemical resolution, and pharmacological evaluation of GAT2800, a racemic compound identified as a positive allosteric modulator (PAM) of α4β2 nAChRs. Enantiomeric resolution yielded the inactive S-enantiomer (GAT2801) and the pharmacologically active R-enantiomer (GAT2802), with their configurations confi

Neuronal hyperactivity in neurons derived from individuals with gray matter heterotopia.

Periventricular heterotopia (PH), a common form of gray matter heterotopia associated with developmental delay and drug-resistant seizures, poses a challenge in understanding its neurophysiological basis. Human cerebral organoids (hCOs) derived from patients with causative mutations in FAT4 or DCHS1 mimic PH features. However, neuronal activity in these 3D models has not yet been investigated. Here we show that silicon probe recordings reveal exaggerated spontaneous spike activity in FAT4 and DCHS1 hCOs, suggesting functional changes in neuronal networks. Piezo1 Channel Activators Yoda1 and Yoda2 in the Context of Red Blood Cells.

Piezo1 is a mechanosensitive non-selective cation channel. Genetic alterations of the channel result in a hematologic phenotype named Hereditary Xerocytosis. With Yoda1 and, more recently, Yoda2, compounds to increase the activity of Piezo1 have become available. However, their concrete effect depends on the nano environment of the channel and hence on the cell type. Here we compare the potency of Yoda1 and Yoda2 in red blood cells (RBCs). We investigate the effect of the compounds on direct channel activity using automated patch clamp, as well as the secondary effects of channel activation on

Perinatal Opioid Exposure Results in Persistent Hypoconnectivity of Excitatory Circuits and Reduced Activity Correlations in Mouse Primary Auditory Cortex.

Opioid use by pregnant women results in neonatal opioid withdrawal syndrome (NOWS) and lifelong neurobehavioral deficits including language impairments. Animal models of NOWS show impaired performance in a two-tone auditory discrimination task, suggesting abnormalities in sensory processing in the auditory cortex. To investigate the consequences of perinatal opioid exposure on auditory cortex circuits, we administered fentanyl to mouse dams in their drinking water throughout gestation and until litters were weaned at postnatal day (P)21. We then used in vivo two-photon Ca2+ imaging in adult an

Functional expression and pharmacological modulation of TRPM3 in human sensory neurons.

BACKGROUND AND PURPOSE: The transient receptor potential (TRP) ion channel TRPM3 functions as a noxious heat sensor, plays a key role in acute pain sensation and inflammatory hyperalgesia in rodents. Despite its potential as a novel analgesic drug target, little is known about the expression, function and modulation in the humans. EXPERIMENTAL APPROACH: We studied TRPM3 in freshly isolated human dorsal root ganglion (hDRG) neurons and human stem cell-derived sensory (hSCDS) neurons. Expression was analysed at the mRNA level using RT-qPCR. Channel function was assessed using Fura-2-based calci

Excitatory effect of bradykinin on intrinsic neurons of the rat heart.

The heart receives sympathetic and parasympathetic innervation through the intrinsic cardiac nervous system. Although bradykinin (BK) has negative inotropic and chronotropic properties of cardiac contraction, the direct effect of BK on the intrinsic neural network of the heart is still unclear. In the present study, the effect of BK on the intracardiac ganglion neurons isolated from rats was investigated using the perforated patch-clamp technique. Under current-clamp conditions, application of 0.1 μM BK depolarized the membrane, accompanied by repetitive firing of action potentials. When BK wa

Identification and functional analysis of novel mutations of the CLCNKB gene in Chinese patients with classic Bartter syndrome.

Mutations in the gene CLCNKB encoding the ClC-Kb chloride channel causes classic Bartter syndrome, which is characterized by hypokalaemic metabolic alkalosis, renal salt loss, hyper-reninaemic hyperaldosteronism and normal blood pressure. We aimed to investigate the underlying mutations in CLCNKB in two Chinese patients with classic Bartter syndrome and then test the effect of the mutations on ClC-Kb chloride channel activity. Mutation analysis of CLCNKB was performed by polymerase chain reaction (PCR) direct sequencing. Expression of the wild-type and mutant ClC-Kb was heterologous in Xenopus

Dopamine modulation of calcium currents in pyloric neurons of the lobster stomatogastric ganglion.

We examined the dopamine (DA) modulation of calcium currents (ICa) that could contribute to the plasticity of the pyloric network in the lobster stomatogastric ganglion. Pyloric somata were voltage-clamped under conditions designed to block voltage-gated Na+, K+, and H currents. Depolarizing steps from -60 mV generated voltage-dependent, inward currents that appeared to originate in electrotonically distal, imperfectly clamped regions of the cell. These currents were blocked by Cd2+ and enhanced by Ba2+ but unaffected by Ni2+. Dopamine enhanced the peak ICa in the pyloric constrictor (PY), lat

Pentobarbital-activated Cl(-) channels in cultured adult and embryonic human DRG neurons.

Developmental differences in pentobarbital-activated Cl(-) currents were studied in adult and embryonic human dorsal root ganglia (DRG) neurons using whole-cell patch-clamp recordings. Pentobarbital-induced Cl(-) conductance was significantly greater in adult DRGs (28.4 pS) than in embryonic cells (19.1 pS). Fluctuation analysis of the spectral density plots of Cl(-) channel activation by pentobarbital showed age differences in the length and number of open time constants (adult cells, tau(1), tau(2), tau(3) were 224, 8. 4, 1.5 ms, respectively; embryonic cells tau(1) and tau (2) were 165 and
