Dissecting the Role of PCDH19 in Clustering Epilepsy by Exploiting Patient-Specific Models of Neurogenesis.

PCDH19-related epilepsy is a rare genetic disease caused by defective function of PCDH19, a calcium-dependent cell-cell adhesion protein of the cadherin superfamily. This disorder is characterized by a heterogeneous phenotypic spectrum, with partial and generalized febrile convulsions that are gradually increasing in frequency. Developmental regression may occur during disease progression. Patients may present with intellectual disability (ID), behavioral problems, motor and language delay, and a low motor tone. In most cases, seizures are resistant to treatment, but their frequency decreases

Patient-specific pediatric silicone heart valve models based on 3D ultrasound.

PURPOSE: Patient-specific heart and valve models have shown promise as training and planning tools for heart surgery, but physically realistic valve models remain elusive. Available proprietary, simulation-focused heart valve models are generic adult mitral valves and do not allow for patient-specific modeling as may be needed for rare diseases such as congenitally abnormal valves. We propose creating silicone valve models from a 3D-printed plastic mold as a solution that can be adapted to any individual patient and heart valve at a fraction of the cost of direct 3D-printing using soft materia

Patient-specific induced pluripotent stem-cell-derived models of LEOPARD syndrome.

The generation of reprogrammed induced pluripotent stem cells (iPSCs) from patients with defined genetic disorders holds the promise of increased understanding of the aetiologies of complex diseases and may also facilitate the development of novel therapeutic interventions. We have generated iPSCs from patients with LEOPARD syndrome (an acronym formed from its main features; that is, lentigines, electrocardiographic abnormalities, ocular hypertelorism, pulmonary valve stenosis, abnormal genitalia, retardation of growth and deafness), an autosomal-dominant developmental disorder belonging to a

Patient-Specific Lumped-Parameter Model for Quantifying Vessel-Specific Remodeling and Predicting Right Ventricular Function in Pulmonary Hypertension.

PURPOSE: Pulmonary hypertension (PH) is a heterogeneous disease with patient-specific variability and vessel-specific remodeling, which eventually lead to right ventricular (RV) failure. The gold standard for RV assessment-pressure-volume (PV) loop acquisition-is invasive and limited to specialized settings. This study aims to develop a patient-specific lumped-parameter model that quantifies vessel-specific remodeling and simulates RV PV loops across PH phenotypes using routine clinical data. METHODS: A lumped-parameter model was calibrated using right heart catheterization and echocardiograp

General versus site specific quantile random forest models for virtual patient-specific quality assurance of VMAT plans.

BACKGROUND: Modern radiotherapy techniques like volumetric-modulated arc therapy (VMAT) require labor-intensive patient-specific quality assurance (PSQA) measurements, which for busy clinics pose a substantial workflow burden. Most of these measurements simply confirm acceptable plans. This inefficiency has motivated the development of machine learning (ML) models for virtual PSQA to predict the gamma passing rate (GPR) and safely reduce the need for pre-treatment measurements. Plan complexity, often quantified by metrics like the small aperture score (SAS) and average leaf pair opening (ALPO)

Patient-specific prediction of regional lung mechanics in patients with ARDS with physics-based models: a validation study.

Lung-protective ventilation significantly influences outcomes in patients with acute respiratory distress syndrome (ARDS), but identifying optimal settings remains a challenge due to pronounced inter- and intrapatient variability in lung anatomy and pathophysiology. This study demonstrates that physics-based computational lung models tailored to individual patients can predict otherwise unobservable local lung states, enabling a quantitative analysis of regional ventilation and the mechanical load experienced by lung parenchyma during ventilation. For seven mechanically ventilated patients wit

Modeling Stent-Graft Deployment in Chronic Type B Aortic Dissection: A Patient-Specific Study on the Impact of Variations in Aortic Wall Thickness and Stent Delivery Procedure Assumptions.

Aortic dissection (AD), particularly type B aortic dissection (TBAD), is a severe vascular condition with complex biomechanical implications that pose challenges for effective treatment. Thoracic endovascular aortic repair (TEVAR) has emerged as the standard approach for acute complicated TBAD; however, its efficacy in chronic cases remains uncertain due to factors such as fibrotic dissection flap and altered aortic wall properties. Current numerical simulations of TEVAR provide valuable insights into stent-graft behavior but lack comprehensive analyses of the effect of variable thickness dist

Patient-specific computational fluid dynamics modeling of anterior communicating artery aneurysms: a study of the sensitivity of intra-aneurysmal flow patterns to flow conditions in the carotid arteries.

BACKGROUND AND PURPOSE: The purpose of this study was to investigate the effects of unequal physiologic flow conditions in the internal carotid arteries (ICAs) on the hemodynamics of anterior communicating artery aneurysms. METHODS: Patient-specific computational fluid dynamics models of 2 cerebral aneurysms were constructed from bilateral 3D rotational angiograms. The flow dynamics of the aneurysm sac were analyzed under the effect of unequal mean flows, phase shifts, and waveforms between the ICAs. A total of 9 simulations were performed for each patient; cine flow velocity simulations and

Numerical investigation of different viscosity models on pulsatile blood flow of thoracic aortic aneurysm (TAA) in a patient-specific model.

Aortic aneurysm is one of the most common aortic diseases that can lead to unfortunate consequences. Numerical simulations have an important role in the prediction of the aftereffects of vascular diseases including aneurysm. In this research, numerical simulation of pulsatile blood flow is performed for a 3-dimensional patient-specific model of a thoracic aortic aneurysm (TAA). Since the choice of blood viscosity model may have a significant impact on the simulation results, the effects of four non-Newtonian models of blood viscosity namely Carreau, Casson, Herschel-Bulkley, power low, and the

Application of Patient-Specific Instrumentation in a Dog Model with Antebrachial Growth Deformity Using a 3-D Phantom Bone Model.

One of the most frequent bone deformities in dogs is antebrachial growth deformity (AGD), which results from malunion of the distal growth plates. The objective of the present study was to re-align the limbs, which can correct the length mismatch and reset the coherence of the joint with the aid of a 3-D phantom model for surgical preplanning. A 14-month-old, intact female Golden Retriever with an angular deformity of the left radius and ulna was selected for the study. The diagnosis was confirmed by orthogonal radiographs. Moreover, computed tomography (CT) scans revealed a multiplane deformi

Evaluating the Predictive Potential of Patient-Specific Biomechanical Models in Class III Protraction Therapy.

Predicting treatment outcomes in Class III protraction therapy remains challenging. Although finite element analysis (FEA) helps in the study of biomechanics and planning of orthodontic treatment, its use in Class III protraction has mainly been in evaluating appliance designs rather than patient-specific anatomy. The predictive accuracy of FEA has not been validated in Class III protration therapy. In this study, ten patients (5 female, 5 male, aged 7-11 years) with Class III malocclusion received either facemask or mentoplate treatment. CT scans from four patients were used to construct simp

In-silico patient-specific modelling of prostate cancer: Predicting PSA dynamics and treatment response.

BACKGROUND AND OBJECTIVE: Prostate cancer remains a significant global health concern, with treatment response varying among patients. Radiotherapy, often combined with hormone therapy, is a key treatment approach, but predicting individual outcomes remains challenging. Computational models have emerged as valuable tools to simulate tumour behaviour and optimise treatment strategies. This study presents a patient-specific computational model designed to predict tumour response by associating Prostate-Specific Antigen (PSA) dynamics with tumour biological behaviour under therapy. METHODS: The

Patient-specific 3D tibial model: transforming meniscal allograft transplantation and surgical planning.

BACKGROUND: Meniscal allograft transplantation (MAT) restores knee function by replacing a damaged or absent meniscus with a healthy allograft, helping to preserve joint stability, distribute the load, and reduce cartilage degeneration. However, traditional 2D imaging techniques fail to fully capture the knee's complex three-dimensional anatomy, making accurate surgical planning challenging. Computed Tomography (CT)-based 3D printing offers a patient-specific solution by generating anatomically precise tibial models, allowing for enhanced preoperative planning. This is particularly valuable in

A postoperative tumor-specific death prediction model for patients with endometrial cancer: a retrospective study.

BACKGROUND: Endometrial cancer (EC) is an epithelial malignancy occurring in the endometrium, with a 5-year mortality rate of above 10%. However, there is currently a lack of studies exploring the potential of a predictive model of tumor-specific death after surgery in these patients. METHODS: From January 2015 to December 2017, data related to 482 patients with EC admitted to the Dushu Lake Hospital Affiliated to Soochow University were analyzed. Patients were divided into death (n=62) and survival (n=420) groups according to whether tumor-specific death occurred at 5 years postoperatively o
