This brief review has attempted to portray the complex involvement of prolactin in pregnancy. Pituitary prolactin production and lactotroph proliferation are markedly affected by oestrogens and pregnancy. Enlargement of pre-existing prolactinomas may occur during pregnancy induced with dopamine agonist drugs, and this can be a major clinical problem with larger tumours. Prolactin is produced by late luteal and decidualized endometrium as well as by the pituitary gland, but much less is known about its secretory regulation or its function. The family of prolactin-related protein hormones is rap

Patients with liver disease have increased plasma concentrations of the endogenous opioid peptides methionine enkephalin and leucine enkephalin. As an initial investigation to determine whether opioid peptides contribute to any of the clinical manifestations of hepatic disease nalmefene, a specific opioid antagonist devoid of agonist activity, was given to 11 patients with cirrhosis. They all experienced a severe opioid withdrawal reaction on starting the drug. In the nine patients with primary biliary cirrhosis pruritus was greatly alleviated, fatigue seemed to improve, and plasma bilirubin c

Endogenous opioid peptides take an active part in the pathogenesis of motion sickness (MS). Some regulatory peptides (substances P, gamma and des-tyr-gamma-endorphins) manifest antiemetic properties, unlike opioid peptides and morphine. Thus, we have examined the vestibulo-protective properties of some regulatory peptides during simulation of MS in cats. Vestibulo-protective properties have been discovered in some peptides: sweet water hydra undecapeptide, substances P, gamma and des-tyr-gamma-endorphins. It has been suggested that regulatory peptides take an active part in the genesis of vest

The application of radioimmunoassay of insulin, C-peptide, gastrin, glucagon, vasoactive intestinal polypeptides (VIP), somatostatin, human pancreatic polypeptides (hPP), substance P and neurotensin to detect endocrine tumors of the pancreas and other organ systems is undoubtedly important in the clinical management of patients suspected of having tumors that secrete these hormones. Radioimmunoassays of the above gut peptides have certain degrees of specificity and sensitivity; however, there are several factors that need to be considered in the interpretation of results since heterogeneity of

The cardiovascular effects of intravenous and intracisternal administration of neurohypophysial peptides were studied in anaesthetized dogs. Intracisternal oxytocin (1 and 10 mU kg-1) increased blood pressure. Intravenous lysine vasopressin (0.1 to 100 mU kg-1) induced a dose-dependent increase in blood pressure with bradycardia. Intracisternal lysine vasopressin (0.01 to 10 mU kg-1) elicited a dose-related decrease in blood pressure but no change in heart rate. These central hypotensive effects of vasopressin were suppressed by intravenous guanethidine, dl-propranolol, prazosin or atropine an

The multilayered plant immune system relies on rapid recognition of pathogen-associated molecular patterns followed by activation of defense-related genes, resulting in the reinforcement of plant cell walls and the production of antimicrobial compounds. To suppress plant defense, fungi secrete effectors, including a recently discovered Zn-metalloproteinase from Fusarium verticillioides, named fungalysin Fv-cmp. This proteinase cleaves class IV chitinases, which are plant defense proteins that bind and degrade chitin of fungal cell walls. In this study, we investigated plant responses to such p

Covalently linking together different proteins can enhance functionality for a range of applications. We have developed the SnoopLigase peptide-peptide conjugation method to easily and specifically link proteins fused to the peptides SnoopTagJr or DogTag via an isopeptide bond. SnoopLigase conjugation has been applied for enhancing enzyme resilience and for antigen oligomerization to enhance vaccine efficacy. Following conjugation, SnoopLigase and unreacted substrates can be removed by solid-phase immobilization of SnoopLigase, yielding purified protein-protein conjugates. Here, we describe pr

The extraordinary capabilities of the biological sense of smell remain unmatched by electronic noses. Here, we present a revolutionary concept for next-generation electronic noses that integrates both specific and cross-reactive recognition principles within a single system. A novel biomimetic opto-electronic nose with enhanced performance based on Surface Plasmon Resonance Imaging (SPRI) was developed, combining cross-reactive peptides with selective Phage Display-derived peptides. Using Phage Display, five highly selective peptides were identified for detecting BTEX compounds (Benzene, Tolue

Hypertension is a common cardiovascular disease closely related to high-risk diseases such as myocardial infarction and diabetes. In this study, LC-MS/MS identification and bioinformatic screening were performed on silkworm pupa protein fermented by Pediococcus acidilactici JP-10, and 11 sequences with potential ACE-inhibitory capacity were obtained. An l-NAME-induced hypertensive rat model was established via gavage. After the model was successfully established, different doses of silkworm pupa ACE inhibitory peptides (SPPs) and captopril were administered to hypertensive rats by gavage at re

Mutations in the cystic fibrosis (CF) transmembrane conductance regulator (CFTR) protein lead to persistent lung bacterial infections, mainly due to Pseudomonas aeruginosa, causing loss of respiratory function and finally death of people affected by CF. Unfortunately, even in the era of CFTR modulation therapies, management of pulmonary infections in CF remains highly challenging especially for patients with advanced stages of lung disease. Recently, we identified antimicrobial peptides (AMPs), namely Esc peptides, with potent antipseudomonal activity. In this study, by means of electrophysiol

Bacterial infection is one of the leading causes of death in young, elderly, and immune-compromised patients. The rapid spread of multi-drug-resistant (MDR) bacteria is a global health emergency and there is a lack of new drugs to control MDR pathogens. We describe a heretofore-unexplored discovery pathway for novel antibiotics that is based on self-targeting, structure-disrupting peptides. We show that a helical peptide, KFF- EcH3, derived from the Escherichia coli methionine aminopeptidase can disrupt secondary and tertiary structure of this essential enzyme, thereby killing the bacterium (i

Both vertebrates and invertebrates employ alpha-helical antimicrobial peptides (AMPs) as an essential component of their innate immune system. However, evolutionary relation of these immune molecules remains unresolved. Venoms, as key weapons of venomous arthropods for prey and defense, receive increasing recognition as an emerging source of such peptides. From a cDNA library prepared from the venom gland of the scorpion Mesobuthus eupeus, clones encoding precursors of two new AMPs, named meucin-13 (IFGAIAGLLKNIF-NH(2)) and meucin-18 (FFGHLFKLATKIIPSLFQ), have been isolated. The precursor of m

Interphotoreceptor retinoid-binding protein (IRBP) is a glycoprotein of 1264 residues (bovine) which localizes specifically in the retina and pineal gland and induces inflammatory changes in these organs (EAU and EAP, respectively) in immunized animals. We report here on differences between the immunological activities in Lewis rats of four IRBP-derived synthetic peptides. Only one of these peptides, designated R14 (residues: 1169-1191) is immunodominant, i.e., it has the capacity to stimulate lymphocytes sensitized against whole IRBP. The remaining peptides, R4 (1158-1180), R8 (1197-1209), an

Preeclampsia, a pregnancy-specific disorder, shares typical pathophysiological features with protein misfolding disorders including Alzheimer's disease. Characteristic for preeclampsia is the involvement of multiple proteins of which fragments of SERPINA1 and β-amyloid co-aggregate in urine and placenta of preeclamptic women. To explore the biophysical basis of this interaction, we investigated the multidimensional efficacy of the FVFLM sequence in SERPINA1, as a model inhibitory agent of β-amyloid aggregation. After studying the oligomerization of FVFLM peptides using all-atom molecular dynam

In recent decades, antimicrobial peptides (AMPs) have emerged as some of the most extensively studied alternatives to conventional antibiotics. To gain deeper insights into their mechanisms of action and development, AFM has played a crucial role as an imaging and force spectroscopic tool. Here, we describe the methods used for examining the effects of AMPs on bacteria using imaging and force spectroscopy techniques. These approaches have proven essential for elucidating their mechanisms of action and identifying potential outcomes.

Oncolytic peptides (OLPs) constitute an emerging class of immunotherapeutics that combine direct cancer cell lysis with immune activation. Indeed, OLPs induce rapid immunogenic cell death by disrupting intracellular membranes, which culminates with the abundant release of malignant cell contents including immunostimulatory danger-associated molecular patterns. This results in the engagement of innate immune sensors that potently activate dendritic cells, leading to the cross-priming of tumor-specific T lymphocytes that mediate systemic anticancer immunity. Through such a dual mechanism, OLPs c

Amyloid fibrils are a common pathological hallmark in multiple neurodegenerative diseases, yet molecular tools to recognize and manipulate them remain scarce. We report FibrilPaints, a family of modular peptides designed for amyloid binding and adaptable chemical functionality. The degenerative amyloid-targeting unit of FibrilPaints, W5P4H3R2, has a high content of π-stacking and aromatic side chains. Systematic sequence variation, altering charge, termini, and residue order, revealed the importance of the composition of the amyloid-targeting unit for high-affinity binding across Tau and Hunti

The potential of peptide therapeutics is high because the targeting is highly specific, toxicity is low, and also the ability to affect complex biological determinants. These things are, however, hampered by issues like low stability, faster corrosion, low bioavailability, and low cell permeability, which limit their effectiveness in clinical conditions. This is why lipidation and peptide stapling approaches to balancing, which are highlighted in this review, are collaborators in challenging these questions. Lipidation is a covalent bonding of fatty acids or hydrophobic components, which incre

A α-helical conformation within Class B1 G protein-coupled receptors agonists has been thought necessary for receptor activation. Gellman et al. suggested that selective replacement of L-amino acids with D-enantiomers could yield biased agonists. The new strategy helical would broaden the tools available for investigating agonist conformations and therapeutic potentials.
