Grafting profoundly influences fruit tree performance, yet the molecular mechanisms underlying rootstock-scion interactions in mulberry (Morus multicaulis) remain poorly understood. To investigate the regulatory networks linking grafting to scion physiology, we performed an integrated analysis combining small RNA sequencing, transcriptome-based KEGG pathway assessment, and qRT-PCR validation of key target genes across five rootstock-scion combinations. High-throughput miRNA profiling of phloem tissues identified 71 conserved and 156 novel miRNAs, which exhibited distinct, genotype-dependent ex

Zinc (Zn) is an essential micronutrient involved in numerous physiological processes in plants, serving catalytic, structural, and regulatory functions. Maintaining its appropriate concentration requires precise control of uptake, transport, distribution, and storage, as both Zn deficiency and excess lead to disruptions in plant growth and development. This paper presents the mechanisms regulating zinc homeostasis in plants, with a particular focus on the role of membrane transporters and chelating compounds. The ZIP, MTP, and HMA protein families play a key role, being responsible for cellula

Anatomical injury of the leaves of the invasive species, Cirsium arvense (L.) Scop., caused by the eriophyid mite Aceria anthocoptes (Nal.), which is the only eriophyid mite that has been recorded on C. arvense worldwide, is described. The injury induced by the mite feeding on the leaves of C. arvense results in visible russeting and bronzing of the leaves. Other conspicuous deformations are folding and distortion of the leaf blade and curling of leaf edge, as well as gradual drying of leaves. The anatomical injury of the mature leaves of field-collected plants was limited to the epidermis of

Phloem-limited bacterial "Candidatus Liberibacter" species are associated with incurable plant diseases worldwide. Antimicrobial treatments for these pathogens are challenging due to the difficulty of reaching the vascular tissue they occupy at bactericidal concentrations. Here, in vitro antimicrobial mechanisms of Zinkicide TMN110 (ZnK), a nonphytotoxic zinc oxide (ZnO)-based nanoformulation, were compared to those of bulk ZnO (b-ZnO) using as a model the only culturable species of the genus, Liberibacter crescens Minimum bactericidal concentration (MBC) determination and time-kill assays sho

In plants, the directed distribution of photosynthetic products from source organs to heterotrophic sink organs forms the material basis for biomass accumulation and crop yield. SWEETs (sugars will eventually be exported transporters) facilitate passive sucrose efflux to link source and sink transport during phloem loading for plant carbon allocation. However, it remains unclear how SWEET-mediated sugar transport coordinates carbon allocation and lipid metabolism. Therefore, this Review integrates the structural evolution, transport mechanisms, and physiological functions of SWEETs. It investi

Whiteflies (Hemiptera: Aleyrodidae) are among the most destructive pests of vegetables and ornamental crops, causing severe yield losses through phloem feeding and virus transmission. Although historically most research has centered on Bemisia tabaci, the greenhouse whitefly Trialeurodes vaporariorum is rapidly emerging as a major pest across sub-Saharan Africa. Its spread in both open-field and protected cropping systems is linked to shifts in land use, climate variability, and intensified production systems. Furthermore, growing insecticide resistance to commonly used insecticides threatens

The lack of sequence similarity among insect effectors necessitates the independent identification of effectors in the Asian citrus psyllid Diaphorina citri, the primary vector of huanglongbing pathogens. In this study, nine candidate effectors were identified from the D. citri salivary gland transcriptome. Among them, DcE13 exhibited the highest FPKM value and was delivered into plants during feeding. Transient expression in tobacco leaves showed that DcE13 is localized in the plasma membrane and around the nuclear envelope of plant cells. It inhibits BAX- and INF1-induced hypersensitive resp

The development of tools to quickly identify the fate of damaged trees after a stress event such as a wildfire is of great importance. In this context, an innovative approach to assess irreversible physiological damage in trees could help to support the planning of management decisions for disturbed sites to restore biodiversity, protect the environment and understand the adaptations of ecosystem functionality. The vitality of trees can be estimated by several physiological indicators, such as cambium activity and the amount of starch and soluble sugars, while the accumulation of ethanol in th

Systemic acquired resistance (SAR) is a form of broad-spectrum resistance induced in response to local infections that protects uninfected parts against subsequent secondary infections by related or unrelated pathogens. SAR signaling requires two parallel branches, one regulated by salicylic acid (SA), and the other by azelaic acid (AzA) and glycerol-3-phosphate (G3P). AzA and G3P function downstream of the free radicals nitric oxide (NO) and reactive oxygen species (ROS). During SAR, SA, AzA and G3P accumulate in the infected leaves, but only a small portion of these is transported to distal

Candidatus Liberibacter asiaticus (CLas) is a phloem-limited, gram-negative, fastidious bacterium that is associated with the development of citrus greening disease, also known as Huanglongbing (HLB). CLas is transmitted by the Asian citrus psyllid (ACP) Diaphorina citri, in a circulative manner. Two major barriers to transmission within the insect are the midgut and the salivary glands. We performed a thorough microscopic analysis within the insect midgut following exposure to CLas-infected citrus trees. We observed changes in nuclear architecture, including pyknosis and karyorrhexis as well

The hollowed root formation and 4'-deoxyflavone accumulation in Scutellaria baicalensis are associated with jasmonate (JAs) signaling, but the upstream regulators integrating these processes with drought tolerance remain unknown. This study investigated whether the 13-lipoxygenase gene SbLOX3 may act as a modulator. Transgenic plants overexpressing SbLOX3 or with RNAi suppression were exposed to polyethylene glycol-simulated and long-term soil drought stress. Metabolite profiling, gene expression analysis, root morphological measurements, and histological observations were conducted. Overexpre

Plants frequently encounter multiple pathogens concurrently, yet the molecular bases of pathogen-pathogen interactions during mixed infections remain largely unresolved. Here we show that co-infection of rice by Rice orange leaf phytoplasma (ROLP) and Rice stripe mosaic virus (RSMV) markedly suppresses RSMV proliferation in both host plants and the insect vector. Electron microscopy and confocal imaging reveal that ROLP and RSMV co-inhabit individual rice phloem cells and undergo close membrane interaction, producing swollen RSMV virions with reduced infectivity. We further demonstrate that th

Honeydew excreted by phloem-feeding insects is a major plant-derived carbohydrate resource in agroecosystems, yet its chemical sensitivity to climate change remains poorly understood. We examined how elevated atmospheric CO2 and water limitation affect honeydew primary metabolites and a honeydew-feeding parasitoid. Using a tritrophic system comprising grapevine (Vitis vinifera), the vine mealybug (Planococcus ficus), and its parasitoid (Anagyrus vladimiri), plants were grown under ambient or elevated CO2 and well-watered or water limitation conditions. Honeydew was quantified, primary metaboli

In plants, the mobile signal for systemic acquired resistance (SAR), an organism-wide state of enhanced defense to subsequent infections, has been elusive. By stimulating immune responses in mosaic tobacco plants created by grafting different genetic backgrounds, we showed that the methyl salicylate (MeSA) esterase activity of salicylic acid-binding protein 2 (SABP2), which converts MeSA into salicylic acid (SA), is required for SAR signal perception in systemic tissue, the tissue that does not receive the primary (initial) infection. Moreover, in plants expressing mutant SABP2 with unregulate

During insect feeding, a complex interaction takes place at the feeding site, with plants deciphering molecular information associated with the feeding herbivore, resulting in the upregulation of the appropriate defenses, and the herbivore avoiding or preventing these defenses from taking effect. Whiteflies can feed on plants without causing significant damage to mesophyll cells, making their detection extra challenging for the plant. However, whiteflies secrete honeydew that ends up on the plant surface at the feeding site and on distal plant parts below the feeding site. We reasoned that thi

Pemphigus spyrothecae Pass. which is a member of the subfamily Pemphiginae is one of the gall-inducing aphids that occurs on poplar trees. Phloem feeding of a founding mother on leaf petiole results in the formation of a new organ, i.e., the spiral gall. This study documents aphid development inside the galls during the growing season and the effect of their feeding on leaf architecture and physiology of the host plant. In particular, leaf length, width, and area were measured, as well as hydrogen peroxide (H2O2) content, electrolyte leakage (EL), malondialdehyde (MDA) concentration, and the a

Diuraphis noxia (Russian wheat aphid, RWA) is a major pest on wheat in South Africa and most other wheat growing countries. Being a probing-sucking insect, RWAs insert their stylets into the phloem sieve elements and feed on the phloem sap. This feeding causes necrotic lesions in resistant varieties, or decoloration of leaves and death in susceptible varieties. In an effort to broaden our understanding on the response of the plant to RWA feeding, we synthesized and analyzed expressed sequence tags (ESTs) from suppression subtractive hybridization (SSH) libraries. These libraries were construct

Plants have evolved a signalling pathway in which, when one root senses local nitrogen (N) deficiency, nitrate uptake by other roots is enhanced in a complementary manner. This long-range communication, known as systemic N-demand signalling, is triggered when the root-to-shoot mobile signal, C-TERMINALLY ENCODED PEPTIDE (CEP), which is induced in roots under N starvation, is perceived by CEP RECEPTOR 1 (CEPR1) expressed in the leaf phloem. However, the molecular components required for CEP-dependent CEPR1 activation remain unknown. Here we identified a leucine-rich repeat receptor kinase that

Huanglongbing (HLB), caused by the phloem-restricted bacterium Candidatus Liberibacter asiaticus (C. Las), is vectored by the Asian citrus psyllid (ACP) and threatens global citrus production. We demonstrate that VU041 (C19H20F3N3O), a small-molecule inhibitor of inward rectifier potassium (Kir) channels, provides a dual-mechanism approach to management of Huanglongbing (HLB) disease by combining acute toxicity with antifeedant activity. Topical and tarsal contact bioassays revealed that VU041 induced rapid mortality to ACP (24 h LD50 = 25 ng/insect) and reduced salivary gland secretion to sup

Achieving optimal flowering time is a pivotal factor in precision plant breeding. Here, in maize, we characterized an EMS-induced additive delayed-flowering mutant, edf1. edf1 and its heterozygotes are late-flowering under both long- and short-day photoperiods in the field, and harbor a splice-donor mutation in ALTERED PHLOEM DEVELOPMENT (APL). This mutation leads to the accumulation of the minor APL-β transcript, a splice variant expressed across a large natural maize population. Expression of the major splice variant APL-α is associated with flowering initiation; APL knockdown delays floweri
