Interfacing Photosynthetic Membrane Protein with Mesoporous WO3 Photoelectrode for Solar Water Oxidation.

Photosynthetic biocatalysts are emerging as a new class of materials, with their sophisticated and intricate structure, which promise improved remarkable quantum efficiency compared to conventional inorganic materials in artificial photosynthesis. To break the limitation of efficiency, the construction of bioconjugated photo-electrochemical conversion devices has garnered substantial interest and stood at the frontier of the multidisciplinary research between biology and chemistry. Herein, a biohybrid photoanode of a photosynthetic membrane protein (Photosystem II (PS II)), extracted from fres

Specific role of phosphatidylglycerol and functional overlaps with other thylakoid lipids in Arabidopsis chloroplast biogenesis.

With phosphate deficiency, the role of phosphatidylglycerol is compensated by increased glycolipid content in thylakoid membrane biogenesis but not photosynthetic electron transport in Arabidopsis chloroplasts. In plants and cyanobacteria, anionic phosphatidylglycerol (PG) is the only major phospholipid in thylakoid membranes, where neutral galactolipids monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG) are predominant. In addition to provide a lipid bilayer matrix, PG plays a specific role in photosynthetic electron transport. Non-phosphorous sulfoquinovosyldiacylglyce

Stimulation of chlororespiration by drought under heat and high illumination in Rosa meillandina.

Rosa meillandina plants were used to study the effects of water deficit on photosynthesis and chlororespiration. Plants showed high tolerance to heat and high illumination in controlled conditions that ensured that there was no water deficit. However, when heat and high illumination were accompanied by low watering photosynthetic linear electron transport was down regulated, as indicated by the reduced photochemistry efficiency of PS II, which was associated with an increase in the non-photochemical quenching of fluorescence. In addition to the effects on the photosynthetic activity, changes w

Effect of (C6H5)3PbCl and (C6H5)3SnCl on delayed luminescence intensity, evolving oxygen and electron transport rate in photosystem II of Chlorella vulgaris.

The effect of the organometallic compounds containing lead, (C6H5)3PbCl , and tin, (C6H5)3SnCl, on Chlorella green algae photosystem II was studied. Suspension of the algae treated with (C6H5)3SnCl at concentrations of 1.0 and 4.0 micromol dm(-3) for 22 h revealed a decrease in most physiological parameters studied, particularly in decasecond component of delayed chlorophyll luminescence, photosynthetic electron transport rate and diluted oxygen concentration, which implies an inhibition of photosynthetic electron transport as well as oxygen evolving system. On the other hand, (C6H5)3PbCl caus

Evaluation of physiological changes induced by the fluoroquinolone antibiotic ciprofloxacin in the freshwater macrophyte species Lemna minor and Lemna gibba.

The worldwide increase in the consumption of antibiotics is becoming a concern for the scientific community, since the presence of their residues in the wild poses specific challenges, especially in ecotoxicological terms. Currently, antibiotics are used for a wide range of purposes, being used against bacterial diseases but also as growth promoters. As a result, their environmental presence can affect wild organisms, especially those from the aquatic environment. This scenario leads to the need of characterizing the toxicity of antibiotics, especially towards non-target organisms. In this stu

[Environmental Effect of Substrate Amelioration on Lake: Effects on Phragmites communis Growth and Photosynthetic Fluorescence Characteristics].

Growth of rooted aquatic macrophytes was affected by the nature and composition of lake bottom sediments. Obviously, it has been recognized as an important ecological restoration measure by improving lake substrate and then reestablishing and restoring aquatic macrophytes in order to get rid of the environmental problem of lake. This study simulated five covering thickness to give an insight into the influence of substrate amelioration on Phragmites communis growth and photosynthetic fluorescence characteristics. The results showed that the total biomass, plant height, leaf length and leaf wid

Evidence for thermosensitivity of the cotton (Gossypium hirsutum L.) immature fiber (im) mutant via hypersensitive stomatal activity.

Thickness of cotton fiber, referred to as fiber maturity, is a key determinant of fiber quality, lint yield, and textile performance. The cotton immature fiber (im) mutant has been used to study fiber maturity since its fiber is thinner than the wild type near isogeneic line (NIL), Texas Marker-1 (TM-1). The im phenotype is caused by a single recessive mutation of a pentatricopeptide repeat (PPR) gene that reduces the activity of mitochondrial complex I and up-regulates stress responsive genes. However, the mechanisms altering the stress responses in im mutant are not well understood. Thus, we

EPR and ENDOR investigation of the primary electron acceptor radical anion QA.- in iron-depleted photosystem II membrane fragments.

Photosystem II (PS II) membrane fragments were treated with trypsin at pH = 7.4 followed by incubation with o-phenanthroline and lithium perchlorate. This procedure removes and/or decouples the non-heme Fe2+ associated with the quinones QA and QB in the PS II reaction center (RC). Treatment of such samples (referred to as iron-depleted) with sodium dithionite or illumination in the presence of dichlorophenol indophenol (DCIP) and sodium ascorbate yielded EPR spectra similar to those of the plastoquinone-9 (PQ-9) radical anion generated in organic solvents. Q-band EPR yielded the principal valu

Improving photosynthesis and crop productivity by accelerating recovery from photoprotection.

Crop leaves in full sunlight dissipate damaging excess absorbed light energy as heat. When sunlit leaves are shaded by clouds or other leaves, this protective dissipation continues for many minutes and reduces photosynthesis. Calculations have shown that this could cost field crops up to 20% of their potential yield. Here, we describe the bioengineering of an accelerated response to natural shading events in Nicotiana (tobacco), resulting in increased leaf carbon dioxide uptake and plant dry matter productivity by about 15% in fluctuating light. Because the photoprotective mechanism that has b

HHL1 and SOQ1 synergistically regulate nonphotochemical quenching in Arabidopsis.

Nonphotochemical quenching (NPQ) is an important photoprotective mechanism that quickly dissipates excess light energy as heat. NPQ can be induced in a few seconds to several hours; most studies of this process have focused on the rapid induction of NPQ. Recently, a new, slowly induced form of NPQ, called qH, was found during the discovery of the quenching inhibitor suppressor of quenching 1 (SOQ1). However, the specific mechanism of qH remains unclear. Here, we found that hypersensitive to high light 1 (HHL1)-a damage repair factor of photosystem II-interacts with SOQ1. The enhanced NPQ pheno

Ultraviolet-B protection of ascorbate and tocopherol in plants related with their function on the stability on carotenoid and phenylpropanoid compounds.

Ascorbate and tocopherol are important hydrophilic or lipophilic antioxidants in plants, while their crucial roles in the antioxidant defense system under ultraviolet B radiation were not well understood. The mutants of Arabidopsis thaliana deficient in ascorbate (vtc1 and vtc2) or tocopherol (vte1) were used to analyze their physiological, biochemical and metabolic change in responses to Ultraviolet B radiation. Results showed that loss of either ascorbate or tocopherol caused reduction in phenylpropanoid and flavonol glycosides compounds, as well as reduction in superoxide dismutase activity

Non-photochemical quenching in cryptophyte alga Rhodomonas salina is located in chlorophyll a/c antennae.

Photosynthesis uses light as a source of energy but its excess can result in production of harmful oxygen radicals. To avoid any resulting damage, phototrophic organisms can employ a process known as non-photochemical quenching (NPQ), where excess light energy is safely dissipated as heat. The mechanism(s) of NPQ vary among different phototrophs. Here, we describe a new type of NPQ in the organism Rhodomonas salina, an alga belonging to the cryptophytes, part of the chromalveolate supergroup. Cryptophytes are exceptional among photosynthetic chromalveolates as they use both chlorophyll a/c pro

Up-regulation of nitrogen metabolism and chlorophyll biosynthesis by hydrogen sulfide improved photosystem photochemistry and gas exchange in chromium-contaminated bean (Phaseolus vulgaris L.) plants.

Hydrogen sulfide (H₂S) is considered as plant growth promoter under heavy metal stress, though its specific effects on photosynthesis are rarely explored. This study investigates the protective effects of exogenous H2S donor sodium hydrosulfide (NaHS) on chlorophyll metabolism and photosystem II (PSII) function in 24-day-old bean plants exposed to 10 μM chromium (Cr) stress. Sodium hydrosulfide (100 μM) reduced Cr accumulation in both roots and leaves, leading to restored plant growth. Concomitantly, H₂S mitigated Cr-induced oxidative damages by decreasing reactive oxygen species levels and fu

Competitive interaction of Mn(II) and Fe(II) cations with the high-affinity Mn-binding site of the photosystem II: evolutionary aspect.

The evolutionary origin of the oxygen-evolving complex (OEC) in the photosystem II (PSII) is still unclear, as is the nature of electron source for the photosystem before the OEC had appeared. Johnson et al. (in PNAS 110:11238, 2013) speculated that Mn(II) cations were the source of electrons for transitional photosystems. However, Archean oceans also contained Fe(II) cations at concentrations comparable or higher than that of Mn(II). Fe(II) cations can bind to the high-affinity (НА) Mn-binding site in the OEC (Semin et al. in Biochemistry 41:5854, 2002). Now we have investigated the competiti

Partial replacement of nitrate by ammonium increases photosynthesis and reduces oxidative stress in tanzania guinea grass exposed to cadmium.

In order to grow and effectively uptake and accumulate cadmium (Cd), plants used for phytoextraction have to cope with toxicity, which may be influenced by the supply of nitrate (NO3-) and ammonium (NH4+). Thus, we evaluated the effect of these nitrogen forms on the photosynthetic and antioxidant enzyme activities of Panicum maximum cv. Tanzania (tanzania guinea grass) under Cd stress. Plants were grown in nutrient solution under greenhouse conditions and subjected to a 3 × 3 factorial experiment. They were supplied with three NO3-/NH4+ ratios (100/0, 70/30 and 50/50) and exposed to three Cd r

Screening of novel chemical compounds as possible inhibitors of carbonic anhydrase and photosynthetic activity of photosystem II.

Thirty novel chemical compounds were designed and synthesized expecting that they would be possible inhibitors. From this number eleven were organic bases, twenty-four were their organic derivatives and fourteen were metal complexes. Screening of these chemicals by their action on photosynthetic electron transfer (PET) and carbonic anhydrase (CA) activity (CAA) of photosystem II (PSII), α-CA, as well as β-CA was done. Several groups were revealed among them. Some of them are capable to suppress either one, two, three, or even all of the measured activities. As example, one of the Cu(II)-phenyl

Covalent immobilization of oriented photosystem II on a nanostructured electrode for solar water oxidation.

Photosystem II (PSII) offers a biological and sustainable route of photochemical water oxidation to O2 and can provide protons and electrons for the generation of solar fuels, such as H2. We present a rational strategy to electrostatically improve the orientation of PSII from a thermophilic cyanobacterium, Thermosynechococcus elongatus , on a nanostructured indium tin oxide (ITO) electrode and to covalently immobilize PSII on the electrode. The ITO electrode was modified with a self-assembled monolayer (SAM) of phosphonic acid ITO linkers with a dangling carboxylate moiety. The negatively char

Lichens survive in space: results from the 2005 LICHENS experiment.

This experiment was aimed at establishing, for the first time, the survival capability of lichens exposed to space conditions. In particular, the damaging effect of various wavelengths of extraterrestrial solar UV radiation was studied. The lichens used were the bipolar species Rhizocarpon geographicum and Xanthoria elegans, which were collected above 2000 m in the mountains of central Spain and as endolithic communities inhabiting granites in the Antarctic Dry Valleys. Lichens were exposed to space in the BIOPAN-5 facility of the European Space Agency; BIOPAN-5 is located on the outer shell o

A novel metabolic form of the 32 kDa-D1 protein in the grana-localized reaction center of photosystem II.

Two forms of the 32 kDa-D1 reaction center protein of photosystem II (PSII), having slightly different mobilities on denaturing polyacrylamide gels, have been resolved in Spirodela oligorrhiza, Glycine max L., Gossypium hirsutum L., Triticum aestivum L., and Zea mays L. The protein band with faster mobility is identified as the 32 kDa-D1 protein, and the less mobile band as a novel form, designated 32*. The two forms are structurally similar based on immunological and partial proteolytic tests. 32* is associated exclusively with the grana and is present in the PSII reaction center. Temporally,
