Mitochondrial calcium uptake orchestrates vertebrate pigmentation via transcriptional regulation of keratin filaments.

Mitochondria regulate several physiological functions through mitochondrial Ca2+ dynamics. However, role of mitochondrial Ca2+ signaling in melanosome biology remains unknown. Here, we show that pigmentation requires mitochondrial Ca2+ uptake. In vitro gain and loss of function studies demonstrate that mitochondrial Ca2+ uniporter (MCU) is crucial for melanogenesis while MCU rheostat, MCUb negatively control melanogenesis. Zebrafish, MCU+/- and MCUb-/- mice models show that MCU complex drives pigmentation in vivo. Mechanistically, MCU silencing activates transcription factor NFAT2 to induce ex

Involvement of miR-495 in the skin pigmentation of rainbow trout (Oncorhynchus mykiss) through the regulation of mc1r.

MicroRNAs (miRNAs) play crucial roles in skin pigmentation in animals. Rainbow trout (Oncorhynchus mykiss) is a key economic fish species worldwide, and skin color directly affects its economic value. However, the functions of miRNAs in rainbow trout skin pigmentation remain largely unknown. Herein, we overexpressed and silenced miR-495 in vitro and in vivo to investigate its functions. The analysis of spatial and temporal expression patterns suggested that miR-495 is a potential regulator during the process of skin pigmentation. In vitro, mc1r was validated as a direct target for miR-495 by d

Changes in facial signs due to age and their respective weights on the perception of age and skin plumpness among differently aged Korean women.

OBJECTIVE: To determine the respective weights of certain facial signs on the assessment of perceived age and plumpness on Korean women of different ages. MATERIAL AND METHODS: Photographs were taken of the faces of 112 Korean women of different ages (18-80 years) under standardized conditions. These photographs allowed to focus and define 19 facial signs, which were then graded by 15 experts and dermatologists, using standardized scales provided by a reference Skin Aging Atlas. The facial signs were dispatched into 5 clusters, namely Wrinkles/Texture, Ptosis/Sagging, Pigmentation disorders,

Gene therapy induces pigmentation in induced pluripotent stem cell-derived retinal pigment epithelium of OCA1A patients and albino rat eye.

Oculocutaneous albinism type 1A (OCA1A), the most severe and prevalent form of albinism, is caused by recessive mutations in the TYROSINASE gene. We provide proof-of-principle of an effective gene therapy-based strategy for future human application in OCA1A patients. Here, we show that an AAV-based TYROSINASE gene delivery under the control of human BESTROPHIN promoter VMD2 can successfully induce pigmentation in human retinal pigment epithelium (RPE) monolayer tissue, derived from OCA1A patients, via induced pluripotent stem cells. Pigmentation was induced without significantly compromising t

Postinflammatory hyperpigmentation: A comprehensive overview: Epidemiology, pathogenesis, clinical presentation, and noninvasive assessment technique.

Postinflammatory hyperpigmentation (PIH) commonly occurs after various endogenous and exogenous stimuli, especially in dark-skinned individuals. PIH is one of the most common complications of procedures performed using laser and other light sources. The severity of PIH is determined by the inherent skin color, degree and depth of inflammation, degree of dermoepidermal junction disruption, inflammatory conditions, and the stability of melanocytes, leading to epidermal and dermal melanin pigment deposition. The depth of melanin pigment is the key factor to predict prognosis and treatment outcome

TRPM2 couples cell-autonomous type-I interferon signaling to pigmentation homeostasis.

Transient Receptor Potential Melastatin 2 (TRPM2), a Ca2+-permeable cation channel, regulates innate and adaptive immunity and has recently been implicated in vitiligo, an autoimmune pigmentary disorder. However, whether TRPM2 exerts cell-autonomous immunoregulatory functions and how such signaling intersects with pigmentation remain unknown. Here, we reveal an unexpected role for TRPM2 as an intrinsic suppressor of pigmentation through type-I interferon (IFN) signaling in melanocytes. Pharmacological inhibition, genetic silencing, and gain-of-function approaches demonstrate that TRPM2 negativ

The Genetics and Evolution of Human Pigmentation.

Human skin pigmentation is one of the most visible and variable traits among populations and has been shaped primarily by natural selection in response to ultraviolet (UV) radiation. This review synthesizes the current understanding of the genetic and evolutionary mechanisms that underlie pigmentation differences across the globe. The roles of key pigmentation-related genes, such as MC1R, SLC24A5, TYR, and OCA2, are examined in terms of how different versions of these genes have been favored in different UV environments to balance the need for photoprotection and vitamin-D synthesis. Evidence

Addisonian Pigmentation - The Great Mimicker - A Review.

Skin colour usually depends upon melanin, haemoglobin, and carotenoids. Pigmentary disorders indicate an increased amount of melanin, leading to a darker colour of the skin, called hypermelanosis. Addison's disease is a rare endocrinal disorder with severe oral and systemic manifestations. Any form of diffuse skin hyper-pigmentation which is more pronounced on sun-exposed areas mimics Addison's disease and is called Addisonian pigmentation. Pigmentation often precedes other manifestations by months to years and hence can lead to early diagnosis and can prevent life-threatening adrenal crisis.

Topical prevention from high energy visible light-induced pigmentation by 2-mercaptonicotinoyl glycine, but not by ascorbic acid antioxidant: 2 randomized controlled trials.

INTRODUCTION: Hyperpigmentation and pigmentary disorders are major clinical consequences of sun exposure. While UV radiation is a known contributor, visible light (VL), particularly High Energy Visible (HEV) light (400-450 nm), also induces long-lasting pigmentation in melanocompetent individuals (Fitzpatrick Phototype III and above), and can worsen pigmentary disorders. Therefore, photoprotection in this wavelengths range is recommended to prevent worsening of hyperpigmentation issues. Efficient solutions rely on the use of pigments, absorbing and diffusing VL. However, tint and opacity of th

[Albinism and the Range of Fundus Hypopigmentation, Macular Hypoplasia, and Nystagmus].

From the ophthalmological view, albinism is a disorder of reduced pigmentation of the retinal and irdial pigment epithelium and the iris and choroid stroma. The reduced pigmentation is accompanied by morphological changes in the retina and the optic nerve. The functional relationship of these morphological changes is not yet well understood. This review summarises the genetic causes of reduced pigment synthesis and impaired pigment distribution, and discusses the variability of expression of albinism symptoms, in the light of other disorders affecting retinal development.

Biology of melanocytes in mammals.

Melanocytes, which originate from the neuroectoderm, are specialized cells responsible for producing pigments and possessing a dendritic morphology. These cells migrate to the epidermis and follicles, contributing to skin and hair pigmentation during embryonic development. The remarkable self-renewal capacity of melanocytes enables them to effectively restore hair and skin pigmentation. The synthesis of melanin to safeguard the skin against damage caused by ultraviolet radiation, as well as the enigmatic immune function of melanocytes, demonstrate their indispensable contributions to maintaini

Diffuse pigmentation of maxillary attached gingiva: four cases of the cultural practice of gingival tattoo.

BACKGROUND: Gingival pigmentation is a common finding, may be of endogenous or exogenous origin, and can have diagnostic significance. Diffuse gingival pigmentation may be physiologic in nature or can be due to environmental factors, drugs, endocrine disorders, or genetic conditions. We present four cases of diffuse gingival pigmentation due to traditional gingival tattooing and review the literature on this practice. METHODS: Four black females (aged 19 to 56 years) of West African origin (Mauritania and Senegal), representing three different ethnic groups (Fulani, Mandinka, and Soninke) pre

Metabolic syndrome and pigmentation: a review of the connecting link.

There is an ongoing interest regarding the association of metabolic syndrome (MetS) and skin diseases. However, the link between MetS and pigmentation has not been extensively explored. In this review, we undertake a detailed discussion on the existing literature on this topic, the various implicated pathomechanisms and/or molecular pathways, and highlight scope for further scientific exploration. A better understanding of the relationship between MetS and disorders of pigmentation contributes to early diagnosis and prevention, as well as providing clues for developing novel therapeutic strate

mTORC1 activity negatively regulates human hair follicle growth and pigmentation.

Dysregulation of the activity of the mechanistic target of rapamycin complex 1 (mTORC1) is commonly linked to aging, cancer, and genetic disorders such as tuberous sclerosis (TS), a rare neurodevelopmental multisystemic disease characterized by benign tumors, seizures, and intellectual disability. Although patches of white hair on the scalp (poliosis) are considered as early signs of TS, the underlying molecular mechanisms and potential involvement of mTORC1 in hair depigmentation remain unclear. Here, we have used healthy, organ-cultured human scalp hair follicles (HFs) to interrogate the rol

Dual Modulatory Effects of Phytochemicals from Iris ×germanica L. var. florentina Dykes Rhizome Extract on Melanogenesis.

Abnormal skin pigmentation can cause significant esthetic concerns and impact quality of life. As melanin determines pigmentation, melanogenesis is a key target to manage pigmentation disorders. This study investigated the effects of extracts from the rhizome of Iris ×germanica L. var. florentina Dykes (often called Iris florentina L.) on melanogenesis. Active phytochemicals were identified by combining LC-MS-MS metabolic profiling with subsequent bioassay-directed fractionation of chromatographic eluent collected into 96-well plates. Fractions 41-43 increased melanin and contained germanaism

Senescent fibroblasts drive ageing pigmentation: A potential therapeutic target for senile lentigo.

Cutaneous ageing is an important extrinsic process that modifies the pigmentary system. Because cellular senescence is a fundamental ageing mechanism, we examined the role of senescent cells in ageing pigmentation. Methods: Biopsies obtained from senile lentigo and perilesional normal skin were assayed for a marker of cellular senescence, p16INK4A. To determine the secretory phenotypes of senescent fibroblasts, we performed microarray, RNA sequencing and methylation array analyses in senile lentigo and senescent fibroblasts. To further investigate the impact of senescent cells on ageing-relate

Cydnidae pigmentation in an adult during monsoon: a case report and review of the literature.

INTRODUCTION AND IMPORTANCE: Cydnidae pigmentation (CP) is a rare and unique pigmentation disorder caused by contact with Cydnidae bug secretions. Only 30 cases of CP have been reported to date, and this is the first case to be reported from Nepal. The unilateral involvement and resistance to alcohol wiping make this case unique, thus adding to the existing literature on CP. CASE PRESENTATION: A 21-year-old patient, student by occupation, presented with multiple hyperpigmented macules on the hand, observed upon waking during the monsoon season. The patient had no prior history of similar skin
