Monolithic methacrylate packed 96-tips for high throughput bioanalysis.

In the pharmaceutical industry the growing number of samples to be analyzed requires high throughput and fully automated analytical techniques. Commonly used sample-preparation methods are solid-phase extraction (SPE), liquid-liquid extraction (LLE) and protein precipitation. In this paper we will discus a new sample-preparation technique based on SPE for high throughput drug extraction developed and used by our group. This new sample-preparation method is based on monolithic methacrylate polymer as packing sorbent for 96-tip robotic device. Using this device a 96-well plate could be handled i

Apomorphine produced more yawning in Sprague-Dawley rats than in F344 rats: a pharmacological study.

Apomorphine induced yawning in both Sprague-Dawley and F344 rats in the same dose range, but F344 rats emitted only about 1/4 as many yawns as did Sprague-Dawley rats. At higher doses, rats of both strains exhibited stereotypic behavior with a comparable intensity. Pretreatment with either SCH 23390 [R(+)-8-chloro-2,3,4,5-tetrahydro-3-methyl-5-phenyl-1H-3-benzazepine-7-o l] or pindolol increased apomorphine-induced yawning further in Sprague-Dawley rats, but had little effect on the low yawning score produced by apomorphine in F344 rats. The low yawning response to apomorphine in F344 rats is,

Characterization of DOI, a putative 5-HT2 receptor agonist in the rat.

DOI (1-100 micrograms/kg i.v.) induced an increase in mean blood pressure in the anaesthetized rat. Similarly, in the pithed rat, DOI (1-100 micrograms/kg i.v.) induced a dose-dependent increase in mean blood pressure, as did 5-HT. However, in contrast to 5-HT, DOI did not change the heart rate in either intact or pithed rats. In the pithed rat, the dose-pressor response curves to both 5-HT and DOI were unaffected by MDL 72222 (5-HT3 receptor antagonist), spiroxatrine or (+/-)-pindolol (5-HT1A receptor antagonists), idazoxan (alpha 2-adrenoceptor blocking agent) and AR-C 239 (alpha 1-adrenocep

Exercise-induced hyperkalaemia: effects of beta-adrenoceptor blocker vs diuretic.

Four groups of eight normotensive male volunteers performed a 60 min bicycle exercise test before and after 2 weeks of either placebo, hydrochlorothiazide (HCTZ, 25 mg day-1), pindolol (PIND, 10 mg day-1) or both drugs in combination using a double-blind, randomized design. During exercise on placebo serum potassium increased by 0.8 mmol l-1. HCTZ significantly decreased potassium levels at rest and during exercise by 0.2 mmol l-1. PIND did not affect resting potassium levels but potentiated the increase by 0.4 mmol l-1 at the end of exercise, and delayed the return to normal of serum potassiu

5-HT1B agonists induce anorexia at a postsynaptic site.

Ru 24969 and two other putative 5-HT1B agonists 1-(3-chlorophenyl)piperazine (mCPP) and 1-[3-(trifluoromethyl) phenyl]piperazine (TFMPP) and RU 24969 dose-dependently decreased food intake over 4 h (and in the case of RU 24969 also over 24 h) in free-feeding male Sprague-Dawley rats. Decreasing the doses of the agonists below the range eliciting anorexia did not cause hyperphagia. The anorexic effect of RU 24969 over 4 h was antagonised by metergoline, (-)pindolol and (+/-)cyanopindolol, but not by ketanserin, spiperone or haloperidol. Metergoline and (+/-)cyanopindolol also antagonised the an

[Exercise-induced tachyarrhythmic syncopes with sinus bradycardia and normal QT-interval at rest (author's transl)].

Two children with exercise-induced syncopes are described. There was no evidence for congenital or acquired cardiac defects, cardiomyopathy or myocarditis in either of them. Both had sinus bradycardia and a consistently normal QT-interval at rest. In both, exercise induced regularly a multiform ventricular tachycardia, additionally in one on occasion a bidirectional, in the other a supraventricular tachycardia. Beta receptor blocking agents prevented tachyarrhythmias in both. Under treatment with beta receptor blocking agents, one child remained without syncopes, the other died suddenly. These

Enhancement of the anticonvulsant effect of fluoxetine following blockade of 5-HT1A receptors.

Serotonin reuptake inhibitors, such as fluoxetine, have been shown to exert anticonvulsant effects in several animal models of epilepsy. In view of recent studies showing that 5-HT1A receptor antagonists (somatodendritic autoreceptor antagonists) enhance the increase in extracellular 5-hydroxytryptamine (5-HT, serotonin) produced by serotonin reuptake inhibitors, it was of interest to determine if these antagonists also enhance the anticonvulsant effect of fluoxetine in Genetically Epilepsy-Prone Rats (GEPRs). The 5-HT1A receptor antagonists (-)-pindolol and LY 206130 (1-[1-H-indol-4-yloxy]-3-

During the first 48 h of HgCl2-induced acute renal failure (ARF), a single intraperitoneal injection of 100-100 micrograms/kg of the beta-adrenergic blockers pindolol, propranolol and practolol caused a significant rise in urinary sodium excretion and a decrease in plasma renin activity (PRA), renal cortical renin activity (RRA) and urinary catecholamine excretion. A single intraperitoneal injection of 3 mg/kg HgCl2 resulted in ARF characterized by progressive azotemia, a transient increase in PRA, a progressive rise in RRA and urinary catecholamine excretion. [Action of a sodium lactate perfusion on the hemodynamic effects of beta-blockaders].

In 15 dogs who had been given propranolol and 5 humans who had been given Pindolol, an infusion of sodium lactate produced: 1) an increased in blood lactate levels and pH; 2) an increase in the cardiac index; 3) a state in which an infusion of isoproterenol (inactive on its own) could act as it would have done had the betaadrenergic receptors not been blocked. A perfusion of lactic acid which was sufficient to cause the variations in pH and blood lactate levels that are seen with exercise had no action on the heart rate nor on the cardiac index of 10 dogs treated with propranolol and could not

The antihypertensive action of several beta-adrenoreceptor-blocking drugs.

The antihypertensive and pulse-slowing effects of racemic propranolol, oxprenolol, pindolol, practolol and d-propranolol were assessed in 54 hypertensive patients. Drug dosage was selected to be proportionate to beta-adrenoreceptor-blocking potency; d-propranolol dosage equalled approximately that of racemic propranolol. D-propranolol had onlyslight antihypertensive effect; the four other drugs were found to have a considerable and approximately equal antihypertensive effect. The degree of slowing of heart rate varied with the different drugs, being greatest with racemic propanolol. The effect

Lack of pineal beta-adrenergic receptor alterations in suicide victims with major depression.

Noradrenergic function may be altered in depressive illness and thereby contribute to decreased pineal production of melatonin levels, as previously described in some depressed subjects. In the present study, the hypothesis was tested that pineal beta-adrenergic binding would be increased in persons committing suicide with evidence of depressive disorders, reflecting diminished noradrenergic input. Initially, post-mortem human pineal glands were obtained at autopsy. Diagnostic and symptomatic information was then systematically collected from family members using standardized interview techniq

[The mechanism of the chronic supraventricular tachycardia: an intracavitary electrophysiological study (author's transl)].

Chronic Supraventricular Tachycardia (CST) or Chronic Atrial Tachycardia is a relatively rare arrhythmia. Because of very poor number of cases studied with electrophysiologic techniques its mechanism is still debated. Three cases of CST, one of persistent type and two of repetitive type, are reported. His bundle electrogram, high and low right atrial electrograms and atrial stimulation were performed in every case, ventricular stimulation in only one case. The Amiodarone, Pindolol, Verapamil and vagal manoeuvres effects were evaluated in all cases. The site of origin of tachycardia, atrial in

[The effect of therapy with adrenergic beta receptor blockers on myocardial beta adrenergic receptors in children with tetralogy of Fallot].

Seventeen children with tetralogy of Fallot undergoing cardiac surgery were investigated for alteration in myocardial beta-adrenergic receptors (BAR) in dependence on the previous chronic administration with the beta-adrenergic blocker metipranolol. Binding parameters, Bmax (maximal binding capacity) and Kd (dissociation constant), in right ventricular tissue were assessed in direct saturation binding studies using 125I-pindolol. It can be concluded that a) chronic administration of metipranolol to nine patients in usual doses did not change either Bmax or Kd of BAR in the samples of right ven

[Treatment-resistant anxiety disorders: A literature review of drug therapy strategies].

Anxiety disorders are widespread psychiatric conditions with significant social and professional disability, poor quality of life, an increased risk of suicide, and frequent attendance of medical services. Serotonin reuptake inhibitors (SRI) and serotonin and norepinephrine reuptake inhibitors (SNRI) have demonstrated a rather robust efficacy for the treatment of most of anxiety disorders. Nevertheless a substantial number of patients are resistant or still suffer from residual symptoms despite this first line treatment. The objective of our paper is to review relevant studies for the pharmaco

Characterization and localization of beta-adrenergic receptors in control and cryptorchidized rat testis by in vitro autoradiography.

beta-adrenergic receptors were localized and characterized in control and cryptorchidized rats to investigate further their role in testicular function. Slide-mounted cryostat sections were incubated with [125I]cyanopindolol, a specific ligand for beta-adrenergic receptors. The subtypes were characterized by the displacement of [125I]cyanopindolol binding by practolol, which binds preferentially to the beta 1 subtype, and zinterol, which is preferential for beta 2 receptors. Labeling was detected by autoradiography, first by exposing sections to Ultrofilm (LKB) and second by dipping slides in

Renal segmental infarction: a cause of reversible malignant hypertension.

Five cases of accelerated or malignant hypertension which occurred in young adults within a few weeks of renal segmental infarction are reported here. Such infarction appears to give rise to a form of renovascular hypertension, the severity and sudden onset of which distinguishes it from renal fibrodysplasia, which is commonly observed in this age group. In three cases hypertension was corrected by nephrectomy. In two cases several months medical treatment was associated with remission of hypertension without surgery. It is therefore suggested that hypertension due to renal infarction, unlike

The effect of adrenergic blockade on potassium concentrations in different conditions.

A moderate increase in serum potassium concentrations has been observed in several controlled clinical trials with beta-blockers. This increase cannot be explained by the retention of potassium in the organism, and is probably caused by the redistribution of potassium from intracellular to extracellular compartments. beta-adrenergic mechanisms seem to be concerned in the extrarenal handling of the potassium-load in man, presumably by inducing an increased uptake of potassium in muscular cells and liver cells. These beta-adrenergic mechanisms are probably of the beta 2-type. In theory there are
