Antifungal activity of different molecular weight chitosans against planktonic cells and biofilm of Sporothrix brasiliensis.

Sporotrichosis, caused by Sporothrix schenckii complex species, is the most prevalent subcutaneous mycosis in many areas of Latin America. Chitosan has been used as an antifungal agent; however the effects of the molecular weight (MW) of chitosan (i.e. high (HMW), medium (MMW) and low (LMW) molecular weight chitosan) on S. brasiliensis has not been well described, particularly on biofilms. Effects on the planktonic form activity of S. brasiliensis were quantified by broth microdilution, while anti-biofilm activity was quantified by measuring metabolic activity via XTT (2,3-bis(2-methoxy-4-nitr

220D-F2 from Rubus ulmifolius kills Streptococcus pneumoniae planktonic cells and pneumococcal biofilms.

Streptococcus pneumoniae (pneumococcus) forms organized biofilms to persist in the human nasopharynx. This persistence allows the pneumococcus to produce severe diseases such as pneumonia, otitis media, bacteremia and meningitis that kill nearly a million children every year. While bacteremia and meningitis are mediated by planktonic pneumococci, biofilm structures are present during pneumonia and otitis media. The global emergence of S. pneumoniae strains resistant to most commonly prescribed antibiotics warrants further discovery of alternative therapeutics. The present study assessed the an

Activities of fluconazole, caspofungin, anidulafungin, and amphotericin B on planktonic and biofilm Candida species determined by microcalorimetry.

We investigated the activities of fluconazole, caspofungin, anidulafungin, and amphotericin B against Candida species in planktonic form and biofilms using a highly sensitive assay measuring growth-related heat production (microcalorimetry). C. albicans, C. glabrata, C. krusei, and C. parapsilosis were tested, and MICs were determined by the broth microdilution method. The antifungal activities were determined by isothermal microcalorimetry at 37°C in RPMI 1640. For planktonic Candida, heat flow was measured in the presence of antifungal dilutions for 24 h. Candida biofilm was formed on porous

Integrated antimicrobial and nonfouling hydrogels to inhibit the growth of planktonic bacterial cells and keep the surface clean.

A new strategy integrating antimicrobial and nonfouling/biocompatible properties is presented. A mild antimicrobial agent (salicylate) was incorporated into a carboxybetaine ester hydrogel, poly(N,N-dimethyl-N-(ethylcarbonylmethyl)-N-[2-(methacryloyloxy)-ethyl]ammonium salicylate) (pCBMA-1 C2 SA) hydrogel, as its anionic counterion. This new hydrogel provides a sustained release of antimicrobial agents to inhibit the growth of planktonic bacteria and create a nonfouling surface to prevent protein adsorption or bacterial accumulation upon the hydrolysis of carboxybetaine esters into zwitterioni

Bioaccumulation patterns of methyl mercury and essential fatty acids in lacustrine planktonic food webs and fish.

Organisms of the planktonic food web convey essential nutrients as well as contaminants to animals at higher trophic levels. We measured concentrations of methyl mercury (MeHg) and essential fatty acids (EFAs, key nutrients for aquatic food webs) in four size categories of planktonic organisms - seston (10-64 microm), micro-(100-200 microm), meso-(200-500 microm), and macrozooplankton (>500 microm) - as well as total mercury (THg) and EFAs in rainbow trout (Oncorhynchus mykiss) in coastal lakes. We demonstrate that, in all lakes during this summer sampling, MeHg concentrations of planktonic or

Effects of aspirin and other nonsteroidal anti-inflammatory drugs on biofilms and planktonic cells of Candida albicans.

Prostaglandins are now known to be produced by Candida albicans and may play an important role in fungal colonization. Their synthesis in mammalian cells is decreased by inhibitors of the cyclooxygenase isoenzymes required for prostaglandin formation. In the present study, a catheter disk model system was used to investigate the effects of nonsteroidal anti-inflammatory drugs (all cyclooxygenase inhibitors) on biofilm formation by three strains of C. albicans. Seven of nine drugs tested at a concentration of 1 mM inhibited biofilm formation. Aspirin, etodolac, and diclofenac produced the great

Comparative antimicrobial activities of antiseptic mouthrinses against isogenic planktonic and biofilm forms of Actinobacillus actinomycetemcomitans.

BACKGROUND: Bacteria contained in biofilms have been shown to have a decreased susceptibility to antimicrobial agents compared to those in planktonic form. Thus, in vitro biofilm models have been developed for screening oral antimicrobial formulations in an effort to produce findings more predictive of clinical activity. This study compared the antimicrobial activity of three mouthrinse formulations when tested against isogenic strains of Actinobacillus actinomycetemcomitans (Aa), one of which was a clinical isolate which forms tenacious biofilms in vitro and the other of which was a spontaneo

A Sampling Method Considering Body Size for Detecting the Associated Microbes in Plankton Populations: A Case Study, Using the Bloom-Forming Cyanobacteria, Microcystis.

Accurately quantifying associated microbes is essential to understand the interactions between microplankton and their associated microbes. Most DNA-based methods, such as high-throughput sequencing, primarily assess the ratio of target objects to references in microplankton samples. However, simple random sampling (SRS) of individuals may lead to deviations in quantifying these ratios at the population level if these characteristics are associated with the reference content of individuals. This study considered group analysis, which involves detecting k groups with n individuals in each group

Role of DOP on the alkaline phosphatase activity of size fractionated plankton in coastal waters in the NW Mediterranean Sea (Toulon Bay, France).

The particulate material was fractionated into 5 size classes (>90μ, 50-90μ, 6-50μ, 1-6μ, and <1μ). DOP was analysed as easily (DOPh, DOPpa) and less easily hydrolysable compounds (DOPox). Based on Vmax, 94% of the high affinity AP activity was due to <50μ cells and 77% to <1μ cells. 83% of the low affinity activity was due to >90μ cells. The high affinity activities were negatively correlated with DOP for the <50μ classes. These correlations came mostly from DOPox. They were more significant when NO3+NO2 concentrations were high, when DIP concentrations were low and when N/P ratio was >10. At

Antifungal activity of green propolis extract against planktonic cells and biofilms of Trichosporon asahii clinical isolates.

Trichosporon spp. have emerged as important agents of disseminated infections in immunocompromised patients, where they can express multiple virulence factors and exhibit reduced susceptibility to commonly used antifungal drugs. In this context, natural products have attracted increasing interest as alternative therapeutic agents. This study evaluated the antifungal and antibiofilm activity of a standardized green propolis ethanolic extract against ten clinical isolates of Trichosporon asahii, alone and in combination with conventional antifungals.Minimum inhibitory concentrations (MICs) and m

6-Aminoflavone Inhibits Planktonic Growth, Disrupts Biofilm Architecture, and Regulates Quorum Sensing in Klebsiella pneumoniae.

Antibiotic resistance has emerged as a major global health challenge, particularly in biofilm-forming pathogens that exhibit enhanced tolerance to antimicrobial therapies. K. pneumoniae, a multidrug-resistant pathogen, is a leading cause of hospital-acquired infections, including pneumonia, septicemia, and device-associated infections. Its robust biofilm-forming capacity facilitates immune evasion, restricts antibiotic penetration, and contributes to recurrent infections. Therefore, the identification of agents capable of targeting both planktonic bacterial growth and biofilm architecture is o

Transcriptomic insights into host transcriptional manipulation by ssDNA and ssRNA viruses in the marine planktonic diatom Chaetoceros tenuissimus.

The marine diatom Chaetoceros tenuissimus coexists in the coastal ocean with two distinct viruses: a circular single-stranded DNA virus and a single-stranded RNA virus. Both viruses are capable of killing the host in vitro experiments; however, the molecular mechanisms by which host genes respond and are utilized by these viruses for replication and proliferation remain unclear. In this study, RNA-seq analyses were conducted to investigate these mechanisms, and the host transcripts specific to each virus were validated by RT-qPCR. In the early phase of infection, RNA-seq analysis revealed that

Exploring plant compounds as enhancers of ciprofloxacin activity against planktonic and biofilm cells of poultry-related enterobacteriaceae.

This study evaluated the antimicrobial and antibiofilm activities of ciprofloxacin (Cip) in combination with trans-cinnamaldehyde (Tc), geraniol (Ger), and eugenol (Eug) against multidrug-resistant Escherichia coli, Proteus mirabilis, and Klebsiella pneumoniae isolated from broiler feces. Disk diffusion and microdilution assays showed that Tc displayed the highest antibacterial activity in disk diffusion (IZD: 30 mm) and microdilution assays (MIC: 312.5 μg/mL), while Ger and Eug revealed smaller inhibition zones (∼9-12 mm) and higher MIC values (2500 μg/mL). Checkerboard assays indicated that

Effect of bovine lactoferrin and lactoferrin-derived peptides on planktonic cells and abiotic surface biofilms of Salmonella enterica.

Salmonella enterica is a Gram-negative bacterium responsible approximately for 155,000 deaths annually. S. enterica is one of the most important foodborne pathogens, affecting mainly people in developed countries. The human immune system produces antibacterial peptides and proteins like lactoferrin (LF). This work addresses the hypothesis that bovine lactoferrin (bLF) and its derivative peptides bLactoferricin17-30, bD-Lactoferricin17-30, bLactoferrampin265-284, bD-Lactoferrampin265-284 and bLF-chimera have antimicrobial activity on planktonic cells and pre-formed biofilms of S. enterica. Spatiotemporal distribution of the planktonic microbiome and antibiotic resistance genes in a typical urban river contaminated by macrolide antibiotics.

The widespread application of macrolide antibiotics has caused antibiotic resistance pollution, threatening the river ecological health. In this study, five macrolide antibiotics (azithromycin, clarithromycin, roxithromycin, erythromycin, and anhydro erythromycin A) were monitored in the Zao River across three hydrological periods (April, July, and December). Simultaneously, the changes in antibiotic resistance genes (ARGs), mobile genetic elements (MGEs), and planktonic bacterial communities were determined using metagenomic sequencing. A clear pollution gradient was observed for azithromycin

Photodynamic inactivation by hypericin-P123 on azole-resistant isolates of the Trichophyton rubrum complex as planktonic cells and biofilm.

INTRODUCTION: The Trichophyton rubrum complex comprises the majority of dermatophyte fungi (DM) responsible for chronic cases of onychomycosis, which is treated with oral or topical antifungals. However, owing to antifungal resistance, alternative therapies, such as photodynamic therapy (PDT), are needed. This study investigated the frequency of the T. rubrum species complex in onychomycosis cases in the northwestern region of Paraná state, Brazil, and evaluated the efficacy of (PDT) using P123-encapsulated hypericin (Hyp-P123) on clinical isolates of T. rubrum in the planktonic cell and biofi

Antimicrobial Photodynamic Inactivation Affects the Antibiotic Susceptibility of Enterococcus spp. Clinical Isolates in Biofilm and Planktonic Cultures.

Enterococcus faecium and Enterococcus faecalis are opportunistic pathogens that can cause a vast variety of nosocomial infections. Moreover, E. faecium belongs to the group of ESKAPE microbes, which are the main cause of hospital-acquired infections and are especially difficult to treat because of their resistance to many antibiotics. Antimicrobial photodynamic inactivation (aPDI) represents an alternative to overcome multidrug resistance problems. This process requires the simultaneous presence of oxygen, visible light, and photosensitizing compounds. In this work, aPDI was used to resensitiz

Photodynamic inactivation of planktonic Staphylococcus aureus by sodium magnesium chlorophyllin and its effect on the storage quality of lettuce.

Photodynamic inactivation (PDI) is a fast and effective non-heat sterilization technology. This study established an efficient blue light-emitting diode (LED) PDI with the photosensitizer sodium magnesium chlorophyllin (SMC) to eradicate Staphylococcus aureus in food. The antibacterial mechanisms were determined by evaluating DNA integrity, protein changes, morphological alteration, and the potency of PDI to eradicate S. aureus on lettuce was evaluated. Results showed that planktonic S. aureus could not be clearly observed on the medium after treatment with 5.0 μmol/L SMC for 10 min (1.14 J/cm

Mechanisms of action of antimicrobial peptides ToAP2 and NDBP-5.7 against Candida albicans planktonic and biofilm cells.

Candida albicans is a major cause of human infections, ranging from relatively simple to treat skin and mucosal diseases to systemic life-threatening invasive candidiasis. Fungal infections treatment faces three major challenges: the limited number of therapeutic options, the toxicity of the available drugs, and the rise of antifungal resistance. In this study, we demonstrate the antifungal activity and mechanism of action of peptides ToAP2 and NDBP-5.7 against planktonic cells and biofilms of C. albicans. Both peptides were active against C. albicans cells; however, ToAP2 was more active and

Photodynamic antimicrobial chemotherapy action of phenothiazinium dyes in planktonic Candida albicans is increased in sodium dodecyl sulfate.

BACKGROUND: Photodynamic antimicrobial chemotherapy (PACT) is an important therapeutic platform for antimicrobial activity, especially due to the resistant strains, however, the physical-chemical properties of the photosensitizers may affect the final outcome. Recently, this research group showed that the control of aggregation potentiates the PACT action of Methylene Blue. In this study, the researchers investigated the PACT action of other phenothiazinium dyes (PDs), such as Azure A, Azure B (AB), and 1,9-dimethyl methylene blue (DMMB), and the effects of different mediums which modulate PD
