Investigation of cytokinin-deficient phenotypes in Arabidopsis by ectopic expression of orchid DSCKX1.

The plant hormone cytokinin plays a major role in regulating plant growth and development. Here we generated cytokinin-reduction Arabidopsis plants by overexpressing a heterologous cytokinin oxidase gene DSCKX1 from Dendrobium orchid. These transgenic plants exhibited reduced biomass, rapid root growth, decreased ability to form roots in vitro, and reduced response to cytokinin in growing calli and roots. Furthermore, the expression of KNAT1, STM, and CycD3 genes was significantly reduced in the transgenic plants, suggesting that cytokinin may function to control the cell cycles and shoot/root

SERK3A and SERK3B could be S-nitrosylated and enhance the salt resistance in tomato seedlings.

Salinity hinders plant growth and development, resulting in reduced crop yields and diminished crop quality. Nitric oxide (NO) and brassinolides (BR) are plant growth regulators that coordinate a plethora of plant physiological responses. Nonetheless, the way in which these factors interact to affect salt tolerance is not well understood. BR is perceived by the BR receptor BRASSINOSTEROID INSENSITIVE 1 (BRI1) and its co-receptor BRI1-associated kinase 1 (BAK1) to form the receptor complex, eventually inducing BR-regulated responses. To response stress, a wide range of NO-mediated protein modif

Crucial Cell Signaling Compounds Crosstalk and Integrative Multi-Omics Techniques for Salinity Stress Tolerance in Plants.

In the era of rapid climate change, abiotic stresses are the primary cause for yield gap in major agricultural crops. Among them, salinity is considered a calamitous stress due to its global distribution and consequences. Salinity affects plant processes and growth by imposing osmotic stress and destroys ionic and redox signaling. It also affects phytohormone homeostasis, which leads to oxidative stress and eventually imbalances metabolic activity. In this situation, signaling compound crosstalk such as gasotransmitters [nitric oxide (NO), hydrogen sulfide (H2S), hydrogen peroxide (H2O2), calc

Analysis of Phytohormone Signal Transduction in Sophora alopecuroides under Salt Stress.

Salt stress seriously restricts crop yield and quality, leading to an urgent need to understand its effects on plants and the mechanism of plant responses. Although phytohormones are crucial for plant responses to salt stress, the role of phytohormone signal transduction in the salt stress responses of stress-resistant species such as Sophora alopecuroides has not been reported. Herein, we combined transcriptome and metabolome analyses to evaluate expression changes of key genes and metabolites associated with plant hormone signal transduction in S. alopecuroides roots under salt stress for 0

Beyond elongation: The multifaceted roles of gibberellins in symbiosis and root development.

Plants regulate root development in response to fluctuating environmental conditions, including establishing symbiotic relationships with arbuscular mycorrhizal fungi and nitrogen-fixing bacteria under nutrient limitation. These processes are orchestrated by plant hormones, particularly gibberellins, and the repressors of gibberellin signalling, DELLA proteins. Gibberellin and DELLAs serve as critical regulators in symbiotic signalling and root organogenesis, integrating hormonal and environmental cues with cellular patterning to direct plant development. This review explores the current under

In vivo Imaging Enables Understanding of Seamless Plant Defense Responses to Wounding and Pathogen Attack.

Plants are exposed to varied biotic stresses, including sequential or simultaneous attack by insects and pathogens. To overcome these complex stresses, plants must perceive each of the stresses, then integrate and relay the information throughout the plant body and eventually activate local and systemic resistance responses. Previous molecular genetic studies identified jasmonic acid and salicylic acid as key plant hormones of wound and immune responses. These hormones, combined with their antagonistic interaction, play critical roles in the initiation and regulation of defense responses again

Exogenous Hormone Treatments in Maize.

Plant hormones have key functions in plant morphology, physiology, and stress responses. Studies on the biology of hormones and their effect on plant physiology and metabolism are greatly facilitated by the exogenous application of these compounds. In general, methods for exogenous hormone application are easy and fast, and provide useful information about their effects in planta. Although hormone effects have been studied in several plant species, the used methods need to be tailored specifically to each species to get robust data. Maize is an established model for basic and applied research,

The MdSLR1b-MdKNOX15-MdGA20ox2/MdGA2ox7 module regulates dwarfing by mediating the gibberellin pathway in apple.

Dwarf and high-density cultivation is a major trend in the apple industry, yet the regulatory mechanisms remain largely unexplored. In this study, we identified MdKNOX15, a KNOX transcription factor highly expressed in spur-type and dwarfing rootstock varieties, as a critical regulator of plant height. Overexpression of MdKNOX15 caused a gibberellin (GA)-deficient dwarf phenotype, whereas VIGS-mediated knockdown increased endogenous GA content and plant height. Exogenous GA alleviated, and uniconazole exacerbated, the dwarfism. Using DAP-seq and RNA-seq, we identified MdGA2ox7 and MdGA20ox2 as

TaEPFL1 gene controls the development of wheat pistils and stamens by regulating ethylene synthesis.

Proper development of floral organs is essential for reproductive success and grain yield in wheat. However, the molecular mechanisms regulating wheat floral organ development remain largely unknown. In this study, we characterized the role of the wheat TaEPFL1 gene in floral organ development and its association with ethylene signaling. TaEPFL1 was highly expressed in immature spikes of the pistillody mutant HTS-1, particularly during the pistil and stamen specification stages. Its expression was responsive to both exogenous ethylene and the ethylene inhibitor 1-Methylcyclopropene (1-MCP). Ov

Transcriptomic and Metabolomic Investigation on Leaf Necrosis Induced by ZmWus2 Transient Overexpression in Nicotiana benthamiana.

WUSCHEL (WUS) is a crucial transcription factor in regulating plant stem cell development, and its expression can also improve genetic transformation. However, the ectopic expression of WUS always causes pleiotropic effects during genetic transformation, making it important to understand the regulatory mechanisms underlying these phenomena. In our study, we found that the transient expression of the maize WUS ortholog ZmWus2 caused severe leaf necrosis in Nicotiana benthamiana. We performed transcriptomic and non-target metabolomic analyses on tobacco leaves during healthy to wilted states aft

Decreased Vascular Bundle 1 affects mitochondrial and plant development in rice.

BACKGROUND: Mitochondria are vital regulators of plant growth and development, constitute the predominant source of ATP, and participate in multiple anabolic and catabolic metabolic pathways. But the mechanism by which dysfunctional mitochondria affect plant growth remains unknown, and more mitochondria-defective mutants need to be identified. RESULTS: A mitochondria-defective mutant decreased vascular bundle 1 (dvb1) was isolated from rice mutant library mutagenized by EMS (ethylmethane sulfonate), which shows dwarfism, narrow leaves, short branches, few vascular bundles, and low fertility.

In Vitro Propagation of Eleutherine palmifolia (L.) Merr.: Optimization of Surface Sterilization and Effects of BA and NAA on Shoot and Root Induction.

Eleutherine palmifolia (L.) Merr., or Wan Hom Daeng, is a medicinal plant valued for its antioxidant, anti-inflammatory, and antibacterial properties. Its bulbs contain flavonoids, phenolics, and naphthoquinones, but slow natural propagation and low seed viability hinder large-scale cultivation and conservation. This study evaluated the optimal Clorox concentration and exposure time for sterilizing E. palmifolia bulb explants and examined the effects of BA and NAA on in vitro shoot and root development. The results revealed that surface sterilization with 20% Clorox for 20 min yielded the high

The Role of PGPB-Microalgae interaction in Alleviating Salt Stress in Plants.

Plant development and yield are severely hampered by climate change. Plants are very prone to a variety of abiotic stressors during growth, making them susceptible to destruction which can reduce the productivity by 20-60%. These stresses generate reactive oxygen species (ROS), which damage lipids, proteins, and nucleic acids. Microalgae and plant growth-promoting bacteria (PGPB) are remarkably effective at reducing the effects of salt stress and promoting plant growth, thereby increasing agricultural yield, and helping ensure global food security. Through a variety of mechanisms, including th

Modulation of plant growth and metabolism in cadmium-enriched environments.

Cadmium (Cd) is a water soluble metal pollutant that is not essential to plant growth. It has attracted attention from soil scientists and plant nutritionists in recent years because of its toxicity and mobility in the soil-plant continuum. Even low levels of Cd (0.1-1 J.!M) cause adverse effects on plant growth and metabolism. Cadmium is known to trigger the synthesis of reactive oxygen species, hinder utilization, uptake and transport of essential nutrients and water, and modify photosynthetic machinery, thereby resulting in plant tissue death. Although the effects of Cd are dose- as well as p

Negative Roles of Strigolactone-Related SMXL6, 7 and 8 Proteins in Drought Resistance in Arabidopsis.

Previous investigations have shown that the SUPPRESSORS OF MAX2 1-LIKE6, 7 and 8 (SMXL6, 7 and 8) proteins redundantly repress strigolactone (SL) signaling in plant growth and development. Recently, a growing body of evidence indicated that SLs positively regulate plant drought resistance through functional analyses of genes involved in SL biosynthesis and positive regulation of SL signaling. However, the functions of the SL-signaling negative regulators SMXL6, 7 and 8 in drought resistance and the associated mechanisms remain elusive. To reveal the functions of these SMXL proteins, we analyze

Salicylic Acid Mediates Chitosan-Induced Immune Responses and Growth Enhancement in Barley.

Chitosan (CS), derived from the partial deacetylation and hydrolysis of chitin, varies in the degree of deacetylation, molecular weight, and origin, influencing its biological effects, including antifungal properties. In plants, CS triggers immune responses and stimulates biomass growth. Previously, we found that the antifungal activity of CS was strongly dependent on its physicochemical properties. This study revealed that the chitosan batch CS_10 with the strongest antifungal activity also effectively activated plant immune responses and promoted biomass growth. Barley treated with CS_10 exh

A salicylic acid-induced rice (Oryza sativa L.) transcription factor OsWRKY77 is involved in disease resistance of Arabidopsis thaliana.

Plant WRKY transcription factors act as either positive or negative regulators of plant basal disease resistance. To comprehensively characterise the complicated functional network, we isolated OsWRKY77 from rice seedlings treated with salicylic acid. OsWRKY77 is a typical WRKY transcription factor, based on in its protein structure analysis, nuclear localisation of the fused OsWRKY77-GFP protein and gel electrophoretic mobility shift assay binding, which demonstrated that OsWRKY77 was able to bind a W-box. Transgenic Arabidopsis lines overexpressing OsWRKY77 repressed growth of Pseudomonas sy
