Gluten Free Wheat: Are We There?

Gluten proteins, major determinants of the bread-making quality of wheat, are related to several digestive disorders. Advances in plant genetic breeding have allowed the production of wheat lines with very low gliadin content through the use of RNAi and gene editing technologies. In this review, we carried out a comprehensive study of the application of these cutting-edge technologies towards the development of wheat lines devoid of immunogenic gluten, and their genetic, nutritional and clinical characterization. One line, named E82, showed outstanding nutritional properties, with very low imm

The effect of enhanced carotenoid content of transgenic maize grain on fungal colonization and mycotoxin content.

Novel strategies that address vitamin A deficiency have been developed, such as high-carotenoid maize, a biofortified transgenic maize line rich in carotenoids generated by genetic transformation. The South African white maize inbred (M37W), which is devoid of carotenoids, was engineered to accumulate high levels of β-carotene (provitamin A), lutein, and zeaxanthin. Maize seeds contaminated with fumonisins and other mycotoxins pose a serious threat to both humans and livestock. During three consecutive harvests, the fungal incidence and the fumonisin and aflatoxin content of maize seeds grown

CRISPR/dCas9-Mediated BRL3 Activation Enhances Growth and Metabolic Resilience Under Osmotic Stress in Nicotiana tabacum.

Brassinosteroids (BRs) are crucial plant hormones that influence growth and stress adaptation. However, the specific function of the BR receptor BRL3 under osmotic stress remains largely unexplored outside Arabidopsis thaliana. In this study, we used a CRISPR/dCas9-based transcriptional activation (CRISPRa) system to upregulate the Nicotiana tabacum BRASSINOSTEROID INSENSITIVE-LIKE 3 receptor (NtBRL3) and assessed its impact on osmotic stress tolerance. Synthetic activation vectors were constructed using Loop Assembly, featuring dCas9-6TAL-VP128 modules driven by either a constitutive (CaMV35S

Validation of a sensitive DNA walking strategy to characterise unauthorised GMOs using model food matrices mimicking common rice products.

To identify unauthorised GMOs in food and feed matrices, an integrated approach has recently been developed targeting pCAMBIA family vectors, highly present in transgenic plants. Their presence is first assessed by qPCR screening and is subsequently confirmed by characterising the transgene flanking regions, using DNA walking. Here, the DNA walking performance has been thoroughly tested for the first time, regarding the targeted DNA quality and quantity. Several assays, on model food matrices mimicking common rice products, have allowed to determine the limit of detection as well as the potent

Inhibition of snake venoms and phospholipases A(2) by extracts from native and genetically modified Eclipta alba: isolation of active coumestans.

We genetically modified Eclipta alba using Agrobacterium rhizogenes LBA 9402, with the aim of producing secondary metabolites with pharmacological properties against phospholipase A(2) and the myotoxic activities of snake venom. Extracts from in natura aerial parts and roots, both native and genetically modified (in vitro), were prepared and analysed by high-performance liquid chromatography. In natura materials showed the coumestan wedelolactone at higher concentration in the aerial parts, while demethylwedelolactone appeared at higher concentration in roots. Among the modified roots, clone 1

DgMYB4 orchestrates anthocyanin modification and acyl-glucose donor biosynthesis in Delphinium grandiflorum.

To elucidate the seasonal variation in flower coloration of Delphinium grandiflorum cv. F1 Super Platinum Blue, we performed transcriptome analysis and heterologous expression in Nicotiana tabacum, and molecular interaction assays. Flowers in early summer (PB1) displayed deeper blue coloration and more than 8-fold higher total anthocyanin content than those in early spring (PB2). In PB1, cyanodelphin was the major anthocyanin, whereas both cyanodelphin and violdelphin accumulated in PB2. Transcriptome data revealed that DgMYB4, an R2R3-MYB, was significantly upregulated in PB1. Ectopic express

A reciprocal inhibitory module for Pi and iron signaling.

Phosphorous (P) and iron (Fe), two essential nutrients for plant growth and development, are highly abundant elements in the earth's crust but often display low availability to plants. Due to the ability to form insoluble complexes, the antagonistic interaction between P and Fe nutrition in plants has been noticed for decades. However, the underlying molecular mechanism modulating the signaling and homeostasis between them remains obscure. Here, we show that the possible iron sensors HRZs, the iron deficiency-induced E3 ligases, could interact with the central regulator of phosphate (Pi) signa

Mitotic instability in wheat x Thinopyrum ponticum derivatives revealed by chromosome counting, nuclear DNA content and histone H3 phosphorylation pattern.

To evaluate the mitotic stability of Triticum aestivum x Thinopyrum ponticum derivatives (BC(2)F(7) and BC(2)F(5) doubled haploids), chromosome counting by both conventional and immunostaining techniques, and measurement of DNA content were performed. The wheat progenitor line, PF 839197, the wheat recurrent parent CEP 19 and the control Chinese Spring were also investigated. In the hybrid derivatives, chromosome number ranged from 2n=36 to 60, with a predominance of chromosome numbers higher than 2n=42, that was confirmed by determination of nuclear DNA content. Chinese Spring' and PF 839197

The loss of function of a C2H2 zinc finger protein alleviates its inhibitory effect on LsRPL15, causing dense red spots in lettuce.

Variegation plays important biological roles and provides sensory quality for ornamental crops. The mechanisms of variegation have been well studied. However, the molecular mechanism underlying the density of variegation remains largely unknown. In this study, we genetically mapped and cloned a gene negatively controlling the density of red spots on lettuce leaves. The causal gene, Sparse Red Spots (SRS), encodes a C2H2 zinc finger protein. SRS interacted physically with a plastid ribosomal protein L15 (LsRPL15). Knocking down LsRPL15 converted dense spots to sparse spots. SRS controlled the s

Genome-wide identification of lipid transfer proteins in Sorghum bicolor and discovery of flower-specific promoters.

Sorghum bicolor is a drought- and heat-tolerant cereal widely cultivated in arid regions. However, like other important grasses, it is susceptible to ergot disease caused by Claviceps species, which infect ovary tissues and reduce grain yield. To combat this disease, targeted biotechnological strategies are needed. In this study, we conducted a genome-wide identification and classification of lipid transfer proteins (LTPs) in S. bicolor, uncovering 68 putative genes grouped into five families. In silico analyses of gene expression and promoter sequences identified two LTP genes, SbLTP1.4 and S

MdBZR1 regulates ABA response by modulating the expression of MdABI5 in apple.

MdBZR1 directly binds to the promoter of MdABI5 and suppresses its expression to mediate ABA response. The plant hormones brassinosteroids (BRs) and abscisic acid (ABA) antagonistically regulate various aspects of plant growth and development. However, the association between BR and ABA signaling is less clear. Here, we identified MdBZR1 in apple (Malus domestica) and demonstrated that it was activated by BRs and could respond to ABA treatment. Overexpression of MdBZR1 in apple calli and Arabidopsis reduced ABA-hypersensitive phenotypes, suggesting that MdBZR1 negatively regulates ABA signalin

Reduced caterpillar damage can benefit plant bugs in Bt cotton.

Bt cotton was genetically modified to produce insecticidal proteins targeting Lepidopteran pests and is therefore only minimally affected by caterpillar damage. This could lead to reduced levels of inherent, systemically inducible defensive compounds in Bt cotton which might benefit other important cotton herbivores such as plant bugs. We studied the effects of plant defense induction on the performance of the plant bug Lygus hesperus by caging nymphs on different food sources (bolls/squares) of Bt and non-Bt cotton which were either undamaged, damaged by Bt tolerant caterpillars, or treated w

Functional characterization of melatonin-responsive GhMYC5 in cotton drought tolerance.

Drought stress imposes severe constraints on cotton yield and fiber quality. Prior research has shown that exogenous melatonin (MT) application effectively enhances drought tolerance in cotton. Through transcriptome sequencing and weighted gene co-expression network analysis (WGCNA), we identified GhMYC5, a basic helix-loop-helix (bHLH) transcription factor, as a melatonin-responsive hub gene governing drought response. To clarify the functional roles and molecular mechanisms of GhMYC5, virus-induced gene silencing (VIGS) was employed to suppress GhMYC5 expression in cotton, alongside the gene

HmWER-like modulates branch-specific anthocyanin flux and floral pigmentation in Hydrangea macrophylla.

Anthocyanins are key flavonoid metabolites determining floral pigmentation, and their biosynthesis is tightly regulated through coordinated transcriptional and metabolic processes. Although R2R3-MYB transcription factors are well recognized as central regulators of anthocyanin biosynthesis, the roles of WER-like MYB members in anthocyanin metabolism remain largely unknown. In this study, we identified and functionally characterized HmWER-like, a WER/MYB114-like R2R3-MYB transcription factor from Hydrangea macrophylla, and investigated its role in anthocyanin metabolism and floral pigmentation.

MdCo31 interacts with an RNA polymerase II transcription subunit 32 to regulate dwarf growth with short internodes in columnar apple.

The dominant Co locus controls the columnar growth phenotype of apple (Malus × domestica) trees. Candidate gene MdCo31, encoding 2-oxoglutarate-dependent dioxygenase, causes dwarf growth with short internodes in transgenic plants by reducing the abundance of biologically active gibberellin. However, the pathway regulating MdCo31 in the dwarfism of apple trees remains unclear. In this study, expression of MdCo31 was proved to be negatively correlated with internode length in F1 populations created by crossing columnar parents, and with dwarfism in transgenic apple plantlets. Yeast (Saccharomyce

Repatterning of the inflorescence meristem in Gerbera hybrida after wounding.

The Asteraceae plant family is characterized by inflorescences, called flower heads or capitula that may combine hundreds of individual florets into a single flower-like structure. The florets are arranged in a regular phyllotactic pattern with Fibonacci numbers of left- and right-winding spirals. Such a pattern may be disrupted due to physical constraints or by wounding occurring during the early meristem development. Recovery from wounding re-establishes patterning although the mechanisms have remained elusive. In this study, we applied Gerbera hybrida as a model system and established metho

Overexpression of GmMYB14 improves high-density yield and drought tolerance of soybean through regulating plant architecture mediated by the brassinosteroid pathway.

MYB transcription factors (TFs) have been reported to regulate the biosynthesis of secondary metabolites, as well as to mediate plant adaption to abiotic stresses, including drought. However, the roles of MYB TFs in regulating plant architecture and yield potential remain poorly understood. Here, we studied the roles of the dehydration-inducible GmMYB14 gene in regulating plant architecture, high-density yield and drought tolerance through the brassinosteroid (BR) pathway in soybean. GmMYB14 was shown to localize to nucleus and has a transactivation activity. Stable GmMYB14-overexpressing (GmM

Comparative proteomics combined with analyses of transgenic plants reveal ZmREM1.3 mediates maize resistance to southern corn rust.

Southern corn rust (SCR), which is a destructive disease caused by Puccinia polysora Underw. (P. polysora), commonly occurs in warm-temperate and tropical regions. To identify candidate proteins related to SCR resistance and characterize the molecular mechanisms underlying the maize-P. polysora interaction, a comparative proteomic analysis of susceptible and resistant maize lines was performed. Statistical analyses revealed 1489 differentially abundant proteins in the resistant line, as well as 1035 differentially abundant proteins in the susceptible line. Manipulating phloem iron transport using the phloem-specific promoter OsSUT1 enhances grain iron accumulation in rice.

Human diets are commonly deficient in iron. Hence, increasing the iron content in rice, a major staple crop globally, will help overcome this iron deficiency. The phloem is an important route for iron distribution. However, little is known about the effects of manipulating iron transport in the phloem on iron content. The phloem-specific OsSUT1 promoter is used to drive the expression of iron transport genes OsYSL9 and OsIRT1. The phloem exudate iron content, iron harvest index and remobilization contribution rate are enhanced in pOsSUT1::OsYSL9 (SY) and pOsSUT1::OsIRT1 (SI) transgenic lines.

Detailed analysis of the expression of an alpha-gliadin promoter and the deposition of alpha-gliadin protein during wheat grain development.

BACKGROUND AND AIMS: Alpha-gliadin proteins are important for the industrial quality of bread wheat flour, but they also contain many epitopes that can trigger celiac (coeliac) disease (CD). The B-genome-encoded alpha-gliadin genes, however, contain very few epitopes. Controlling alpha-gliadin gene expression in wheat requires knowledge on the processes of expression and deposition of alpha-gliadin protein during wheat grain development. METHODS: A 592-bp fragment of the promotor of a B-genome-encoded alpha-gliadin gene driving the expression of a GUS reporter gene was transformed into wheat.
