Response to infusions of polyelectrolyte fractionated human factor VIII concentrate in human haemophilia A and von Willebrand's disease.

Factor VIII was purified from cryoprecipitate by ion exchange chromatography on solid phase polyelectrolyte E-5 (PE-E5). The product was highly purified (3.5 u VIII:C/mg protein) compared to conventional concentrate (0.3 u VIII:C/mg protein) with low fibrinogen, low isoagglutinin titre, and a ratio of factor VIII coagulant activity (VIII:C) to factor VII related antigen (VIIIR:Ag) of 16:1. Trial infusions of this material (PE VIII) were given to three patients with severe haemophilia A and one patient with homozygous von Willebrand's disease. These patients also each received separate infusion

ROS-responsive chitosan/hyaluronan polyelectrolyte nanogels for targeted chemo-ferroptosis therapy against breast cancer.

Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has emerged as a promising strategy for combination with chemotherapy in cancer treatment. However, the rational design of delivery systems capable of simultaneously inducing ferroptosis, enhancing chemotherapy efficacy, and reducing systemic toxicity remains a substantial challenge. Herein, we developed a reactive oxygen species (ROS)-responsive, ionically crosslinked chitosan/hyaluronan polyelectrolyte nanogel for targeted chemo-ferroptosis combination therapy. In this system, hyaluronic acid (HA) was f

Novel polyelectrolyte functionalized mesoporous silica for sustainable and ultraselective adsorption of Re(VII) and 99Tc(VII) remediation.

The selective recovery of perrhenate (ReO4-) and radio-toxic pertechnetate (99TcO4-) from complex and hypersaline matrices remain challenging task for nuclear waste management and strategic metal valorization. High-performance novel anionic sorbent (QA-APMA-Si), is engineered by anchoring high-density cationic poly-additives, covalently bonded onto a mesoporous silica via surface-initiated redox polymerization. The resulting material features with superior capacity and selectivity toward Re and 99Tc. Adsorption experiments show high sorption efficiency. The kinetic data fitted well to the PFOR

A highly sensitive and drift-free iontronic sensor enabled by a polarity-balanced polyelectrolyte for surgical force sensing.

Iontronic pressure sensors featuring high sensitivity are promising across different emerging fields. In applications requiring high-fidelity sensing such as surgeries, reliable pressure monitoring necessitates sensitivity and stability at the same time to ensure safety. However, existing iontronic sensors often face a trade-off between them-soft ionic materials with high conductivity often present pronounced viscoelasticity, resulting in a low sensitivity-to-drift-rate (S/D) ratio. Here, we report a polyelectrolyte-based iontronic pressure sensor that achieves a nearly one order-of-magnitude

Bio-inspired polyphenol-polyelectrolyte WRAP system for geometrically controlled AAV delivery in diverse anatomical sites.

Systemic delivery of therapeutics frequently requires elevated dosing to achieve effective local concentrations, often resulting in unintended toxicity and insufficient drug levels at the target site. To address this inherent limitation, localized drug and gene delivery strategies have been developed to deposit therapeutic payloads directly within diseased microenvironments. Herein, we report the development of a WRAP (Writable, Retentive, Adhesive, Permeable) drug delivery scaffold system, a biomaterial-mediated polyelectrolyte complex (PEC) film formed through spontaneous ionic complexation

Oral administration of polyelectrolyte modified curcumin-loaded egg protein nanoparticles for suppression of inflammation in colitis mice.

Ulcerative colitis (UC) is a chronic inflammatory disease affecting the large intestine, resulting in inflammation to the mucosa and submucosa of the colon. Overproduction of reactive oxygen species (ROS) plays an important role in contributing to the exacerbation of inflammation and strategies aimed at reducing ROS in the inflamed colon offer potential therapeutic benefits for managing UC. Curcumin possesses antioxidant and anti-inflammatory effects; however, it faces many challenges, such as rapid metabolism and low solubility. In this study, we developed a curcumin-loaded egg protein nanopa

Large-scale super-resolution optoacoustic imaging facilitated by FeNP/ICG-loaded coreless polyelectrolyte microcapsules.

Rationale: Localization optoacoustic tomography (LOT) enhances imaging of deep-tissue microvasculature by leveraging flowing contrast particles. However, achieving high-resolution, large-scale imaging requires contrast agents with strong per-particle signal, good biocompatibility, and prolonged circulation time. This study introduces coreless polyelectrolyte microcapsules (MCs) encapsulating indocyanine green (ICG) and iron oxide nanoparticles (FeNP) to overcome current limitations in LOT imaging. Methods: MCs were engineered using a layer-by-layer technique by depositing polyelectrolytes, FeN

Three-dimensional conductive conjugated polyelectrolyte gels facilitate interfacial electron transfer for improved biophotovoltaic performance.

Living biophotovoltaics represent a potentially green and sustainable method to generate bio-electricity by harnessing photosynthetic microorganisms. However, barriers to electron transfer across the abiotic/biotic interface hinder solar-to-electricity conversion efficiencies. Herein, we report on a facile method to improve interfacial electron transfer by combining the photosynthetic cyanobacterium Synechococcus elongatus PCC 7942 (S. elongatus) with a conjugated polyelectrolyte (CPE) atop indium tin oxide (ITO) charge-collecting electrodes. By self-assembly of the CPE with S. elongatus, soft

Polyelectrolyte-coated liposomes microfluidically assembled in one-step for enhancing cell endocytosis and in-vivo immune responses.

To enhance the cellular uptake of liposomes, we prepared conventional liposomes with targeting molecules and surface-charged liposomes and evaluated their potential as nano-carriers and vaccine adjuvants by comparing their endocytosis efficiencies using immune cells. Surface-charged liposomes were synthesized via a one-step microfluidic method, which provided a novel, simple, fast, and highly reproducible method for preparing liposomes. Flow cytometry revealed that cationic polyelectrolyte-coated liposomes exhibited higher endocytosis efficiencies (of up to a factor of 100) in A774A.1 cells an

Co-encapsulation and release of apigenin and ascorbic acid in polyelectrolyte multilayer capsules for targeted polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS), a prevalent endocrine disorder in women of reproductive age, is linked to hormonal imbalances and oxidative stress. Our study investigates the regenerative potential of apigenin (AP, hydrophobic) and ascorbic acid (AC, hydrophilic) encapsulated within poly (allylamine hydrochloride) and dextran sulfate (PAH/DS) hollow microcapsules for PCOS. These microcapsules, constructed using a layer-by-layer (LbL) assembly, are found to be 4 ± 0.5 μm in size. Our research successfully demonstrates the co-encapsulation of AP and AC in a single PAH/DS system with high encap

An Enteric-Coated Polyelectrolyte Nanocomplex Delivers Insulin in Rat Intestinal Instillations when Combined with a Permeation Enhancer.

The use of nanocarriers is being researched to achieve oral peptide delivery. Insulin-associated anionic polyelectrolyte nanoparticle complexes (PECs) were formed that comprised hyaluronic acid and chitosan in an optimum mass mixing ratio of 5:1 (MR 5), followed by coating with a pH-dependent polymer. Free insulin was separated from PECs by size exclusion chromatography and then measured by HPLC. The association efficiency of insulin in PECs was >95% and the loading was ~83 µg/mg particles. Dynamic light scattering and nanoparticle tracking analysis of PECs revealed low polydispersity, a negat

Mechanistic basis for unexpected bioavailability enhancement of polyelectrolyte complexes incorporating BCS class III drugs and carrageenans.

The objective of this study was to investigate the potential of λ-carrageenan to work as an absorption modifying excipient in combination with formulations of BCS class 3 substances. Trospium chloride was used as a model BCS class 3 substance. Polyelectrolyte complexes of trospium and λ-carrageenan were produced by layer-by-layer complexation. A λ-carrageenan-containing formulation was administered either in capsules size 9 to rats by gavage or directly into ligated intestinal loops of rats. Exceptionally strong variations were observed in the plasma concentrations of the rats that received λ-

Hybrid 3D Printing of Interfacial Polyelectrolyte Complex Formed between Hyaluronic Acid and Poly-L-lysine Hydrogels.

Hyaluronic acid (HA), a principal component of the extracellular matrix (ECM), exhibits excellent biocompatibility, high water-retention capacity, and inherent biodegradability. α-Poly-L-lysine (PLL), a cationic polypeptide with favorable biocompatibility, is widely employed for surface modification to promote cell adhesion and proliferation. Electrostatic interactions between oppositely charged polyelectrolytes enable the formation of interfacial polyelectrolyte complexes (IPC) in the absence of chemical cross-linking agents. In this study, IPC hydrogels were fabricated from HA and PLL and pr

Designing cryogels through cryostructuring of β-chitin nanofibers to reinforce polyelectrolyte complexes for non-compressible hemorrhage control.

Non-compressible hemorrhage remains a major clinical challenge, requiring hemostatic materials with rapid blood absorption and sufficient mechanical robustness. Conventional carboxymethyl chitosan (CMCS)/sodium alginate (SA) polyelectrolyte complex (PEC) sponges exhibit high swelling capacity but limited uptake kinetics and mechanical strength. Here, β-chitin nanofibers (β-ChNFs) were incorporated into the CMCS/SA system to establish a freeze-thaw reinforcement strategy depended on β-ChNFs. Repeated freeze-thaw cycles had little effect on the storage modulus (G') of CMCS/SA alone, whereas G' p
