Smart Polymeric Biomaterials for Clinical Use.

As biomaterials transition from static, inert objects into dynamic scaffolds that augment the healing process, the ability to design large-scale changes in architecture in response to environmental changes is increasingly valuable. To address this need, stimuli-responsive, or "smart," biomaterials are employed in clinical applications more frequently. These materials are capable of undergoing large and abrupt changes in shape in response to small changes in the environment, such as temperature, pH, mechanical forces, light, magnetic fields, or enzymes. Smart polymeric biomaterials include shap

Polymerizing laminins: Assembly, functions and disorders.

Laminins initiate basement membrane (BM) assembly by adhering to cells, establishing cytoskeletal links through integrin and dystroglycan receptors, and by polymerizing to form a sheet-like provisional matrix that recruits other BM components to complete assembly. Laminin polymerization is mediated by the LN domain tips of the three short arms that bind together to form "polymer nodes" connecting adjacent laminins. The first step of this assembly is the low affinity binding of a β-LN domain to a γ-LN domain, followed by a higher affinity binding of an α-LN domain to the β-γ complex. Negative s

Polymeric micelle as multifunctional pharmaceutical carriers.

A number of formulations have been developed and reported in literature as a carrier for drug, gene, and diagnostic agents. Amphiphilic block copolymers have achieved increasing attention due to their high stability both in vitro and in vivo and their good biocompatibility. In comparison to other long-circulating nanocarriers, micelles possess a number of advantages such as tissue penetrability, reduced toxicity, and controlled drug release. The characteristic features of micelle as carriers, such as particle size, shape, drug loading, cellular internalization, stability, and release kinetics

Encapsulation of dexamethasone into biodegradable polymeric nanoparticles.

The present paper concerns both the optimization of dexamethasone (DXM) entrapment and its release from biodegradable poly(D,L-lactide-co-glycolide) (PLGA) nanoparticles prepared by the solvent evaporation process. Since the addition of DXM induced the formation of drug crystals beside the nanoparticle suspension, the influence of several parameters on DXM encapsulation was investigated such as the type of organic solvent and polymer, the DXM initial mass, the evaporation rate of the solvent, the continuous phase saturation and the incorporation of a lipid in the polymer. Nanoparticle size and

Polymeric micelles as carriers of diagnostic agents.

This review deals with diagnostic applications of polymeric micelles composed of amphiphilic block-copolymers. In aqueous solutions these polymers spontaneously form particles with diameter 20-100 nm. A variety of diagnostic moieties can be incorporated covalently or non-covalently into the particulates with high loads. Resulting particles can be used as particulate agents for diagnostic imaging using three major imaging modalities: gamma-scintigraphy, magnetic resonance imaging and computed tomography. The use of polyethyleneoxide-diacyllipid micelles loaded with chelated (111)In/Gd(3+) as we

Release of drugs from polymeric hydrogels.

Hydrogels were prepared from 2-hydroxyethyl methacrylate and ethylene glycol dimethacrylate as cross-linking agent. Two different polymerization techniques were followed, namely bulk polymerization initiated by dibenzoyl peroxide and solution polymerization, in water, using a redox initiator system. Differences in the water desorption from swollen samples between the two types of the above products were recorded and attributed to the fact that those hydrogels were obtained in glassy or rubbery state, according to the polymerization technique. Also, the delivery of salicylic acid from samples o

Inflammatory activity of polymeric photoproducts of chlorpromazine.

To confirm the inflammatory activity of polymeric photoproducts (CPZ-polymers) of chlorpromazine (CPZ), which were obtained by gel filtration of a UV-pre-irradiated CPZ aqueous solution, the histamine release from rat peritoneal exudate cells was studied and the paw-inflammation in mice induced by these CPZ-polymers was examined. CPZ-polymers induced a dose-dependent histamine release at concentrations of 1, 3 and 10 mg/ml. This effect was approximately one-tenth of that of compound 48/80. Furthermore, CPZ-polymers markedly induced lasting paw-edema in a dose-dependent manner, the swellings re

Polymeric adhesion to dentin: contrasting substrates.

Clinical research has shown that resin dentin adhesive materials often fail prematurely despite encouraging laboratory-derived data. Current adhesives are reported to react with dentin surfaces by chemical and/or micro-mechanical mechanisms. The ionic bonding to calcium and the covalent coupling to collagen are representative chemical strategies employed today. Recent findings suggest that the mechanical interlocking of resins into open dentinal tubules may play an even more important role with present adhesive systems. Dentin surfaces, such as cervical abrasions, undergo changes in the oral c

Ouzo polymerization: A bottom-up green synthesis of polymer nanoparticles by free-radical polymerization of monomers spontaneously nucleated by the Ouzo effect; Application to molecular imprinting.

HYPOTHESIS: Top-down fabrication of polymer nanoparticles from preformed polymers by spontaneous colloid formation under influence of the Ouzo effect is a widely applied concept whereas bottom-up free-radical polymerization of monomers nucleated under the Ouzo regime have found limited application after a seminal report almost half a century ago. We hypothesized that the approach would be of value today as a versatile method for green synthesis of polymer nanoparticles, including molecularly imprinted ones. EXPERIMENTS: Ternary mixtures of pentaerythritol triacrylate, ethanol, and water were

pH-Responsive polymeric vesicles prepared via polymerization-induced self-assembly for controlled 5-fluorouracil delivery.

Stimuli-responsive polymeric vesicles are promising nanocarriers for the controlled release of chemotherapeutic agents. Although pH-responsive poly(2-(diisopropylamino)ethyl methacrylate) (PDPA) vesicles have been widely studied for the delivery of drugs such as doxorubicin hydrochloride, their use in 5-fluorouracil (5-FU) delivery has not been reported. Here, we demonstrate for the first time the application of PDPA-based vesicles for pH-triggered release of 5-FU. The vesicles were prepared via polymerization-induced self-assembly (PISA) using reversible addition-fragmentation chain transfer

Programming Hydrogel Mechanics via Sequence-Controlled Polymerization Using Peptide Self-Assembly.

Hydrogels often have poor mechanical properties due to their high water content and low polymer concentration, which limits their utility in applications that require them to withstand applied forces. Inspired by natural biopolymers such as collagen and actin, which form highly extended fibrillar networks that stiffen biological tissues, we developed a modular strategy that utilizes self-assembling peptides to direct the formation of covalently polymerized diacetylene networks in hydrogels. By systematically tuning peptide sequences, we precisely controlled the supramolecular organization and
