Phylogenetic and molecular evidence for allochronic speciation in gall-forming aphids (Pemphigus).

Sympatric populations can diverge when variation in phenology or life cycle causes them to mate at distinctly different times. We report patterns consistent with this process (allochronic speciation) in North American gall-forming aphids, in the absence of a host or habitat shift. Pemphigus populi-transversus Riley and P. obesinymphae Aoki form a monophyletic clade within the North American Pemphigus group. They are sympatric on the eastern cottonwood, Populus deltoides (Salicaceae), but have distinctly different life cycles, with sexual stages offset by approximately six months. Field evidenc

Three-dimensional model construction and wind simulation of different tree species in farmland shelter.

Protective forests are the ecological barriers of oases in arid sand areas and can effectively prevent and control wind and sand hazards. The structural characteristics of individual trees, as the basic unit of protective forests, are the key factors affecting the protective benefits. With the typical leafless tree species of Ulan Buh Desert oasis, i.e., Populus alba var. pyramidalis, Populus nigra var. thevestina, and Populus popularis, as the research objects, and by using the ground-based LiDAR and through computational fluid dynamics (CFD), we fully explored the structural characteristics

A wider spectrum of avoidance and tolerance mechanisms explained ozone sensitivity of two white poplar ploidy levels.

BACKGROUND AND AIMS: Polyploidization can improve plant mass yield for bioenergy support, yet few studies have investigated ozone (O3) sensitivity linked to internal regulatory mechanisms at different ploidy levels. METHODS: Diploid and triploid Populus tomentosa plants were exposed to ambient and ambient plus 60 ppb [O3]. We explored their differences in sensitivity (leaf morphological, physiological and biochemical traits, and plant mass) as well as mechanisms of avoidance (stomatal conductance, xanthophyll cycle, thermal dissipation) and tolerance (ROS scavenging system) in response to O3

Remote sensing of cytotype and its consequences for canopy damage in quaking aspen.

Mapping geographic mosaics of genetic variation and their consequences via genotype x environment interactions at large extents and high resolution has been limited by the scalability of DNA sequencing. Here, we address this challenge for cytotype (chromosome copy number) variation in quaking aspen, a drought-impacted foundation tree species. We integrate airborne imaging spectroscopy data with ground-based DNA sequencing data and canopy damage data in 391 km2 of southwestern Colorado. We show that (1) aspen cover and cytotype can be remotely sensed at 1 m spatial resolution, (2) the geographi

Transcriptome comparison of different ploidy reveals the mechanism of photosynthetic efficiency superiority of triploid poplar.

Triploid poplars have obvious growth advantages, especially in leaf development and photosynthetic characteristics, but the molecular mechanism has not been revealed yet. In order to better understand the regulation mechanisms of leaf and chlorophyll development in the triploid poplars, we combined the leaf phenotypic data with the transcriptomic data of the 5th, 10th, and 25th leaves from triploid and diploid poplars, using weighted gene co-expression network analysis (WGCNA), and revealed that PpnGRF5-1 had a strong correlation with leaf development and net photosynthetic rate (Pn). Poplar root exudates contain compounds that induce the expression of MiSSP7 in Laccaria bicolor.

Communication between organisms is crucial for their survival, especially for sessile organisms such as plants that depend upon interactions with mutualistic organisms to maximize their nutrient acquisition. This communication can take the form of the exchange of volatile compounds, metabolites or effectors - small protein signals secreted from the colonizing cell that change the biology of the host cell.  We recently characterized the first mutualistic effector protein from an ectomycorrhizal fungus, a small secreted protein named MiSSP7 encoded by Laccaria bicolor.  Ectomycorrhizal fungi are

Isolation and characterization of hemicelluloses extracted by hydrothermal pretreatment.

The dewaxed sample from Triploid of Populus tomentosa Carr. was extracted by using organic alkaline solvent (Dimethylformamide, DMF) via hydrothermal pretreatment. Neutral sugar compositions and molecular weight analysis demonstrated that the hemicellulosic fractions with a higher Uro/Xyl ratio, namely the more branched hemicelluloses, had higher molecular weights. Interestingly, these results were different from the previous report, in which the ratio of Uro/Xyl in the water-soluble hemicellulosic fraction was more than that of the alkali-soluble hemicellulosic fraction. Spectroscopy (FTIR, (

Mass balance studies of volatile chlorinated hydrocarbon phytoremediation.

In field-scale mass balance studies of poplar remediation of carbon tetrachloride (CT), more than 95% of the mass of CT was degraded with all of the CT chlorine accountable as chloride ion accumulation in the soil. Atmospheric loss of CT through leaf transpiration and trunk diffusion was insignificant. These findings are consistent with previously reported uptake and degradation of trichloroethylene (TCE) by poplar. Poplar phytoremediation of CT and TCE results in little decrease in aqueous concentration, since water is taken up at about the same rate as the chlorinated compounds. From this re

Secreted Effector Proteins of Poplar Leaf Spot and Stem Canker Pathogen Sphaerulina musiva Manipulate Plant Immunity and Contribute to Virulence in Diverse Ways.

Fungal effectors play critical roles in manipulating plant immune responses and promoting colonization. Sphaerulina musiva is a heterothallic ascomycete fungus that causes Septoria leaf spot and stem canker disease in poplar (Populus spp.) plantations. This disease can result in premature defoliation, branch and stem breakage, increased mortality, and plantation failure. However, little is known about the interaction between S. musiva and poplar. Previous work predicted 142 candidate secreted effector proteins in S. musiva (SmCSEPs), 19 of which were selected for further functional characteriz

Polyploidy and growth-defense tradeoffs in natural populations of western quaking Aspen.

Polyploidy, the expression of more than two sets of chromosomes, is common in plants, and is thought to influence plant trait expression and drive plant species evolution. The degree to which polyploidy influences interactions among physiological processes such as growth and defense in natural populations through its effect on phenotypic variability is poorly understood. We link broad plant genotypic features (including polyploidy) to phenotypic expression of growth and chemical defense in natural populations of quaking aspen (Populus tremuloides) to examine patterns in resource allocation tha

First report of the poplar leaf miner, Phyllonorycter populifoliella (Treitschke) (Lepidoptera: Gracillariidae) from India.

Here we report about the discovery of the poplar leaf miner, Phyllonorycter populifoliella (Treitschke) in India. The mines of this micromoth were found in noticeable density on the leaves of poplar, Populus sp. (Salicaceae) in the northern mountainous region Ladakh in 2017-2018. We provide short morphological diagnosis, describe bionomics and analyze molecular data of Ph. populifoliella from India comparing sequences with those from other countries in Eurasia where the species is known as native. We also illustrate male and female genitalia, an adult of the moth, the leaf mines and the infest

Gasification treatment of poplar biomass produced in a contaminated area restored using plant assisted bioremediation.

Remediation of polluted soils using phytoremediation techniques is an effective strategy. However, the use of the biomass from these soils for energy purposes may raise efficiency and pollution emission problems and there is currently little research on this issue. In this work, the main results of a fluidized-bed gasification treatment conducted on poplar biomass pruning residues from a multi-contaminated area are presented. The samples were collected from an experimental site in which a plant-assisted bioremediation (PABR) technology has been applied since 2013 to reduce the soil heavy metal

Comparative Proteomic Analysis of Plant-Pathogen Interactions in Resistant and Susceptible Poplar Ecotypes Infected with Botryosphaeria dothidea.

Poplar are important forestry species in China, but the Botryosphaeria dothidea pathogen causes serious economic losses worldwide. To identify candidate B. dothidea resistance proteins and explore the molecular mechanisms involved in poplar-pathogen interactions, proteomic responses of stem samples from resistant and susceptible poplar ecotypes to B. dothidea were investigated using nanoflow liquid chromatography-tandem mass spectrometry with label-free quantitative analysis. We identified 588 proteins, divided into 21 biological process categories including 48 oxidoreductases, 72 hydrolytic e

Genetics-based interactions of foundation species affect community diversity, stability and network structure.

We examined the hypothesis that genetics-based interactions between strongly interacting foundation species, the tree Populus angustifolia and the aphid Pemphigus betae, affect arthropod community diversity, stability and species interaction networks of which little is known. In a 2-year experimental manipulation of the tree and its aphid herbivore four major findings emerged: (i) the interactions of these two species determined the composition of an arthropod community of 139 species; (ii) both tree genotype and aphid presence significantly predicted community diversity; (iii) the presence of

Tangential single-cell spatial transcriptomics resolves early ray lineage trajectory in poplar.

Vascular cambium drives secondary growth in woody plants, yet its closely related stem-cell populations-fusiform and ray initials-are difficult to distinguish, limiting resolution of early ray fate specification. In Populus tremula × Populus alba clone 717, we integrated a 2-yr biweekly field survey of xylem ray anatomy with time-series stem transcriptomes and single-cell-resolution tangential spatial transcriptomics to resolve early ray-lineage states in situ. Across 2 growing seasons, ray cell number and size/area traits showed reproducible dynamics and were most strongly associated with day

Unravelling the novel sex determination genotype with 'ZY' and a distinctive 2.15-2.95 Mb inversion among poplar species through haplotype-resolved genome assembly and comparative genomics analysis.

Populus tomentosa, an indigenous tree species, is widely distributed and cultivated over 1,000,000 km2 in China, contributing significantly to forest production, ecological conservation and urban-rural greening. Although a reference genome is available for P. tomentosa, the intricate interspecific hybrid origins, chromosome structural variations (SVs) and sex determination mechanisms remain confusion and unclear due to its broad and even overlapping geographical distribution, extensive morphological variations and cross infiltration among white poplar species. We conducted a haplotype-resolved

Genome-Wide Identification of WRKY Family Genes and the Expression Profiles in Response to Nitrogen Deficiency in Poplar.

The fast-growing arbor poplar is widely distributed across the world and is susceptible to nitrogen availability. The WRKY transcription factor is an important regulatory node of stress tolerance as well as nutrient utilization. However, the potential response mechanism of WRKY genes toward nitrogen is poorly understood. Therefore, the identification of WRKY genes on the Populus trichocarpa genome was performed, and 98 PtWRKYs (i.e., PtWRKY1 to PtWRKY98) were identified. Phylogenetic analysis and the promoter cis-acting element detection revealed that PtWRKYs have multiple functions, including

Techno-Economic Analysis of Producing Glacial Acetic Acid from Poplar Biomass via Bioconversion.

Most of the current commercial production of glacial acetic acid (GAA) is by petrochemical routes, primarily methanol carbonylation. GAA is an intermediate in the production of plastics, textiles, dyes, and paints. GAA production from biomass might be an economically viable and sustainable alternative to petroleum-derived routes. Separation of acetic acid from water is a major expense and requires considerable energy. This study evaluates and compares the technical and economic feasibility of GAA production via bioconversion using either ethyl acetate or alamine in diisobutylkerosene (DIBK) as

Separating ontogenetic and environmental determination of resistance to herbivory in cottonwood.

We used narrowleaf cottonwood, Populus angustifolia, and the gall-forming aphid, Pemphigus betae, to determine the extent to which ontogenetic variation in resistance to herbivory is due to endogenous, stable genetic influences. In a three-year common garden trial using ramets propagated from the top, middle, and bottom of mature trees, we found that the resistance of trees to aphids was significantly higher in top vs. bottom source ramets, supporting the hypothesis of a stable, genetically programmed component to aphid resistance. The magnitude of ontogenetically based variation in resistance
