[Lateral thalamic infarction. 22 cases].

Isolated lateral thalamic infarcts (LThl) are rare. They often produce lacunar syndromes, and their main cause is thought to be an hypertensive arteriolopathy. To verify these data, we reviewed 639 cerebral infarcts demonstrated by CT and/or MRI and included in a hospital stroke registry over a 4-year period. We identified 22 cases (3.5 p. 100) of isolated LThl (right LThl: 15; left LThl: 7). Nineteen had MRI study. There were 13 men and 9 women of mean age 65 years. None had the complete Dejerine-Roussy syndrome: sensory disturbances 21 cases, hemiparesis 7, hemiataxia 5, involuntary movement

[Cerebral hemosiderosis related to hereditary ceruloplasmin deficiency. Clinical familial case study].

A 59-year-old patient progressively developed dementia, hallucinations and facial dyskinesia. Brain T and T2-weighted MRI images showed low signal intensity on basal ganglia specially striatum, posterior thalamic and dentate nuclei. He had no evidence of ceruloplasmin and a high level of ferritin in the serum. Liver biopsy confirmed accumulation of iron in the cytoplasm of many hepatocytes. Similar clinical and biological signs were also observed in two brothers. All the three siblings were homozygous for a hereditary ceruloplasmin deficiency. This new clinico-pathological entity, first descri

Changes in the regional shape and volume of subcortical nuclei in patients with tinnitus comorbid with mild hearing loss.

PURPOSE: Tinnitus, the perception of sound without an external source, is a prevalent disease, but its underlying mechanism has not been fully elucidated. Recent studies have suggested the involvement of subcortical nuclei in tinnitus generation. We investigated changes in the local shape and volume of subcortical nuclei in relation to tinnitus. METHODS: The participants included 53 patients with tinnitus and 52 age- and gender-matched normal controls. Individual 3D T1-weighted structural images were obtained using 3-T magnetic resonance imaging. Surface-based vertex analysis (SVA) was perfor

Comprehensive mapping of the thalamic commissures in the rodent brain.

Brain commissures are antiparallel white matter fiber bundles that connect both hemispheres across the midsagittal plane at different anatomical levels. In the mammalian brain, the best-known commissures are the corpus callosum, the anterior and the posterior commissures. Recent studies have identified a new white matter pathway, the thalamic commissures (TCs), that connect the cortex to the contralateral thalamus in rodents and primates. However, the specific cortical regions that project via the TCs and their target thalamic nuclei are still unknown. To thoroughly and accurately map this pro

Neural systems underlying episodic memory: insights from animal research.

Two strategies used to uncover neural systems for episodic-like memory in animals are discussed: (i) an attribute of episodic memory (what? when? where?) is examined in order to reveal the neuronal interactions supporting that component of memory; and (ii) the connections of a structure thought to be central to episodic memory in humans are studied at a level of detail not feasible in humans. By focusing on spatial memory (where?) and the hippocampus, it has proved possible to bring the strategies together. A review of lesion, disconnection and immediate early-gene studies in animals reveals t

Thalamic degeneration.

A man of 61 died after an illness of 20 years duration. The clinical picture displayed progressive dementia, loss of memory, apathy, inertia, emotional lability, temporal and local disorientation, visual hallucination and apraxia. Neuropathological findings predominated in the thalamus and displayed extensive nerve cell loss and gliosis in the anterior, medial, and posterior formations, whereas the lateral formations were somewhat better preserved. The lateral amygdala were severely affected. The cortex showed widespread moderate damage in the third or fourth layer which are interpreted as tra

Detailed mapping of CGRP mRNA expression in the rat central nervous system: comparison with previous immunocytochemical findings.

The localization of CGRP mRNA in neurons of the rat brain and spinal cord was assessed by in situ hybridization histochemistry (ISH) using a radiolabeled synthetic 57-mer oligodeoxynucleotide probe complementary to the rat prepro CGRP mRNA. Results were compared with previously published findings of CGRP-immunoreactive (CGRP-IR) cell bodies revealed by an indirect immunofluorescence technique. The highest numbers of CGRP mRNA expressing neurons as well as the greatest intensity of staining were found in the lateral hypothalamic area, the parabrachial nuclei, and among the cranial motor nuclei,

Prefrontal system in the rat visualized by means of labeled deoxyglucose--further evidence for functional heterogeneity of the neostriatum.

An epileptic focus was formed in the anteromedial part of the prefrontal cortex in rats by means of intracortical injection of penicillin, and radioactively labeled deoxyglucose was immediately injected intravenously. Auto-radiograms revealed that, in addition to the injected cortical area, a number of formations increased their consumption of deoxyglucose. The labeled formations were the posterior medial and perirhinal cortical areas, the anteromedial part of the neostriatum, the anterior and intermediodorsal part of globus pallidus, the entopeduncular nucleus, the reticular portion of the su

There is a topographic organization in human cortico-pontine connectivity.

Of the three largest outputs of the cerebral cortex, two have been extensively studied and mapped. Topographic maps of cortico-thalamic and cortico-striatal functional connectivity in humans are well established. However, for the third largest cerebral output, to the pontine nuclei, which connect the cerebrum to the cerebellum, the existence of such an organized connectivity pattern in humans is unknown. Here, using high-resolution functional MRI and a large sample size, we found a topographically organized pattern of functional connectivity between the human cerebral cortex and pons. Our resu

Positron emission tomography assessment of cerebral glucose metabolic rates in autism spectrum disorder and schizophrenia.

Several models have been proposed to account for observed overlaps in clinical features and genetic predisposition between schizophrenia and autism spectrum disorder. This study assessed similarities and differences in topological patterns and vectors of glucose metabolism in both disorders in reference to these models. Co-registered 18fluorodeoxyglucose PET and MRI scans were obtained in 41 schizophrenia, 25 ASD, and 55 healthy control subjects. AFNI was used to map cortical and subcortical regions of interest. Metabolic rates were compared between three diagnostic groups using univariate and

Cortical, subcortical and brain stem connections of the cerebellum via the superior and middle cerebellar peduncle in the rat.

The role of cerebellum in coordination of somatic motor activity has been studied in detailed in various species. However, experimental and clinical studies have shown the involvement of the cerebellum with various visceral and cognitive functions via its vast connections with the central nervous system. The present study aims to define the cortical and subcortical and brain stem connections of the cerebellum via the superior (SCP) and middle (MCP) cerebellar peduncle using biotinylated dextran amine (BDA) and Fluoro-Gold (FG) tracer in Wistar albino rats. 14 male albino rats received 20-50-nl

Corticothalamic projections from postcruciate area 4 in the dog.

Corticothalamic projections from postcruciate area 4, located on the rostral part of the posterior sigmoid gyrus, were traced with the autoradiographic technique in the dog. Injections of tritiated amino acids were made into the lateral and medial parts of area 4 in regions corresponding to the forelimb and hindlimb areas of the primary motor cortex, respectively. In cases with injections placed in the lateral part of area 4, dense accumulations of label were present in the lateral part of the ventral anterior nucleus (VA), the central part of the ventral lateral nucleus (VL), the ventral half

Limb apraxia in patients with damage confined to the left basal ganglia and thalamus.

Limb apraxia was investigated with standardised tests in 14 patients whose CT scan provided evidence of a vascular lesion confined to the left basal ganglia, or the thalamus, or both, and not involving the cortex or adjacent white matter. Five patients were severely impaired in imitating movements and pantomiming object use. Four of them also performed poorly when tested with real objects. In two patients the lesion was primarily thalamic and in three the lesion was primarily in the lenticular nucleus and the posterior limb of the internal capsule. Patients without apraxia generally had smalle

Retrograde transport of brain-derived neurotrophic factor (BDNF) following infusion in neo- and limbic cortex in rat: relationship to BDNF mRNA expressing neurons.

Brain-derived neurotrophic factor (BDNF) was the second member of the nerve growth factor (NGF) family to be isolated. The ability of BDNF to be retrogradely transported following intraparenchymal infusion represents a unique neurobiological tool to determine the location of putative neuron-specific BDNF-responsive neuronal systems. In the present study, we infused recombinant human (rh) BDNF into the rodent neo- and limbic cortex and used a turkey anti-BDNF antibody to determine specific populations of neurons which retrogradely transport this neurotrophin. Frontal cortex infusion retrogradel

Dorsal column input to thalamic VL neurons: an intracellular study in the cat.

This study investigated the role of the ventral lateral (VL) nucleus of the thalamus as a lemniscal relay to motor cortex. Intracellular recordings were obtained from thalamic VL relay neurons in cats anesthetized with chloralose, following stimulation of the dorsal column nuclei. VL neurons were identified by their short-latency input from the cerebellar nuclei, their antidromic activation from motor cortex and their anatomical location. A total of 105 neurons was studied. The occurrence of temporal facilitation to double volleys was also examined. It was found that 80/105 (75%) neurons respo

Reduced thalamic resting-state functional connectivity and impaired cognition in acute abstinent heroin users.

As a critical component of cortico-striato-thalamo-cortical loop in addiction, our understanding of the thalamus in impaired cognition of heroin users (HU) has been limited. Due to the complex thalamic connection with cortical and subcortical regions, thalamus was divided into prefrontal (PFC), occipital (OC), premotor, primary motor, sensory, temporal, and posterior parietal association subregions according to white matter tractography. We adopted seven subregions of bilateral thalamus as regions of interest to systematically study the implications of distinct thalamic nuclei in acute abstine

Anatomical and physiological basis of continuous spike-wave of sleep syndrome after early thalamic lesions.

OBJECTIVE: Early neonatal thalamic lesions account for about 14% of continuous spike-wave of sleep (CSWS) syndrome, representing the most common etiology in this epileptic encephalopathy in children, and promise useful insights into the pathophysiology of the disease. METHODS: We describe nine patients with unilateral neonatal thalamic lesions which progressed to CSWS. Longitudinal whole-night and high-density electroencephalograms (EEGs) were performed, as well as detailed imaging and clinical evaluation. Visual evoked potentials were used to probe cortical excitability. RESULTS: Thalamic v

Multiple heteroreceptors on limbic thalamic axons: M2 acetylcholine, serotonin1B, beta 2-adrenoceptors, mu-opioid, and neurotensin.

Ligand binding to many transmitter receptors is much higher in layer Ia of rat posterior cingulate cortex than it is in other layers, and this is where most axons from the anterior thalamus terminate. The present study explores the possibility that a number of receptors may be expressed on axons from limbic thalamic nuclei that terminate in layer Ia. Unilateral thalamic lesions were placed in rats and, 2 weeks later, five ligand binding protocols, coverslip autoradiography, and single grain counting techniques were used to quantify binding in control and ablated hemispheres. Binding to the fol

Development of the human motor-related thalamic nuclei during the first half of gestation, with special emphasis on GABAergic circuits.

This study analyzed the expression of differentiation markers (Calbindin D28K: CaBP; parvalbumin: PARV; calretinin: CalR), gamma-aminobutyric acid (GABA) markers (GABA, glutamic acid decarboxylases: GAD65, GAD67; and GABA transporters: GAT1, GAT3), and other markers (neurotensin: NT, and neurofilament-specific protein: SMI32) in the human thalamus at 8-23 gestation weeks (g.w.), focusing on the motor-related nuclei. From 8-13 g.w. mainly CaBP was expressed in the cells while fiber bundles traversing the thalamus in addition to CaBP expressed all GABA markers except GAD67. CaBP and PARV express
