Algal-bacterial bioremediation of cyanide-containing wastewater in a continuous stirred photobioreactor.

This study reports the isolation and characterization of highly resistant bacterial and microalgal strains from an Egyptian wastewater treatment station to cyanide-containing compounds. The bacterial strain was identified as Bacillus licheniformis by 16S rRNA gene sequencing. The isolate removed up to 1 g L-1 potassium cyanide, 3 g L-1 benzonitrile, and 1 g L-1 sodium salicylate when incubated as 10% v/v in MSM at 30 ℃. However, it failed to degrade potassium thiocyanate at all tested concentrations. The microalgal isolate was identified by electron microscopy as a strain of Chlorella spp.. Al

A pilot study in the swine model of lethal cyanide intoxication indicates efficacy of a platinum-methionine complex countermeasure.

Cyanide presents a chemical security threat because of its wide availability, due to industrial uses, and significant toxicity. Platinum-thioether complexes are a recent addition to candidate antidotes, showing efficacy in mouse models of hydrogen cyanide inhalation and reversing toxicity measures in rabbits. This pilot study evaluated the effectiveness of a platinum-methionine complex (Met2Pt), a potent lead candidate, in a lethal swine model, which provides a solid foundation for future research. Potassium cyanide (0.2 mg/kg/min; IV) was infused into anesthetized swine until 6 min post-apnea

Intramuscular sodium tetrathionate as an antidote in a clinically relevant swine model of acute cyanide toxicity.

Background: Cyanide is a metabolic poison used in multiple industries and is a high threat chemical agent. Current antidotes require intravenous administration, limiting their usefulness in a mass casualty scenario. Sodium tetrathionate reacts directly with cyanide yielding thiosulfate and the non-toxic compound thiocyanate. Thiosulfate, in turn, neutralizes a second molecule of cyanide, thus, per mole, sodium tetrathionate neutralizes two moles of cyanide. Historical studies examined its efficacy as a cyanide antidote, but it has not been evaluated in a clinically relevant, large animal model

Seizures and selective CA-1 hippocampal lesions induced by an excitotoxic cyanide metabolite, 2-iminothiazolidine-4-carboxylic acid.

Excitatory amino acid (EAA)-like and excitotoxic properties of the secondary metabolite of cyanide, 2-iminothiazolidine-4-carboxylic acid, (2-ICA) were evaluated because of its possible role in cyanide-induced neurotoxicity. Intracerebroventricular (i.c.v.) injections of 2-ICA in mice produced wild-running seizures that were qualitatively and quantitatively similar to seizures observed with glutamate. 2-ICA, kainate and proline seizures were prevented by both the NMDA and non-NMDA antagonists, MK-801 and CNQX, respectively. In contrast, NMDA-induced seizures were prevented by MK-801, but not C

Encapsulation of thiosulfate: cyanide sulfurtransferase by mouse erythrocytes.

Murine carrier erythrocytes, prepared by hypotonic dialysis, were employed in the encapsulation of several compounds including [14C]sucrose, [3H]inulin, and bovine thiosulfate:cyanide sulfurtransferase (rhodanese), a mitochondrial enzyme which converts cyanide to thiocyanate. Approximately 30% of the added [14C]sucrose, [3H]inulin, and rhodanese was encapsulated by predialyzed erythrocytes, and a decrease in the mean corpuscular volume and mean corpuscular hemoglobin was observed. In the encapsulation of rhodanese a recovery of 95% of the erythrocytes was achieved and an 85% equilibrium was es

Evaluation of aqueous dimethyl trisulfide as an antidote to a highly lethal cyanide poisoning in a large swine model.

BACKGROUND: Cyanide is a rapid acting, lethal, metabolic poison and remains a significant threat. Current FDA-approved antidotes are not amenable or efficient enough for a mass casualty incident. OBJECTIVE: The objective of this study is to evaluate short and long-term efficacy of intramuscular aqueous dimethyl trisulfide (DMTS) on survival and clinical outcomes in a swine model of cyanide exposure. METHODS: Anesthetized swine were instrumented and acclimated until breathing spontaneously. Potassium cyanide infusion was initiated and continued until 5 min after the onset of apnea. Subsequent

Removal of cyanide by woody plants.

Hydrogen cyanide is a high volume production chemical that causes severe environmental problems. The toxicity of potassium cyanide (KCN) to basket willow trees (Salix viminalis) was tested. In aqueous solution, 2 mg CN l(-1) as KCN depressed the transpiration after 72 h about 50%. Trees exposed to 0.4 mg CN l(-1) in aqueous solution showed initially a depression of transpiration, but recovered. Doses of 8 and 20 mg CN l(-1) in aqueous solution were quickly mortal to the trees. At the end of the test, almost all cyanide had disappeared from the solutions. Levels of cyanide in plants were relate

[Effect of practolol on experimental hypertension in dogs].

In the dog, the intra-venous injection of practolol (1 to 3 mg/kg) reduces the hypertension provoked by vagotomy and sino-aortic nerves section, or by intra-venous administration of potassium cyanide. But practolol differs from propranolol ; he does not modify hypertension observed after electrical stimulation of central end of saphene, laryngeal superior, or vagus nerf.

Differences in Michaelis-Menten kinetics for different cultivars of maize during cyanide removal.

Knowledge of the kinetic parameters, the half-saturation constant (K(m)) and the maximum metabolic capacity (v(max)), is very useful for the characterization of enzymes and biochemical processes. Little is known about rates of which vegetation metabolizes environmental chemicals. It is known, however, that vascular plants possess an enzyme system that detoxifies cyanide by converting it into the amino acid asparagine. This study investigated the differences in Michaelis-Menten kinetics of cyanide removal by different cultivars of maize. Detached leaves (1.0 g fresh weight) of seven different c

Severe hypoxia produced by concomitant intoxication with sublethal doses of carbon monoxide and cyanide.

It has long been known that a number of tissue hypoxicants are generated in the fire scenario. However, until recently few investigators have undertaken to correlate smoke inhalation deaths with the simultaneous exposure to histotoxic hypoxicants. Carbon monoxide and hydrogen cyanide are two histotoxic hypoxicants that are generated in nearly every fire. Prior studies performed in our laboratory have demonstrated that death can result from concomitant exposure to otherwise sublethal concentrations of carbon monoxide and cyanide. Since most smoke inhalation victims exhibit acid/base anomalies,

Effects of hypoxia, cyanide, and ischaemia on myocardial contraction: observations in isolated muscle and intact heart.

Using potassium cyanide (KCN) to stimulate hypoxia, the effects of intracoronary injections of KCN were compared with total occlusions of the same vessel. Imparied contraction as measured by segment length gauges was of equally abrupt onset following both interventions. The magnitude of systolic expansion at one minute was more marked following total occlusion than after KCN administration.

The membrane potential of the cellular slime mold Dictyostelium discoideum is mainly generated by an electrogenic proton pump.

Trans membrane potential or ionic current changes may play a role in signal transduction and differentiation in the cellular slime mold dictyostelium discoideum. Therefore, the contribution of electrogenic ion pumps to the membrane potential of D. discoideum cells was investigated. the (negative) peak-value of the rapid potential transient, seen upon microelectrode impalement, was used to detect membrane potential changes upon changes in the external pH in the range of 5.5 to 8.0. The membrane potential was close to the Nernstian potential for protons over the pH range 5.5 to 7.5. The acid-ind

Kinetic study of the slow cyanide binding to Glycera dibranchiata monomer hemoglobin components III and IV.

Compared to other monomeric heme proteins and the heme peroxidases, the Glycera dibranchiata monomer hemoglobin components III and IV exhibit very slow cyanide binding kinetics. This is agreement with the previously reported behavior of component II. Similar to component II, components III and IV have been studied under pseudo-first-order conditions at pH 6.0, 7.0, 8.0, and 9.0 by using a 100-250-fold excess of potassium cyanide at each pH. At 20 degrees C with micromolar protein concentrations, kobs for component III varies between 7.08 x 10(-5) s-1 at pH 6.0 and 100-fold cyanide excess and 1

[Oxidation Destruction of Cu(CN)32- by Persulfate].

In recent years, cyanide contamination problem is becoming more and more serious. Cyanides widely occur in the wastewater from metallurgical industry and electroplating industry. The efficiency for cyanides removal of conventional physical and chemical methods is not very satisfactory. Therefore, it is urgent to develop highly efficient and environmental friendly methods for the removal of cyanides. As an environmental-friendly oxidant, potassium persulfate is widely used in the treatment of organic pollutants. The destruction of Cu(CN)32- was investigated by homogeneous K2S2O8 under alkaline

Potassium induced potential changes in rat diaphragm muscle.

The effect of different potassium concentrations on the membrane potential and membrane resistance of rat diaphragm muscle fibres was measured by means of a double sucrose gap method and a microelectrode technique. Concentration measurements showed that the muscle fibres gained sodium and lost potassium in the equilibration period. In the absence of external chloride changing the external potassium concentration from 2.8 mM to potassium-free caused a depolarization of the membrane of about 30 mV and a small increase in membrane resistance. This K-dependent potential change (K-response) was ind

Effect of sub-acute oral cyanide administration in rats: protective efficacy of alpha-ketoglutarate and sodium thiosulfate.

Chronic toxicity of cyanide in humans and animals has been previously described. Alpha-ketoglutarate (alpha-KG) and sodium thiosulfate (STS) are known to confer remarkable protection against acute cyanide poisoning in rodents. Their efficacy against sub-acute or chronic cyanide exposure is not known. The objective of the present study was to assess the sub-acute toxicity of potassium cyanide (KCN) in female rats following oral administration of 7.0 mg/kg (0.5 LD50) for 14 d. The effect of alpha-KG (oral; 1.0 g/kg) and/or STS (intraperitoneal, 1.0 g/kg) on cyanide toxicity was also evaluated. V

Mechanism and scope of the cyanide-catalyzed cross silyl benzoin reaction.

In this work, cross silyl benzoin addition reactions between acylsilanes (1) and aldehydes (2) catalyzed by metal cyanides are described. Unsymmetrical aryl-, heteroaryl-, and alkyl-substituted benzoin adducts can be generated in moderate to excellent yields with complete regiocontrol using potassium cyanide and a phase transfer catalyst. From a screen of transition metal cyanide complexes, lanthanum tricyanide was identified as an improved second-generation catalyst for the cross silyl benzoin reaction. A study of the influence of water on the KCN-catalyzed cross silyl benzoin addition reveal

Cyanide sensitivity and induction of the microsomal oleoyl-CoA desaturase of potato tuber.

1. The properties of oleoyl-CoA desaturase, induced in microsomal fractions by the 'ageing' treatment of potato tuber slices (aeration of slices for 3-18 h), were investigated to study the effect of cyanide on desaturation and cycloheximide on the induction of the desaturase. 2. The electrons needed for the desaturation can be supplied either by NADH or NADPH, but ascorbate can also drive the reaction; experiments with CO suggested that cytochrome P-450 was not involved in the desaturation. 3. A strong inhibition of the desaturation by potassium cyanide was observed with each of the electron d

Therapeutic efficacy of sodium nitrite and 4-dimethylaminophenol or hydroxylamine co-administration against cyanide poisoning in rats.

1. The therapeutic efficacy of combined treatment of sodium nitrite (NaNO2) and 4-dimethylaminophenol (DMAP) or hydroxylamine (H2NOH) was investigated in potassium cyanide (KCN) intoxication in male rats. 2. Therapy with NaNO2 (0.27 mmol kg-1) + DMAP (0.09 mmol kg-1) or NaNO2 + H2NOH (0.09 mmol kg-1, produced a protection index (ratio of LD50 of KCN in rats receiving therapy to an LD50 of KCN in rats given only 0.9% saline) of 2.5 and 2.0 respectively. 3. Both the regimens exhibited a beneficial effect in terms of improving the survival time and postural defects in rats exposed to 2 LD50 KCN.
