Gene expression profiling of depression and suicide in human prefrontal cortex.

Mood disorders are a major cause of disability. Etiology includes genetic and environmental factors, but the responsible genes have yet to be identified. Using DNA microarrays, we have conducted a large-scale gene expression analysis, in two regions of the human prefrontal cortex from post-mortem matched groups of subjects with major depression who had died by suicide, and control subjects who died from other causes and were free from psychiatric disorders. Bioinformatic analysis was used to investigate molecular and cellular pathways potentially involved in depression and suicidal behavior. W

Volumetric analysis of the prefrontal cortex, amygdala, and hippocampus in major depression.

Magnetic resonance imaging (MRI) studies in depressed subjects report smaller volumes of amygdala, hippocampus, inferior anterior cingulate, and the orbital prefrontal cortex (OPFC), components of the limbic-cortico-thalamic circuit. Major depression occurs more commonly in women, raising the possibility of an additional psychopathological process affecting women and not men. We sought to determine whether volumetric differences related to mood disorders are dependent on sex. Eight male and 10 female depressed subjects, meeting DSM III R criteria for a major depressive episode, and eight male

The role of prefrontal cortex in verbal episodic memory: rTMS evidence.

Long-term, episodic memory processing is supposed to involve the prefrontal cortex asymmetrically. Here we investigate the role of the dorsolateral prefrontal cortex (DLPFC) in encoding and retrieval of semantically related or unrelated word pairs. Subjects were required to perform a task consisting of two parts: a study phase (encoding), in which word pairs were presented, and a test phase (retrieval), during which stimuli previously presented had to be recognized among other stimuli. Consistently with our previous findings using pictures, repetitive transcranial magnetic stimulation (rTMS) h

Interactions of prefrontal cortex during eyeblink conditioning as a function of age.

Changes in regional cerebral blood flow (rCBF) in eleven elderly subjects during pairings of tone and air puff were compared to rCBF changes during pairings in young subjects. Although all subjects reported being aware of the relationship between tone and air puff, elderly subjects did not condition as well as young subjects and their rCBF measures were attenuated. Covarying the performance differences between young and old subjects did not change this conclusion suggesting that differences in neural activation during learning are related to binding of CS-US information prior to the impact of

Schizophrenia: glutathione deficit in cerebrospinal fluid and prefrontal cortex in vivo.

Schizophrenia is a major psychiatric disease, which affects the centre of the personality, with severe problems of perception, cognition as well as affective and social behaviour. In cerebrospinal fluid of drug-free schizophrenic patients, a significant decrease in the level of total glutathione (GSH) by 27% (P<0.05) was observed as compared to controls, in keeping with the reported reduced level of its metabolite gamma-glutamylglutamine. With a new non-invasive proton magnetic resonance spectroscopy methodology, GSH level in medial prefrontal cortex of schizophrenic patients was found to be 5

Evidence for the importance of dopamine for prefrontal cortex functions early in life.

There is considerable evidence that dorsolateral prefrontal cortex subserves critical cognitive abilities even during early infancy and that improvement in these abilities is evident over roughly the next 10 years. We also know that (a) in adult monkeys these cognitive abilities depend critically on the dopaminergic projection to prefrontal cortex and (b) the distribution of dopamine axons within dorsolateral prefrontal cortex changes, and the level of dopamine increases, during the period that infant monkeys are improving on tasks that require the cognitive abilities dependent on prefrontal c

The role of the prefrontal cortex in self-consciousness: the case of auditory hallucinations.

Many patients with schizophrenia report hallucinations in which they hear voices talking to them or about them. Behavioural and physiological studies show that this experience is associated with processes occurring in auditory language systems associated with both the production and the reception of speech. I propose that hallucinations are experienced because patients have difficulty in distinguishing sensations caused by their own actions from those that arise from external influences. This distinction can be made by predicting the sensations that will result from executive commands (forward

Effects of attention on visuomotor activity in the premotor and prefrontal cortex of a primate.

We examined neuronal activity in the primate premotor (PM) and prefrontal (PF) areas during a demanding spatial matching task. On each behavioral trial, a rhesus monkey moved its forelimb when a visual stimulus, called the "prime stimulus," reappeared at a previously cued location. Because it triggered a movement, the part of space cued by the prime stimulus had to be either remembered or attended during the time between prime stimulus presentations. Between the first and second appearances of the prime stimulus, behaviorally irrelevant visual stimuli could appear at one or several locations o

Behavioural changes after ablation of subdivisions of the rat prefrontal cortex.

Sham operated controls and four groups of rats subjected to ablation of various parts of their frontal cortex were compared in food and water intake and four behavioural tasks. The ablations were aimed at removing (1) the ventral prefrontal cortex, (2) the dorsal part of the medial prefrontal cortex, (3) the total medial prefrontal cortex, and (4) the anterior dorsolateral (non-prefrontal) cortex. Only two groups had a significantly impaired food and water intake: the ventral prefrontal and the non-prefrontal anterolateral. The latter group was not adipsic. Paratenial thalamus engages in reciprocal and broadcast circuits with the prefrontal cortex.

The dorsal anterior midline thalamus (aMT) consists of several closely packed nuclei that are important for motivated and emotional behavior. Previous work on aMT has focused on cells and synapses in the paraventricular thalamus (PVT), and little is known about the adjacent paratenial thalamus (PT). Here we examine neural circuits involving PT using a combination of molecular profiling, anatomical tracing, electrophysiology, and optogenetics. We first find that Protein Kinase C-delta (PKCd) selectively labels thalamocortical (TC) cells concentrated in PT but largely absent from neighboring PVT

Microplastic Neurotoxicity in the Prefrontal Cortex: A Review of Mechanisms and Neuropsychiatric Associations.

The escalating crisis of plastic pollution has positioned microplastics (MPs) as globally pervasive environmental contaminants, with a documented presence across aquatic, terrestrial, and atmospheric ecosystems, as well as within biological organisms. A growing body of evidence suggests that MPs not only threaten ecological integrity but may also induce multifaceted neurotoxic effects in humans, particularly targeting the functional architecture of the prefrontal cortex (PFC). As the central regulator of cognition, emotional processing, and behavioral control, PFC dysfunction has been hypothes

BDNF relates to prefrontal cortex activity in the context of physical exercise.

Exercise has been shown to support brain health, cognitive function, and increase levels of brain-derived neurotrophic factor (BDNF). While BDNF is known to support the central nervous system through improved brain metabolism, vasculature, neurotransmission and synaptic plasticity, the association between exercise-induced changes in BDNF concentrations and exercise-related cognitive improvements is still unclear. This study investigated the relationship between exercise-induced changes in plasma BDNF (pBDNF) and serum BDNF (sBDNF), and haemodynamic indicators of prefrontal cortex function in s

On the Causal Role of the Right Lateral Prefrontal Cortex in Active Forgetting.

Extensive research has demonstrated that people can intentionally forget by inhibiting the retrieval of unwanted memories, a phenomenon known as suppression-induced forgetting (SIF). Although neuroimaging studies have linked retrieval suppression to the right lateral prefrontal cortex (LPFC), direct evidence for the causal role this region plays in supporting SIF is still lacking. In this registered report, our aim is putting to a strong empirical test such an idea by using cathodal transcranial direct current stimulation in a standard think/no-think procedure. Across two experiments, we will

Sex differences in noradrenergic regulation of the medial prefrontal cortex in mice.

BACKGROUND: Norepinephrine (noradrenaline; NE) is a stress signal released from the locus coeruleus (LC) into the prefrontal cortex (PFC) to govern arousal, attention, and cognition. The LC is sexually dimorphic, and PFC NE dysfunction contributes to alcohol use disorder and several stress-related neuropsychiatric disorders that manifest differently in men and women. However, most preclinical studies of the medial PFC (mPFC) NE system have only used male subjects. Additionally, even though each mPFC subregion and layer forms unique circuits that mediate different aspects of cognitive behavior,

Chronic pain is associated with greater brain entropy in the prefrontal cortex.

Chronic pain is a debilitating clinical condition and a severe public health issue that demands to be addressed. Neuroimaging-based techniques have been widely adopted to investigate the neural underpinnings of chronic pain. Despite the efforts the complex nature of pain experience as well as the heterogeneity of chronic pain have made the identification of neuroimaging-based biomarkers extremely challenging. In this study, resting-state fMRI-based brain entropy, a measure reflecting the "irregularity" of brain activity, was adopted as a biomarker of chronic pain by comparing individuals with

Exploring the Role of Prefrontal Cortex tDCS in Hypertension: A Mini-Review.

Arterial Hypertension (HTN) is the leading cause of cardiovascular diseases, which, in turn, are the primary cause of mortality worldwide. The success rates in Blood Pressure (BP) control among the general population remain unacceptably low. HTN etiology is multifactorial, but ample evidence has shown an essential role of the Autonomic Nervous System (ANS) dysfunction in its physiopathology. Concurrently, studies have pointed to the promising effect of non-invasive cortical stimulation techniques, such as transcranial Direct Current Stimulation (tDCS), on modulating blood pressure and the ANS.

Neural responses in macaque prefrontal cortex are linked to strategic exploration.

Humans have been shown to strategically explore. They can identify situations in which gathering information about distant and uncertain options is beneficial for the future. Because primates rely on scarce resources when they forage, they are also thought to strategically explore, but whether they use the same strategies as humans and the neural bases of strategic exploration in monkeys are largely unknown. We designed a sequential choice task to investigate whether monkeys mobilize strategic exploration based on whether information can improve subsequent choice, but also to ask the novel que

The Interaction of the Dorsolateral and Ventromedial Prefrontal Cortex During Mind Wandering.

BACKGROUND AND AIMS: Mind wandering refers to spontaneously occurring, often disruptive thoughts during an ongoing task or resting state. The ventromedial prefrontal cortex (vmPFC) and dorsolateral prefrontal cortex (dlPFC) are two main cortical areas which are involved in this process. This study aimed to explore the interaction of these areas during mind wandering by enhancing specific oscillatory activity of these areas via transcranial alternating current stimulation (tACS) in the theta frequency range. MATERIAL AND METHODS: Eighteen healthy adults participated in a randomized, single-bli
