1. In ovariectomized rats pre-treated with oestrogen and progesterone, the intensity of the catecholamine fluorescence was determined by micro-fluorimetry in the cell bodies of the tubero-infundibular dopamine neurones (situated in the arcuate and periventricular hypothalamic nuclei). The concentrations of follicle-stimulating (FSH) and luteinizing hormone (LH) in serum and pituitary were estimated by bio-assay.2. The characteristic changes in fluorescence intensity of the tuberal catecholamine-containing nerve cell group that are induced by unilateral intermittent electrical stimulation of th

This study investigated the impact of multiple nerve block methods (local anesthesia, conventional radiofrequency thermocoagulation [CRF], and pulsed radiofrequency [PRF]) on thermoregulation. Focusing on hypothalamic function, the effects of local anesthesia, CRF, and PRF on central and peripheral temperatures were analyzed and compared. Our findings revealed that all three nerve block groups cause a decrease in central temperature, with the CRF group exhibiting the most pronounced effect. Furthermore, immunostaining analysis showed decreased neural activity in the preoptic area, suggesting t

Sexually naïve female mice do not display high levels of sexual receptivity in their first sexual experience; they require around 4-5 sexual encounters to display the full receptive response, assessed by the lordosis reflex. In this study, we evaluated if repeated sexual stimulation with the same male is associated with changes in synaptic remodeling evaluated by synaptophysin (SYP) in brain structures involved in the control of sexual behavior such as the main and accessory olfactory bulbs (MOB and AOB, respectively), medial preoptic area (MPOA), ventromedial hypothalamus (VMH), and amygdala

AIM: We hypothesized that hydrogen sulphide (H2 S), acting specifically in the anteroventral preoptic region (AVPO - an important integrating site of thermal and cardiorespiratory responses to hypoxia in which H2 S synthesis has been shown to be increased under hypoxic conditions), modulates the hypoxic ventilatory response.
METHODS: To test this hypothesis, we measured pulmonary ventilation (V˙E) and deep body temperature of rats before and after intracerebroventricular (icv) or intra-AVPO microinjection of aminooxyacetate (AOA; CBS inhibitor) or Na2 S (H2 S donor) followed by 60 min of hypo

Kainic acid (KA) and muscimol were injected into the lateral preoptic area (LPO) of the rat to study their effects on drinking behavior. A low dose (5 ng) of KA, which stimulates neurons, decreased the amount of water intake induced by hypertonic saline (IP) and angiotensin II (SC). Injection of 2 ng muscimol, a potent GABAA receptor agonist that suppresses neurons, facilitated drinking responses induced by hypertonic saline, but did not affect angiotensin II-induced drinking. Rats injected with a high dose (150 ng) of KA, which destroys neurons, showed marked polydipsia accompanied by increas

Based on the chicken hypothalamic analysis using the Nissl, Golgi-Cox and silver staining methods, a first attempt to make the cyto-, dendro- and fibro-architectonic atlases of the frontal planes at the same level was performed. In the preparations stained with Nissl method, the hypothalamus was subdivisible basically into the preoptic, rostral and caudal hypothalamic regions, each of which was composed of medial and lateral parts respectively. A well-defined boundaries between the preoptic and rostral hypothalamic regions was decided at the level of the rostral margin of the commissura rostra

The ovarian steroids estradiol (E(2)) and progesterone (P) act on target neurons in the hypothalamus and preoptic area to coordinate the expression of female reproductive behaviors with the timing of the preovulatory luteinizing hormone (LH) surge. This chapter will summarize evidence that E(2) and P facilitation of the receptive component of female reproductive behavior, lordosis, involves changes in both the expression of and intracellular signal transduction pathways engaged by alpha(1)-adrenergic receptors in the hypothalamus and preoptic area. The alpha(1)-adrenoceptors are thought to med

Amylin, a 37-aa pancreatic peptide, was found to be expressed in the preoptic area of mother rats in our recent microarray study. Here, we report a marked increase in amylin expression around parturition and show that amylin mRNA level remains elevated as long as the pups are not removed from the dams. Amylin expression is also induced in maternally behaving (sensitized) nonlactating but not in nonsensitized nulliparous females or in females that did not become maternal despite the sensitization procedure. Immunohistochemistry verified the increased amylin peptide expression in maternally beha

Angiotensin II (AII; 1.0 microliter of 10(-10) M) injected bilaterally into the anterior hypothalamus-preoptic area of the brain produced a 100% increase in mean whole blood LH concentrations in ovariectomized rats treated with estradiol and progesterone. Injections of equal volumes and concentrations of the peptide into the third cerebral ventricle or into the posterior hypothalamus did not affect blood LH values. The effects of AII administration into the rostral hypothalamus were dose-related and specific. Equimolar injections of angiotensin I or angiotensin III into this region did not sig

The overall projection pattern of a tiny bed nuclei of the stria terminalis anteromedial group differentiation, the dorsomedial nucleus (BSTdm), was analyzed with the Phaseolus vulgaris-leucoagglutinin anterograde pathway tracing method in rats. Many brain regions receive a relatively moderate to strong input from the BSTdm. They fall into eight general categories: humeral sensory-related (subfornical organ and median preoptic nucleus, involved in initiating drinking behavior and salt appetite), neuroendocrine system (magnocellular: oxytocin, vasopressin; parvicellular: gonadotropin-releasing

The connections of the olfactory bulb were traced using horseradish peroxidase. A homologue of the medial olfactory tract in gnathostomes projects to the ipsilateral septal nucleus, preoptic area and, possibly, the rostral striatum. A homologue of the lateral olfactory tract projects to the ipsilateral lateral pallium, dorsal pallium and, possibly, the medial pallium, as well as to the posterior diencephalon. A component of the lateral olfactory tract decussates in the habenular and posterior commissures and distributes to the contralateral hemisphere and caudal diencephalon. A dorsal componen

Neurons expressing luteinizing hormone-releasing hormone (LHRH), found in the septal-preoptic nuclei and hypothalamus, control the release of gonadotropic hormones from the anterior pituitary gland and facilitate reproductive behaviour. LHRH-expressing neurons are also found in the nervus terminalis, a cranial nerve that is a part of the accessory olfactory system and which projects directly from the nose to the septal-preoptic nuclei in the brain. During development, LHRH-immunoreactivity is detected in the peripheral parts of the nervus terminalis before it is found in the brain. Using a com

Growth retardation induced by dietary restriction in the lamb results in a low GnRH pulse frequency, and thus, puberty is delayed. In our experimental model, in which ovariectomized lambs are maintained at weaning weight (approximately 20 kg BW), hypothalamic GnRH is present and releasable, suggesting that central mechanisms limit the release of GnRH during chronic growth restriction. Our study compared the number and distribution of GnRH-containing neurons in growth-restricted (n = 5) and rapidly growing (n = 5) ovariectomized prepubertal female lambs at 24 weeks of age (normal age of puberty

In postmortem examination of brains of four patients with chronic paranoid schizophrenia, above-normal norepinephrine levels were measured in the ventral septum, the bed nucleus of the stria terminalis, the nucleus accumbens, and the mammillary bodies. No changes were detected in other limbic forebrain regions, including the hypothalamus and the medial olfactory (preoptic) area. The results point to the possibility of a malfunction of limbic noradrenergic mechanisms in schizophrenia, especially the paranoid variety.

An array of guide tubes to accommodate concentric push-pull cannulae was implanted chronically within the diencephalon of the rhesus or other species of macaque monkey which was accustomed to a primate chair. Colonic and skin temperatures were monitored continuously during each experiment in which a circumscribed site in the monkey's hypothalamus had been labelled by microinjection of 50-100 muCi serotonin (3H-5-HT) or 50-100 muCi or norepinephrine (3H-NE). Consecutive push-pull perfusions with an artificial CSF were carried out for 10 min at a rate of 50 mul/min at 20 min intervals. Under the

Damage to the medial preoptic area (MPOA) was produced in one-day-old female rats by bilateral electrolytic lesions or by aspiration of preoptic tissue. Both procedures resulted in similar advancement of vaginal opening and the first ovulation. A further group of rats was lesioned or aspirated in the MPOA and immediately bilaterally implanted with medial preoptic tissue collected by the puncture method from 17-19-day-old female rat fetuses. After autopsy, tissue destruction and implants located in the MPOA could be identified in 5 females that showed significantly later onset of puberty than b

Sex hormone binding globulin (SHBG) is found in the brain and acts directly on plasma membrane-associated receptors in the prostate gland. Infusing SHBG into the medial preoptic area or medial basal hypothalamus of female rats increases their female sexual receptivity. SHBG, SHBG plus estradiol (SHBG-E), and SHBG-E plus oxytocin all significantly increased female sexual receptivity over vehicle or estradiol plus oxytocin infused controls, as measured by lordosis quotients and receptivity scores, at 40, and 90 min after their infusions into the medial preoptic area. When infused into the medial

Neural correlates of mating calling can be recorded from isolated brainstems of male, Northern leopard frogs after the circuits for this behavior have been triggered by electrical stimulation of the preoptic area. Correlates can be evoked reliably and by a stimulus of low amplitude. However, such correlates can be evoked only rarely from female brainstems, and then only by a much larger stimulus. The sensitivity to triggering in female brainstems can be masculinized by previous treatment of the intact frog with testosterone propionate or dihydrotestosterone, but not by estradiol benzoate. In many mammalian species, females exhibit higher sociability and gregariousness than males, presumably due to the benefit of group living for maternal care. We have previously reported that adult female mice exhibit contact-seeking behaviors upon acute social isolation via amylin-calcitonin receptor (Calcr) signaling in the medial preoptic area (MPOA). In this study, we examined the sex differences in the behavioral responses to acute social isolation and reunion, and the levels of amylin and Calcr expression in the MPOA. We found that male mice exhibited significantly less contact-seeking up

It is well established that sexually dimorphic brain regions, which are critical for reproductive physiology and behavior, are organized by steroid hormones during the first 2 weeks after birth in the rodents. In our recent observation, neonatal exposure to estradiol-17β (E2) in the female rat revealed increase in cyclooxygenase 2 (COX-2) level, sexually dimorphic nucleus (SDN)-pre-optic area (POA) size and down-regulation of synaptogenesis related genes in POA in the adult stage. In the present study, using the same animal model, the protein profile of control and neonatally E2-treated POA wa
