Predictive Factors for Poor Progression-free Survival in Patients with Non-small Cell Lung Cancer Treated with Nivolumab.

BACKGROUND: Nivolumab has shown promising effects in patients with non-small-cell lung cancer (NSCLC) as a second- or later-line treatment. This study aimed to identify patients who would not experience any benefit from nivolumab treatment. MATERIALS AND METHODS: In this study, data for 201 patients treated with nivolumab during 17 December 2015 to 31 July 2016 at three respiratory medical centers in Japan were retrospectively reviewed. We collected clinical data at the time of nivolumab treatment commencement. We investigated the relationship between progression-free survival (PFS) and patie

Polymorphisms in ERCC1 C8092A predict progression-free survival in metastatic/recurrent nasopharyngeal carcinoma treated with cisplatin-based chemotherapy.

OBJECTIVES: We evaluated whether DNA repair gene polymorphisms had an effect on clinical outcomes in metastatic/recurrent nasopharyngeal carcinoma (NPC) patients treated with cisplatin-based chemotherapy. MATERIALS AND METHODS: Clinical data of 101 patients with metastatic/recurrent NPC between 2004 and 2011 were reviewed. Five potentially functional polymorphisms (ERCC1 Asn118Asn, ERCC1 C8092A, XPD Lys751Gln, XRCC1 Arg399Gln and XRCC1 Arg280His) were genotyped using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. RESULTS: The ERCC1 C8092A polymorphism was an in

Primary Tumor Localization is Associated with Progression-free Survival and Overall Survival after Selective-Internal-Radio-Therapy of Liver Metastases in Metastatic Colorectal Cancer.

This retrospective study aimed to investigate whether the differing biology of right-sided primary tumors (RSP) and left-sided primary tumors (LSP) in metastatic colorectal cancer (mCRC) affects survival outcomes following selective internal radiation therapy (SIRT).A cohort of 140 mCRC patients with liver metastases treated with SIRT between 2009 and 2016 was analyzed. Patients were stratified by tumor localization (RSP: proximal colon; LSP: distal colon and rectum), clinical, histopathological, and molecular data, were collected. Survival outcomes (OS and PFS) were evaluated using Kaplan-Mei

Influence of treatment modalities on survival and progression free survival in primary spinal cord glioblastoma: a systematic review and individual participant data meta-analysis.

INTRODUCTION: Glioblastoma (GBM) is the most common primary malignant tumor of the central nervous system, primarily affecting the brain. It infrequently occurs in the spinal cord, accounting for approximately 1.5% of all spinal cord tumors. Primary spinal cord GBM (sGBM) is associated with a poor prognosis, with a median overall survival (OS) of 12-15 months and a median progression-free survival (PFS) of 11.5 months. This study performed a systematic review and a individual participant data meta-analysis to evaluate the impact of various therapeutic strategies on OS and PFS in patients with

Prolonged progression-free survival and overall survival are associated with diabetes mellitus but inversely associated with levels of blood glucose in patients with lung cancer.

BACKGROUND: Previous studies have provided conflicting evidence about the increased overall survival (OS) in lung cancer patients with diabetes mellitus (DM) compared with those without DM. This study assessed progression-free survival (PFS)/OS in lung cancer patients with or without DM and tentatively analyzed the impact of blood glucose levels on PFS/OS in lung cancer patients. METHODS: Data were collected from lung cancer patients based upon admission records from January 2010 to January 2012 and follow-up records from January 2010 to January 2015 in the Department of Pulmonary Medicine, Z

A novel severe toxicity-free, and progression-free survival endpoint predicts outcomes after CD19 chimeric antigen receptor-T cell therapy in large B-cell lymphoma.

CD19-directed chimeric antigen receptor-T cell (CAR-T) therapies have transformed treatment for relapsed/refractory large B-cell lymphoma (LBCL), yet heterogeneity in toxicity and efficacy persists. To address this, we developed a novel composite endpoint: severe toxicity-free and progression-free survival at 6 months (TPFS6), defined as absence of severe CRS, ICANS, progressive/stable disease (PD/SD), and non-relapse mortality (NRM) within 6 months of CAR-T therapy. In our cohort, 37% achieved TPFS6. Events contributing to TPFS6 failure included severe CRS (6.8%), ICANS (30.3%), NRM (1.7%), a

Secondary malignant neoplasms, progression-free survival and overall survival in patients treated for Hodgkin lymphoma: a systematic review and meta-analysis of randomized clinical trials.

Treatment intensification to maximize disease control and reduced intensity approaches to minimize the risk of late sequelae have been evaluated in newly diagnosed Hodgkin lymphoma. The influence of these interventions on the risk of secondary malignant neoplasms, progression-free survival and overall survival is reported in the meta-analysis herein, based on individual patient data from 9498 patients treated within 16 randomized controlled trials for newly diagnosed Hodgkin lymphoma between 1984 and 2007. Secondary malignant neoplasms were meta-analyzed using Peto's method as time-to-event ou

Contemporary phase III clinical trial endpoints in advanced ovarian cancer: assessing the pros and cons of objective response rate, progression-free survival, and overall survival.

Among gynecologic cancers, ovarian cancer provides the greatest challenge because 75% to 80% of patients present with stage III/IV disease. Over the last 40 years, a series of large trials conducted by the Gynecologic Oncology Group and other cooperative groups has produced striking improvements in patient outcome; but the majority still dies of their disease. Further research in both the laboratory and the clinic is essential to continued improvement in patient management. Clinical trials, however, have become a major challenge because of issues with trial endpoints. Historically, overall sur

An FDA analysis of the association of tumor growth rate, overall survival, and progression-free survival in patients with metastatic non-small cell lung cancer.

BACKGROUND: Despite recent advances and an improved treatment paradigm, the prognosis in patients with advanced non-small-cell lung cancer (NSCLC) remains poor. Novel endpoints to evaluate antitumor activity are needed to expedite safe and effective drug development. METHODS: We performed a retrospective pooled analysis of 10,495 patients with NSCLC using radiological tumor measurements across 22 randomized clinical trials fit to a regression-growth model to derive the g value. We evaluated the g value across both type and line of therapy. RESULTS: The g value is inversely associated with su

Metastatic Lymph Node Status Is Associated With Progression-Free Survival in Oral Cavity Squamous Cell Carcinoma Treated With Pembrolizumab.

BACKGROUND: The prognostic implications of regional lymph node metastases at the initiation of immune checkpoint inhibitor (ICI) therapy in oral cavity squamous cell carcinoma (OCSCC) are not well defined. This study evaluated whether the presence of metastatic cervical lymph nodes at the start of pembrolizumab therapy was associated with progression-free or overall survival among patients with recurrent or metastatic (R/M) OCSCC. METHODS: A retrospective cohort study was conducted at a tertiary academic medical center including patients with R/M OCSCC who initiated pembrolizumab between May

Correlation of Tumor Absorbed Dose with Progression-Free Survival in Metastatic Castration-Resistant Prostate Cancer Treated with 177Lu-PSMA.

Today's common practice of empiric activity prescription in radiopharmaceutical therapy is inconsistent with the modern philosophy of personalized medicine. We investigate the radiobiologic relationship between tumor mean absorbed dose (D mean) and progression-free survival (PFS) in 177Lu-PSMA I&T therapy guided by personalized predictive dosimetry in metastatic castration-resistant prostate cancer. Methods: We conducted a single-center retrospective study of the first 20 patients treated with 177Lu-PSMA I&T at our institution. PFS was calculated from the day a patient was assessed by a nuclea

A functional prognostic model predicts progression-free survival in patients with relapsed chronic lymphocytic leukemia treated with ibrutinib + venetoclax.

Targeted therapies have improved outcomes for patients with chronic lymphocytic leukemia (CLL); yet, not all achieve durable responses. Existing prognostic models, developed for chemoimmunotherapy, have limited predictive value for newer targeted combinations. Here, we used data from a randomized phase II clinical trial to identify predictors of progression-free survival (PFS) in response to Bruton tyrosine kinase (BTK) + B-cell lymphoma 2 (Bcl-2) inhibitor combination therapy. We studied the functional, genetic, and clinical characteristics of patients with relapsed/refractory (R/R) CLL treat

Meningiomas classified as grade 2 due to brain invasion alone have better progression-free survival than other atypical meningiomas.

BACKGROUND: Brain invasion (BI) was incorporated as a major criterion for grade 2 meningiomas in the 2016 WHO guidelines. However, its prognostic value remains controversial due to variability in surgical sampling and histopathologic interpretation, as well as BI may be the consequence of mechanical forces related more with the size of the tumor than the biologic presence of an innately more aggressive biologic lesion. Despite advances in molecular characterization of meningiomas, the relevance of BI as an independent prognostic indicator remains unresolved. We sought to determine whether BI a
