Molecular therapy : the journal of the American Society of Gene Therapy2020eng
Autoimmune destruction of pancreatic beta cells is the characteristic feature of type 1 diabetes mellitus. Consequently, both short- and intermediate-acting insulin analogs are under development to compensate for the lack of endogenous insulin gene expression. Basal insulin is continuously released at low levels in response to hepatic glucose output, while post-prandial insulin is secreted in response to hyperglycemia following a meal. As an alternative to multiple daily injections of insulin, glucose-regulated insulin gene expression by gene therapy is under development to better endure postp

The exchange of 125I-insulin, 125I-glucagon, 125I-proinsulin, 125I-growth hormone, 131I-albumin, 14C-inulin, and 14C-dextran across isolated rat mesentery was studied in a diffusion cell. The passage of immunoprecipitable porcine 125I-insulin (0.88 ng./ml.) was not affected by porcine proinsulin (145 ng./ml.), crystalline porcine insulin (17.4 ng./ml.), human growth hormone (87 ng./ml.), bovine serum albumin (4.5 mg./ml.), or normal guinea pig serum (840 mug. protein/ml.). However, the rate of insulin exchange was reduced by guinea pig anti-insulin antiserum and partially purified human serum-

KEY POINTS: Type 2 diabetes partitioned polygenic scores capture metabolic profiles for obesity, β-cell with positive proinsulin, and lipodystrophy clusters in CKD. The type 2 diabetes obesity genetic cluster was associated with higher risk of overall mortality among individuals with CKD.
BACKGROUND: Type 2 diabetes (T2D) exhibits biologic and pathophysiologic heterogeneity, which may contribute to variations in diabetes-related complications, particularly in high-risk populations such as individuals with CKD. Prior research has explored T2D mechanisms using partitioned polygenic scores (pPGS

Type 1 diabetes (T1D) is related to the autoimmune destruction of β-cells, leading to their almost complete absence in patients with longstanding T1D. However, endogenous insulin secretion persists in such patients as evidenced by the measurement of plasma C-peptide. Recently, a low level of insulin has been found in non-β islet cells of patients with longstanding T1D, indicating that other islet cell types may contribute to persistent insulin secretion. The present study aimed to test the ability of various antibodies to detect insulin in insulin-deficient islets of T1D patients. Pancreatic a

Human insulin (INS) gene diverged from the ancestral genes of invertebrate and mammalian species millions of years ago. We previously found that mouse insulin gene (Ins2) isoforms are expressed in brain choroid plexus (ChP) epithelium cells, where insulin secretion is regulated by serotonin and not by glucose. We further compared human INS isoform expression in postmortem ChP and islets of Langerhans. We uncovered novel INS upstream open reading frame isoforms and their protein products. In addition, we found a novel alternatively spliced isoform that translates to a 74-amino acid (AA) proinsu

Autoantigen-specific immunological tolerance represents a central objective for prevention of type 1 diabetes (T1D). Previous studies demonstrated mucosal antigen administration results in expansion of Foxp3+ and LAP+ regulatory T cells (Tregs), suggesting oral delivery of self-antigens might represent an effective means for modulating autoimmune disease. Early preclinical experiments using the non-obese diabetic (NOD) mouse model reported mucosal administration of T1D-related autoantigens [proinsulin or glutamic acid decarboxylase 65 (GAD)] delayed T1D onset, but published data are conflictin

It has long been known that prolactin (PRL) induces proliferation of the mucosal epithelial lining of the pigeon crop-sac. This system was used to study possible interactions of other hormones with PRL's mitogenic effect directly on the crop-sac cells. Injection of a sheep pituitary powder into the loose skin between the leg and abdomen of pigeons increased the responsiveness of the crop-sac to direct, local application of PRL. Similar systemic injections of anterior pituitary hormones, ACTH and TSH (alone or together), or a combination of LH and FSH, were without effect on the local response

Islet amyloid polypeptide (IAPP) messenger RNA levels, biosynthesis, processing, and secretion were studied in cultured mouse beta TC3 insulinoma cells. Northern blot analysis revealed that the size of IAPP mRNA (0.9 kb) in beta TC3 cells was the same as that in normal mouse islets; IAPP mRNA was approximately 60% of the level of insulin mRNA in beta TC3 cells. However, the ratio of synthesis of insulin to IAPP was approximately 6:1, suggesting that IAPP mRNA is not translated efficiently in these cells. Metabolic labeling of beta TC3 cells with [3H]leucine revealed the synthesis of both a pre

Studies of the biosynthesis of insulin in a human insulinoma beginning in 1965 provided the first evidence for a precursor of insulin, the first such prohormone to be identified. Further studies with isolated rat islets then confirmed that the precursor became labeled more rapidly than insulin and later was converted to insulin by a proteolytic processing system located mainly within the secretory granules of the beta cell and was then stored or secreted. The precursor was designated "proinsulin" in 1967 and was isolated and sequenced from beef and pork sources. These structural studies confir

OBJECTIVE: Polypyrimidine tract-binding protein 1 (PTBP1) promotes stability and translation of mRNAs coding for insulin secretion granule proteins and thereby plays a role in β-cells function. We studied whether common genetic variations within the PTBP1 locus influence insulin secretion, and/or proinsulin conversion.
METHODS: We genotyped 1,502 healthy German subjects for four tagging single nucleotide polymorphisms (SNPs) within the PTBP1 locus (rs351974, rs11085226, rs736926, and rs123698) covering 100% of genetic variation with an r(2)≥0.8. The subjects were metabolically characterized b

Recent studies have shown that severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection may induce metabolic distress, leading to hyperglycemia in patients affected by coronavirus disease 19 (COVID-19). We investigated the potential indirect and direct effects of SARS-CoV-2 on human pancreatic islets in 10 patients who became hyperglycemic after COVID-19. Although there was no evidence of peripheral anti-islet autoimmunity, the serum of these patients displayed toxicity on human pancreatic islets, which could be abrogated by the use of anti-interleukin-1β (IL-1β), anti-IL-6, and

We investigated the contributions made by the entero-insular axis, proinsulin and the fractional hepatic extraction of insulin to the hyperinsulinaemia characteristic of polycystic ovarian syndrome (PCOS). We measured plasma glucose, gastric inhibitory polypeptide (GIP), glucagon-like peptide-1 (7-36 amide) (GLP-1(7-36) amide), immunoreactive insulin (IRI), intact proinsulin (IPI), and C-peptide concentrations during a 75 g oral glucose tolerance test in seven normal weight women with PCOS and eight healthy women. Women with PCOS had higher fasting (P = 0.05) and integrated (P < 0.01) IRI conc

STUDY OBJECTIVES: Obstructive sleep apnea (OSA) is associated with maladaptive changes to glucose homeostasis. Chronic intermittent hypoxia (IH) modeling OSA in rodents causes similar patterns of dysglycemia. Time-restricted eating (TRE), limiting caloric intake to a fixed daily window, improves the metabolic profile in rodents and humans, though it is unknown whether TRE improves IH-induced dysglycemia in an obese OSA model.
METHODS: C57BL/6J mice were fed a high-fat diet for 4 weeks, and then divided into four groups: Half in intermittent air (IA) or IH, and half from each group fed either

Diabetes mellitus, a chronic metabolic disorder, is characterized by high blood glucose levels. External insulin administration, along with diet and exercise, is recommended to the patients. The increase in the number of diabetics and the adoption of oral insulin uptake (higher dosage) has led to an enhancement in insulin demand. Although insulin has been expressed in Pseudomonas fluorescens-based expression system, medium optimization, a crucial step in bioprocess for product titer and yield improvement, has not been reported. In this study, medium components were screened using the Placket-B

Hybrid insulin peptides (HIPs) formed through covalent cross-linking of proinsulin fragments to secretory granule peptides are detectable within murine and human islets. The 2.5HIP (C-peptide-chromogranin A [CgA] HIP), recognized by the diabetogenic BDC-2.5 clone, is a major autoantigen in the nonobese diabetic mouse. However, the relevance of this epitope in human disease is currently unclear. A recent study probed T-cell reactivity toward HIPs in patients with type 1 diabetes, documenting responses in one-third of the patients and isolating several HIP-reactive T-cell clones. In this study,
