Nanoplastics (NPs) are emerging global contaminants that can exacerbate the animal toxicity and cytotoxicity of cadmium (Cd). However, the mechanisms by which NPs influence the toxic effects of Cd on key functional proteins within the body remain unknown. In this study, trypsin, a protein that is prone to coexist with NPs in the digestive tract, was selected as the target protein. The effects and mechanisms of NPs on Cd2+-induced structural damage at multiple levels and alterations in the biological function of trypsin were investigated using multi-spectroscopy techniques, enzyme activity assa

Personalized protein corona significantly influences the biodistribution and therapeutic efficacy of nanomedicines, generating unique profiles that can impact treatment outcomes. Here, we demonstrate that pegylated liposomal doxorubicin (PLD) exhibits increased tumor accumulation and enhanced antitumor immunity in obese mice bearing ovarian tumor, inducing a greater capacity to inhibit tumor growth compared to normal mice. Mechanistically, the protein corona, particularly enriched with complement component 1q (C1q) in the plasma of obese mice, significantly enhances the internalization of PLD

Nanoparticle rigidity influences cellular uptake and in vivo behavior, yet the underlying mechanisms remain unclear. Here we used lipids with identical head groups but different acyl chains to prepare liposomes with comparable surface composition, size and zeta potential, but distinct mechanical properties. Rigid liposomes showed much higher uptake than soft ones, mediated by distinct endocytic mechanisms. Proteomic analysis revealed that rigid and soft liposomes adsorbed very different coronas upon dispersion in serum. RNA interference and competition studies indicated that the enhanced uptak

The architecture of a nanoparticles' surface formed due to a modification with a ligand and protein corona formation in biofluids is critical for interactions with cells in vivo. Here we studied interactions of immune cells with magnetic nanoparticles (MNPs) covalently modified with polyethylene glycol (PEG) and their counterparts conjugated with peptides: a pH (low) insertion peptide (pHLIP) and cycloRGD as a targeting ligand in human serum. The conjugation of MNPs-PEG with pHLIP, but not with cycloRGD, enhanced the association of these particles with mononuclear phagocytic cells in vitro and

The protein corona formed upon systemic administration critically modulates the pharmacokinetics, biodistribution, and therapeutic efficacy of the nanomedicines. While emerging evidence links obesity to heightened chemosensitivity, the underlying nanobio-interfacial mechanisms remain poorly understood. Herein, we demonstrate that pegylated liposomal doxorubicin (PLD) exhibits significantly enhanced antitumor and antimetastatic efficacy in obese breast tumor-bearing mice compared to normal controls. Mechanistic investigations reveal that obesity confers PLD with prolonged systemic circulation a

In this study, protein/polysaccharide core-shell-corona (PP-CSC) nano delivery systems (NDS) were prepared by using bovine serum albumin (BSA) and chitosan (CS). The potential of PP-CSC-NDS for increasing the stability and bio-accessibility of ε-poly-l-lysine (ε-PL) as effective therapy for gastric Helicobacter pylori was investigated. The presence of CS-shell increased the size (223 ± 1.7 nm) of BSA-core as compared to BSA-shell on CS-core (191 ± 2.6 nm). Conversely, encapsulation efficiency of ε-PL was reduced for C(B)NDS [CS-shell on BSA-core] to 73 ± 1% than 82 ± 2% for B(C)NDS [BSA-shell

The surface of proteins is heterogeneous with sophisticated but precise hydrophobic and hydrophilic patches, which is essential for their diverse biological functions. To emulate such distinct surface patterns on macromolecules, we used rigid spherical synthetic dendrimers (polyphenylene dendrimers) to provide controlled amphiphilic surface patches with molecular precision. We identified an optimal spatial arrangement of these patches on certain dendrimers that enabled their interaction with human adenovirus 5 (Ad5). Patchy dendrimers bound to the surface of Ad5 formed a synthetic polymer coro

Gene therapy holds great potential for treating almost any disease by gene silencing, protein expression, or gene correction. To efficiently deliver the nucleic acid payload to its target tissue, the genetic material needs to be combined with a delivery platform. Lipid nanoparticles (LNPs) have proven to be excellent delivery vectors for gene therapy and are increasingly entering into routine clinical practice. Over the past two decades, the optimization of LNP formulations for nucleic acid delivery has led to a well-established body of knowledge culminating in the first-ever RNA interference

Nanoparticles in physiological environments acquire a biomolecular corona that defines their biological identity, mediating immune system recognition and accelerating blood clearance of the nanoparticles. Typically, low-fouling materials are chosen to minimize protein adsorption and thereby immune system responses, contributing to stealth in blood. However, absolute prevention of the biomolecular corona remains tantalizingly out of reach. Herein, it is proposed to leverage the biomolecular corona rather than preventing its formation, in order to overcome immune responses toward nanoparticles.

The affinity of zeolite nanoparticles (diameter of 8-12 nm) possessing high surface area and high pore volume towards human plasma proteins has been investigated. The protein composition (corona) of zeolite nanoparticles has been shown to be more dependent on the plasma protein concentrations and the type of zeolites than zeolite nanoparticles concentration. The number of proteins present in the corona of zeolite nanoparticles at 100% plasma (in vivo state) is less than with 10% plasma exposure. This could be due to a competition between the proteins to occupy the corona of the zeolite nanopar

Nanoparticles entering the human body instantly become coated with a "protein corona" that influences the effects and distribution of the particles in vivo. Yet, whether nanoparticles may bind to other organic compounds remains unclear. Here we use an untargeted metabolomic approach based on ultra-performance liquid chromatography and quadruple time-of-flight mass spectrometry to identify the organic compounds that bind to mineral nanoparticles formed in human body fluids (serum, plasma, saliva, and urine). A wide range of organic compounds is identified, including fatty acids, glycerophosphol

Nanoparticle-enhanced laser-induced breakdown spectroscopy (NELIBS) is an optical emission technique based on the laser-induced plasma (LIP) on a sample after the deposition of plasmonic nanoparticles (NPs) on its surface. The employment of the NPs allows an enhancement of the signal with respect to the one obtained with the conventional laser-induced breakdown spectroscopy (LIBS) enabling an extremely high sensitivity and very low limits of detection compared with the LIBS performance. Recently, NELIBS was used for monitoring the NP protein corona formation. As a matter of fact, the NPs in th

Surface functionalization of nanoparticles with polyethylene glycol (PEG) has been widely demonstrated as an anti-opsonization strategy to reduce protein corona formation which is one of the major concerns affecting target receptor recognition. However, excessive surface passivation with PEG can lead to the strong inhibition of cellular uptake and less efficient binding to target receptors, resulting in reduced potential of targeted delivery. To improve specific cell targeting while reducing the nonspecific protein adsorption, a secondary packaging of the nanoparticles with shorter PEG chains,

Infants may ingest large quantities of micro- and nanoplastics (MNP) due to the heating and sterilization of the feeding bottles, which promotes the generation and migration of MNP into the milk or milk formula. The effect of MNP on the digestion of proteins, which are crucial for the growth and development of infants remain unknown. The current study investigated the in vitro digestion of cow's milk proteins (1 mg/mL) with or without polypropylene (PP) microplastics (MP, pristine or oxidized) (20 mg/mL and 63-180 μm) in simulated gastric fluids (SGF) using an infant (pH 5.0; pepsin activity;

Engineered albumin interfaces can modify the biological identity and performance of non-albumin nanoparticles in cancer nanomedicine, but their independent contribution is often obscured by multicomponent designs. This critical review evaluates studies in which albumin or an albumin-derived construct was deliberately positioned at the outer interface of a non-albumin nanoparticle or drug nanocrystal before biological exposure. Architectures included adsorbed layers, covalent or crosslinked coatings, albumin-stabilized nanocrystal interfaces, and hybrid or modified constructs. Across 35 primary

Activated platelets provide a promising target for imaging inflammatory and thrombotic events along with site-specific delivery of a variety of therapeutic agents. Multifunctional protein micelles bearing targeting and therapeutic proteins were now obtained by one-pot transpeptidation using an evolved sortase A. Conjugation to the corona of a single-chain antibody (scFv), which binds to the ligand-induced binding site (LIBS) of activated GPIIb/IIIa receptors, enabled the efficient detection of thrombi. The inhibition of thrombus formation was subsequently accomplished by incorporating the cata

Controlling particle-level heterogeneity arising from postformation modification of biomolecular coronas remains a central challenge in bionano interaction studies. Variability in corona modifications can alter the biological identity of individual nanoparticles (NPs), compromising the accuracy, reproducibility, and interpretability of both in vivo and in vitro experiments. Despite its significant impact, particle-level uniformity is rarely measured directly. This issue is particularly critical for corona fluorescent labeling, where bulk fluorescence readouts can suggest efficient modification

Environmental pollution with plastic polymers has become a global problem, leaving no continent and habitat unaffected. Plastic waste is broken down into smaller parts by environmental factors, which generate micro- and nanoplastic particles (MNPPs), ultimately ending up in the human food chain. Before entering the human body, MNPPs make their first contact with saliva in the human mouth. However, it is unknown what proteins attach to plastic particles and whether such protein corona formation is affected by the particle's biophysical properties. To this end, we employed polystyrene MNPPs of t

The adsorption of serum proteins on biomaterial surfaces is a critical determinant for the outcome of medical procedures and therapies, which involve inserting materials and devices into the body. In this study, we aimed to understand how surface topography at the nanoscale influences the composition of the protein corona that forms on the (bio)material surface when placed in contact with serum proteins. To achieve that, we developed nanoengineered model surfaces with finely tuned topography of 16, 40, and 70 nm, overcoated with methyl oxazoline to ensure uniform outermost chemistry across all
