Harnessing reactive oxygen species for precision medicine: ROS-Activatable PROTACs for lung Cancer.

Reactive oxygen species (ROS) play a pivotal role in maintaining cellular balance, functioning both as essential messengers in signaling pathways and as agents of oxidative damage. In cancer their influence is paradoxical: moderate ROS levels can foster tumor growth, whereas excessive accumulation triggers cell death. In lung cancer-the foremost cause of cancer-related deaths globally-disrupted ROS regulation contributes to DNA instability, abnormal pathway activation, and therapeutic resistance. Traditional approaches that manipulate ROS, such as antioxidant treatments, enzyme inhibition, or

Methotrexate-based PROTACs as DHFR-specific chemical probes.

Methotrexate (MTX) is a tight-binding dihydrofolate reductase (DHFR) inhibitor, used as both an antineoplastic and immunosuppressant therapeutic. MTX, like folate undergoes folylpolyglutamate synthetase-mediated γ-glutamylation, which affects cellular retention and target specificity. Mechanisms of MTX resistance in cancers include a decrease in MTX poly-γ-glutamylation and an upregulation of DHFR. Here, we report a series of potent MTX-based proteolysis targeting chimeras (PROTACs) to investigate DHFR degradation pharmacology and one-carbon biochemistry. These on-target, cell-active PROTACs s

The synthetic landscape of cereblon (CRBN) binders: from thalidomide to emerging chemotypes.

Thalidomide, initially marketed as a sedative and anti-emetic, was withdrawn in the 1960s due to catastrophic teratogenic effects. Decades later, thalidomide was reconsidered as an anticancer agent, resulting in FDA approval against multiple myeloma. This success spurred the development of structural analogs lenalidomide and then pomalidomide as cancer therapies (immunomodulatory imide drugs, IMiDs). Subsequent landmark studies have revealed that IMiDs act as molecular glue degraders, co-opting the E3 ubiquitin ligase cereblon (CRBN) to induce the polyubiquitination and proteasomal degradation

Glycoengineered Host-Guest Nanoparticles Potentiate Alzheimer's Disease Therapy via Lesion-Specific Modulation of Tau Pathology.

Tau pathology is a principal driver of cognitive impairment in Alzheimer's disease (AD), but the therapeutic targeting of tau has been hindered by poor brain delivery and a lack of lesion-confined activity. Here, we delineate a pathogenic cascade wherein the impaired dephosphorylation of hyperphosphorylated tau (p-tau) leads to its aggregation, which is amplified by microglia-mediated propagation. To combat this p-tau cascade, we developed a glycoengineered proteolysis targeting chimera (PROTAC) nanoparticle for lesion-specific p-tau modulation therapy. We first synthesized a library of p-tau

Vepdegestrant: the first FDA-approved oral PROTAC and a milestone in targeted protein degradation.

Vepdegestrant (ARV-471, Veppanu) represents an important milestone in targeted protein degradation (TPD) as the first FDA-approved heterobifunctional proteolysis-targeting chimera (PROTAC). Designed to selectively degrade estrogen receptor α (ERα) through recruitment of the cereblon E3 ubiquitin ligase, vepdegestrant provides an event-driven approach to targeting ERα, including clinically relevant estrogen receptor-1 (ESR1)-mutated forms associated with endocrine therapy resistance. Its oral bioavailability and pharmacological properties enabled clinical development in patients with ER-positiv

An Oral PROTAC Targeting HPK1 Degradation Potentiates Anti-Solid Tumor Immunity.

Hematopoietic progenitor pinase1 (HPK1) knockout has been identified as an efficient route to enhance anti-tumor immune response. Here, this work develops an oral proteolysis targeting chimera (PROTAC) targeting HPK1 to efficiently and selectively degrade HPK1 to augment immunotherapeutic outcomes. In a postoperative tumor model of human cervical cancer in NSG mice, the orally-administrated PROTAC can reach tumors, down-regulate HPK1 levels in locally-administrated CAR-T cells, and promote their efficiency in inhibiting solid tumor recurrence, achieving 50% partial response (PR) and 50% comple

PhenoDEL: A Novel Screening Strategy Based on Intracellular Protein Degradation Activity.

Targeted protein degradation (TPD), including proteolysis targeting chimeras (PROTACs) and molecular glue degraders (MGDs), is a promising therapeutic approach. However, systematic discovery of such small molecules remains a major challenge. Here, we present PhenoDEL, a novel phenotypic DNA-encoded library (DEL) screening platform that integrates one-bead one-compound DEL (OBOC-DEL) with the Beacon optofluidic system for single-cell analysis. By coculturing individual OBOC-DEL beads and engineered reporter cells in nanoliter-scale chambers, PhenoDEL enables time-resolved, single-cell phenotypi

Discovery of an Androgen Receptor Degrader Featuring a Pyridazinyl Glutarimide CRBN-Binding Motif for Transdermal Treatment of Androgenetic Alopecia.

Androgenetic alopecia (AGA) is primarily driven by overexpression of androgen receptor (AR) in hair follicles, leading to androgen hypersensitivity and progressive follicular miniaturization. Although PROTAC degraders offer the potential for sustained AR downregulation, their application in AGA has been limited by poor solubility and skin permeability. In this study, we incorporated pyridazinyl glutarimide (PDG) moiety as a novel CRBN ligand and developed dAR-6-1, a PROTAC with both potent AR degradation activity (DC50 = 0.90 nM, Dmax = 91% in LNCaP; DC50 = 0.23 nM, Dmax = 90% in hDPC) and exc

Dual Targeting of Nucleotidase-Dependent and -Independent Functions via PROTAC-Mediated CD73 Degradation.

CD73 (ecto-5'-nucleotidase) drives immunosuppressive and tumor progression through both enzymatic adenosine production and nonenzymatic mechanisms, limiting the efficacy of current CD73-targeted therapies, including small-molecule enzymatic inhibitors and antibodies. Here, we reported NUCC-0227579 (C79), a first-in-class proteolysis-targeting chimera (PROTAC) that degraded CD73 via the VHL E3 ligase-dependent proteasomal and lysosomal pathways. C79 eliminated CD73 at the cell surface and intracellular compartments across multiple human cancer cell lines, abolishing nucleotidase activity and mo

Targeted degradation of kidney injury molecule 1 ameliorates acute kidney injury.

Acute kidney injury (AKI) is a global public health issue with a high incidence and mortality rate, without effective treatments currently available. Kidney injury molecule 1 (KIM1), an FDA-approved biomarker for AKI, is dramatically induced on the membrane of injured renal tubular epithelial cells. However, whether it works as a therapeutic target remains unclear. Herein, by using a modified Proteolysis Targeting Chimera (PROTAC) strategy, we constructed KIM1-targeted degradable liposome (KIM1-TDL) by conjugating a KIM1-targeting peptide TKP4 that we previously designed, and an E3 ligase liga

Click. Screen. Degrade. A Miniaturized D2B Workflow for Rapid PROTAC Discovery.

Targeted protein degradation is one of the fastest developing fields in medicinal chemistry and chemical biology. Despite significant development in assay technologies and inhibitor discovery, the development of PROTACs remains a challenging endeavor since rational design approaches remain widely elusive. Our workflow eliminates the rate-limiting step of classic synthesis, namely compound purification, and pairs it with high-throughput, semi-automated plate-based synthesis, and direct cellular assay evaluation. We applied this direct-to-biology approach to four diverse targets, demonstrating t

Dual-function PROTAC suppresses ferroptosis and restores neuronal function via brain-targeted delivery.

Targeted protein degradation (TPD) via proteolysis-targeting chimeras (PROTACs) offers a promising strategy for modulating disease-associated proteins, yet effective brain-preferred protein degradation remains challenging. Herein, we report a dual-function PROTAC, dACSL4, and its nose-to-brain delivery for brain-preferred protein degradation and therapeutic suppression of ferroptosis in neurodegeneration. dACSL4 selectively degrades acyl-CoA synthetase long-chain family member 4 (ACSL4) while concurrently activating peroxisome proliferator-activated receptor γ (PPARγ), thereby coordinating lip

Targeting Bruton's Tyrosine Kinase in CLL: Selectivity, Resistance Mechanisms, and Emerging Therapeutic Strategies.

Kinases are highly explored drug targets due to their central role in cell growth, differentiation, and cell death, as well as their relationship to cancer initiation and progression. However, with more than 500 kinases in the human kinome and significant structural similarities among them, kinases present a major selectivity challenge. Bruton's tyrosine kinase (BTK), a highly studied kinase, plays an important role in the B-cell receptor pathway. Overexpression of BTK in B cells has been linked to the development of B-cell lymphomas, like chronic lymphocytic leukemia (CLL), as well as certain

A Novel PROTAC Confers a Dual Benefit Against Amyloid and Tau Pathology in Alzheimer's Disease via DAPK1 Degradation.

Alzheimer's disease (AD) is a neurodegenerative disorder that is caused by multiple factors, characterized by a progressive decline in cognitive ability, extracellular amyloid-β (Aβ) plaques, and intracellular neurofibrillary tangles composed of hyperphosphorylated tau. Current treatment strategies can provide only symptomatic treatment or limited efficacy, highlighting the need to intervene in the upstream regulatory factors that drive both amyloid and tau pathologies. Death-associated protein kinase 1 (DAPK1) is a key driver upstream of both amyloid precursor protein processing and tau phosp

Vepdegestrant: First Approval.

Vepdegestrant (VEPPANU; formerly ARV 471) is a novel PROteolysis TArgeting Chimera (PROTAC) that is able to simultaneously bind the estrogen receptor (ER) and an E3 ubiquitin ligase complex, resulting in polyubiquitination of the ER and its subsequent degradation via a proteasome. It is being developed by Arvinas and Pfizer primarily as a treatment for ER+, HER2- breast cancer. In May 2026, vepdegestrant received its first global approval in the USA for adults with ER+, HER2-, ESR1-mutated advanced or metastatic breast cancer, as detected by an FDA-authorized test, with disease progression fol

Discovery of a Potent and Selective KRAS G12D Degrader based on PROTAC Degradation.

KRAS-G12D has long been regarded as an intractable therapeutic target due to the flat binding pocket and its strong affinity for GTP/GDP. Proteolysis-targeting chimera (PROTAC) is a revolutionary drug discovery strategy that, by virtue of its unique pharmacological mode of action, provides more options for targeting undruggable targets. In this study, we designed and synthesized 20 novel KRAS G12D PROTACs based on the MRTX1133 derivative. Through systematic exploration of linker structure-activity relationship and multi-cell line screening, compound VI-1 exhibited significant KRAS G12D degrada

Intracellular sclerostin promotes tumor progression and metastasis as a potential therapeutic target in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) urgently requires promising therapeutic targets. This study identifies sclerostin, an osteocyte-derived secretory protein traditionally linked to bone homeostasis, as an unexpected intracellular oncogenic driver in TNBC. Although genetic ablation of sclerostin markedly suppresses tumor progression and lung metastasis, neither its antibody nor recombinant protein exerts any effects, excluding the role of extracellular sclerostin in TNBC. Genetic and pharmacological approaches (sclerostin aptamer-based proteolysis-targeting chimera with potent intracellular s

De novo pyrimidine biosynthesis inhibition synergizes with BCL-XL targeting in pancreatic cancer.

Oncogenic KRAS induces metabolic rewiring in pancreatic ductal adenocarcinoma (PDAC) characterized, in part, by dependency on de novo pyrimidine biosynthesis. Pharmacologic inhibition of dihydroorotate dehydrogenase (DHODH), an enzyme in the de novo pyrimidine synthesis pathway, delays pancreatic tumor growth; however, limited monotherapy efficacy suggests that compensatory pathways may drive resistance. Here, we use an integrated metabolomic, proteomic and in vitro and in vivo DHODH inhibitor-anchored genetic screening approach to identify compensatory pathways to DHODH inhibition (DHODHi) an

A General Photoredox Platform for the Hydroarylation of Olefins.

Alkene hydroarylation provides a direct strategy for merging aryl groups with alkene-derived C(sp3)-rich fragments, yet general methods that employ abundant aryl bromides across diverse alkene classes remain elusive. Herein, we report XAT-Heck, a radical hydroarylation platform that couples aryl bromides with alkenes under mild photoredox conditions through halogen-atom transfer (XAT)-enabled aryl radical generation. The method accommodates terminal, internal, and gem-disubstituted alkenes, as well as a broad range of heteroaryl and aryl bromides. Chemical-space mapping further supports the br

Precision Ferroptosis Amplification via SLC7A11-Directed Proteasomal Degradation for Enhanced Cancer Therapy.

Ferroptosis is a regulated cell death pathway driven by the iron-dependent accumulation of reactive oxygen species (ROS) and lipid hydroperoxides. A major factor limiting the effectiveness of ferroptosis induction is the antioxidant activity of glutathione peroxidase 4 (GPX4). Herein, we reported SLC7A11-targeting proteolysis targeting chimeras (PROTACs) to deplete GPX4 and to augment oxidative stress within cancer cells. A bifunctional PROTAC, namely dSLC7A11, ws designed by conjugating the SLC7A11 inhibitor sulfasalazine to the CRBN ligand pomalidomide via an alkyl linker. The rational desig
