In patients with cardiovascular abnormalities or immunological disorders, an increased number of circulating leukocyte-platelet aggregates is observed. Leukocyte-platelet aggregates play an essential role in linking the hemostatic and immune systems. High shear stress and pro-coagulant and pro-inflammatory stimulants are known to activate platelets and promote the formation of aggregates. Pulsatile blood flow under low shear stress can also induce platelet activation in comparatively mild conditions. However, the effect of such events on leukocyte-platelet aggregates has not yet been investiga

BACKGROUND: Cerebral small vessel disease (SVD) causes up to 45% of dementias and 25% of ischemic strokes, but the understanding of vascular pathophysiology is limited. We aimed to investigate the contribution of pulsatility of intracranial arteries, veins, and cerebrospinal fluid (CSF) and cerebral blood flow to long-term imaging and clinical outcomes in SVD.
METHODS: We prospectively recruited participants in Edinburgh/Lothian, Scotland, with lacunar or nonlacunar ischemic stroke (modified Rankin Scale score ≤2, as controls) and assessed medical and brain magnetic resonance imaging characte

Arterial walls stiffen with age, cardiovascular risk factors, and various vascular diseases, which may lead to less damping of the arterial blood flow pulse, subsequent microvascular damage, and enlarged perivascular spaces (PVS). However, the exact interplay between these processes is unclear. This study aimed to investigate the relation between blood flow velocity pulsatility in the small lenticulostriate arteries and their supplying middle cerebral artery and the respective damping factor (DF), with the number of MRI-visible PVS in elderly subjects. Blood flow velocity waveforms were obtain

A great need exists for the development of a more representative in-vitro model to efficiently screen novel thrombolytic therapies. We herein report the design, validation, and characterization of a highly reproducible, physiological scale, flowing clot lysis platform with real-time fibrinolysis monitoring to screen thrombolytic drugs utilizing a fluorescein isothiocyanate (FITC)-labeled clot analog. Using this Real-Time Fluorometric Flowing Fibrinolysis assay (RT-FluFF assay), a tPa-dependent degree of thrombolysis was observed both via clot mass loss as well as fluorometrically monitored rel

We study the dynamics of a binary fluid, where the two fluids are flowing parallel to each other in a cylindrical geometry, and driven by a pulsatile pressure gradient. One of the fluids is a low viscosity Newtonian fluid, the other one is viscoelastic. In order to be able to apply the model to different biofluids, we consider that the viscoelastic fluid has several characteristic times. We characterize the dynamics of the fluids as generalized Darcy's laws, with linear response functions to pulsatile pressure gradients, whose parameters are coupled for both fluids through the fluid-fluid boun

OBJECTIVE: Transcranial Doppler sonography (TCD) can detect visual evoked blood flow responses non-invasively and continuously with high temporal resolution. While the mean flow velocity response to visual stimuli is well documented, the response of the pulsatile characteristics of the waveform is less well known.
METHOD: We examined the changes of blood flow velocity and pulsatility index (PI) in the posterior cerebral artery (PCA) in 50 healthy volunteers. TCD responses were measured in response to metabolic activation of the visual cortex by visual stimulation.
RESULTS: A specific, stimul

OBJECTIVES: To determine gestational age-specific reference ranges and develop a Z-score equation for the fetal ductal pulsatility index (PI), between 24 and 33 + 6 weeks of gestation.
METHODS: A prospective cross-sectional study conducted from 2023 to 2024, 226 singleton fetuses between 24 and 33 + 6 weeks were enrolled. The ductus arteriosus (DA) was assessed in the longitudinal plane of the fetus using color Doppler to confirm its course and flow direction, and pulsed Doppler sampling was placed at the proximal DA with an insonation angle <15°. Three consecutive measurements of PI were rec

The flow convergence method includes calculation of the proximal isovelocity surface area (PISA) and is widely used to classify mitral regurgitation (MR) with echocardiography. It constitutes a primary decision factor for determination of treatment and should therefore be a robust quantification method. However, it is known for its tendency to underestimate MR and its dependence on user expertise. The present work systematically compares different pulsatile flow profiles arising from different regurgitation orifices using transesophageal echocardiographic (TEE) probe and particle image velocim

Nonsurgical bleeding occurs in a significant proportion of patients implanted with continuous-flow ventricular assist devices (CF-VADs) and is associated with nonphysiologic flow with diminished pulsatility. An in vitro vascular pulse perfusion model seeded with adult human aortic endothelial cells (HAECs) was used to identify biomarkers sensitive to changes in pulsatility. Diminished pulsatility resulted in an ~45% decrease in von Willebrand factor (vWF) levels from 9.80 to 5.32 ng/ml (n = 5, p < 0.05) and a threefold increase in angiopoietin-2 (ANGPT-2) levels from 775.29 to 2471.93 pg/ml (n

Our current understanding of flow through the circuit of left ventricular assist device (LVAD), left ventricle and ascending aorta remains incompletely understood. Computational fluid dynamics, which allow for analysis of flow in the cardiovascular system, have been used for this purpose, although current simulation models have failed to fully incorporate the interplay between the pulsatile left ventricle and continuous-flow generated by the LVAD. Flow-through the LVAD is dependent on the interaction between device and patient-specific factors with suboptimal flow patterns evoking increased ri

BACKGROUND: Bleeding is an important cause of morbidity and mortality in patients with continuous-flow left ventricular assist devices (LVADs). Reduced pulsatility has been implicated as a contributing cause. The aim of this study was to assess the effects of different degrees of pulsatility on the incidence of nonsurgical bleeding.
METHODS AND RESULTS: The Utah Transplantation Affiliated Hospitals (U.T.A.H.) heart failure and transplant program databases were queried for patients with end-stage heart failure who required support with the continuous-flow LVAD HeartMate II (Thoratec Corp, Plea

Aortic stiffness increases with age and vascular risk factor exposure and is associated with increased risk for structural and functional abnormalities in the brain. High ambient flow and low impedance are thought to sensitize the cerebral microcirculation to harmful effects of excessive pressure and flow pulsatility. However, haemodynamic mechanisms contributing to structural brain lesions and cognitive impairment in the presence of high aortic stiffness remain unclear. We hypothesized that disproportionate stiffening of the proximal aorta as compared with the carotid arteries reduces wave re

The use of experimental relations to approximate the efficient thermophysical properties of a nanofluid (NF) with Cu nanoparticles (NPs) and hybrid nanofluid (HNF) with Cu-SWCNT NPs and subsequently model the two-dimensional pulsatile Casson fluid flow under the impact of the magnetic field and thermal radiation is a novelty of the current study. Heat and mass transfer analysis of the pulsatile flow of non-Newtonian Casson HNF via a Darcy-Forchheimer porous channel with compliant walls is presented. Such a problem offers a prospective model to study the blood flow via stenosed arteries. A fini

The hemodynamics of elastic cerebral aneurysms are complicated by phenomena that affect the initiation and the progress of each aneurysm. The blood vessel deforms with pulsatile flow. In a phantom, however, it remains unclear whether the wall compliance can be neglected. In our previous study, the flow structure at another plane oriented perpendicular to the median plane was not clarified. In the approach presented here, an identical phantom is used for both the rigid and elastic wall models by adjusting the surrounding fluid when immersed in a bath. For this purpose, the full-scale phantom of

Aortic aneurysm is one of the most common aortic diseases that can lead to unfortunate consequences. Numerical simulations have an important role in the prediction of the aftereffects of vascular diseases including aneurysm. In this research, numerical simulation of pulsatile blood flow is performed for a 3-dimensional patient-specific model of a thoracic aortic aneurysm (TAA). Since the choice of blood viscosity model may have a significant impact on the simulation results, the effects of four non-Newtonian models of blood viscosity namely Carreau, Casson, Herschel-Bulkley, power low, and the

INTRODUCTION: Veno-arterial extracorporeal life support (ECLS) is increasingly being used to treat rapidly progressing or severe cardiogenic shock. However, it has been repeatedly shown that increased afterload associated with ECLS significantly diminishes left ventricular (LV) performance. The objective of the present study was to compare LV function and coronary flow during standard continuous-flow ECLS support and electrocardiogram (ECG)-synchronized pulsatile ECLS flow in a porcine model of cardiogenic shock.
METHODS: Sixteen female swine (mean body weight 45 kg) underwent ECLS implantati

With the advent of noninvasive clinical techniques which can measure blood flow velocities (Doppler ultrasound), it is suggested that a fundamental knowledge of the axial flow velocity patterns in the pulmonary artery, and the changes caused by stenosis, may be used to support accurate diagnosis of valvular pulmonic stenosis. The present study was designed to characterize the axial flow velocity patterns in an in vitro model of a human adult pulmonary artery with varying degrees of valvular pulmonic stenosis. A two-dimensional laser Doppler anemometer (LDA) system was used to map the flow fiel
