Nutritional Factors That Affect the Formation of 5-Aminolevulinic Acid, a Key Intermediate of Heme Biosynthesis.

5-Aminolevulinic acid (ALA) is a key intermediate of heme biosynthesis, which is an essential component of the respiratory chain. Therefore, nutrients that affect ALA biosynthesis eventually affect ATP production, which is the basis of mitochondrial function. Although the effects of various non-nutrient components that affect ALA after biosynthesis have been reported, there are few reports on the effects of dietary amino acids/protein on ALA formation and the effects of dietary vitamins that are involved in amino acid metabolism. In mitochondria, ALA is synthesized from succinyl-CoA and glycin

Association between vitamin B6 metabolism and the prevalence of age-related macular degeneration: A cross-sectional analysis of National Health and Nutrition Examination Survey (NHANES).

B vitamins may be involved in the development of age-related macular degeneration (AMD). This investigation intended to examine the association between vitamin B6 catabolic status and the prevalence of AMD. Individuals ≥ 40 years old in the 2005 to 2008 National Health and Nutrition Examination Survey who had complete vitamin B6 measurement and AMD diagnostic data were included in this cross-sectional study. The catabolic status of vitamin B6 was evaluated using serum levels of pyridoxal 5'-phosphate (PLP) (the active form) and its metabolite 4-pyridoxic acid (4-PA). The analysis of 4-PA, PLP,

Vitamin B6 catabolism and psoriasis risk: a cross-sectional study.

BACKGROUND: Psoriasis is a common autoimmune inflammatory disease. Vitamin B6 is crucial for the body's inflammatory response, yet the relationship between 4-pyridoxic acid (4-PA), pyridoxal 5'-phosphate (PLP) and vitamin B6 turnover (4-PA/PLP) in psoriasis remains unexplored. OBJECTIVES: To investigate the relationship of 4-PA, PLP and vitamin B6 catabolism with the risk of psoriasis. METHODS: This cross-sectional study analysed records for 7540 participants from the US National Health and Nutrition Examination Survey. Vitamin B6 catabolism was assessed via the serum 4-PA to PLP ratio (4-PA

Postpartum fluoxetine increased maternal inflammatory signalling and decreased tryptophan metabolism: Clues for efficacy.

Perinatal depression (PND) affects approximately 15% of women, and de novo postpartum depression affects approximately 40% of PND cases. Selective serotonin reuptake inhibitors (SSRIs) are a common class of antidepressants prescribed to treat PND. However, the safety and efficacy of SSRIs have been questioned in both clinical and preclinical research. Here, using a preclinical rodent model of de novo postpartum depression, we aim to better understand neuroinflammatory cytokines and tryptophan mechanisms that may be related to SSRI efficacy. Rat dams were treated with high corticosterone (CORT;

Nephrotoxicity of S-(2-chloro-1,1,2-trifluoroethyl)glutathione and S-(2-chloro-1,1,2-trifluoroethyl)-L-cysteine, the glutathione and cysteine conjugates of chlorotrifluoroethene.

The glutathione and cysteine conjugates of the nephrotoxin chlorotrifluoroethene, S-(2-chloro-1,1,2-trifluoroethyl)glutathione (CTFG) and S-(2-chloro-1,1,2-trifluoroethyl)cysteine (CTFC), are potent nephrotoxins in male rats. Morphological changes in the kidneys were observed 1.5 hr after giving 100 mumol/kg of CTFG (i.v.), and severe damage to the proximal tubules was evident 24 hr after treatment; this dose of CTFG caused a 100-fold increase in urine glucose excretion, a 10-fold increase in urine protein excretion and a 4-fold increase in blood urea nitrogen concentrations 24 hr after admini

Direct Asymmetric α-C Conjugate Addition of Aminomethylphosphonate Enabled by Carbonyl/Iminium Double Organoactivation.

As important bioisosteres of α-amino acids, chiral α-aminophosphonic acids exhibit broad bioactivities and are widely employed in pharmaceuticals. Direct asymmetric α-C conjugate addition of aminomethylphosphonate to α,β-unsaturated ketones is an intriguing strategy to make bioactive chiral phosphonic 1-pyrrolines. However, this transformation remains challenging due to the low α-C-H acidity of the aminomethylphosphonate, significant steric hindrance from the phosphonate moiety and interference by side reactions. In this work, we report the direct enantioselective α-C conjugate addition of NH2

Characterization of a haemolytic factor from Candida albicans.

The culture supernatant of Candida albicans promoted the disruption of human red blood cells (RBCs). The haemolytic activity was detected in a sugar-rich fraction (about 200 kDa) from Sephacryl S-100 chromatography. As the haemolytic activity was adsorbed by concanavalin A-Sepharose, the haemolytic factor may be a mannoprotein. The activity was inactivated by periodate oxidation, indicating that the sugar moiety of the mannoprotein played an important role in the haemolysis. The structure of the sugar moiety of the mannoprotein was identified as a cell-wall mannan by 1H-NMR analysis, and purif

Toward an Understanding of the Mechanistic Differences Between the Hydroxylating and Desaturating Pyridoxal 5'-Phosphate-Dependent L-Arginine Oxidases.

Pyridoxal-5'-phosphate (PLP)-dependent L-arginine oxidases (PDAOs) catalyze 4-electron oxidations of L-arginine in the context of secondary metabolite biosynthesis. PDAOs are separated into two classes based on whether they hydroxylate or desaturate L-arginine. A detailed mechanistic understanding of these enzymes will extend our knowledge of natural products biosynthesis. Here, we present detailed transient kinetic analyses of representative hydroxylating and desaturating PDAOs: MppP from Streptomyces wadayamensis (SwMppP) and the Ind4 homolog from Pseudoalteromonas luteoviolacea (Plu4). Crystal Structure and Functional Characterization of Acetylornithine Aminotransferase from Corynebacterium glutamicum.

The amino acids l-arginine and l-ornithine are widely used in animal feed and as health supplements and pharmaceutical compounds. In arginine biosynthesis, acetylornithine aminotransferase (AcOAT) uses pyridoxal-5'-phosphate (PLP) as a cofactor for amino group transfer. Here, we determined the crystal structures of the apo and PLP complex forms of AcOAT from Corynebacterium glutamicum (CgAcOAT). Our structural observations revealed that CgAcOAT undergoes an order-to-disorder conformational change upon binding with PLP. Additionally, we observed that unlike other AcOATs, CgAcOAT exists as a tet

Epilepsy in inborn errors of metabolism.

Epilepsies associated with inborn errors of metabolism (IEM) represent a major challenge. Seizures rarely dominate the clinical presentation, which is more frequently associated with other neurological symptoms, such as hypotonia and/or cognitive disturbances. Although epilepsy in IEM can be classified in various ways according to pathogenesis, age of onset, or electroclinical presentation, the most pragmatic approach is determined by whether they are accessible to specific treatment or not. The main potentially treatable causes comprise vitamin B6 (pyridoxine deficiency), biotine, and GLUT1 d

Evidence for increased catabolism of vitamin B-6 during systemic inflammation.

BACKGROUND: Plasma concentrations of PL 5'-phosphate (PLP), which is the active coenzyme form of vitamin B-6, are reduced during inflammation. The underlying mechanisms may include altered tissue distribution or increased catabolism via pyridoxal (PL) to pyridoxic acid (PA). Recently, we showed that catabolic enzyme activity could be assessed by substrate product ratios measured in plasma. OBJECTIVE: We evaluated the ratios PA:PL, PA:PLP, and PA:(PL + PLP) as possible markers of vitamin B-6 catabolism. DESIGN: Cross-sectional and longitudinal data were derived from the Western Norway B-Vitam

Cycloserine inhibits glutamate decarboxylase from Mycobacterium tuberculosis.

Glutamate decarboxylase (GadB) is a pyridoxal phosphate (PLP)-dependent enzyme that contributes to intracellular pH homeostasis and supports the survival of Mycobacterium tuberculosis (Mtb) in lung tissue, granulomas, and within host macrophages. Here, we demonstrate that glutamate decarboxylase from Mycobacterium tuberculosis (MtbGadB) is a multimeric enzyme exhibiting maximal activity under acidic conditions. MtbGadB is inhibited by the clinical antibiotic d-cycloserine as well as l-cycloserine in vitro. Both cycloserine enantiomers break the internal aldimine bond between PLP and the conser
