Emerging treatments in fungal keratitis.

Fungal keratitis is a major cause of corneal blindness, particularly in tropical and low-resource settings, and remains challenging to manage due to delayed diagnosis, limited drug penetration, emerging resistance, and host-mediated tissue damage. Conventional antifungal therapies, including natamycin and voriconazole, are often inadequate in deep, severe, or refractory infections, underscoring the need for alternative strategies. This narrative review summarizes and critically appraises emerging treatments in fungal keratitis, with a focus on recent pharmacological advances, innovative drug d

Glycoregulation of E3(SCF) ubiquitin ligases in unicellular eukaryotes.

Skp1 is an essential adaptor within the Skp1/Cul1/F-box (SCF) class of E3 polyubiquitin ligases that regulate protein degradation in all eukaryotes. Skp1 is also a target of a 5-enzyme glycosylation pathway in parasites and other unicellular eukaryotes. Glycosylation of Skp1 is contingent upon oxygen-dependent hydroxylation of a critical Pro residue by a homolog of the HIFα PHD2 oxygen sensor of animals. The resulting hydroxyproline is modified by a series of soluble, cytoplasmic, sugar nucleotide-dependent glycosyltransferases that vary among branches of protist evolution, and are evolutionar

Sudden Wilt of Melons in Southern Israel: Fungal Agents and Relationship with Plant Development.

Fungi belonging to five genera, Monosporascus sp., Pythium aphanidermatum, Rhizoctonia solani, Olpidium sp., Fusarium solani, and F. proliferatum, were the species most frequently isolated from the root systems of wilted melon. Diseased plants were collected from 24 fields in the northern and central Arava region of southern Israel during the fall seasons of 1994 and 1995. In pathogenicity tests conducted under field conditions, in artificially inoculated microplots, the first wilt symptoms were observed at various stages of fruit maturation. High mortality levels (73 to 97%) were recorded for

Conservation tillage and organic farming induce minor variations in Pseudomonas abundance, their antimicrobial function and soil disease resistance.

Conservation tillage and organic farming are strategies used worldwide to preserve the stability and fertility of soils. While positive effects on soil structure have been extensively reported, the effects on specific root- and soil-associated microorganisms are less known. The aim of this study was to investigate how conservation tillage and organic farming influence the frequency and activity of plant-beneficial pseudomonads. Amplicon sequencing using the 16S rRNA gene revealed that Pseudomonas is among the most abundant bacterial taxa in the root microbiome of field-grown wheat, independent

Phylogeny of HCN synthase-encoding hcnBC genes in biocontrol fluorescent pseudomonads and its relationship with host plant species and HCN synthesis ability.

Hydrogen cyanide (HCN) is a broad-spectrum antimicrobial compound involved in biological control of root diseases by many plant-associated fluorescent pseudomonads. The HCN synthase is encoded by three biosynthetic genes (hcnA, hcnB, and hcnC), but little is known about the diversity of these genes in fluorescent Pseudomonas spp. and in other bacteria. Here, the partial hcnBC sequence was determined for a worldwide collection of biocontrol fluorescent Pseudomonas spp. Phylogenies based on hcnBC and deduced protein sequences revealed four main bacterial groups, but topological incongruences wer

Hyphae, pseudohyphae, yeasts, spherules, spores, and more: A review on the morphology and pathology of fungal and oomycete infections in the skin of domestic animals.

Fungi are among the most common infectious agents affecting the skin of animals. The skin can serve as a port of entry for fungal infections, which can eventually become disseminated. In some regions of the world, oomycetes, such as Pythium and Lagenidium, are also responsible for a significant number of severe cutaneous infections. Histologic evaluation of fungal morphology, including size, shape, septation, branching, and budding characteristics, combined with the distribution of inflammatory infiltrates within different skin layers can potentially identify etiologic agents, guiding selectio

Pitchers are the unique structures of carnivorous plants used for the trapping of insects and other small invertebrates. The digestion of captured prey here is assisted by the bacteria, which have been associated with pitchers. These bacterial communities can therefore expect to have a variety of plant beneficial functions. In this study, the bacterial isolate NhPBG1 from the pitcher of Nepenthes hamblack was screened for activity against Pythium aphanidermatum, Rhizoctonia solani, Fusarium oxysporum, and Colletotrichum accutatum and was found to have the inhibitory activity towards all the te

ROS and Oxidative Response Systems in Plants Under Biotic and Abiotic Stresses: Revisiting the Crucial Role of Phosphite Triggered Plants Defense Response.

Phosphite (Phi) is a chemical analog of orthophosphate [HPO4 3-]. It is a systemic pesticide generally known to control the prevalence of oomycetes and soil-borne diseases such as Phytophthora, Pythium, and Plasmopora species. Phi can also control disease symptoms and the spread of pathogenic bacteria, fungi, and nematodes. Phi plays critical roles as a fungicide, pesticide, fertilizer, or biostimulator. Overall, Phi can alleviate the severity of the disease caused by oomycete, fungi, pathogenic bacteria, and nematodes (leave, stem, fruit, tuber, and root) in various plants (vegetables, fruits

Trichoderma biocontrol: signal transduction pathways involved in host sensing and mycoparasitism.

Fungi of the genus Trichoderma are used as biocontrol agents against several plant pathogenic fungi like Rhizoctonia spp., Pythium spp., Botrytis cinerea and Fusarium spp. which cause both soil-borne and leaf- or flower-borne diseases of agricultural plants. Plant disease control by Trichoderma is based on complex interactions between Trichoderma, the plant pathogen and the plant. Until now, two main components of biocontrol have been identified: direct activity of Trichoderma against the plant pathogen by mycoparasitism and induced systemic resistance in plants. As the mycoparasitic interacti

A Modular Fusion Protein Strategy Confers Broad-Spectrum Resistance Against Phytophthora infestans, Alternaria solani, and Ralstonia solanacearum.

Potato production is severely threatened by the oomycete Phytophthora infestans, the fungus Alternaria solani, and the bacterium Ralstonia solanacearum; however, strategies capable of simultaneously managing these three pathogen classes remain relatively scarce. In this study, we designed a series of signal peptide (SP)-fused recombinant constructs containing four functional modules with distinct antimicrobial and immune-inducing properties: GAFP1-2×FYVE, an oomycete-inhibiting antimicrobial protein; BbAFP1-ErBD, a fungal-suppressive protein fragment; the pathogen-associated molecular pattern

The Integrated Management of Sugar-Beet Soilborne Diseases Through Rhizosphere Microbiome Strategies: A Critical Review of Agronomic Evidence and Application Gaps.

Soilborne diseases caused by Rhizoctonia solani, Fusarium oxysporum f. sp. betae, Aphanomyces cochlioides, and Pythium spp. constrain sugar-beet production worldwide, with yield losses exceeding 50% in severely affected fields. These pathogens frequently co-occur, yet most biological control and microbiome studies address them individually, and recommendations from model crops often fail to translate to sugar beet's distinctive root biology and rotation systems. This critical review synthesises evidence on the rhizosphere microbiome as a plant protection resource, organising the literature by
