Synthesis and DNA cleavage studies of novel quinoline oxime esters.

New 2-chloro-3-formyl quinoline oxime esters were synthesized by the reaction of 2-chloro-3-formyl quinoline oximes with various benzoyl chlorides in the presence of triethyl amine and dichloromethane at 0°C. The DNA photo cleavage studies of some new oxime esters were investigated by neutral agarose gel electrophoresis at different concentrations (40μM and 80μM). Analysis of the cleavage products in agarose gel indicated that few of quinoline oxime esters (3d-i) converted into supercoiled pUC19 plasmid DNA to its nicked or linear form.

Arene- and quinoline-sulfonamides as novel 5-HT7 receptor ligands.

Novel arene- and quinolinesulfonamides were synthesized using different solutions and a solid-support methodology, and were evaluated for their affinity for 5-HT(1A), 5-HT(2A), 5-HT(6), and 5-HT(7) receptors. Compound 54 (N-Ethyl-N-[4-(1,2,3,4,4a,5,6,7,8,8a-decahydroisoquinolin-2-yl)butyl]-8-quinolinesulfonamide) was identified as potent 5-HT(7) antagonist (K(i)=13 nM, K(B)=140 nM) with good selectivity over 5-HT(1A), 5-HT(2A), 5-HT(6) receptors. In the FST in mice, it reduced immobility in a manner similar to the selective 5-HT(7) antagonist SB-269970.

Aciclovir protects against quinolinic-acid-induced oxidative neurotoxicity.

AIDS-related encephalopathy, including AIDS dementia complex (ADC) and the opportunistic disease, herpes simplex encephalitis (HSE), are postulated to arise due to the release of neurotoxic products, such as quinolinic acid (QUIN), by activated microglial cells in the brain. QUIN causes a cascade of events to occur, which leads to the production of reactive oxygen species (ROS), these being ultimately responsible for oxidative neurotoxicity. The antiherpes antiviral aciclovir has been reported to protect against neuron loss in HSE, but the mechanism for this neuroprotection is unknown. Therefo

The regulation of quinolinic acid in human immunodeficiency virus-infected monocytes.

Quinolinic acid (Quin) is thought to underlie cognitive and motor dysfunctions for a variety of neurological disorders. Specifically, in human immunodeficiency virus (HIV)-associated dementia, Quin levels correlate with the degree of neurological dysfunction observed in affected individuals. Since recent data from our laboratories suggest that both HIV-1 infection and activation of brain macrophages are required for the development of neurotoxicity we examined Quin production during virus infection and immune activation. HIV-1 infection of monocytes induced low levels of Quin while lipopolysac

Systemic approaches to modifying quinolinic acid striatal lesions in rats.

Quinolinic acid (QA) is an endogenous excitotoxin present in mammalian brain that reproduces many of the histologic and neurochemical features of Huntington's disease (HD). In the present study we have examined the ability of a variety of systemically administered compounds to modify striatal QA neurotoxicity. Lesions were assessed by measurements of the intrinsic striatal neurotransmitters substance P, somatostatin, neuropeptide Y, and GABA. Histologic examination was performed with Nissl stains. The antioxidants ascorbic acid, beta-carotene, and alpha-tocopherol administered s.c. for 3 d pri

An Efficient One-Pot Synthesis of Densely Functionalized Fused-Quinolines via Sequential Ugi4CC and Acid-Mediated Povarov-Type Reaction.

A divergent synthesis of fused-quinolines has been explored by performing Ugi four-component condensation and sulfuric acid promoted deprotection/Povarov-type reaction in one-pot. The process involves Ugi condensation of propiolic acids, aldehydes/ketones, aminoaldehyde acetals and isocyanides followed by sulfuric acid promoted deprotection and Povarov-type reaction with anilines in ethanol. This method enables straightforward access to the structurally diverse 2,3-dihydro-1H-pyrrolo[3,4-b]quinolin-1-ones (DHPQ), 3,4-dihydrobenzo[b][1,6]naphthyridin-1(2H)-ones (DHBN), and 2,3,4,5-tetrahydro-1H

Ag(I)-Promoted Cascade Cyclization of 1-Isocyano-2-((2-alkoxyphenyl)ethynyl)benzenes to Access Benzofuro[2,3-b]quinolines.

This work discloses an alternative synthetic route to access benzofuro[2,3-b]quinolines. The reaction proceeds via Ag(I)-assisted cascade cyclization of 1-isocyano-2-((2-alkoxyphenyl)ethynyl)benzenes, derived from the corresponding N-(2-((2-alkoxyphenyl)ethynyl)phenyl)formamide derivatives, allowing a one-pot, two-step assembly of the benzofuroquinoline scaffolds with four fused rings. The salient features of the synthetic method are broad substrate scopes, good to excellent yields, and ease of experimental procedure. A gram-scale synthesis demonstrated the scalability and practical applicabil

Selective Synthesis of 2-Substituted Quinolines via Amine-Assisted Heck Reaction of 2-Iodoanilines with α,β-Unsaturated Aldehydes.

2-Substituted quinoline has been identified as a significant structural scaffold, with prevalence in a variety of biologically active molecules. In this study, we propose an amine-assisted Heck reaction as a novel approach for the selective synthesis of 2-substituted quinolines while inhibiting the formation of 3-benzyl-1H-indole derivatives and 4-substituted quinoline isomers. A variety of α,β-unsaturated aldehydes and 2-iodoanilines were compatible with this protocol, affording the desired quinoline products in good-to-excellent yields. Moreover, the reaction can be effectively scaled up to

Catalytic utility of PNN-based MnI pincer complexes in the synthesis of quinolines and transfer hydrogenation of carbonyl derivatives.

This manuscript describes the synthesis of a triazolyl-pyridine-based phosphine, N-((diphenylphosphaneyl)methyl)-N-methyl-6-(1-phenyl-1H-1,2,3-triazol-4-yl)pyridin-2-amine, [2,6-{(PPh2)CH2N(Me)(C5H3N)(C2HN3C6H5)}] (1) (here onwards referred to as PNN) and its cationic and neutral MnI complexes and catalytic applications. The reaction of 1 with Mn(CO)5Br afforded a cationic complex [Mn(CO)3(PNN)]Br (2), which is highly stable in solid state, but in solution it gradually loses one of the CO groups to form a neutral complex [Mn(CO)2(PNN)Br] (3). Complex 2 on treatment with AgBF4 also yielded a ca

Cu-catalyzed Synthesis of Quinolines by Dehydrogenative Reaction of 2-Aminobenzyl Alcohol and Ketones: A Combined Experimental and Computational Study.

Quinoline derivatives are important moieties in bioactive molecules and advanced materials. However, an efficient strategy to synthesize quinoline derivatives remains challenging. Herein, we describe an efficient and practical method for the synthesis of quinolines by Cu-catalyzed cyclization of 2-amino benzyl alcohol with ketones (or secondary alcohols) via an acceptorless dehydrogenation pathway. Interestingly, a range of highly functionalized quinolines is prepared in good yields using low catalyst loading under relatively mild conditions. Furthermore, density functional theory (DFT) calcul

Quinoline/thiazole compounds as selective acetylcholinesterase inhibitors: synthesis and biological assessment.

Acetylcholine (ACh), acetylcholinesterase enzyme (AChE), and AChE inhibition are of great importance in the treatment of neurodegenerative diseases. When it comes to AChE inhibition, FDA-approved AChE inhibitors such as donepezil are actively used in the treatment of neurodegenerative disorders. However, there is still a need for novel and unique inhibitors for the radical treatment of these diseases. The aim of this study was to design and evaluate new inhibitors for the treatment of neurodegenerative diseases. To meet this need, a series of new quinoline/thiazole derivative compounds were de

Discovery of a quinoline-based senomorphic agent for the treatment of chronic kidney disease.

Senescence is a risk factor for chronic diseases, making anti-aging interventions a promising strategy for geriatric conditions. While a non-antibacterial derivative C1 of enrofloxacin has been proposed to extend lifespan in Caenorhabditis elegans (C. elegans), its unresolved cytotoxicity in healthy mammalian cells limits its combinatory potential and safety as an anti-aging agent. With the aim of preserving anti-aging activity while enhancing safety, we here performed structural optimization of C1 and created 37 derivatives. Among them, C36, which derived from introducing an isopropoxy group

Photopharmacology of Quinoline and Benzimidazole Azobenzene-Based Photoswitchable β‑Hematin Inhibitors.

Recent advances in antimicrobial drug design have incorporated photopharmacology to control drug behavior through light activation, alongside traditional phenotypical strategies including molecular hybridization and metal incorporation. The quinoline and benzimidazole scaffolds are important chemical frameworks identified by the Medicines for Malaria Venture and are known to exert their antiplasmodial activity by inhibiting hemozoin formation, an important target in antimalarial drug design. These scaffolds form part of growing libraries in drug discovery, aimed at developing novel and potent

Quinolin(on)es and pyridin(on)es as promising scaffolds against mycobacterial infections.

Mycobacterial infections pose a significant global health challenge. In 2024, 1.23 million deaths were associated with M. tuberculosis (M.tb). In recent years, the number of recorded cases of non-tuberculous mycobacterial infections has risen, particularly among elderly patients or people with underlying pulmonary diseases. Due to the emergence of antibioresistance, the discovery of new drugs which target essential enzymes in mycobacteria is required to move towards new therapeutic options for future years. Some current clinical drugs contain promising scaffolds, which serve as a basis for oth
