Pyrroloquinoline quinone prevents knee osteoarthritis by inhibiting oxidative stress and chondrocyte senescence.

Accumulating evidence suggests that oxidative stress plays an important role in the progression of osteoarthritis (OA), and pyrroloquinoline quinone (PQQ) is considered a strong antioxidant. However, it is unclear whether PQQ can prevent the progression of OA by inhibiting oxidative stress. In this study, anterior cruciate ligament transection (ACLT)-induced OA mice received a diet supplemented with/without PQQ, and were compared with each other and with sham-operated mice. Our results showed that in PQQ-untreated OA mice, articular surfaces collapsed, while the thickness of articular cartilag

Formation of dopamine quinone-DNA adducts and their potential role in the etiology of Parkinson's disease.

The neurotransmitter dopamine is oxidized to its quinone (DA-Q), which at neutral pH undergoes intramolecular cyclization by 1,4-Michael addition, followed by oxidation to form leukochrome, then aminochrome, and finally neuromelanin. At lower pH, the amino group of DA is partially protonated, allowing the competitive intermolecular 1,4-Michael addition with nucleophiles in DNA to form the depurinating adducts, DA-6-N3Ade and DA-6-N7Gua. Catechol estrogen-3,4-quinones react by 1,4-Michael addition to form the depurinating 4-hydroxyestrone(estradiol)-1-N3Ade [4-OHE1(E2)-1-N3Ade] and 4-OHE1(E2)-1

Quinone reductase inducing activity of the dichloromethane /ethanol extract of the roots of Pulsatilla chinensis.

Agents with phase II enzyme inducing activities play important roles in intervening in the carcinogenic process. In the present study, the quinone reductase (QR) inducing activities of nine known triterpene saponins from the dichloromethane/ethanol extract of the roots of Pulsatilla chinensis were tested. The oleanane saponins exhibited more potent QR inducing activities than the lupane saponins, and the CD value of the compound with the most potent QR inducing activity was 1.1 microM. The chemopreventive activity of the dichloromethane/ethanol extract was also evaluated using the DMBA-induced

Cellular fatty acid compositions and isoprenoid quinone contents of 23 Legionella species.

The cellular fatty acid compositions and ubiquinone contents of 182 Legionella strains representing 23 species were determined by capillary gas-liquid chromatography and reverse-phase high-performance liquid chromatography, respectively. Except for the type strain of Legionella erythra (ATCC 35303T), all Legionella species contained large (40 to 90%) amounts of branched-chain fatty acids and only trace to small (less than 0.5 to 5%) amounts of ester-linked hydroxy acids. The 23 species were placed in three major fatty acid groups on the basis of differences in the relative amounts of 14-methyl

Identification of the quinone species in cyanide-sensitive and cyanide-insensitive mitochondria of Moniliella tomentosa.

Moniliella tomentosa was investigated for the presence of different quinones that might be involved in the cyanide-sensitive and/or cyanide-insensitive electron-transport pathways. The naturally occurring quinone in Moniliella tomentosa was found to be ubiquinone-45. Other quinone species could not be detected. The concentration of ubiquinone-45 in mitochondria is not related to the presence or absence of the alternative oxidase activity.

Prodrug-like Quinone Containing Two Redox-Active Centers Induces Ferroptosis in Prostate Cancer Cells with Cytotoxic Action Partially Depending on NAD(P)H Quinone Dehydrogenase 1 (NQO1).

Naphthoquinones are redox-active compounds with antitumor potential, often linked to NQO1 overexpression in cancer cells. This study evaluated the cytotoxic activity of two synthetic 1,4-naphthoquinones, compound 1 and its dual redox analog, compound 4, against NQO1-overexpressing DU-145 prostate cancer cells through integrated in vitro, enzymatic, and computational (docking and quantum) analyses. Compound 4 exhibited over 2-fold greater cytotoxicity against DU-145 cells compared to nontumor MRC-5 cells, while maintaining low hemolytic activity. Evidence of membrane damage, ROS generation, lip

Removal of Sulfonamide Antibiotics in Peracetic Acid-Mediated Natural Polyphenol Systems via an Overlooked Polymerization Pathway: Role of ortho-Quinones.

Natural polyphenols can be oxidized into reactive quinones, which might play a key role in the removal of specific contaminants in natural polyphenol-related advanced oxidation processes (AOPs). In this study, peracetic acid (PAA) was employed in combination with natural protocatechuic acid (PCA) to remove sulfonamide antibiotics (SAs) from water. More than 95% removal of sulfamethoxazole (SMX) and other SAs was observed in the PCA/PAA system, and neutral pH conditions (5.0-8.0) were more conducive to the removal of SMX. The PCA/PAA system exhibited a great anti-interference ability against co

Dopamine-derived quinones affect the structure of the redox sensor DJ-1 through modifications at Cys-106 and Cys-53.

The physiological role of DJ-1, a protein involved in familial Parkinson disease is still controversial. One of the hypotheses proposed indicates a sensor role for oxidative stress, through oxidation of a conserved cysteine residue (Cys-106). The association of DJ-1 mutations with Parkinson disease suggests a loss of function, specific to dopaminergic neurons. Under oxidative conditions, highly reactive dopamine quinones (DAQs) can be produced, which can modify cysteine residues. In cellular models, DJ-1 was found covalently modified by dopamine. We analyzed the structural modifications induce

Acute toxicity and tissue concentrations of 6PPD-quinone in Arctic grayling (Thymallus arcticus, Thymallinae).

Studies of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPDQ) toxicity have identified sensitive and resistant species within the subfamily Salmoninae, but thus far fewer than 10 species, all in two genera (Oncorhynchus and Salvelinus), have shown sensitivity in experiments. The Arctic grayling (Thymallus arcticus, Salmonidae, Thymallinae) was extirpated from its range in the Great Lakes region of the United States, but because of the importance of this species to the Native American tribes of Michigan, a reintroduction effort is underway. Because high 6PPDQ concentrations have

Quinone-heme π-π interactions bridge the biotic-abiotic interface for enhanced methanogenesis.

Quinone-based conductive materials promote extracellular electron transfer and methanogenesis during anaerobic digestion, in which the materials mediates the transfer of electrons produced by exoelectrogens to electrotrophic methanogens. However, the electron transfer mechanism between quinone and electrotrophic methanogens at the biotic-abiotic interface remains elusive. In this study, an interface between reduced anthraquinone and Methanosarcina barkeri is established in a microbial electrolysis cell. Biomimetic experiments and characterization of isolated extracellular polymeric substances

A role for the antioxidants coenzyme Q10 and pyrroloquinoline quinone in mitigating obesity-associated reproductive dysfunction†.

Obesity is a rising public health concern in the United States, with significant implications for maternal and fetal health during pregnancy. Maternal obesity is associated with increased risks of miscarriage, reduced implantation and pregnancy rates, fetal anomalies, abnormal fetal growth, stillbirth, preeclampsia, gestational diabetes, hypertensive disorders, labor and wound complications, and maternal depression. Current strategies for managing these risks include pre-pregnancy weight loss, nutritional counseling and weight management during pregnancy, lifestyle modifications, and low-dose

Pyrroloquinoline Quinone Reprograms the Single-Cell Landscape of Immune Aging in Hematopoietic Immune System.

Aging is an inevitable biological process, driven in part by increased oxidative stress, which accelerates cellular damage and contributes to immune system dysfunction. Therefore, targeting oxidative stress has emerged as a potential strategy. Pyrroloquinoline quinone (PQQ), a potent antioxidant, has demonstrated significant efficacy in reducing oxidative stress and modulating immune responses, making it a promising therapeutic candidate. In this study, we investigated the effects of aging on the hematopoietic immune system (HIS) through single-cell RNA sequencing (scRNA-seq) of spleen and bon

Neuroprotective role of pyrroloquinoline quinone in folate deficiency-induced blood-brain barrier disruption.

Healthy neurodevelopment requires adequate folates (vitamin B9), which are critical for key biosynthetic and homeostatic processes in the central nervous system (CNS). In the brain, folate transport is mediated by three major pathways: folate receptor alpha (FRα), proton-coupled folate transporter (PCFT), and reduced folate carrier (RFC). Folate uptake primarily occurs at the blood-cerebrospinal fluid barrier (BCSFB) by concerted actions of FRα and PCFT. Alterations in this transport pathway can result in cerebral folate deficiency (CFD), a rare but devastating pediatric condition associated w

Antimicrobial activity of natural quinone-methide triterpenoids celastrol and pristimerin against pathogenic Neisseria.

Neisseria gonorrhoeae and Neisseria meningitidis are closely related and important human pathogens that cause distinct diseases of clinical significance. While N. meningitidis infrequently demonstrates antibiotic resistance, the rapid emergence of multidrug-resistant N. gonorrhoeae isolates emphasizes the urgency of novel therapeutics against this pathogen. Recently, repurposing of approved drugs and the therapeutic potential of natural compounds have been explored as alternative strategies to treat gonococcal infections. We screened 54 natural bioactive compounds and identified celastrol and

Poisoning by butylated hydroxytoluene quinone methide acting as a superwarfarin: first reported case in humans.

Superwarfarins are anticoagulant rodenticides nearly 100-fold potent than the parent compound. Since their development, accidental and intentional cases of superwarfarin poisoning have been reported. We report the first human case of poisoning by butylated hydroxytoluene (BHT) quinone methide acting as a superwarfarin otherwise reported to be a well tolerated food additive and preservative and used as an antioxidant, stabilizer, anti-skinning agent in various industries. We aim to highlight the possible underlying cause of this previously unreported and potentially lethal BHT-related complicat

Design, Synthesis, and Structure-Activity Relationship Studies of New Quinone Derivatives as Antibacterial Agents.

Resistance to antibacterial agents is a growing global public health problem that reduces the efficacy of available antibacterial agents, leading to increased patient mortality and morbidity. Unfortunately, only 16 antibacterial drugs have been approved by the FDA in the last 10 years, so it is necessary to develop new agents with novel chemical structures and/or mechanisms of action. In response to this, our group takes up the challenge of designing a new family of pyrimidoisoquinolinquinones displaying antimicrobial activities against multidrug-resistant Gram-positive bacteria. Accordingly,

Discovery of Bis-sulfonamides as Novel Inhibitors of Mitochondrial NADH-Quinone Oxidoreductase (Complex I).

Mitochondrial oxidative phosphorylation (OXPHOS) is an essential cellular metabolic process that generates ATP. The enzymes involved in OXPHOS are considered to be promising druggable targets. Through screening of an in-house synthetic library with bovine heart submitochondrial particles, we identified a unique symmetric bis-sulfonamide, KPYC01112 (1) as an inhibitor targeting NADH-quinone oxidoreductase (complex I). Structural modifications of KPYC01112 (1) led to the discovery of the more potent inhibitors 32 and 35 possessing long alkyl chains (IC50 = 0.017 and 0.014 μM, respectively). A ph
