The prognostic value of RASGEF1A RNA expression and DNA methylation in cytogenetically normal acute myeloid leukemia.

BACKGROUND: Acute myeloid leukemia (AML) is a significantly heterogeneous malignancy of the blood. Cytogenetic abnormalities are crucial for the prognosis of AML. However, since more than half of patients with AML are cytogenetically normal AML (CN-AML), predictive prognostic indicators need to be further refined. In recent years, gene abnormalities are considered to be strong prognostic factors of CN-AML, already having clinical significance for treatment. In addition, the relationship of methylation in some genes and AML prognosis predicting has been discovered. RASGEF1A is a guanine nucleot

RNA methylation reading protein YTHDF2 relieves myocardial ischemia-reperfusion injury by downregulating BNIP3 via m6A modification.

BNIP3 is reported to be involved in hypoxia-induced mitochondrial defect and cell death in cardiomyocytes. However, little is known about the specific function and molecular mechanism of BNIP3-mediated mitophagy in myocardial ischemia-reperfusion injury (MIRI). Herein, this study explored the mechanism regulating BNIP3-modulated mitophagy in MIRI. Rat cardiomyocytes (H9c2 cells) underwent transfection and hypoxia/reoxygenation (H/R) treatment, followed by cell viability and apoptosis detection. Gain-of-function assays were conducted in rats before MIRI modeling, followed by the monitoring of c

Alterations of m6A RNA methylation regulators contribute to autophagy and immune infiltration in primary Sjögren's syndrome.

N6-methyladenosine (m6A) RNA modification is a new epigenetic regulation mechanism on eukaryotic mRNA. Few autoimmune diseases focused on the role of m6A in their pathogenies, and m6A modulation in the pathological process of primary Sjögren's syndrome (pSS) is still unknown. In this work, three microarray datasets of pSS patients were downloaded from the GEO database: datasets #1 and #2 from the whole peripheral blood (PB) samples, dataset #3 from the labial salivary gland tissue samples, as well as a PB cohort collected from our hospital. Six differentially expressed m6A regulators were iden

m6A-Finder: Detecting m6A methylation sites from RNA transcriptomes using physical and statistical properties based features.

N6-methyladenosine (m6A) is one of the abundant post-transcription modification in cellular RNA. It regulates different biological processes, such as, protein synthesis, X-chromosome inactivation, cell stability, cell-reprogramming and miRNA regulation etc. Most recently, various studies claimed that mutations in m6A sites are linked with various diseases, such as, brain-tumor, heart attack, obesity and cancer. The correct identification of m6A sites is essential to overcome these diseases. However, the state-of-the-art predictors face many challenges for precise detection of m6A sites. Even f

Gluten-induced RNA methylation changes regulate intestinal inflammation via allele-specific XPO1 translation in epithelial cells.

OBJECTIVES: Coeliac disease (CD) is a complex autoimmune disorder that develops in genetically susceptible individuals. Dietary gluten triggers an immune response for which the only available treatment so far is a strict, lifelong gluten free diet. Human leucocyte antigen (HLA) genes and several non-HLA regions have been associated with the genetic susceptibility to CD, but their role in the pathogenesis of the disease is still essentially unknown, making it complicated to develop much needed non-dietary treatments. Here, we describe the functional involvement of a CD-associated single-nucleot

m5C methylation of mitochondrial RNA and non-coding RNA by NSUN3 is associated with variant gene expression and asexual blood-stage development in Plasmodium falciparum.

BACKGROUND: Malaria is caused by Plasmodium spp. and is a prevalent parasitic disease worldwide. To evade detection by the immune system, by switching variant gene expression, the malaria parasite continually establishes new patterns displaying a single variant erythrocyte surface antigen. The distinct surface molecules encoded by clonally variant gene families include var, rif, stevor, Pfmc-2tm, and surfins. However, the mechanism behind the exclusive expression of a single member of the variant gene family is still not clear. This study aims to describe the molecular process of variant gene

The landscape of implantation and placentation: deciphering the function of dynamic RNA methylation at the maternal-fetal interface.

The maternal-fetal interface is defined as the interface between maternal tissue and sections of the fetus in close contact. RNA methylation modifications are the most frequent kind of RNA alterations. It is effective throughout both normal and pathological implantation and placentation during pregnancy. By influencing early embryo development, embryo implantation, endometrium receptivity, immune microenvironment, as well as some implantation and placentation-related disorders like miscarriage and preeclampsia, it is essential for the establishment of the maternal-fetal interface. Our review f

Transgenerational effect of mutants in the RNA-directed DNA methylation pathway on the triploid block in Arabidopsis.

BACKGROUND: Hybridization of plants that differ in number of chromosome sets (ploidy) frequently causes endosperm failure and seed arrest, a phenomenon referred to as triploid block. In Arabidopsis, loss of function of NRPD1, encoding the largest subunit of the plant-specific RNA polymerase IV (Pol IV), can suppress the triploid block. Pol IV generates short RNAs required to guide de novo methylation in the RNA-directed DNA methylation (RdDM) pathway. Recent work suggests that suppression of the triploid block by mutants in RdDM components differs, depending on whether the diploid pollen is de

2'-O-Methyl-guanosine RNA fragments antagonize TLR7 and TLR8 to limit autoimmunity.

Recognition of RNA fragments by Toll-like receptor 7 (TLR7) and TLR8 helps to initiate the innate immune response against pathogens. An outstanding question is why RNA fragments generated during clearance of apoptotic cells fail to activate TLR7 and TLR8 signaling. Here we show that select 2'-O-methyl (2'-OMe) guanosine RNA fragments, including those derived from host RNAs, function as potent TLR7 and TLR8 antagonists and reduce TLR7 sensing in vivo. Mechanistically, these fragments bind to an antagonistic site on these proteins via their 5'-end 2'-OMe guanosine. These findings indicate that h

A Comparative Analysis of the Methylation Status of Non-Coding RNA Promoters in Fibroid and Matched Myometrium.

Uterine fibroids exhibit dysregulated expression of non-coding RNAs (ncRNAs), although the underlying mechanisms remain incompletely understood. We investigated promoter DNA methylation and its relationship with ncRNA expression in fibroids. Genomic DNA from eight paired fibroid and matched myometrial tissues was analyzed using MeDIP-chip to identify differentially methylated ncRNA promoters. Selected candidates were validated by methylation-specific PCR (MSP) in 16 paired samples, and transcript expression was assessed by qRT-PCR in 68-94 paired specimens. MeDIP-chip identified 538 lncRNAs an

The involvement of m6A RNA methylation in the transgenerational inheritance of hepatotoxicity induced by paternal cadmium exposure.

Cadmium (Cd), a widespread environmental pollutant, has linked with various adverse health effects. However, the paternally inherited transgenerational effects of cadmium exposure on hepatotoxicity inoffspring and the underlying mechanism remained undefined. To address this issue, male mice (F0) were administrated with 100 mg/L cadmium chloride (CdCl2) in drinking water for 3 months. Then, F0 males were mated with healthy female mice to produce F1 and F2 offspring.Liver function was assessed at age of 6 weeks and 6 months, respectively. Herein, we showed ancestral cadmium exposure led to liver

The translational potential of m6A RNA methylation in DKD: emerging biomarkers for early detection and targeted therapies.

Diabetic Kidney Disease (DKD) is a primary cause of end-stage renal disease, characterized by podocyte dysfunction. While current clinical biomarkers including albuminuria and estimated glomerular filtration rate are foundational for diagnosis, they often function as lagging indicators and fail to detect early renal injury. Recently, N6-methyladenosine (m6A) RNA methylation has emerged as a key epitranscriptomic link between hyperglycemia and podocyte injury, offering a novel frontier for diagnostic biomarker discovery. This review shifts the traditional focus from basic molecular mechanisms t

Chromones from Saposhnikovia divaricata modulate m6A RNA methylation-mediated macrophage polarization by targeting CBLL1 to ameliorate RA.

BACKGROUND: Over-activated M1 macrophages, which destroyed the balance of macrophage phenotype in the synovial microenvironment, play a pivotal role in rheumatoid arthritis (RA) progression. Increasing evidence indicates that m6A RNA methylation serves as a crucial epigenetic regulatory mechanism in modulating the transition from M0 to M1 macrophages. Saposhnikovia divaricata (Turcz.) Schischk treats RA through immune regulation. In our previous study, chromones (CHR) from Saposhnikovia divaricata could alleviate RA by inhibiting the pathological manifestations of synovial inflammation, but th

Transcriptome-wide study of mRNAs modified by m6A RNA methylation in the testis development of dairy goats.

N6-methyladenosine (m6A) is an important epigenetic modification in RNA, playing a crucial role in regulating the production and aging of animal testicular sperm. This study extracted mRNA from the testicular tissue of male goats before and after sexual maturity, generating a methylation map through preliminary experiments and methylation immunoprecipitation sequencing. The results showed that during the development of dairy goats, the expression levels of marker genes related to testicular development and methylation-related enzymes changed significantly. A total of 36,602 peaks and 11,223 ge

Profiling of RNA N6-Methyladenosine methylation reveals the critical role of m6A in betaine alleviating hepatic steatosis.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a major predisposing factor of metabolic dysfunction-associated steatohepatitis, hepatic fibrosis and hepatocellular carcinoma. Abnormal RNA m6A modification leads to the disturbance of lipid metabolism, insulin resistance, and chronic inflammation, all of which are critical for the onset and progression of MASLD. It has been shown that betaine supplementation has an ameliorative effect on symptoms associated with MASLD. The purpose of this study is to explore the role of RNA m6A modification in the alleviation of MASLD by bet

N6-methyladenosine RNA methylation is a novel epitranscriptomic regulator of excessive alcohol drinking and vulnerability to relapse.

While internal RNA modifications have been known for decades, the contribution of epitranscriptomics to alcohol use disorder (AUD) remains unexplored. Here we investigated, for the first time, the role of the most abundant mRNA modification, N6-methyladenosine (m6A) in the regulation of alcohol drinking and alcohol mediated gene expression. The fat mass and obesity associated gene (FTO) plays a key role in erasing m6A RNA methylation marks. We generated brain m6A hypermethylated mice with selective deletion of FTO in neurons by crossing FTO-floxed mice (FTO-fl/fl) with the synapsin-1-CRE delet

Epitranscriptomic regulation in cardiovascula disease: mechanistic roles and clinical implications of m6A RNA methylation in cardiac pathophysiology.

N6-methyladenosine (m6A) RNA methylation has emerged as a pivotal epitranscriptomic regulator influencing cardiovascular development, homeostasis, and disease progression. As the most abundant internal modification in eukaryotic mRNA, m6A dynamically modulates RNA stability, translation, splicing, and degradation through the coordinated actions of "writers," "readers," and "erasers." Recent advances demonstrate that dysregulated m6A modifications contribute to major cardiovascular disorders, including cardiac hypertrophy, heart failure, arrhythmias, atherosclerosis, ischemia reperfusion injury

Non-coding RNA-mediated endothelial-to-mesenchymal transition in human diabetic cardiomyopathy, potential regulation by DNA methylation.

AIMS: Diabetic cardiomyopathy (DCM) is a major complication of diabetes and a risk factor for cardiovascular disease. Endothelial dysfunction is central to DCM, and endothelial-to-mesenchymal transition (EndMT) is a key form of endothelial dysfunction in diabetes. EndMT in DCM has been well-studied in model systems and has been found to be epigenetically regulated by non-coding RNAs (ncRNAs). However, EndMT in DCM and its associated epigenetic changes need further characterization in human patients. It is also not known if ncRNAs are affected by changes in DNA methylation in DCM. This study ai
