Application of random forest based on semi-automatic parameter adjustment for optimization of anti-breast cancer drugs.

The optimization of drug properties in the process of cancer drug development is very important to save research and development time and cost. In order to make the anti-breast cancer drug candidates with good biological activity, this paper collected 1974 compounds, firstly, the top 20 molecular descriptors that have the most influence on biological activity were screened by using XGBoost-based data feature selection; secondly, on this basis, take pIC50 values as feature data and use a variety of machine learning algorithms to compare, soas to select a most suitable algorithm to predict the I

A hybrid random forest deep learning classifier empowered edge cloud architecture for COVID-19 and pneumonia detection.

COVID-19 is a global pandemic that mostly affects patients' respiratory systems, and the only way to protect oneself against the virus at present moment is to diagnose the illness, isolate the patient, and provide immunization. In the present situation, the testing used to predict COVID-19 is inefficient and results in more false positives. This difficulty can be solved by developing a remote medical decision support system that detects illness using CT scans or X-ray images with less manual interaction and is less prone to errors. The state-of-art techniques mainly used complex deep learning

Improving Ovine Behavioral Pain Diagnosis by Implementing Statistical Weightings Based on Logistic Regression and Random Forest Algorithms.

Recently, the Unesp-Botucatu sheep acute pain scale (USAPS) was created, refined, and psychometrically validated as a tool that offers fast, robust, and simple application. Evidence points to an improvement in pain diagnosis when the importance of the behavioral items of an instrument is statistically weighted; however, this has not yet been investigated in animals. The objective was to investigate whether the implementation of statistical weightings using machine learning algorithms improves the USAPS discriminatory capacity. A behavioral database, previously collected for USAPS validation, o

Side effect prediction based on drug-induced gene expression profiles and random forest with iterative feature selection.

One in every ten drug candidates fail in clinical trials mainly due to efficacy and safety related issues, despite in-depth preclinical testing. Even some of the approved drugs such as chemotherapeutics are notorious for their side effects that are burdensome on patients. In order to pave the way for new therapeutics with more tolerable side effects, the mechanisms underlying side effects need to be fully elucidated. In this work, we addressed the common side effects of chemotherapeutics, namely alopecia, diarrhea and edema. A strategy based on Random Forest algorithm unveiled an expression si

Analysis and prediction of hand, foot and mouth disease incidence in China using Random Forest and XGBoost.

Hand, foot and mouth disease (HFMD) is an increasingly serious public health problem, and it has caused an outbreak in China every year since 2008. Predicting the incidence of HFMD and analyzing its influential factors are of great significance to its prevention. Now, machine learning has shown advantages in infectious disease models, but there are few studies on HFMD incidence based on machine learning that cover all the provinces in mainland China. In this study, we proposed two different machine learning algorithms, Random Forest and eXtreme Gradient Boosting (XGBoost), to perform our analy

Comparison of random forest and parametric imputation models for imputing missing data using MICE: a CALIBER study.

Multivariate imputation by chained equations (MICE) is commonly used for imputing missing data in epidemiologic research. The "true" imputation model may contain nonlinearities which are not included in default imputation models. Random forest imputation is a machine learning technique which can accommodate nonlinearities and interactions and does not require a particular regression model to be specified. We compared parametric MICE with a random forest-based MICE algorithm in 2 simulation studies. The first study used 1,000 random samples of 2,000 persons drawn from the 10,128 stable angina p

Weighted Random Forests to Improve Arrhythmia Classification.

Construction of an ensemble model is a process of combining many diverse base predictive learners. It arises questions of how to weight each model and how to tune the parameters of the weighting process. The most straightforward approach is simply to average the base models. However, numerous studies have shown that a weighted ensemble can provide superior prediction results to a simple average of models. The main goals of this article are to propose a new weighting algorithm applicable for each tree in the Random Forest model and the comprehensive examination of the optimal parameter tuning.

Dimensionality reduction and metabolite panel derivation in urinary metabolomics based on Random Forest with Gini index feature selection.

Untargeted urinary metabolomics represents a promising approach for investigating metabolic alterations associated with oncogenic processes such as breast cancer (BC). However, the stable selection of informative m/z features remains a central challenge in biomarker-oriented studies, particularly in the context of early BC detection and population-level screening. Urine samples from two independent cohorts (n = 50 and n = 75) and analyzed using distinct UHPLC-QTOF-ESI⁺ mass spectrometry workflows, yielded 224 and 129 aligned m/z features, respectively. Classification and embedded feature selec

Enhancing Alzheimer's disease prediction using random forest: A novel framework combining backward feature elimination and ant colony optimization.

BACKGROUND: Alzheimer's disease (AD) represents a significant global health challenge due to its increasing prevalence and the limitations of current diagnostic approaches. Early detection is crucial as pathological changes occur 10-15 years before clinical symptoms manifest, yet current diagnostic methods typically identify the disease at moderate to advanced stages. Machine learning techniques offer promising solutions for early prediction, but face challenges related to feature selection and hyperparameter optimization. OBJECTIVE: To develop an enhanced predictive model for Alzheimer's dis

Iterative random forest-based identification of a novel population with high risk of complications post non-cardiac surgery.

Assessing the risk of postoperative cardiovascular events before performing non-cardiac surgery is clinically important. The current risk score systems for preoperative evaluation may not adequately represent a small subset of high-risk populations. Accordingly, this study aimed at applying iterative random forest to analyze combinations of factors that could potentially be clinically valuable in identifying these high-risk populations. To this end, we used the Japan Medical Data Center database, which includes claims data from Japan between January 2005 and April 2021, and employed iterative
