Segregation of calcium signalling mechanisms in magnocellular neurones and terminals.

Every cell or neuronal type utilizes its own specific organization of its Ca(2+) homeostasis depending on its specific function and its physiological needs. The magnocellular neurones, with their somata situated in the supraoptic and paraventricular nuclei of the hypothalamus and their nerve terminals populating the posterior hypophysis (neural lobe) are a typical and classical example of a neuroendocrine system, and an important experimental model for attempting to understand the characteristics of the neuronal organization of Ca(2+) homeostasis. The magnocellular neurones synthesize, in a ce

Renal Myeloid Mechanisms, Cross-Talk in Hypertension.

Hypertension involves an impairment of renal sodium excretion along with activation of pro-inflammatory responses in the kidney. Dendritic cells (DCs) are professional antigen-presenting cells that sense disturbances in cell and tissue homeostasis to orchestrate T cell activation and recruitment. Renal DCs distribute as a cellular network throughout the renal interstitium, forming an interface between innate and adaptive immunity. Here, we explore what is known about renal cross-talk following prohypertensive stimuli, which modify renal DCs, T cell polarization, and renal function, leading to

Mitochondrial DNA in necrosis-like cell death in failing hearts: evidence and mechanisms fostering damage.

Mitochondrial DNA (mtDNA) is increasingly recognized as a damage-associated molecular pattern (DAMP) that signals mitochondrial injury and the consequent pro-death molecular signature. Such mtDNA release triggers programmed necrotic pathways, specifically necroptosis and pyroptosis, both of which are central to the pathogenesis of heart failure (HF). Mechanistically, mtDNA liberated from dysfunctional mitochondria engages cytosolic nucleic acid sensors and is associated with activation of necroptotic pathways via the Z-DNA binding protein 1 (ZBP1)-receptor-interacting protein kinase 3 (RIP3)-m

Cancer-induced nerve injury unveils a sympathetic-to-sensory nerve axis in a mouse model of head and neck squamous cell carcinoma.

Oral squamous cell carcinoma (OSCC) is one of the most painful cancers, with patients frequently reporting spontaneous, neuropathic-like pain. Sympathetic and sensory nerves have been individually implicated in cancer progression, but whether and how these systems interact to drive pain and tumor growth have remained unclear. Here, we integrated prospective human data with reverse-translational mouse models to reveal that cancer-induced nerve injury unveils cross-talk between sympathetic postganglionic neurons and trigeminal sensory afferents in the tumor microenvironment. In patients, circula

Modulation of beta2- and beta3-adrenoceptor-mediated relaxation of rat oesophagus smooth muscle by protein kinase C.

Although a prominent role for protein kinase C (PKC) in the cross-talk between the phosphoinositide pathway and beta2-adrenoceptor signalling has been indicated, modulation of beta3-adrenoceptor function by PKC has not been studied thus far. In the present study, we have compared the relative capacity of PKC in modulating beta2- and beta3-adrenoceptor-mediated relaxation of methacholine-contracted rat oesophagus smooth muscle. To this purpose the effects of the PKC-inhibitor GF 109203X (2-[1-(3-dimethylaminopropyl)-1H-indol-3-yl]-3-(1H-indol-3-yl)-maleimide) on relaxation induced by fenoterol,

RANKL-RANK signaling in osteoclastogenesis and bone disease.

Hundreds of millions of people worldwide are affected by bone-related diseases, such as osteoporosis and rheumatoid arthritis. Understanding the molecular mechanisms of bone metabolism is crucial for developing novel drugs for treating such diseases. In particular, genetic experiments showing that the receptor activator of NF-kappaB (RANK), its ligand RANKL, and the decoy receptor OPG are essential, central regulators of osteoclast development and osteoclast function were significant turning points in our understanding of bone diseases. RANKL-RANK signaling activates a variety of downstream si

ATGL-dependent white adipose tissue lipolysis controls hepatocyte PPARα activity.

In hepatocytes, peroxisome proliferator-activated receptor α (PPARα) orchestrates a genomic and metabolic response required for homeostasis during fasting. This includes the biosynthesis of ketone bodies and of fibroblast growth factor 21 (FGF21). Here we show that in the absence of adipose triglyceride lipase (ATGL) in adipocytes, ketone body and FGF21 production is impaired upon fasting. Liver gene expression analysis highlights a set of fasting-induced genes sensitive to both ATGL deletion in adipocytes and PPARα deletion in hepatocytes. Adipose tissue lipolysis induced by activation of the

Activation of epidermal growth factor receptor via CCR3 in bronchial epithelial cells.

We have previously found that bronchial epithelial cells express CCR3 whose signaling elicits mitogen-activated protein (MAP) kinase activation and cytokine production. Several investigators have focused on the signaling crosstalk between G protein-coupled receptors (GPCRs) and epidermal growth factor receptor (EGFR) in cancer cells. In this study, we investigated the role of EGFR in CCR3 signaling in the bronchial epithelial cell line NCI-H292. Eotaxin (1-100 nM) induced dose-dependent tyrosine phosphorylation of EGFR in NCI-H292 cells. Pretreatment of the cells with the EGFR inhibitor (AG147

Glucocorticoid-Dependent Mechanisms of Brain Tolerance to Hypoxia.

Adaptation of organisms to stressors is coordinated by the hypothalamic-pituitary-adrenal axis (HPA), which involves glucocorticoids (GCs) and glucocorticoid receptors (GRs). Although the effects of GCs are well characterized, their impact on brain adaptation to hypoxia/ischemia is still understudied. The brain is not only the most susceptible to hypoxic injury, but also vulnerable to GC-induced damage, which makes studying the mechanisms of brain hypoxic tolerance and resistance to stress-related elevation of GCs of great importance. Cross-talk between the molecular mechanisms activated in ne

The role of inflammation in cholestasis: clinical and basic aspects.

Hepatobiliary transport systems are essential for the uptake and excretion of a variety of compounds including bile acids. Disruption and dysregulation of this excretory pathway result in cholestasis, leading to the intrahepatic accumulation of bile acids and other toxic compounds with progression of liver pathology. Cholestasis induced by inflammation is a common complication in patients with extrahepatic infections or inflammatory processes, generally referred to as sepsis-associated cholestasis. Microbial products, including endotoxin, induce signaling pathways within hepatocytes either dir

The Wnt/frizzled/GSK-3 beta pathway: a novel therapeutic target for cardiac hypertrophy.

An excessive hypertrophic response of the heart to an increased workload is a leading cause of heart failure. At present, cardiac hypertrophy is treated with inhibitors of the renin-angiotensin system or with beta-adrenoceptor antagonists. These current therapeutic strategies inhibit prohypertrophic signaling pathways, but this therapy is inadequate in a substantial number of patients. However, the hypertrophic response of the heart is the net result of activation of prohypertrophic and antihypertrophic pathways. Glycogen synthase kinase-3 beta (GSK-3 beta) has a powerful antihypertrophic effe

Mechanism of tumor cells escaping from immune surveillance of NK cells.

Natural killer (NK) cells play an important role in anti-tumor and anti-infection, and perform their immune surveillance function in various ways. However, no matter what kind of cancer, the functional activity of NK cells in the tumor microenvironment (TME) is suppressed. Understanding the relationship between tumor cells and NK cells is very critical for tumor immunotherapy. This review discusses the mechanism of tumor cells escaping the immune surveillance of NK cells. These include a variety of factors that inhibit the activity of NK cells, an imbalance of activating receptors and inhibiti

Mg-Cross-Linked Alginate Hydrogel Induces BMSC/Macrophage Crosstalk to Enhance Bone Tissue Regeneration via Dual Promotion of the Ligand-Receptor Pairing of the OSM/miR-370-3p-gp130 Signaling Pathway.

Macrophages play a pivotal role in the crosstalk between the immune and skeletal systems, while Mg-based biomaterials demonstrate immunomodulatory capabilities in this procedure. However, the mechanism of how Mg2+ promotes osteogenesis through the interplay of bone marrow-derived mesenchymal stem cells (BMSCs) and macrophages remains undescribed. Here, we demonstrated that a Mg-cross-linked alginate hydrogel exerted a dual enhancement of BMSCs osteogenic differentiation through the ligand-receptor pairing of the OSM/miR-370-3p-gp130 axis. On the one hand, Mg2+, released from the Mg-cross-linke

The rise of selective estrogen receptor modulators (SERMs) in breast cancer therapy: a promising horizon.

Despite advancements in breast cancer treatment, breast cancer is still among the major causes of cancer-related deaths worldwide, with most breast cancers being estrogen receptor positive (ER+). Selective estrogen receptor modulators (SERMs) have been the core drug class used in endocrine therapy for ER+ breast cancer through tissue-specific conformational changes to the estrogen receptor. The present study critically evaluates the pharmacologic development, thermodynamics, and clinical effectiveness of SERMs. We systematically analyze approved agents (e.g., tamoxifen, raloxifene, bazedoxifen

Ang-(1-7) and ET-1 Interplay Through Mas and ETB Receptor Interaction Defines a Novel Vasoprotective Mechanism.

BACKGROUND: Ang-(1-7) (angiotensin (1-7)) via MasR (Mas receptor) opposes vaso-injurious actions of Ang II (angiotensin II) as shown in models of pulmonary hypertension. The underlying mechanisms remain unclear. We hypothesized cross talk between Ang-(1-7) and the protective arm of the ET-1 (endothelin-1) system involving MasR and ETBR (endothelin receptor type B). METHODS: To address this, we studied multiple models: in vivo, in a mouse model of ET-1-associated vascular injury (hypoxia-induced pulmonary hypertension); ex vivo, in isolated mouse arteries; and in vitro, in human endothelial ce

Synergy between the classical and alternative pathways of complement is essential for conferring effective protection against the pandemic influenza A(H1N1) 2009 virus infection.

The pandemic influenza A(H1N1) 2009 virus caused significant morbidity and mortality worldwide thus necessitating the need to understand the host factors that influence its control. Previously, the complement system has been shown to provide protection during the seasonal influenza virus infection, however, the role of individual complement pathways is not yet clear. Here, we have dissected the role of intact complement as well as of its individual activation pathways during the pandemic influenza virus infection using mouse strains deficient in various complement components. We show that the

Exercise-induced signalling in alleviating neuronal insulin resistance.

Exercise contributes to a multitude of positive changes within the body and brain with regard to glucose homeostasis, insulin sensitivity, synaptic plasticity, neuroprotection and neurogenesis, among other effects. It provides a non-pharmaceutical alternative for addressing metabolic disorders in individuals with type 2 diabetes, who also face an increased risk of developing Alzheimer's disease. A number of molecules are evoked upon exercise and circulate through the bloodstream, transmitting the wide-reaching advantages of exercise. The ensuing cross-talk has been shown to improve conditions

Pattern recognition receptor signaling in otitis media: immune crosstalk and pathogenic mechanisms.

Otitis media (OM) remains a prevalent and multifactorial inflammatory disease of the middle ear, especially in pediatric populations. The immune system, particularly pattern recognition receptors (PRRs), plays a central role in detecting microbial pathogens and initiating host defenses. TLR2 and TLR4 mediate bacterial clearance but exhibit subtype-specific dysregulation in chronic OM forms. NOD1, NOD2, and NLRP3 modulate intracellular pathogen sensing and inflammasome activation, while RIG-I governs antiviral immunity. C-type lectin receptors (CLRs) are emerging as modulators of both innate an

S1P-sPRR axis in renal ischemia-reperfusion injury: inflammation via a cut.

Inflammation is a determining factor in acute kidney injury (AKI) and subsequent kidney repair, yet its regulation is complicated and incompletely understood. In this issue, Feng and colleagues report that site-1 protease (S1P)-dependent cleavage of the (pro)renin receptor (PRR/ATP6AP2) generates soluble PRR (sPRR), which functions as an inflammation amplifier in ischemic AKI. Remarkably, S1P inhibition pharmacologically or genetically suppresses sPRR production and attenuates macrophage infiltration and activation, blunting AKI. The work elevates sPRR from a putative biomarker to a mechanisti
