ON HYPERTROPHY AND REGENERATION OF THE ISLANDS OF LANGERHANS.

1. Hypertrophy of the islands of Langerhans occurs in two forms: (1) the simple type which is nothing more than an increase in size of preexisting islands; and (2) the columnar type, in which the islands are composed of anastomosing columns of cylindrical cells. 2. Regeneration of the islands of Langerhans takes place by means of a budding off of columnar cells from the ducts. The process is analogous to the development of the islands in the fetal pancreas. 3. Newly formed islands of Langerhans are composed of cylindrical cells similar in all respects to those constituting hypertrophied island

Effect of guided tissue regeneration using bioresorbable membranes on periodontal regeneration in grade II furcation defects.

Grade II furcation defects pose a significant therapeutic challenge in periodontology due to their complex anatomy and limited accessibility for effective regeneration. Therefore, it is of interest to evaluate the efficacy of guided tissue regeneration (GTR) using a bioresorbable collagen membrane compared to open flap debridement (OFD) alone in 48 patients with mandibular molar Grade II furcation defects. Clinical parameters (probing depth, clinical attachment level, horizontal probing depth and gingival recession) and radiographic bone fill were assessed at baseline and 6 months postoperativ

Bone regeneration adjacent to titanium dental implants using guided tissue regeneration: a report of two cases.

The biologic principle of guided tissue regeneration was applied to regenerate alveolar bone in conjunction with the placement of titanium dental implants. In one case, complete osseointegration of an implant was achieved by the placement of a Teflon membrane over an implant that had been inserted into an alveolus immediately following tooth extraction. In a second case, the same biologic principle was used to increase the volume (height and width) of a resorbed, edentulous alveolar ridge to provide adequate bone dimensions for implant installation. In both cases, the membranes appear to have

The shh limb enhancer is activated in patterned limb regeneration but not in hypomorphic limb regeneration in Xenopus laevis.

Xenopus young tadpoles regenerate a limb with the anteroposterior (AP) pattern, but metamorphosed froglets regenerate a hypomorphic limb after amputation. The key gene for AP patterning, shh, is expressed in a regenerating limb of the tadpole but not in that of the froglet. Genomic DNA in the shh limb-specific enhancer, MFCS1 (ZRS), is hypermethylated in froglets but hypomethylated in tadpoles: shh expression may be controlled by epigenetic regulation of MFCS1. Is MFCS1 specifically activated for regenerating the AP-patterned limb? We generated transgenic Xenopus laevis lines that visualize th

Electrical stimulation accelerates and enhances expression of regeneration-associated genes in regenerating rat femoral motoneurons.

1. In this study we investigated whether electrical stimulation accelerates the upregulation of Talpha1-tubulin and GAP-43 (regeneration-associated genes; RAGs) and the downregulation of the medium-molecular-weight neurofilament (NFM), in concert with stimulation-induced acceleration of BDNF and trkB gene expression and axonal regeneration. 2. Two weeks prior to unilateral femoral nerve transection and suture, fluorogold (Fluorochrome Inc., Denver) or fluororuby (Dextran tetramethylrhodamine, Mol. Probes, D-1817, Eugene, OR) was injected into quadriceps muscles of the left and right hindlimbs

Unlocking the Secrets of the Regenerating Fish Heart: Comparing Regenerative Models to Shed Light on Successful Regeneration.

The adult human heart cannot repair itself after injury and, instead, forms a permanent fibrotic scar that impairs cardiac function and can lead to incurable heart failure. The zebrafish, amongst other organisms, has been extensively studied for its innate capacity to repair its heart after injury. Understanding the signals that govern successful regeneration in models such as the zebrafish will lead to the development of effective therapies that can stimulate endogenous repair in humans. To date, many studies have investigated cardiac regeneration using a reverse genetics candidate gene appro

Tendon Regeneration Ability in Axolotl Limbs.

Tendon injuries in humans often lead to significant functional impairments, necessitating effective regeneration methods. While mammalian tendon healing typically results in scar formation and functional loss, certain species such as axolotls exhibit remarkable regenerative abilities. This study investigates axolotl tendon regeneration following complete transection (tenotomy) and volumetric loss (tenectomy). Axolotl tendons, identified through histological and gene expression analyses, consist of collagens and exhibit specific tendon markers. Our results indicated complete regeneration of tra

Shape-Based Morphoelasticity for Jellyfish Regeneration.

Many animals exhibit remarkable regenerative abilities, yet the mechanical processes that drive large-scale tissue reshaping remain challenging to characterize. Direct measurements of stress during such remodeling are difficult, while changes in morphology are readily observed. Motivated by wound healing in Clytia jellyfish, we introduce a theoretical framework that infers mechanical stress directly from shape evolution. We model the transformation as a morphoelastic process in quasi-two-dimensional geometry and exploit conformal mappings to obtain analytical expressions for the stress distrib

Acute Inflammation Drives Corneal Pathological Regeneration.

PURPOSE: This study aimed to characterize transcriptional and epigenetic remodeling in the cornea induced by acute inflammation. METHODS: Limbal-corneal tissues were collected on days 5 and 10 after intrastromal administration of lipopolysaccharide or PBS. ScRNA-seq and scATAC-seq were used to delineate the transcriptomic and epigenomic atlas of the inflamed cornea, following the Seurat and Signac pipelines. Immunofluorescence was performed to assess changes in corneal epithelial markers and immune cell infiltration. RESULTS: We identified 16 cell clusters, including ten epithelial cell subp

[Neural regulation mechanism in bone regeneration].

The global aging population has intensified the incidence of degenerative bone diseases and the therapeutic demand for traumatic bone injuries, thereby making bone regenerative medicine a research focus. There is a close connection and interaction between the skeletal system and the nervous system, and innervation plays an indispensable regulatory role in the process of bone regeneration: the sympathetic nervous system exerts a negative regulatory effect during bone regeneration, while the parasympathetic nervous system plays a positive regulatory role in this process. Nerve fibers within bone

Liposome-Enabled Nanomaterials for Muscle Regeneration.

Muscle regeneration is a vital biological process that is crucial for maintaining muscle function and integrity, particularly for the treatment of muscle diseases such as sarcopenia and muscular dystrophy. Generally, muscular tissues can self-repair and regenerate under various conditions, including acute or chronic injuries, aging, and genetic mutation. However, regeneration becomes challenging beyond a certain threshold, particularly in severe muscle injuries or progressive diseases. In recent years, liposome-based nanotechnologies have shown potential as promising therapeutic strategies for

Modeling proximalisation in axolotl limb regeneration.

The axolotl (Ambystoma mexicanum) possesses a remarkable ability to regenerate tissues. Following limb amputation, a blastema of progenitor cells forms, expands, and reconstructs all distal structures, implying that mature cells near the wound retain positional memory along the proximal-distal (PD) axis. Key regulators of positional identity, such as Prod1 and Tig1, promote proximalisation-a shift toward a more proximal identity-when overexpressed, but the mechanisms governing this process remain unclear. In this study, we tracked changes in cellular density along the PD axis of regenerating a

Proteasome-mediated ubiquitination in neural regeneration.

The proteasome, the principal intracellular protein-degradation system, is essential for nerve regeneration after injury. This review summarizes recent evidence that the proteasome orchestrates axonal regeneration by modulating growth-cone formation, microtubule dynamics, reactive oxygen species (ROS) levels, and neuroinflammation. Proteasomes are delivered to growth cones via retrograde axonal transport, and their localization and activity depend on neuronal maturation and axon length. They further promote growth-cone assembly and axonal extension by regulating tubulin expression and polymeriza

Mechanisms underpinning spontaneous spinal cord regeneration.

Vertebrates exhibit a range of regenerative capacities following spinal cord injury. At one end of the spectrum are chief regenerators, including teleost fish and urodele amphibians. At the other end, most mammalian species exhibit limited repair and multicellular complications following spinal cord injury. Pro-regenerative immune, glial and neuronal injury responses underlie innate spinal cord repair in highly regenerative vertebrates. In many instances, fundamental mechanisms of spinal cord repair represent ancestral neuroprotection mechanisms that are conserved but become overwhelmed by ant

Serotonin neuromodulation directs optic nerve regeneration.

Optic nerve (ON) regeneration in mammalian systems is limited by an overshadowing dominance of inhibitory factors. This has severely hampered the identification of pro-regenerative pathways. Here, we take advantage of the regenerative capacity of larval zebrafish to identify pathways that promote ON regeneration. From a small molecule screen, we identified modulators of serotonin (5-HT) signaling that inhibit ON regeneration. We find that several serotonin type-1 (5-HT1) receptor genes are expressed in retinal ganglion cells during regeneration and that inhibiting 5-HT1 receptors or components

Overcoming big bottlenecks in vascular regeneration.

Bioengineering and regenerative medicine strategies are promising for the treatment of vascular diseases. However, current limitations inhibit the ability of these approaches to be translated to clinical practice. Here we summarize some of the big bottlenecks that inhibit vascular regeneration in the disease applications of aortic aneurysms, stroke, and peripheral artery disease. We also describe the bottlenecks preventing three-dimensional bioprinting of vascular networks for tissue engineering applications. Finally, we describe emerging technologies and opportunities to overcome these challe

Drug-Loaded Bioscaffolds for Osteochondral Regeneration.

Osteochondral defect is a complex tissue loss disease caused by arthritis, high-energy trauma, and many other reasons. Due to the unique structural characteristics of osteochondral tissue, the repair process is sophisticated and involves the regeneration of both hyaline cartilage and subchondral bone. However, the current clinical treatments often fall short of achieving the desired outcomes. Tissue engineering bioscaffolds, especially those created via three-dimensional (3D) printing, offer promising solutions for osteochondral defects due to their precisely controllable 3D structures. Multimodal decoding of human liver regeneration.

The liver has a unique ability to regenerate1,2; however, in the setting of acute liver failure (ALF), this regenerative capacity is often overwhelmed, leaving emergency liver transplantation as the only curative option3-5. Here, to advance understanding of human liver regeneration, we use paired single-nucleus RNA sequencing combined with spatial profiling of healthy and ALF explant human livers to generate a single-cell, pan-lineage atlas of human liver regeneration. We uncover a novel ANXA2+ migratory hepatocyte subpopulation, which emerges during human liver regeneration, and a corollary s

Optic nerve regeneration: Potential treatment approaches.

The optic nerve, predominantly constituted by the axons of retinal ganglion cells (RGCs), lacks the ability to regenerate and re-establish function after injury. RGCs are crucial for visual function, and thus, RGC death contributes to the development of numerous progressive neurodegenerative optic neuropathies including glaucoma, ischemic optic neuropathy, and optic neuritis. Regenerating optic nerve axons poses numerous challenges due to factors such as the intricate and inhibitory conditions that exist within their environment, intrinsic breaks to regeneration, and the geometric tortuosity t
