Effects of hunger and VI value on VI pacing.

In two experiments, each involving four rats, responses preceded by an inter-response time between 8 and 10 sec in duration were intermittently reinforced. In Experiment I, final performance was compared under two hunger levels, while the frequency of reinforcement was held constant by a VI 5 schedule. In Experiment II, hunger was held constant and VI 3 was compared with VI 8. Both hunger and frequency of reinforcement increased the over-all rate of response, but the exact effects of these operations on temporal discrimination were different for different rats. Usually, a peak "response probab

Measuring Cannabis Reinforcement among Young Adults: A Mixed Methods Examination.

INTRODUCTION: Increasing reinforcement received from cannabis-free activities, relative to reinforcement from cannabis-related activities, is one way to reduce harmful cannabis use. Thus, accurate measurement of cannabis reinforcement is important. Using convergent mixed methods, we developed the Adolescent Reinforcement Survey Schedule-Cannabis Use Version (ARSS-CUV). ARSS-CUV, adapted from the alcohol use version, measures cannabis reinforcement by asking individuals how frequently they engaged in, and how much they enjoyed, different activities when using and not using cannabis. METHOD: Yo

Time to choose: impact of intertrial interval on selecting between methamphetamine and food reinforcement in male and female rats.

RATIONALE: A central component of substance use disorder is the maladaptive choice of the drug over natural reinforcers. Compared to other drugs of abuse, methamphetamine (METH) choice has received limited study. OBJECTIVE: We sought to characterize the role of intertrial interval on METH choice behavior. METHODS: We examined the choice of METH versus food, across multiple METH doses (0.05-0.2 mg/kg/infusion), between male and female rats, employing a fixed ratio (FR1) reinforcement schedule with intertrial intervals (ITIs) of 20 and 600 s. Rats learned to lever-press for either the METH or

Effects of triadimefon on a multiple schedule of fixed-interval performance: comparison with methylphenidate, d-amphetamine and chlorpromazine.

Triadimefon is a fungicide that has recently been shown to increase motor activity and rates of schedule-controlled responding. These findings indicate that triadimefon resembles psychomotor stimulants and in this respect is a unique pesticide. The present experiment was designed to evaluate triadimefon's effects on performance maintained by a multiple schedule of reinforcement and to compare triadimefon to known psychomotor stimulants. Four rats were trained to perform under a mult FI 1-min FI 5-min schedule of milk reinforcement. They then received a series of dosages of triadimefon (10-170

Schedule induction of drug intake: differential responsiveness to agents with abuse potential.

In previous research, two groups of rats exposed to a fixed time (FT) 1-min schedule of food-pellet delivery exhibited equal schedule-induced polydipsia for water vehicle and a 0.05 mg/ml solution of midazolam, respectively. Exposure to FT schedule values greater than 1 min (3 and 5 min) resulted in greater milliliter per pellet intakes of midazolam than of water, perhaps reflecting the reinforcing efficacy of the drug. To determine if drugs with unequivocal reinforcing efficacy display similar elevations at the larger FT values, solutions of cocaine and ethanol, but also of the equivocally re

Operant Costs Modulate Dopamine Release to Self-Administered Cocaine.

The costs associated with obtaining illicit drugs can fluctuate depending upon the relative drug availability. As a consequence of the changing costs, the effort that one must exert to obtain drugs is dynamic. Considerable evidence illustrates a critical role for dopamine in the ventral medial striatum in mediating drug reinforcement. However, little is known regarding how dopamine release is affected by changes in the costs associated with earning drugs. We used fast-scan cyclic voltammetry to determine how changes in the operant requirement affected dopamine release to self-administered coca

Effects of antimuscarinic antiparkinsonian drugs on brightness discrimination performance in rats.

Biperiden (BPR) and trihexyphenidyl (THP), the current antimuscarinic drugs of choice in the management of parkinsonism, have been shown to exert anticonvulsant effects induced by poisoning by the organophosphorus compound soman. The present study was undertaken to evaluate the effects of these drugs on performance of a simple light-intensity discrimination task in rats under a tandem schedule of fixed-ratio (FR) reward/ differential-reinforcement-of-low-rate (DRL) nonreward contingencies, for water reinforcement in 2-h experimental sessions. Both BPR (0.125-2.0 mg/kg, SC) and THP (0.25-8.0 mg

A preliminary procedure for predicting the positive and negative effects of reinforcement-based procedures.

In the current investigation, a modification was made to the preference assessment described by Pace, Ivancic, Edwards, Iwata, and Page (1985) to predict the effects of stimuli when used in a differential-reinforcement-of-other-behavior (DRO) schedule for 2 clients with severe self-injurious behavior (SIB) and profound mental retardation. Based on the results of the preference assessment, three types of stimuli were identified: (a) high-preference stimuli associated with high rates of SIB (HP/HS), (b) high-preference stimuli associated with relatively lower rates of SIB (HP/LS), and (c)low-pre

Clonidine suppression and its adrenoreceptor mediation in schedule-induced polydipsia.

Rats were trained in a fixed-interval, one-minute (FI 1 min) food reinforcement schedule for 1 hour daily at reduced body weight until their lever presses, licks and water intake all became stabilized for 6 days. Two experiments were performed to examine the function of sympathetic activity in schedule-induced polydipsia. In experiment 1, intracerebroventricular injection of clonidine (0.75-37.5 nmol) produced a dose-related suppression of schedule-induced drinking and licking and schedule-dependent lever pressing; these effects were later attenuated by yohimbine (5 nmol) pretreatment. Effects of haloperidol on schedule-induced polydipsia.

In dose-related amounts, the drug haloperidol attenuated schedule-induced drinking by rats prefed with 0.01-mg drug added to 0, 25, 50, 75 or all of 100 Noyes 45-mg pellets. Drug pellets also induced less drinking than did regular Noyes pellets by rats that obtained these pellets at 1-min intervals by bar pressing. Haloperidol also reduced bar pressing and, temporarily, rate of reinforcement. The results appeared not to be due to a general sedative effect of haloperidol but to its selective power to reduce angiotensin-induced drinking. Thus, schedule-induced drinking, which is abnormal in not

Effects of an acute cannabidiol treatment on cocaine self-administration and cue-induced cocaine seeking in male rats.

Cannabidiol is a non-psychoactive compound that is the second most abundant component of cannabis. It has been shown to have a potential therapeutic value for a wide range of disorders, including anxiety, psychosis, and depression. Recently, it was suggested that cannabidiol might be a potential treatment for heroin craving and relapse. Here we investigated the effects of an acute treatment with cannabidiol on cocaine self-administration and cue-induced cocaine seeking in rats. Rats were trained to press a lever to self-administer cocaine (0.5 mg/kg/infusion), first under a fixed interval 20 s

Dopaminergic and opiate agonists and antagonists differentially decrease multiple schedule responding maintained by sucrose/ethanol and sucrose.

Similar neurobiological mechanisms are hypothesized to influence ethanol- and food-related reinforcement processes. This study examined the ability of compounds with dopaminergic or opiate activity to selectively alter responding maintained by a sucrose/ethanol solution in comparison to a sucrose solution. Long-Evans rats were trained to press a lever using 5% sucrose/10% ethanol and 5% sucrose as the reinforcers on a multiple Fixed Ratio 4 Fixed Ratio 4 schedule of reinforcement. When stable responding was established, the effects of intraperitoneally administered amphetamine (0.0-3.0 mg/kg),

Integrating Finite Element Simulation with Actual GTAW Weld Profiles to Optimize Root Height in Stainless Steel 316L Pipe Joints.

Weld root reinforcement is a critical geometric parameter governing stress concentration and structural performance in thin-walled stainless-steel piping systems designed to ASME B31.3. While current codes specify permissible dimensional limits, they do not explicitly quantify how incremental variations in root height influence stress distribution under realistic service loading conditions. This study integrates finite element analysis (FEA) with experimentally validated GTAW weld profiles to evaluate the structural influence of weld root height in 316L stainless-steel pipe joints. An experime

NMR-based Metabolomics and Fatty Acid Profiles to Unravel Biomarkers in Preclinical Animal Models of Compulsive Behavior.

Compulsivity is a key manifestation of inhibitory control deficit and a cardinal symptom of psychopathological conditions such as obsessive-compulsive and attention-deficit hyperactivity disorders, in which metabolic alterations have raised attention as putative biomarkers for early identification. The present study assessed the metabolic profile in a preclinical model of a compulsive phenotype of rats. We used the schedule-induced polydipsia (SIP) method to classify male Wistar rats into high drinkers (HDs) or low drinkers (LDs) according to their compulsive drinking rate developed by exposur

The effects of amphetamine, butorphanol, and their combination on cocaine self-administration.

There have been recent calls to examine the efficacy of drug-combination therapies in the treatment of substance use disorders. The purpose of the present study was to examine the ability of a novel stimulant-opioid combination to reduce cocaine self-administration, and to compare these effects to those of each drug administered alone. To this end, male Long-Evans rats were implanted with intravenous catheters and trained to self-administer cocaine under positive reinforcement contingencies. Once self-administration was acquired, rats were divided into four different groups and treated chronic

Re-evaluating the predictive value of reinforcement motivation on outcome in anorexia nervosa: The role of affective symptoms.

BACKGROUND: Reinforcement motivation, particularly heightened punishment sensitivity and its imbalance relative to reward sensitivity ("motivational bias"), can predict outcomes in anorexia nervosa (AN). However, previous studies did not account for affective symptoms, which are intertwined with reinforcement processes and illness trajectory. This study examined whether affective symptoms explain outcomes beyond reinforcement motivation. METHODS: Adolescent girls and young women with AN (n = 192; Mage = 16.33 years, SD = 2.61) completed self-report measures of punishment/reward sensitivity, d

Incorporating discriminative stimuli into functional communication training with augmentative and alternative communication devices: a tutorial.

Functional communication training (FCT) is a commonly used and effective treatment for problem behavior maintained by social reinforcement (e.g., an individual engages in self-injurious behavior to gain access to adult attention). FCT involves teaching an individual to emit an appropriate communication response to access the reinforcer maintaining problem behavior (e.g., pressing a "Play, please" symbol on a device to gain the communication partner's attention) and withholding that reinforcer following problem behavior (e.g., the communication partner minimizes attention-following problem beha

Effects of positive or negative reinforcement history and extinction arrangement on resurgence.

The recurrence of previously reduced behavior following successful differential reinforcement has been termed resurgence. Across studies, resurgence has occurred following either positive or negative reinforcement, but no direct within-experiment comparisons have been conducted. The primary purpose of the current experiments was to address this gap by comparing resurgence following a history of positive or negative reinforcement with human participants responding to earn or avoid losing money. Across experiments, we manipulated the arrangement of the resurgence test following negative reinforc

Reinforcing effectiveness of midazolam, ethanol, and sucrose: behavioral economic comparison of a mixture relative to its component solutions.

Benzodiazepines (BZs) are relatively safe when administered alone. However, these drugs can produce severe side effects when coadministered with ethanol. Despite these adverse consequences, rates of concurrent BZ and ethanol misuse are increasing, and it is unclear whether this behavior is maintained by an enhanced reinforcing effect of the mixture. To address this issue, the current study compared the reinforcing effectiveness of sucrose solutions mixed with midazolam, ethanol, or both. Eight male rats were trained to orally self-administer solutions of either sucrose (S), sucrose+midazolam (

Drug discrimination under a concurrent fixed-interval fixed-interval schedule.

Pigeons were trained to discriminate 5.0 mg/kg pentobarbital from saline under a concurrent fixed-interval (FI) FI schedule of food presentation on which, after pentobarbital administration, responses on one key were reinforced with food under an FI 60-s component and responses on the other key were reinforced under an FI 240-s component. After saline administration, the schedule contingencies on the two keys were reversed.
