Modulation of Presynaptic Inhibitory Retinal Circuits for Retinal Ganglion Cell Neuroprotection in Glaucoma.

Glaucoma is a leading cause of irreversible blindness, yet the circuit-level mechanisms contributing to retinal ganglion cell (RGC) degeneration remain incompletely defined. Although multiple neuroprotective strategies have been explored, clinical translation remains limited. Here, we identify early dysfunction of inner retinal inhibitory circuitry as a modifiable contributor to RGC vulnerability during ocular hypertensive stress. In mouse ocular hypertension models, inner nuclear layer neurons exhibited increased c-fos expression and enhanced oscillatory potential-associated differential elec

Melanopsin ganglion cells are the most resistant retinal ganglion cell type to axonal injury in the rat retina.

We report that the most common retinal ganglion cell type that remains after optic nerve transection is the M1 melanopsin ganglion cell. M1 ganglion cells are members of the intrinsically photosensitive retinal ganglion cell population that mediates non-image-forming vision, comprising ∼2.5% of all ganglion cells in the rat retina. In the present study, M1 ganglion cells comprised 1.7±1%, 28±14%, 55±13% and 82±8% of the surviving ganglion cells 7, 14, 21 and 60 days after optic nerve transection, respectively. Average M1 ganglion cell somal diameter and overall morphological appearance remaine

Blocking apoptosis promotes survival and alters developmental dynamics of human retinal ganglion cells in retinal organoids.

Retinal ganglion cells (RGCs) are the projection neurons connecting the retina to the brain. In many species, a substantial proportion of RGCs are eliminated by programmed cell death during development to regulate their final number, but how cell death impacts human RGC development remains poorly understood. Here, we characterized cell death in human fetal retinas and retinal organoids. Both retinas and organoids exhibited two waves of apoptosis: an early wave targeting neurogenic retinal progenitor cells and neuronal precursors and a late wave affecting RGCs and other neurons. Additionally, o

Human Retinal Ganglion Cells Respond to Evolutionarily Conserved Chemotropic Cues for Intra Retinal Guidance and Regeneration.

Retinal ganglion cells (RGCs) connect the retina with the higher centers in the brain for visual perception. Their degeneration leads to irreversible vision loss in patients with glaucoma. The mechanism underlying human RGCs (hRGCs) axon growth and guidance remains poorly understood because hRGCs are born during development and connections with the central targets are established before birth. Here, using RGCs directly generated from human embryonic stem cells, we demonstrate that hRGCs express a battery of guidance receptors. These receptors allow hRGCs to read the spatially arrayed chemotrop

Photodisruption of the Inner Limiting Membrane Promotes Retinal Engraftment of Stem-Cell Derived Retinal Ganglion Cells.

PURPOSE: The purpose of this study is to address the challenge of limited donor retinal ganglion cell (RGC) migration into the retina after transplantation, which is largely due to donor neuron accumulation at the inner limiting membrane (ILM). We present a minimally invasive technique, ILM photodisruption, to locally ablate the ILM and thereby promote RGC engraftment. METHODS: ILM photodisruption uses indocyanine green as a photosensitizer, which is delivered to the ILM and activated with ultra-short laser pulses. This process generates vapor nanobubbles (VNBs) that, upon collapse, create lo

SARS-CoV-2 infects and replicates in photoreceptor and retinal ganglion cells of human retinal organoids.

Several studies have pointed to retinal involvement in COVID-19, yet many questions remain regarding the ability of SARS-CoV-2 to infect and replicate in retinal cells and its effects on the retina. Here, we have used human pluripotent stem cell-derived retinal organoids to study retinal infection by SARS-CoV-2. Indeed, SARS-CoV-2 can infect and replicate in retinal organoids, as it is shown to infect different retinal lineages, such as retinal ganglion cells and photoreceptors. SARS-CoV-2 infection of retinal organoids also induces the expression of several inflammatory genes, such as interle

Spatial Adaptation of Primate Retinal Ganglion Cells Between Artificial and Natural Stimuli.

The retina encodes a broad range of stimuli, adapting its computations to features like brightness, contrast, and motion. However, it is unclear whether it also adapts when switching between natural scenes and white noise (WN). To address this, we analyzed the neural activity of male marmoset retinal ganglion cells (RGCs) in response to WN and naturalistic movies. We trained linear-nonlinear models on both stimuli, evaluated their performance, and compared their receptive fields across stimulus domains. We found that models with spatial filters trained on one stimulus ensemble were less accura

Tracking Satb2-positive retinal ganglion cells in zebrafish unveils developmental functional reorganization.

Retinal ganglion cells (RGCs) relay visual information to the brain, and their functional properties are crucial for visually guided behavior. However, functional maturation of RGCs across development remains elusive. Here, we investigated the developmental maturation of a genetically defined RGC type expressing Satb2 in zebrafish, which allowed single-layer-resolved analysis of the tectal neuropil. Using in vivo calcium imaging, we characterized visual response properties of Satb2-positive RGC axon terminals at two developmental stages: 6-7 and 10-11 days post-fertilization. At the early stag

PURPOSE: Retinal ganglion cells (RGCs) are essential for visual signal transmission, yet they are vulnerable to injury and degeneration. Gene modulation in RGCs using adeno-associated virus (AAV) offers a promising avenue for neuroprotection and regeneration, but promoters lack sufficient RGC specificity, limiting precision needed for preclinical studies. This study aims to identify novel promoter-enhancer combinations (PECs) to achieve gene expression preferentially in RGCs. Although extensive research has been conducted on neuroprotection for retinal ganglion cells, there is still no treatment for clinical use. Recent evidence shows that extracellular vesicles isolated from a variety of stem cells are efficacious in retinal ganglion cell neuroprotection. In this study, we tested the novel extracellular vesicle source of the retinal progenitor R-28 cell line in vitro and in vivo .

A Cell-Type Selective Stimulation and Recording System for Retinal Ganglion Cells.

Future retinal implants will require a stimulation selectivity between different sub-types of Retinal Ganglion Cells (RGCs) to evoke natural perceptions rather than phosphenes in patients. To achieve this, a cell-type specific stimulation pipeline is required that identifies target RGC sub-types from recorded input images and extracts the specific stimulation parameters to activate this cell-type selectively. Promising biological experiments showed that ON-/OFF- sustained/transient RGCs could be selectively activated by modulating repetition rate and amplitude of an electrical stimulation curr

Differential enrichment of retinal ganglion cells underlies proposed core neurodegenerative transcription programs.

In a published Correction 1 , a revised analysis updated two "core transcription programs" proposed to underlie axon injury-induced retinal ganglion cell (RGC) neurodegeneration. Though extensive, the Correction purported to leave the two principal conclusions of its parent study 2 unaltered. The first of those findings was that a core program mediated by the Activating Transcription Factor-4 (ATF4) and its likely heterodimeric partner does not include numerous canonical ATF4 target genes stimulated by RGC axon injury. The second was that the Activating Transcription Factor-3 (ATF3) and C/EBP

Inferring light responses of primate retinal ganglion cells using intrinsic electrical signatures.

Objective. Retinal implants are designed to stimulate retinal ganglion cells (RGCs) in a way that restores sight to individuals blinded by photoreceptor degeneration. Reproducing high-acuity vision with these devices will likely require inferring the natural light responses of diverse RGC types in the implanted retina, without being able to measure them directly. Here we demonstrate an inference approach that exploits intrinsic electrophysiological features of primate RGCs.Approach.First, ON-parasol and OFF-parasol RGC types were identified using their intrinsic electrical features in large-sc

Intravitreal Injection of AAV for the Transduction of Mouse Retinal Ganglion Cells.

The injection of therapies into the eye is common practice, both clinically and pre-clinically. The most straightforward delivery route is via an intravitreal injection, which introduces the treatment into the largest cavity at the posterior of the eye. This technique is frequently used to deliver gene therapies, including those containing recombinant adeno-associated viral vectors (AAVs), to the back of the eye to enable inner retinal targeting. This chapter provides detailed methodology on how to successfully perform an intravitreal injection in mice. The chapter covers vector preparation co

Timing is Everything: Stochastic Optogenetic Stimulation Reduces Adaptation in Retinal Ganglion Cells.

Optogenetics gives us unprecedented power to investigate brain connectivity. The ability to activate neural circuits with single cell resolution and its ease of application has provided a wealth of knowledge in brain function. More recently, optogenetics has shown tremendous utility in prosthetics applications, including vision restoration for patients with retinitis pigmentosa. One of the disadvantages of optogenetics, however, is its poor temporal bandwidth, i.e. the cell's inability to fire at a rate that matches the optical stimulation rate at high frequencies (>30 Hz). This research propo
