Quantitative analysis of retinal ganglion cell survival with Rbpms immunolabeling in animal models of optic neuropathies.

PURPOSE: To investigate whether a recently described retinal ganglion cell (RGC) marker Rbpms (RNA binding protein with multiple splicing) could be used for RGC quantification in various models of RGC degeneration. METHODS: Optic nerve crush, excitotoxicity, and elevated intraocular pressure (IOP) rat models were used. Topographic analysis of Rbpms immunolabeling was performed on retinal wholemounts. Retrograde labelings with Fluorogold (FG) and III β-tubulin immunohistochemistry were compared. RESULTS: In the optic nerve crush model, 37%, 87%, and 93% of Rbpms-positive cells were lost 1, 2,

Off-target myocardial necrosis due to incomplete balloon occlusion during retrograde coronary venous ethanol ablation.

BACKGROUND: It is assumed that complete balloon occlusion is necessary for effective lesion formation in retrograde coronary venous ethanol infusion for ventricular arrhythmias. OBJECTIVE: This study aimed to evaluate the myocardial effects of ethanol infusion under conditions of incomplete balloon occlusion. METHODS: 12 swine were randomized into acute (n = 6) and chronic (n = 6) cohorts. Ethanol was infused into different coronary veins using single over-the-wire balloons sized to achieve incomplete occlusion. Myocardial injury was assessed by gross pathology, histology, and ultrastructura

[Hereditary spastic paraplegias].

Hereditary spastic paraplegias (HSP) are characterised by symmetric spastic paraplegia, pallhypaesthesia, and urinary dysfunction (uncomplicated HSP). Complicated HSP is present if uncomplicated HSP additionally presents with epilepsy, dementia, cataract, extrapyramidal dysfunction, amyotrophy, polyneuropathy, or ichthyosis. Clinically, HSP are similar but genetically even more heterogeneous. The disease course is slowly progressive and, the earlier the disease onset, the slower the course. Causes of HSP are mutations in 20 different genes, of which eight have been identified so far. A single

Immunohistochemical evidence for degeneration of cholinergic neurons in the forebrain of the rat following injection of AF64A-picrylsulfonate into the dorsal hippocampus.

The cholinergic neurotoxin, AF64A-picrylsulfonate, was unilaterally infused into the dorsal hippocampus of Wistar rats (2 nmol/2 microliters/4 min; A 6.2, Ls 1.5, H 6.5, Paxinos and Watson). After 19 days the for immunohistochemical staining of choline acetyltransferase (ChAT). Morphometry and counting of ChAT-immunoreactive profiles revealed shrinkage and disappearance of cholinergic neurons in the medial septum and diagonal band of Broca at the lesioned brain side. These data indicate a retrograde degeneration of cholinergic neurons following injection of AF64A-picrylsulfonate into the dorsa

Reptilian somatosensory midbrain: identification based on input from the spinal cord and dorsal column nucleus.

A midbrain somatosensory area was identified in reptiles Caiman crocodilus, by orthograde degeneration techniques after midcervical spinal cord transections or after dorsal column lesions. Terminations of these somesthetic inputs were located in a caudal lateral part of the central nucleus of the torus semicircularis and contiguous dorsal mesencephalon. Dorsal column projections were contralateral while spinal connections were mainly ipsilateral. In order to confirm the results of the anterograde degeneration experiments independently, injections of horseradish peroxidase were placed in this m

[Management of post-traumatic tibiotalar osteoarthritis following a failed osteosynthesis : retrograde plantar nailing].

Tibiotalar osteoarthritis, uncommon and often of post-traumatic origin, leads to functional limitations, chronic pain and significantly impairs the quality of life. Both tibiotalar arthrodesis and arthroplasty have been proven effective. Tibiotalocalcaneal (TTC) arthrodesis is generally reserved for ankle osteoarthritis associated with severe degeneration of the subtalar joint and hindfoot. Nevertheless, new data showed good outcomes regardless of the subtalar joint state. We present the clinical case of an 86-year-old female patient with severe tibiotalar osteoarthritis after a failed osteosy

Delayed diffuse upper motor neuron syndrome after compressive thoracic myelopathy.

A 54-year-old man developed progressive spastic paraparesis beginning 2 weeks after a back injury caused by a subacute compressive thoracic myelopathy attributable to a post-traumatic arachnoid cyst. Three to 18 months after surgical decompression of the thoracic arachnoid cyst, the patient developed a diffuse predominantly upper motor neuron syndrome characterized by spastic quadriparesis, pseudobulbar paresis, and pseudobulbar affect. Retrograde corticospinal tract degeneration and upper motor neuron death after spinal cord injury is recognized. This case suggests that focal upper motor neur

Stent-graft treatment of a large internal carotid artery vein graft aneurysm.

Aneurysm degeneration of internal carotid artery interposition vein grafts can occur, and their repair can be complicated by repeated neck dissection and difficulty in obtaining distal artery control. We used a covered stent graft to exclude a large internal carotid artery interposition vein graft aneurysm. Retrograde flow was induced in the internal carotid artery so that an embolization protection balloon could be placed through the aneurysm. The stent graft was then placed with embolization protection. The repair was performed without complication, and the aneurysm remained successfully exc

Organization of cingulo-ponto-cerebellar connections in the cat.

This study deals with three different aspects of the organization of connections from the cingulate gyrus to the cerebellum. (1) With the use of wheat germ agglutinin-horseradish peroxidase as a retrograde tracer, the distribution of cingulate neurons projecting to the pontine nuclei was studied. Retrogradely labeled cells were found in layer 5 in all parts of the cingulate gyrus. Average densities of cingulo-pontine cells were similar in the different cytoarchitectonic subdivisions, although some density gradients were observed. The projection was found to be remarkably strong. Average densit

Novel role of SARM1 mediated axonal degeneration in the pathogenesis of rabies.

Neurotropic viral infections continue to pose a serious threat to human and animal wellbeing. Host responses combatting the invading virus in these infections often cause irreversible damage to the nervous system, resulting in poor prognosis. Rabies is the most lethal neurotropic virus, which specifically infects neurons and spreads through the host nervous system by retrograde axonal transport. The key pathogenic mechanisms associated with rabies infection and axonal transmission in neurons remains unclear. Here we studied the pathogenesis of different field isolates of lyssavirus including r

Nimodipine promotes regeneration and functional recovery after intracranial facial nerve crush.

The calcium flow inhibitor, nimodipine, has been shown to promote motor neuron survival in the facial nucleus after intracranial facial nerve transection. However, it has not been known whether the neuroprotective effects primarily involve survival of nerve cell bodies or outgrowth and/or myelination of nerve fibers. Here, we studied the effects of nimodipine in a different injury model in which the facial nerve was unilaterally crushed intracranially. This lesion caused complete anterograde degeneration and partial retrograde degeneration that were studied with a combination of several stereo

Light and electron microscopic identification of monoaminergic terminals in the central nervous system.

A brief critical survey of methods used for light and electron microscopic examination of amine-containing pathways within the CNS. Light microscopic techniques such as fluorescence histochemistry, immunocytochemistry, autoradiography, silver degeneration techniques, and retrograde tracing technique are suitable for studying the topography of pathways but, due to limits of resolution, they are inadequate for identifying terminals. Electron microscopy which is adequate to visualize terminals does not provide an overall view. This review considers various methods which have been devised to speci

Hybrid repair of aortic arch dissections.

OBJECTIVE: Hybrid interventions combining debranching of supra-aortic branch vessels with stent grafting of the aortic arch have become an attractive alternative to open repair for aortic arch pathologies. However, results in patients with dissections of the aortic arch remain unclear. We present our experience with hybrid aortic arch repair for acute and chronic type B aortic dissections (TBAD) involving the distal part of the arch and aortic dissections distal to previous repair of the ascending aorta. METHODS: Between January 2004 and December 2011, hybrid arch repair with supra-aortic bra

Longitudinal evidence for anterograde trans-synaptic degeneration after optic neuritis.

In multiple sclerosis, microstructural damage of normal-appearing brain tissue is an important feature of its pathology. Understanding these mechanisms is vital to help develop neuroprotective strategies. The visual pathway is a key model to study mechanisms of damage and recovery in demyelination. Anterograde trans-synaptic degeneration across the lateral geniculate nuclei has been suggested as a mechanism of tissue damage to explain optic radiation abnormalities seen in association with demyelinating disease and optic neuritis, although evidence for this has relied solely on cross-sectional

Optic nerve in globoid leukodystrophy (Krabbe's disease). Ultrastructural changes.

Globoid leukodystrophy (Krabbe's disease) was diagnosed in an infant in whom a progressive neurological disorder and optic atrophy developed. At autopsy, ultrastructural examination of the optic nerves and cerebral white matter revealed characteristic tubular inclusions in globoid-epithelioid cells. Thinning of the nerve fiber and ganglion cell layers of the retina appeared to be due to retrograde degeneration of the optic nerve related to the abnormal metabolism of myelin. We discuss the pathogenesis of the clinical and pathological ocular findings, with regard to the inherited absence of the

Degenerative and regenerative processes in the olfactory system of homing pigeons.

Experimental resection of the olfactory nerve in the homing pigeon induces a total degeneration of the nerve and olfactory epithelium. The orthograde degenerative process starts before the retrograde one. Ten days after resection, new neurons begin to differentiate from the basal cells. The axon forms earlier than the distal dendritic process, and the speed of growth increases slowly. The regenerated axons only reach the bulb in the 5th month. Two months after resection the olfactory epithelium is similar to that of the intact control side. The ultrastructural features of the mucosa and olfact

Head Vessel Reintervention Post-Type A Dissection Repair With a Hybrid Stent Prosthesis.

BACKGROUND: Adverse aortic remodeling may occur after the repair of acute type A aortic dissection (ATAAD), with branch vessel involvement being implicated due to retrograde perfusion of the false lumen (FL). CASE SUMMARY: A 61-year-old man presented with adverse aortic remodeling following ATAAD repair with the AMDS Hybrid Prosthesis, with maximal growth in zone 2 of 9 mm over 1 year. Tears were identified in the right common carotid artery and innominate artery, which were treated with endovascular stenting. There was no further growth in zones 0 to 3 three years postprocedure. DISCUSSION:

The PKG Inhibitor CN238 Affords Functional Protection of Photoreceptors and Ganglion Cells against Retinal Degeneration.

Hereditary retinal degeneration (RD) is often associated with excessive cGMP signalling in photoreceptors. Previous research has shown that inhibition of cGMP-dependent protein kinase G (PKG) can reduce photoreceptor loss in two different RD animal models. In this study, we identified a PKG inhibitor, the cGMP analogue CN238, which preserved photoreceptor viability and functionality in rd1 and rd10 mutant mice. Surprisingly, in explanted retinae, CN238 also protected retinal ganglion cells from axotomy-induced retrograde degeneration and preserved their functionality. Furthermore, kinase activ

Intramuscular viral delivery of paraplegin rescues peripheral axonopathy in a model of hereditary spastic paraplegia.

Degeneration of peripheral motor axons is a common feature of several debilitating diseases including complicated forms of hereditary spastic paraplegia. One such form is caused by loss of the mitochondrial energy-dependent protease paraplegin. Paraplegin-deficient mice display a progressive degeneration in several axonal tracts, characterized by the accumulation of morphological abnormal mitochondria. We show that adenoassociated virus-mediated (AAV-mediated) intramuscular delivery of paraplegin halted the progression of neuropathological changes and rescued mitochondrial morphology in the pe
