The Granular Retrosplenial Cortex Is Necessary in Male Rats for Object-Location Associative Learning and Memory, But Not Spatial Working Memory or Visual Discrimination and Reversal, in the Touchscreen Operant Chamber.

The retrosplenial cortex (RSC) is a hub of diverse afferent and efferent projections thought to be involved in associative learning. RSC shows early pathology in mild cognitive impairment and Alzheimer's disease (AD), which impairs associative learning. To understand and develop therapies for diseases such as AD, animal models are essential. Given the importance of human RSC in object-location associative learning and the success of object-location associative paradigms in human studies and in the clinic, it would be of considerable value to establish a translational model of object-location l

Voluntary wheel exercise improves learning and memory impairment caused by hippocampal Hb-α deficiency by reducing microglial activation and reversing synaptic damage.

Decreased hemoglobin (Hb) levels in peripheral blood may be a risk factor for Alzheimer's disease (AD). Hb-α is a monomeric form of Hb that exists in the central nervous system. Our previous RNA sequencing results revealed a decrease in the expression of the Hb-α gene in the hippocampus of AD model mice. However, the effects of Hb-α deficiency in the hippocampus on cognitive function and the underlying mechanism are unclear. Running exercise has been shown to improve cognition, but whether it can reverse the damage caused by Hb-α deficiency in the hippocampus needs to be further researched. In

Algorithmically defined therapeutic targets: integrating single-cell transfer learning frameworks with small molecule drugs to reverse disease-associated cell fates.

The high heterogeneity of the disease microenvironment is a critical factor contributing to therapeutic failure and the emergence of drug resistance; however, predicting drug responses with precision at single-cell resolution remains a substantial challenge. Traditional pharmacogenomic studies are constrained by averaged signals at the population level, which frequently obscure rare yet lethal resistant subpopulations. This article reviews a closed-loop strategy that integrates computational pharmacology with cell biology to address this dilemma. First, we explore computational frameworks base

Methamphetamine exposure during gestation and lactation periods impairs the learning and memory of offspring mice, which is reversed by melatonin: the role of oxidative stress and acetylcholinesterase.

BACKGROUND AND PURPOSE: Melatonin is a product of the pineal gland, which regulates the circadian cycle. Neurotoxicity is the most important side effect of methamphetamine (Met) abuse during pregnancy. This study aimed to explore the effect of Met exposure during gestation and lactation periods on the learning and memory of offspring mice. The protective effect of melatonin and the role of oxidative stress and acetylcholinesterase were also investigated. EXPERIMENTAL APPROACH: The pregnant mice were randomly divided into 2 groups. Saline or Met (5 mg/kg) was injected daily during pregnancy an

Mice deficient for the synaptic vesicle protein Rab3a show impaired spatial reversal learning and increased explorative activity but none of the behavioral changes shown by mice deficient for the Rab3a regulator Gdi1.

Rab proteins are small GTPases involved in intracellular trafficking. Among the 60 different Rab proteins described in mammals, Rab3a is the most abundant in brain, where it is involved in synaptic vesicle fusion and neurotransmitter release. Rab3a constitutive knockout mice (Rab3a(-/-)) are characterized by deficient short- and long-term synaptic plasticity in the mossy fiber pathway and altered circadian motor activity, while no effects on spatial learning have been reported so far for these mice. The goals of this study were to analyse possible behavioral consequences of the lack of synapti

FFPM, a PDE4 inhibitor, reverses learning and memory deficits in APP/PS1 transgenic mice via cAMP/PKA/CREB signaling and anti-inflammatory effects.

Thus far, phosphodiesterase-4 (PDE4) inhibitors have not been approved for application in Alzheimer's disease (AD) in a clinical setting due to severe side effects, such as nausea and vomiting. In this study, we investigated the effect of FFPM, a novel PDE4 inhibitor, on learning and memory abilities, as well as the underlying mechanism in the APP/PS1 mouse model of AD. Pharmacokinetic studies have revealed that FFPM efficiently permeates into the brain, and reached peak values in plasma 2 h after orally dosing. A 3-week treatment with FFPM, at doses of 0.25 mg/kg and 0.5 mg/kg, significantly

Zika Virus Testing Considerations: Lessons Learned from the First 80 Real-Time Reverse Transcription-PCR-Positive Cases Diagnosed in New York State.

The performance and interpretation of laboratory tests for Zika virus (ZKV) continue to be evaluated. Serology is cross-reactive, laborious, and frequently difficult to interpret, and serum was initially solely recommended for molecular diagnosis. ZKV testing was initiated in January 2016 in New York State for symptomatic patients, pregnant women, their infants, and patients with Guillain-Barré syndrome who had traveled to areas with ZKV transmission. Subsequently, eligibility was expanded to pregnant women with sexual partners with similar travel histories. Serum and urine collected within 4

Vitamin D, Folic Acid and Vitamin B12 Can Reverse Vitamin D Deficiency-Induced Learning and Memory Impairment by Altering 27-Hydroxycholesterol and S-Adenosylmethionine.

The cholesterol-oxidized metabolite 27-hydroxycholesterol (27-OHC) is synthesized by CYP27A1, which is a key factor in vitamin D and oxysterol metabolism. Both vitamin D and 27-OHC are considered to play important roles in Alzheimer’s disease (AD). The study aims to research the effects of co-supplementation of vitamin D, folic acid, and vitamin B12 on learning and memory ability in vitamin D-deficient mice, and to explore the underlying mechanism. In this study, C57BL/6J mice were fed a vitamin D-deficient diet for 13 weeks to establish a vitamin D-deficient mice model. The vitamin D-deficien

Abnormal modulation of reward versus punishment learning by a dopamine D2-receptor antagonist in pathological gamblers.

RATIONALE: Pathological gambling has been associated with dopamine transmission abnormalities, in particular dopamine D2-receptor deficiency, and reversal learning deficits. Moreover, pervasive theoretical accounts suggest a key role for dopamine in reversal learning. However, there is no empirical evidence for a direct link between dopamine, reversal learning and pathological gambling. OBJECTIVE: The aim of the present study is to triangulate dopamine, reversal learning, and pathological gambling. METHODS: Here, we assess the hypothesis that pathological gambling is accompanied by dopamine-

Reverse AIEE/ACQ Engineering in a Carbon Dot-Quantum Dot Ratiometric Nanosensor: Machine-Learning-Driven Quantitative and Recognition Detection of Biogenic Amines for Food Freshness Monitoring.

Simultaneous profiling of biogenic amines, including spermine, putrescine, cadaverine, histamine, and tyramine, is crucial for assessing food freshness, tracking spoilage progression, and preventing associated foodborne illnesses and adverse health effects. Herein, we report a chemically unmodified dual-emissive nanosensor constructed by noncovalent integration of green carbon dots (G-CDs) and near-infrared CdTe quantum dots (QD700). The sensing mechanism arises from competitive modulation of excited-state relaxation pathways via multiple noncovalent interactions (electrostatic attraction, mul

Permutationally invariant polynomial regression for energies and gradients, using reverse differentiation, achieves orders of magnitude speed-up with high precision compared to other machine learning methods.

Permutationally invariant polynomial (PIP) regression has been used to obtain machine-learned potential energy surfaces, including analytical gradients, for many molecules and chemical reactions. Recently, the approach has been extended to moderate size molecules with up to 15 atoms. The algorithm, including "purification of the basis," is computationally efficient for energies; however, we found that the recent extension to obtain analytical gradients, despite being a remarkable advance over previous methods, could be further improved. Here, we report developments to further compact a purifie
